Enzyme immunoassay for cuprozinc-superoxide dismutase in serum and urine.

A highly sensitive enzyme immunoassay system for the determination of cuprozinc-superoxide dismutase (Cu,Zn-SOD) in serum and urine by using beta-galactosidase as a labeling enzyme is reported. This assay had a greater sensitivity than that of previously reported radioimmunoassay methods and could measure from 0.05 to 10.0 ng of Cu,Zn-SOD with good reproducibility. Coefficients of variation for this enzyme immunoassay were 2.8-8.3% within a day and 5.6-16.1% between days. The average recoveries were 96-103% from sera and 96-105% from urines, respectively. Using this enzyme immunoassay, serum Cu,Zn-SOD concentrations increased significantly in patients with renal diseases and were slightly elevated in patients with liver diseases. Urinary Cu,Zn-SOD appeared to be the highest in all renal diseases. By immunofluorescent staining, Cu,Zn-SOD was located in the thickened portions of the glomerular basement membrane and proliferating mesangial cells of the kidney tissue from a patient with membranous glomerulonephritis.

Enzyme immunoassay for manganese-superoxide dismutase in serum and urine.

An enzyme immunoassay has been developed to measure human manganese-superoxide dismutase (Mn-SOD) in serum and urine. The assay can be done in less than 5 h, is highly sensitive (detecting limit, 0.1 ng of Mn-SOD) and simple. There is no interference from factors in serum or urine under assay conditions, and the method is specific for human Mn-SOD. Serum Mn-SOD concentrations were markedly increased in patients with liver diseases, but not in renal diseases. On the other hand, urinary Mn-SOD levels were elevated in a few patients with nephrotic syndrome and lung cancer, but were decreased in patients with hypertension. Furthermore, investigations were conducted on the clinical course of serum Mn-SOD levels in a case of alcoholic hepatitis, and on correlations between serum Mn-SOD and the conventional liver function tests. The localization of Mn-SOD in liver was also explored using immunofluorescent staining. The fluorescence was intense in the degenerated portions of liver tissue from a patient with drug-induced hepatitis.

Original surface replacement total hip prosthesis with alumina ceramic prosthetic head without cement.

We have developed a surface replacement total hip prosthesis of H.D.P. socket to ceramic head type having a polycrystal alumina ceramic prosthetic head and without bone cement for fixation. The strength test was done on this hip prosthesis, and comparative studies were made between the metal prosthetic head and the ceramic prosthetic head, using calculation models prepared in consideration of friction and the prosthetic head's inside diameter. Besides, the cadaver femur was replaced by the prosthetic head, and the impact test and the fatigue test by means of repeated loading were done. As we could consequently confirm that it could be fully used for clinical practice, we have manufactured various instruments to be used for surgery, and we are now actually practicing it.

[Experimental studies on antitumor effects of lysozyme-II. Effect of lysozyme on immunopotentiation].

Effects of lysozyme (LY) on immuno-responses were examined. Tumor-bearing mice were used on most of experiments and normal mice on several cases of experiments. Cytostatic activity on the Peyer's patch cells in Meth-A bearing Balb/c mice was enhanced at 6 to 12 hrs after oral administration of LY. Furthermore, LY showed to recover from the decline of blastogenic activity, which was observed on the lymph node cells and the spleen cells of tumor-bearing mice, at 8 and 14 days after tumor inoculation. In the normal ICR mice, it was not observed that LY increased the foot-pad reaction against the sheep red blood cells (SRBC). On the other hand, the decrease of the foot-pad reaction, which was observed in the tumor bearing mice at 20 days after Sarcoma-180 (S-180) inoculation, was recovered by the administration of LY. A decrease of the number of plaque-forming cells (PFC) against SRBC which was induced with tumor inoculation in the spleen cells was not observed at 20 days after S-180 inoculation, but LY caused a significant increase of PFC response in spleens on both of normal and tumor bearing mice. From these results, it was suggested that potentiation of systemic immuno-response derived from the activation of Peyer's patches brought about antitumor effects of LY.

[Experimental studies on antitumor effects of lysozyme].

Effects of lysozyme(LY) alone and combination with various antitumor agents were examined using syngeneic tumors mainly Meth-A and MH 134 transplanted into Balb/c and C3H/He mice, respectively. The effects of LY on pulmonary metastases were also examined in BDF1 mice bearing Lewis lung carcinoma (3LL). LY inhibited the growth of Meth-A tumor and enhanced antitumor effects of mitomycin C (MMC). bleomycin (BLM) and 5-fluorouracil (5-FU), LY activated antitumor effects of MMC, BLM and 5-FU on MH 134 tumor, however, LY alone did not show any significant antitumor effect on it. The combination treatments using LY with MMC or 5-FU showed a marked inhibition on pulmonary metastases of 3LL. The pre-treatment of LY inhibited the tumor growth of Sarcoma-180 in ICR mice.

The renal and hemodynamic effects of furosemide in acute myocardial infarction.

Renal and hemodynamic effects of furosemide were studied in 30 patients with acute myocardial infarction. When marked congestion of lungs was absent, the effects of furosemide were somewhat different from those of recent reports. The changes of pulmonary arterial pressure and wedge pressure were biphasic; there was first pressure rise in many occasions followed by reduced pressure; the latter is commonly believed to be the beneficial effect of furosemide. In these patients without marked congestion but in an acute state of myocardial infarction, within 5 min of furosemide administration, there was marked increase in urine volume, sodium and potassium excretion along with increases in creatinine clearance which is commonly found in the other conditions, but is in contrast to the recent reports on patients with acute myocardial complicated with pulmonary congestion.

[Traumatic posterior fossa epidural hematoma--especially the value of CT scan (author's transl)].

Epidural hematoma in the posterior fossa is a rare clinical entity which requires the early diagnosis and treatment, and the difficulty of the diagnosis has been pointed out. However, more recently CT scan began to use to make the valuable diagnostic method for traumatic posterior fossa hematomas. Epidural hematoma may be classified as acute, subacute and chronic, according to the time of free interval or lucid interval after injury. In this paper three acute cases and two subacute cases are reported. CT findings in acute cases show two different types. "Type I" shows crescent or lenticular high density area which is not enhanced after contrast infusion. "Type II" shows lenticular low density area with membranous high density region in its medial side after contrast infusion. In subacute cases plain CT scan shows lenticular iso or low density area with membranous high density region in its medial side. Forty five cases of posterior fossa epidural hematoma in the review of the literature of this country are discussed. Disturbances of the consciousness are the most predominant symptoms in acute cases, while in subacute cases cerebellar signs, vomiting, headache and choked disc are noted. Angiographical examinations may not always be valuable in collecting the direct information of the existence of the epidural hematoma. Liquor cavity in the posterior fossa which is thought to serve as a buffer action of hematoma is about 20 ml, so we discuss about the volume of hematoma, especially of 20 ml, associated with clinical course and prognosis. Volume of epidural hematoma is one of the most important factors affecting clinical course and prognosis. In summary of these our experiences, we again emphasize the value of CT scan as the rapid, noninvasive, accurate radiological examination in the diagnosis of traumatic posterior fossa epidural hematoma.

Studies on the synthesis of chemotherapeutics. 10. Synthesis and antitumor activity of N-acyl- and N-(alkoxycarbonyl)-5-fluorouracil derivatives.

A number of N-acyl and N-(alkoxycarbonyl)-5-fluorouracil derivatives possessing, for example, benzoyl, o-toluyl, acetyl, propionyl, heptanoyl, ethoxycarbonyl, phenoxycarbonyl, and benzyloxycarbonyl groups as N1 and/or N3 substituents were synthesized, and their antitumor activities were evaluated. The synthesis was achieved by a direct and two-step acylation of 5-fluorouracil and by selective N1-deacetylation of N1-acetyl-N3-substituted-5-fluorouracil under appropriate reaction conditions. Several N3-benzoyl- and N3-o-toluyl-5-fluorouracil derivates and showed significant activity against experimental tumor, and N1-acetyl-N3-o-toluyl-5-fluorouracil was found to be most promising among them. Further investigation revealed 12 to retain higher activity toward various tumors, with lower toxicity and good blood level, than either 1 or FT-207, even for oral administration.
