A 14q+ chromosome in adult T-cell leukemia.

Chromosome studies were conducted on two patients with adult T-cell leukemia. In both patients, a marker chromosome 14q+ and a structural change involving chromosome 1 with trisomy of the q arm were found in peripheral blood leukocytes. The 14q+ marker chromosome had resulted from translocation from #5p in one patient and #5q in the other patient. The present and previous studies suggest that the donor chromosomes involved in the 14q+ translocation are variable. This indicates that the 14q+ marker chromosome rather than the donor chromosome is intimately related with adult T-cell leukemia.

Antitumor agents. II. Effect of 5-fluorouracil and cyclophosphamide on immunological parameters and liver microsomes of tumor-bearing rats.

An attempt has been made to clarify immunosuppressive properties of anti-tumor agents by studying the effect of the agents on the thymus, the reticulo-endothelial system (RES) and hepatic drug-metabolizing enzyme activities of tumor (an ascites hepatoma, AH 130 cells)-bearing rats. A drastic decrease in the thymus weight and the total number of the lymphocytes and an enhanced activity of thymus alkaline phosphatase were detected by injecting either 5-fluorouracil (5FU) or cyclophosphamide (CP) (30 mg each/kg weight, i.p.) daily for 5 days to tumor-bearing rats. The agents, however, did not induce any conspicuous damage in microsomal mixed function oxidase system or the RES. The presence of 10-day-old tumor resulted in an extreme decrease in the weight and lymphocytes of thymus and a partial decrease in the microsomal drug metabolizing enzyme activities and the RES. Thus, these antitumor agents may lead to the decline of host-mediated immune mechanism. The multiplication of the tumor cells also appears to depress the immune functions and the host resistance.

Antitumor agents. I. Effect of 5-fluorouracil and cyclophosphamide on liver microsomes and thymus of rat.

Effects of antitumor agents on rat liver microsomal drug-metabolizing enzyme activities and thymus lymphocytes were studied in male Wistar rats. High doses of 5-fluorouracil (5-FU) and cyclophosphamide (CP) given parenterally for 6 days caused a partial decrease in whole body weight and the microsomal enzyme content such as cytochrome P-450 and cytochrome b5. Aniline p-hydroxylase and aminopyrine N-demethylase activities also decreased in rats dosed for 5 days decreased compared with the control. Both compounds in the high concentrations produced spectral change of "modified type II". However, the magnitude of the spectral changes observed was independent of the the concentration of substrate added. The addition of NADPH to the microsomes-substrate mixture modified the spectral change. Both drugs caused a considerable decrease in thymus weight and the number of thymus lymphocytes, while the alkaline phosphatase activity was enhanced in 5-FU groups, indicating that the agents cause a significant involution of the thymus. Decrease in the total number of the lymphocytes was greater than that in the blood leucocytes.

K-ras gene mutations in non-small cell lung cancer in Japanese.

To evaluate the association of mutations in the K-ras gene with the incidence of non-small cell lung cancer (NSCLC) in Japanese patients, 410 patients treated surgically were studied. DNA was extracted from frozen specimens, and polymerase chain reaction-sequence specific oligonucleotide probe (PCR-SSOP) hybridization assay was used to examine K-ras mutations in codons 12, 13 and 61. K-ras mutations were found in 33 patients (8.0%), and all were smokers or ex-smokers. There were no significant differences in clinical or pathological stage between wild-type cases and mutant cases. The most frequently identified mutation was a G-->T transversion (25/33, 75.8%) that resulted in the substitution of a glycine for a cysteine or a valine. Previous studies have shown that the majority of K-ras mutations among smokers are G-->T transversion. In our study, K-ras mutations were found only in smokers, and there was a high frequency of G-->T transversions. A clear correlation exists between smoking and K-ras gene mutations.

Analysis of natural coloring matters in food. III. Application of methylation with diazomethane for the detection of lac color.

A new method has been proposed for detection of lac color in food. Lac color is a natural color additive derived from a secretion of the insect Coccus Laccae (Laccifer lacca Kerr). It is extracted from food with methanolic oxalic acid and eluted from a column of Amberlite XAD-2 with the same solvent. The fraction containing the lac color is treated with diazomethane to produce 2 reddish-orange markers. The marker species in the reaction mixture are detected by both thin-layer chromatography and reverse-phase liquid chromatography.
