[The dependence of the incidence of encountering cells with micronuclei in the experimental metastases of rat rhabdomyosarcoma on the duration of the growth and on the size of these metastases].

Cell suspensions of rat transplantable rhabdomyosarcoma RA-2 were injected to white rats. The experimentally induced lung metastases (ELM) were estimated according to the frequency of micronucleated cells (MNC) that appeared 9-20 days after injection (a. i.). The frequency of MNC in ELM decreased from 2.7% (9 days a. i.) to 0.7% (20 days a. i.). The ELM which were of the same size but were examined at different time after injection had equal frequencies of MNC. At each investigated time after injection the least frequencies of MNC were observed in the ELV of the biggest size. The data provided allow to infer that the growth time and site of ELM may influence the spontaneous karyotype instability rates.

[Clonal analysis of karyotypic instability and metastatic potential in tumor cell populations].

Rat organospecific transplantable RA-2 rhabdomyosarcoma strains RA-2H and RA-2L were investigated using single cell cloning technique for "Micronuclear containing cell frequency" (MCF) and "Metastatic potential" (MP). The RA-2H was obtained from RA-2 during selection for increasing metastatic potential, whereas the RA-2L was the result of selection in opposite direction. MP was assessed by lung colony formation technique and in the case of RA-2H, it was one hundred times greater than in the case of RA-2L (2490 +/- 280 and 22 +/- 11 lung colonies i.v. after injection of 10 cells, respectively). Average MCE for RA-2H was 3 and for RA-2L 5.3%. Also, significant differences were observed between these substrains in percentage of anaphase and telophase cells with bridges and fragments (24 +/- 4 for RA-2H and 56 +/- 5% for RA-2L, P less than 0.05). The selection of RA-2H clones with high level of MCF was successful. In the population obtained MCF was increased up to 7.8% and MP decreased (79 +/- 44). So, the selection for decreasing of MP led to significant increase in genome instability characteristics and the selection for increasing of MCF--to the abrupt decline of MP, which proved the existence of negative correlation between these two characters in populations studied. It is concluded that correlation between malignancy and genome instability in tumor populations with different levels of malignancy is not, as a rule, positive.

[Cytogenetic instability, the ability to produce interferon and immunoreactivity of blood lymphocytes in patients with schizophrenia].

In schizophrenia patients, the level of T-lymphocytes with cytogenetic aberrations is elevated, and the ability of these cells for PGA-blast-transformation and the interferon synthesis is reduced. The elevation of the lymphocytes ability for blast-transformation in the presence of the native preparation of DNA and for lysis of the erythrocytes proper is marked in these patients. The observed changes were especially marked in schizophrenia with an interrupted course.

[Complex study of various pathogenetic and physiologic parameters of the health status of watchmen in the oil industry].

Complex examination of oil industry workers and those engaged in nonproductive area (control group) was undertaken. It was established that in both groups there were persons with a high level of cells with cytogenetic disturbances (micronuclear test). However, as opposed to control group, oil industry workers had higher levels of systolic and pulse pressure. Besides it was shown that after 12 hours of work at an oil field under winter conditions in the north area of the Tomsk Region they had higher body temperature. It appeared that these persons primarily had the longest term of professional service. Further examination of persons with especially high level of micronuclei cells showed that they had elevated lymphocyte amount with chromosome impairments and some parameters of T- and B-immunoreactivity, phagocytosis and activity of normal killer cells were changed.

[Mechanisms of DNA repair disorders in human cells. Genome instability in lymphocytes of patients with myopathy related to DNA repair disorders].

Increased level of cytogenetic damages was observed in lymphocytes of 10 patients with muscular dystrophy (Duchenne, Erba, Landuzi--Degerin). Analysis of DNA repair synthesis revealed inhibition of this process in lymphocytes of 10 patients observed. On the other hand, decreased reactivation of vaccinia virus and increased level of virus mutagenesis induced by 4-nitroquinoline-1-oxide and bleomycin was noted in the experiments with lymphocytes of 3 patients observed.

[Immunological elimination of cytogenetically aberrant cells induced by infectious factors].

Injection of measles virus and streptolysin-O into the cultures of human fibroblasts (HF) significantly increased the number of cells with cytogenetic aberrations. Injection of non-immune autologous T-lymphocytes resulted in the decrease in the number of aneuploid HF leading to the intact condition. Immune homologous T-lymphocytes, in contrast to non-immune autologous ones, were shown to remove cells with structural chromosome damage, without affecting the number of polyploid and aneuploid cells. Two equally possible suggestions can be made concerning non-immune autologous T-lymphocytes: 1) T-lymphocytes remove HF having virus antigens on their surface or 2) T-lymphocytes define and eliminate fibroblasts with the cell surface changed, as a result of virus-induced damage.

Cytogenetic effects of the addition of leukocytes and blood sera on human fibroblasts exposed to measles virus or to streptolysin-O.

Infection with measles virus and also introduction of streptolysin-O induced a significant increase in the level of cells with cytogenetic disturbances in the culture of human fibroblasts (HF). A decrease to intact condition of the number of HF with aneuploid and polyploid sets of chromosomes was observed after the introduction of non-immune autologous T-lymphocytes into the cultures. Immune homologous T-lymphocytes, unlike non-immune autologous T-lymphocytes, eliminated cells with structural disturbances of the chromosomes from the culture, but did not influence the level of aneuploid and polyploid cells. The ability of immune T-lymphocytes to exert antimutagenic effect can obviously be explained by their cytolytic action on virus-infected cells. As for non-immune autologous T-lymphocytes, two ways are equally probable: T-lymphocytes eliminate HF with virus antigens on their surface, or T-lymphocytes determine and destroy fibroblasts with changed cell surface developing as a result of cytogenetic disturbances induced by infectious factors. Specificity of the cytolytic reaction of T-lymphocytes concerning cells with some types of cytogenetic disturbances has been demonstrated.

[Applicability of micronucleus analysis for testing the aneuploid cell level in mouse bone marrow subjected to the action of different mutagenic factors].

The results of parallel studies in murine bone marrow of aneuploid cells' level and of micronuclei cells under the action of different mutagenic factors (X-irradiation, ethanol, measles virus and colchicine) are presented. The correlation analysis demonstrated the absence of correlation for the indices studied, in most cases. Possible reasons for this phenomenon are discussed.

[Age-dependent characteristics of immune cytolysis of mouse cells with cytogenetic changes induced by streptolysin O].

It has been determined that streptolysin-O induces cytogenetic alterations in the cell culture of murine kidneys in all the mice examined, the most pronounced alterations being noted in the cells of newborn and, especially, old mice. The injection of syngeneic lymphocytes from newborn and sexually mature mice of the middle age group has led to a considerable decrease in the number of cells with altered chromosome number in cultures. The lymphocytes from old mice have no antimutagenic activity.

Cytogenetic disturbances and immunoreactivity in patients with measles and influenza.

The number of lymphocytes with cytogenetic aberrations is increased in patients with measles and influenza. Considerable changes in immunoreactivity of the organism can be observed during measles and influenza. Correlation analysis suggests that in measles and influenza there exists an inversely proportional connection between the number and functional activity of T lymphocytes and the level of cytogenetic disturbances in them. Indices of B immune system and the level of cytogenetic aberrations in influenza were characterized by directly proportional correlation while in measles the type of correlation depended on the intervals of observation.

[Antimutagenic action of cellular and humoral immunity factors in cytogenetic disorders of cultured fibroblasts induced by a vaccinal strain of the measles virus].

In the human fibroblast culture media infected with measles virus (strain Leningrad-16), the injection of non-immune autogenic leucocytes and T-lymphocytes was shown to lead to a substantial decrease in fibroblast frequency with alterations in chromosome number and, in a lesser degree, with break in the structure of chromosomes. Immune homogenic leucocytes render an opposite effect. The injection of immune sera stabilized the cell frequency with chromosome aberrations. Non-immune homogenic leucocytes have no antimutagenic activity.

[Effect of nonviral parasitic agents on the cytogenetic apparatus of animals and man].

On the basis of literature survey, it is shown that many non-viral microorganisms (Rickettsiae, Mycoplasmata, Bacteria, Protozoa) and some Helminths are able to induce substantial aberrations in the chromosome number and the structure of human and animal cells. The data concerning possible factors of aggressiveness of the agents favouring cytogenetic disturbances are presented. Close association between immunoreactivity and the ability of parasitic agents to induce mutations in the organism conditions is emphasized.

[Comparative characteristics of different methods of studying the cytogenetic disturbances in the bone marrow cells of Macaca rhesus monkeys infected with the poliomyelitis virus].

It has been testified that to study the influence of viruses on cytogenetic structures of bone marrow cells of monkeys (Macaca mulatta), it is possible to use not only the traditional methods, i.e. the analysis of chromosome set and the pathologic division, but the micronuclear test as well, based on the study of polychromatophylous erythrocytes with micronuclei in the bone marrow of animals.
