Antigenic alterations of carcinoembryonic antigen involved in the secretion from various human tumor cell lines.

Antigenic variations of carcinoembryonic antigens (CEAs) produced by 8 human tumor cell lines of various organ origins--4 derived from colonic, one from gastric, one from pancreatic, and 2 from lung cancers--were investigated on the basis of reactivity with several monoclonal antibodies recognizing either the peptide or the carbohydrate moiety of the CEA molecule by using a sandwich-type solid-phase radioimmunoassay (RIA). CEAs from all the cell lines revealed nearly uniform reactivities with the monoclonal antibodies recognizing the peptide epitopes, and no difference was observed between the reactivities of CEAs released into culture medium (m-CEAs) and of those extracted from cells (c-CEAs). On the other hand, with each of 4 monoclonal antibodies recognizing the carbohydrate epitopes, the reactivities of CEAs were found to be highly variable from one cell line to another, and a marked difference was detected between the reactivities of m-CEA and of c-CEA in every cell line. In general, c-CEA reacted much more strongly than m-CEA with these antibodies. The reactivities of some of the CEAs with 2 out of 4 antibodies to sugar epitopes increased significantly after treatment of CEAs with neuraminidase. These results suggest that distinct structural changes in the carbohydrate moiety may occur during the releasing process of CEA from cells, and that, in some cases, some epitopes may be masked by attachment of sialic acid residues.

Craniofacial and oral malformations in an autopsy population of Japanese human fetuses and newborns.

A review of the autopsy records of the Department of Pathology of Nagasaki University and the Department of Geneticopathology of Hiroshima University from 1944 through 1982 revealed that during this period there were 11,050 cases of fetuses and newborns, including 432 cases with craniofacial and oral malformations and 22 cases with amniogenous malformations. Among the cranial malformations, there were 202 (46.8%) anencephaly cases, 38 (8.8%) holoprosencephaly, 28 (6.48%) hydrocephaly, 27 (6.25%) meningoencephalocele, and 13 other cranial malformations (seven microcephaly and two each of macrocephaly, dolichocephaly, and iniencephaly). Among the oral malformations, there were 25 (5.78%) cleft lip cases, 23 (5.32%) cleft palate, and 76 (17.6%) cleft lip and palate. Among the facial malformations, there were 12 (2.78%) anomicrophthalmia cases, 37 (8.56%) ear malformations, 15 (3.47%) micrognathia, and 8 (1.85%) nose malformations. One cranial malformation was found that was complicated with anencephaly and holoprosencephaly. Of 307 cranial malformations, 38 (12.4%) were complicated with oral malformations and 4 (4.6%) with facial malformations. Among the 124 cases of oral malformations, 38 (30.6%) were complicated with cranial malformations. The complicating cranial malformations were anencephaly in 16 cases, holoprosencephaly in seven, hydrocephaly in six, meningoencephalocele in seven, other cranial malformations in two, and with facial malformations in 19 cases. Among the 72 cases of facial malformations, 14 were complicated with cranial malformations and 19 with oral malformations. Four cases showed three or more cranial, facial, and/or oral malformations at the same time. Recently, experimental embryological studies have shown that the neural crest cell-derived mesectoderm participates largely in the morphogenesis of the face and the cardiovascular system. It may be said that neural crest cells are deeply involved in the teratogenesis.

Morphogenesis of human cardiac outflow.

The developmental anatomy of human cardiac outflow was studied in a series of 16 normal embryos (gestational days 29-39, crown-rump length 6-20 mm, stages 14-19). Structural features and kinetics during truncal septation (TS) were described from external photographs, serial histological sections, and computer graphic reconstructions of selected tissues. Early in the period studied, the tubular myocardium ensheathed the single cardiac lumen and spiralling conotruncal ridges, which were filled with mesenchymal cells during days 31-33. As TS began (late stage 16), the aorticopulmonary (AP) septum appeared across the dorsal wall of the aortic sac between arches IV and VI. Mesenchymal condensations formed within the AP septum, crossing the lumen bifurcation to extend along the truncal ridges to the myocardium. During days 35-37, the cephalic margin of the myocardium grew or folded in toward these mesenchymal condensations between the developing valves and within the nearby conal ridges, which appeared to fuse to separate the subvalvular outflow channels by day 39. These observations are consistent with studies in chicks and rats which suggest that mesenchymal condensations or cell death foci interact with the distal myocardial rim during TS to form a structural septation complex dividing the two arterial streams.

Treatment of inferior vena cava obstruction producing Budd-Chiari syndrome.

We treated a patient who had an inferior vena cava (IVC) obstruction associated with Budd-Chiari syndrome. All of the right, middle, and left hepatic veins were completely obstructed. The IVC was obstructed by a membranous substance and thrombus at the hepatic portion and was completely occluded by a fibrous septum at the site of a suprahepatic coarctation. A cavotomy was performed transversely at the suprahepatic level and then longitudinally to the level just above the renal veins, and the obstructing tissue was removed. An additional vertical incision was made in the IVC over the coarctation, and an autologous pericardial patch was sutured in place to widen the IVC. The patient was discharged with the patency of the IVC restored.

Isolation and characterization of N-long chain acyl aminoacylase from Pseudomonas diminuta.

N-Long chain acyl aminoacylase II (Enzyme II) catalyzing the hydrolysis of N-long chain acyl amino acids was purified about 2,000-fold from the cell extracts of Pseudomonas diminuta with 1.8% of activity yield. The purified enzyme was homogeneous on polyacrylamide gel electrophoresis and the molecular weight was 220,000. Enzyme II differed from N-long chain acyl aminoacylase I (Enzyme I) in molecular weight, in substrate specificity, and in behavior toward temperature and pH. Enzyme II showed broader substrate specificity than Enzyme I and catalyzed the hydrolysis of lipoamino acids containing various amino acid residues, although Enzyme I was almost specific to the lipoamino acids containing L-glutamate. The extent of hydrolysis by Enzyme II reaction varied depending on the kinds of lipoamino acids and were: 100% for palmitoyl-L-glutamate, 91% for myristoyl-L-glutamate, 85% for lauroyl-L-glutamate, 54% for lauroyl-L-aspartate, 28% for stearoyl-L-glutamate and 17.5% for lauroyl-glycine.

Biochemical characterization of eel virus european.

The polypeptide and RNA compositions of purified virions of eel virus European were compared to those of three strains (VR-299, SP and AB) of infectious pancreatic necrosis virus (IPNV) isolated from trout. All three IPNV strains could be distinguished by the relative mobilities of either the virion polypeptides or the double-stranded RNA genome segments. The eel virus had a similar polypeptide profile to strain AB IPNV but differences between the two viruses in the migration of genome segments indicated each was unique.

A case with leiomyoma of iris extirpated with cryosurgery and its pathological findings.

This report describes surgical observation and histopathological findings in a case of a 44-year-old male with leiomyoma of the iris in his left eye which had initiated from uveitis. In view of clinical efficiency and electron microscopic detection of leiomyoma, it is recommended that the tumour be excised after freezing coagulation. For confirmative diagnosis of leiomyoma of the iris, an electron microscopic detection is necessary to demonstrate the characteristic findings including cells with cigar-shaped nuclei, cytoplasmic and plasma-lemmal density, fine filaments, surface-connected vesicles and basement membrane.

Induction of DNA ligase during stimulation of DNA synthesis in intact rat liver by a dietary manipulation.

After a nutritional shift from a protein-free to a diet containing 50% casein, the activity of DNA ligase increases in intact rat liver in correlation with the induction of hepatic DNA replication. The treated rat liver as well as control rat liver contains a single species of DNA ligase having a sedimentation coefficient of about 5.5 S. The administration of cycloheximide in vivo completely inhibits the increase in DNA ligase activity and in DNA synthesis, indicating that DNA ligase is induced in the hepatic cells replicating DNA. In contrast to DNA ligase, DNA kinase is unchanged in the activity level by the dietary manipulation.
