Radial maze performance in three strains of mice: role of the fimbria/fornix.

Three strains of mice were tested on an 8-arm radial maze, an index of hippocampus-dependent spatial memory. Levels of performance differed between strains with C57Br/cj greater than Balb/cj greater than C57Bl/6j. Lesions of the fimbria/fornix disrupted performance in the C57Br and Balb strains: the C57Bl mice never performed better than chance before or after surgery. Choline acetyltransferase activity in hippocampus was not correlated with radial maze performance. These findings suggest a possible genetic contribution towards radial maze behavior.

Mucoepidermoid carcinomas of the lacrimal sac.

Mucoepidermoid carcinoma is a malignant tumor containing both neoplastic mucin-producing cells and epidermoid cells. To the only reported case arising in the lacrimal sac, we add three additional cases. All three were locally aggressive and required orbital exenterations. No deaths from metastatic disease occurred.

A prognostic factor study of disease-free interval and survival following enucleation for uveal melanoma.

Prognostic indicators of uveal melanoma disease-free interval and survival following enucleation were evaluated for 267 patients. The median follow-up time was 17 years. Analysis involved Kaplan-Meier survival curves based on time to tumor-related deaths and multivariate proportional hazards analysis, which provides an estimate of the "force of mortality." Prognostic indicators considered included demographic, clinical, and histopathological factors. Cell type was classified according to the number of epithelioid cells present per high-power field (HPF) on light microscopy. Various classifications of size of the tumor were compared for prognostic value. The five leading predictors of survival in order of importance were as follows: (1) number of epithelioid cells per HPF, (2) largest dimension of the tumor, (3) location of the anterior margin of the tumor, (4) invasion to the line of transection, and (5) degree of pigmentation. Risk categories for survival were constructed based on the three leading prognostic factors.

Further efficacy evaluation of doxpicomine for postoperative pain.

In this single-dose, double-blind study, the analgesic activity of 400 and 200 mg doxpicomine was compared with 100 and 50 mg meperidine and placebo when given intramuscularly in 102 subject patients experiencing severe postoperative pain. Results indicate that 400 mg doxicomine is similar to 100 mg meperidine in analgesic activity, onset, and duration of action. Side effects were of the same order as those produced by other centrally acting analgesics.

Serologic and immunopathologic findings in temporal arteritis.

In 104 patients undergoing biopsies for temporal arteritis, lymphocyte characterization identified both T4 helper/inducer and T8 cytotoxic/suppressor lymphocytes in approximately equal numbers. B lymphocytes were absent. Deposition of IgM and IgG was observed in three of 16 positive biopsy specimens. Antinuclear antibodies were present in ten of 16 biopsy-proven cases of temporal arteritis compared with 19 of 55 in the control group with negative biopsy specimens. Anti-smooth-muscle, anti-DNA, and antimitochondrial antibodies were not useful in distinguishing between the two groups.

Airway management in the transfer of the unconscious patient.

Thirty-one consecutive unconscious patients admitted to a specialist neurosurgical centre for computerised axial tomography and further management were reviewed with emphasis upon initial airway management. Fourteen patients had inadequate airway control on arrival and needed immediate intubation. Six of this group developed pulmonary complications. The implications of this are discussed and certain recommendations made.

Ultrastructural study of enzymes in reactive astrocytes: clarification of astrocytic activity.

Enzymes in reactive of the cerebral cortex were examined at the ultrastructural level in an attempt to resolve some conflicting aspects of astrocytic activity. Correlations between morphological and enzyme changes after injury established that the apparent increase in oxidative enzyme activity was exclusively mitochondrial and not an artefactual reaction product resulting from anoxic cellular damage. Pronounced glucose-6-phosphatase activity within cisternae of an increased amount of the granular endoplasmic reticulum was related to increased glycogen. Further evidence from acid phosphatase activity indicated that astrocytes played a minimal role in phagocytosis.

Estimation of osmotic parameters accompanying zeatin-induced growth of detached cucumber cotyledons.

Water potential (psi), the osmotic potential (psi(pi)), and the pressure potential (psi(p)) of detached cotyledons isolated from Cucumis sativus L. cv Marketer seedlings after 0, 1.5, and 3 days growth with and without zeatin were determined. From zero time to 3 days, cotyledons incubated without exogenous zeatin exhibited a slight decrease in psi (from -0.4 to -1.0 bars), while those grown with zeatin developed even more negative values (about -4 bars). Both groups showed rising psi(pi) values (decreases in solutes per unit volume), but this rise was more dramatic in those treated with zeatin. These data indicate that the capacity of zeatin-treated cotyledons to take up water more rapidly than controls and thus expand faster must be due to wall loosening, as reflected in psi(p) values which declined during 3 days from about +11 bars to about +1.4 bars.It was also found that freshly detached cotyledons or those grown without exogenous zeatin exhibited osmoregulation in polyethylene glycol (PEG) solutions. That is, while cotyledons initially lost H(2)O into certain PEG solutions, their psi values decreased over time and they began absorbing water after 1 to 4 hours. After 3 days growth, zeatin-treated cotyledons had lost most of this capacity of osmoregulate. It seems likely that osmoregulation in cotyledons not treated with zeatin is due to wall loosening rather than changes in psi(pi). Zeatin-treated cotyledons with already loosened walls may not have this option to deal with water stress and thus simply come to equilibrium with external PEG solutions.

Brain phagocytes: source of high acid phosphatase activity.

A stab wound was produced in the adult rat cerebral cortex, and the progress of enzyme cytochemistry of phagocytosis was studied over 450 days. Light- and electron-microscopic observations were made to establish the origin of high acid phosphatase activity commonly seen at the site of brain lesions. Cells with phagocytic potential became differentiated and activated by the presence of degenerating neurons. The Golgi-lysosomal system of the phagocytes became elaborated, as evidenced by thiamine pyrophosphatase and acid phosphatase activities, the synthesis of acid phosphatase was increased, and the enzyme then secreted into the digestive vacuoles containing dead cells to be digested. Progress of the digestive process resulted in the accumulation of large amounts of acid phosphatase reaction product within the digestive vacuoles. The results showed that the phagocytes were the only detectable source of increased acid phosphatase activity at the site of injury in the cerebral cortex. In contrast to the phagocytes, newly formed multi-nucleated giant cells exhibited weak acid phosphatase, and intense cytochrome oxidase activities, the difference between the two cells reflecting the functional characteristics of each.

Ultrastructural demonstration of glucose 6-phosphatase in cerebral cortex.

A two-stage fixation technique has been developed to obtain morphological preservation and retention of glucose 6-phosphatase (G6-Pase) activity for its demonstration in rat cerebral cortex. The technique was then employed to localize the enzyme in the cortex where it produced a dense reaction over the well developed granular endoplasmic reticulum cisternae in nerve cells and oligodendrocytes which contrasted with a thin reaction in astrocytes. Other membranous organelles showed no reaction.

The relationship of malignant melanoma, basal and squamous skin cancers to indoor and outdoor work.

An analysis of occupational incidence data for malignant melanomas and squamous-and basal-self carcinomas of the skin in England and Wales from 1970 to 1975 is reported. The occupational pattern for melanomas of the trunk and limbs differed markedly from the pattern for melanomas of the head, face and neck. Office work was associated with a large excess of melanomas of the trunk and limbs. In contrast, outdoor work was associated with an excess of melanomas of the head, face and neck; and was also associated with an excess of squamous-and basal-cell carcinomas of the skin. This suggests that prolonged occupational exposure to sunlight is an important cause of squamous-and basal-cell carcinomas and of melanomas of the head, face and neck, but not of melanomas on other parts of the body. The high rate of lesions on the trunk and limbs in office workers may reflect their sunbathing or other recreational habits; but it contrasts clearly with other indoor work, where there is a generally low rate of all forms of skin cancer.

Doxpicomine in postoperative pain.

In a single-dose study the analgesic activity of 400 mg doxpicomine was compared with 4 and 8 mg morphine administered intramuscularly. Our subjects wee 43 men with moderate or severe postsurgical pain on the first postoperative day. Results of a double-blind trial indicate the analgesic activity of 400 mg intramuscular doxpicomine to be within the range of 8 mg morphine. There was no statistical significance between the effects of 400 mg doxpicomine and 8 mg morphine with the exception of half-hour data where doxpicomine was more effective than morphine. Doxpicomine has a rapid onset of analgesic action, the duration of which is equivalent to that of 8 mg morphine.

Trends in pelvic inflammatory disease in England and Wales.

Hospital admissions for pelvic inflammatory disease increased by 8.7% a year among women aged 15-44 in England and Wales between 1966 and 1976. Acute disease was three times as frequent as chronic disease, although admissions for the chronic condition are increasing more rapidly than for the acute. Peak incidence of acute disease is at ages 20-24 and of chronic disease at ages 25-29. Divorced women had the highest rates of both forms. Single women had higher rates of acute disease than married women but the latter had higher admission rates for chronic disease. In general, the pattern of acute disease follows that of sexually transmitted disease. The pattern of chronic disease in turn reflects the long-term effects of acute disease. The increase in chronic disease is of concern because hospital discharge rates are increasing more rapidly than for acute disease, and in 1976 one in every eight cases mentioned infertility in association with the disease.

Value of blood-pressure measurement in relatives of hypertensive patients.

First-degree relatives of hypertensive patients are more likely to comply with a request to visit their general practitioner for a screening blood-pressure measurement than are the relatives of normotensive controls. On average they have higher blood-pressure levels and are more likely to be started on treatment as a result of screening. In the majority of cases treatment can be maintained long-term and blood pressure falls to acceptable levels. The relatives of people already on treatment for hypertension include a high proportion of potential candidates for treatment, and screening of this group may be worth while.

Intermittent pressure cuffing for oedematous digits.

A double-walled rubber cylinder, made of cheap, readily available materials, is described. This device enables intermittent positive pressure to be applied to individual fingers during the course of management of post-surgical or post-traumatic finger oedema. It has a number of advantages over the standard total hand splint currently used.

Failure to detect abnormal fatty acid profiles in serum lipoproteins in Friedreich's ataxia.

A recent report indicated an association between the spinocerebellar degeneration of Friedreich's ataxia and abnormal fatty acid profiles of the cholesterol ester fraction in both total plasma and high-density lipoprotein. In an effort to substantiate these findings in lipoproteins, we estimated fatty acid profiles of total high-density lipoprotein and the cholesterol esters of high- and low-density lipoproteins. Those profiles in 10 Friedreich's ataxia patients compared well with previously reported normal values and did not differ significantly from results in 10 age-matched controls.
