[Nephelometric inhibition immunoassay (NIIA), a new methodology for the determination of phenobarbital and phenytoin concentration in serum (author's transl)].

Phenobarbital and phenytoin concentration was determined in serum by the aid of the nephelometric inhibition immunoassay (NIIA) and the homogeneous enzyme immunoassay (EMIT). Both methods showed a good precision and accuracy. The comparison of the two methods in 77 sera for phenobarbital (3--68 micrograms/ml) and 39 sera for phenytoin (1.3--44 micrograms/ml) showed a very close correlation. The correlation coefficient for phenobarbital was 0,9525 and for phenytoin 0,9714. The special merits of the NIIA-method were precision, less of interference and low costs in reagent material.

[Normal values of IgA, IgG, IgM, a1-antitrypsin, C2-complement, C4-complement, ceruloplasmin, haptoglobin and transferrin in serum of healthy, matured newborns and comparison with adults. Method: kinetic nephelometry. (author's transl)].

Concentrations of IgA, IgG, IgM, a1-antitrypsin, C3-complement, C4-complement, ceruloplasmin, haptoglobin and transferrin were determined by the kinetic nephelometric method in fasting blood serum of 88 healthy, matured newborns (male=46, female=42) and of 50 healthy adults (male=12, female=38). The age specific comparison showed that all the nine examined proteins had significantly lower levels in the group of newborns. Sex specific differences were not detected in the group of newborns.

[Non-invasive determination of oxygen saturation with the oxygenmet pulse wave oximeter. Comparison with the invasive reflectometric method (author's transl)].

Oxygen saturation was determined non-invasively with the oxygenmet pulse wave oximeter 1471 and at the same time with the invasive reflectometric method. The determinations were carried out on 49 children (males = 24, females = 25) during 52 catherizations of the heart. Among these children aged between one week and 15 years were 11 children suffering from cyanotic heart disease. Depending on age and health condition 1-8 blood samples from different arterial regions were analysed reflectometrically. The comparison between the oxygen saturations determined non-invasively with the oxygenmet oximeter and the oxygen saturations obtained invasively with the aid of the reflectometric method, showed a very close correlation (r = 0.923), however the differences were statistically significant. The comparison of the calculated regression line with the ideal regression line revealed that the values of the two methods differed considerably, especially in the range of low oxygen saturations. The causes for these differences, especially the absorption effect of the skin and the subcutaneous tissue of cyanotic patients, were discussed.

[Study of the concentration of triiodothyronine, thyroxine and thyreotropin in blood serum of euthyroid children (author's transl)].

Concentrations of triiodothyronine, thyroxine and thyreotropin were determined using the radioimmunoassay method in blood serum of 369 (male = 231, female = 138) euthyroid children. Beyond the newborn age no significant difference were found for the triiodothyronine and the thyreotropin concentration. Compilation of the values resulted in a physiological triiodothyronine concentration of 1.96 +/- 1.11 (1s) ng/ml -3.02 +/- 1.71 (1s) nmol/l- and in a thyreotropin concentration of 1.50 +/- 1.48 (1s) microunits/ml for infants, children and juveniles. In opposition to these results the thyroxine concentration showed a clear age-specific dependence. The thyroxine concentration decreased continuously from the infant age- (120 +/- 27 (1s) ng/ml, 154.8 +/- 34.8 (1s) to the juvenile age (69.1 +/- 16.4 (1s) ng/ml, 89.1 +/- 21.2 (1s) nmol/l).

[Amino acid losses in human sweat. Influence of oral contraceptives, physical exercise and sex on the excretion of amino acids in sweat (author's transl)].

UNLABELLED: With the aid of ion exchange column chromatography we determined quantitatively the free amino acids in thermal sweat of the total human body surface. Sweat of 60 healthy men and 63 healthy women (aged 18-31 years) collected during 12 min of sauna bathing was investigated. Thirty men and 31 women were well trained, 33 women were taking oral, hormonal contraceptives. Sweat was deproteinised by adding an equal volume of 5% sulphosalicylic acid. RESULTS: 1. The comparison of the women taking oral contraceptives with the women not taking oral contraceptives showed no qualitative or significant quantitative differences in the excretion of amino acids in thermal sweat collected from the total body surface. 2. The comparison of the trained women with the untrained women neither revealed qualitative or significant quantitative differences in the excretion of amino acids in thermal sweat collected from the total body surface. 3. As compared to the trained men, untrained men excreted in thermal sweat collected from the total body surface significantly more alanine, arginine, citrulline, cystine, glycine, histidine, isoleucine, leucine, lysine, phenylalanine, serine, threonine, tyrosine, and valine. 4. The sex-specific comparison of the amino acid excretion in sweat showed that men had a significantly increased excretion of cystine, histidine, leucine, and taurine. Proline was detected only in the sweat of women. 5. The quantitative results obtained for the amino acid excretions during 12 min of sauna bathing suggest that persons exposed to hard, physical activity and/or high environmental temperatures have quite equal amino acid losses in sweat and in 24 h urine.

A quantitative study of free amino acids in eccrine sweat collected from the forearms of healthy young men during sauna bathing.

Using ion-exchange column chromatography we measured the free amino acids in eccrine sweat collected from the forearms of 20 healthy young men at intervals of 15, 30, and 45 min during sauna bathing. In comparison with the amino acid concentrations determined after 15 min of sweating (100%) a significant and uniform decrease occurred in the concentrations of all amino acids examined after a further 15 min (65%), with a further decrease after another 15 min (45%). These results indicate that either the mechanism of excretion of amino acids in the secretory cells of the eccrine sweat glands, or the reabsorption of amino acids by the ductular cells, or the combination of both mechanisms are similar for all the amino acids examined here.

[Study on the determination of IgA, IgG, IgM, a1-antitrypsin, haptoglobin and transferrin with the kinetic nephelometric method. Comparison with the radial immunodiffusion method (author's transl)].

The concentration of IgA, IgG, IgM, a1-antitrypsin, haptoglobin and transferrin was determined by the kinetic nephelometric method (Beckman immunochemistry analyzer) and by the radial immunodiffusion method (Behring partigen immunodiffusion plates). The investigation was carried out in 151 normal and pathological sera. The results obtained with the radial immunodiffusion method were for IgA, IgG and IgM in middle about 15%, for a1-antitrypsin about 50% and for haptoglobin about 80% higher as compared to the results determined with the kinetic nephelometric method. An acceptable correspondence between the two methods was found for the results of transferrin.

Quantitative study of free amino acids in human eccrine sweat excreted from the forearms of healthy trained and untrained men during exercise.

The free amino acids in eccrine sweat collected from the forearms of 20 healthy trained and 20 healthy untrained men during controlled exercise were determined quantitatively using ion exchange column chromatography. Sweat was deproteinized by adding an equal volume of 5% sulphosalicylic acid. The amino acid concentrations showed a constant qualitative pattern in sweat and large individual differences. Essential amino acids, such as isoleucine, leucine, lysine, methionine, phenylalanine, and valine were excreted in relatively small amounts. As compared to the trained men, untrained men showed statistically significantly higher concentrations in sweat for the following amino acids: Alanine, arginine, glycine, histidine, isoleucine, leucine, lysine, ornithine, phenylalanine, serine, taurine, threonine, tyrosine, and valine. No significant differences were found for citrulline, cystine, ethanolamine, and methionine. The comparison of the amino acid excretions in sweat obtained under controlled exercise and in urine showed that the amounts of amino acids excreted in sweat under controlled exercise were comparable to the losses of amino acids in urine.

[Non-invasive determination of oxygen saturation with the oxygenmet pulse wave oximeter on fingers, metacarpus and wrist of infants. Comparison with the oxygen saturation calculated from the pH and pO2 of the blood gas analysis (author's transl)].

Oxygen saturation was determined non-invasively with the oxygenmet pulse wave oximeter 1471 and mathematically from the pH and pO2 of the blood gas analysis. The determination of the oxygen saturation by the aid of the oximetric method was carried out on fingers, metacarpus and wrist of 80 infants (male = 44, female = 36 - ages: 1 day - 1 year and 8 months). Blood gas analysis was done just behind the oximetric analysis and oxygen saturation was calculated from the pH and pO2. The comparison of the oxygen saturations determined non-invasively on fingers, metacarpus and wrist among themselves, showed no statistical significant differences. Also no statistical significant differences were observed between the results obtained with the pulse wave oximeter and the results calculated from the blood gas analysis. The comparison of the oxygen saturations determined with the oxygenmet oximeter (mean of fingers, metacarpus, wrist) with the oxygen saturations calculated from the blood gas analysis showed a very close correlation. The correlation coefficient was 0.930.

[Effect of 17-alpha-hydroxyprogesteronecapronate on sodium excretion and the renin-angiotensin-aldosterone-system in humans].

In seven healthy male subjects, a natriuretic effect of 17 alpha-hydroxyprogesterone caproate (17 alpha-OHPC) was demonstrated. Three of these subjects were kept on an uncontrolled diet and were examined over a period of 12 days. To the remaining four subjects, a single dose of 250 mg 17 alpha-OHPC was given intramuscularly after four days of intake of a chemically defined diet (Vivasorb). In this second test procedure, blood samples were taken in the recumbent position every two hours throughout a period of 12 h after the injection. For two more days and during the days before the administration of 17 alpha-OHPC, blood was taken at 8 a.m. before getting up from bed in same intervals, urine was collected for analysis of sodium and potassium excretion. During the first 12 h after the injection of 17 alpha-OHPC, the urinary sodium/potassium ratio significantly increased in all subjects. Plasma renin activity showed no characteristic changes at this time, whereas the plasma concentrations of aldosterone and cortisol decreased. The decrease of cortisol concentration started immediately after the injection and was more pronounced than that of plasma aldosterone. During the following 36 h, renin activity as well as aldosterone and cortisol concentrations in plasma showed an increase; in contrast, the sodium/potassium ratio decreased. On the basis of these results, the following effects of 17 alpha-OHPC are discussed: (1) an acute natriuresis which may be due to a competitive inhibition of aldosterone at the renal tubules, and (2) an inhibition of pituitary ACTH secretion or of adrenal steroid biosynthesis.

[Study of the glucose concentration in five vascular regions -- aorta, arteria pulmonalis, vena cava inferior, vena cava superior, vena hepatica -- in childhood (author's transl)].

Using the Beckman glucose analyzer, glucose concentration was determined in the serum of the fasting blood in five vascular regions -- aorta, arteria pulmonalis, vena cava inferior, vena cava superior, vena hepatica -- of 52 children (ages 2,5 months up to 14 years and 3 months, x = 6 years and 3 months, male = 28, female = 24) with a healthy metabolism. The comparison of the average values of the glucose concentrations determined in five vascular regions in childhood showed that the glucose concentration in the vena hepatica was statistically significant higher than in the other vascular regions. Moreover the comparison of the glucose concentrations revealed that the values in the vena cava inferior were statistically significant higher as compared to the values in the vena cava superior. The glucose concentrations in the arteria pulmonalis, the vena cava superior and the aorta were nearly equal. Compared with the older children, the younger children had in all the investigated vascular regions lower glucose concentrations.

[gamma-Glutamyl transpeptidase in human eccrine sweat (author's transl)].

UNLABELLED: The concentration of gamma-glutamyl transpeptidase was determined in eccrine, thermal sweat of 56 healthy men (ages 20--60 years) and 48 healthy women (ages 17--55 years). Samples of sweat were collected from the chest and back. RESULTS: The concentrations of gamma-glutamyl transpeptidase showed great individual variations. The sex specific comparison of the concentrations of gamma-glutamyl transpeptidase revealed that the women excreted in the sweat from the chest and the back nearly the double amount of this enzyme. The gamma-glutamyl transpeptidase concentrations determined in the sweat from the chest of the examined men and women were three times higher as compared with those excreted simultaneous from the back of the same persons. In the group of the women was this differences statistically significant.

[Refractometric measurement of total serum protein, comparison of refractometry and biuret test (author's transl)].

Study on the Refractometric Determination of Total Protein in Serum, Comparison of the Refractometric and Biuret Method. Total protein concentration in serum was determined by the aid of the Abbé-refractometer and the biuret method. Both methods showed a good precision and accuracy. The investigation was carried out in 241 sera with normal bilirubin (up to 1 mg/100 ml), cholesterol (up to 200 mg/100 ml) and urea (up to 23,0 mg/100 ml) concentration, in 43 sera with increased (10,6-26,6 mg/100 ml) bilirubin concentration, in 129 sera with increased (200-520 mg/100 ml) cholesterol concentration and in 43 sera with increased (23,0-155,3 mg/100 ml) urea concentration. The comparison of the refractometric values with the values obtained by the biuret method in the 241 sera with normal bilirubin, cholesterol and urea concentration (correlation coefficient = 0,971) showed a close correlation and in the 43 sera with increased bilirubin concentration (correlation coefficient = 0,958) an acceptable correlation. However no close correlations were observed in the 129 sera with increased cholesterol concentration and in the 43 sera with increased urea concentration. The correlation lines diverged proportional with the increase of cholesterol and urea concentration from the expected correlation lines.

[Normal range of free amino acids in cerebrospinal fluid of children (author's transl)].

UNLABELLED: Free amino acids were determined quantitatively in cerebrospinal fluid of 80 boys and 69 girls (ages 1 month-14years). On the basis of the cliniclal criterions these children were considered to be ""healthy''. The total number of examined children was 2000. Cerebrospinal fluid was mixed with an equal volume of 5% sulphosalicylic acid and the amino acids were separated by ion exchange column chromatography. RESULTS: 1. The sexspecific investigation of the 1 month -2 years old children showed that the boys had a statistical significant hgher concentration of lysine and that no statistical significant differences existed between the groups of 2-14 years old children. 2. The age specific comparison revealed that the amino acid concentrations ofaspartic acid, citrulline, histidine, lysine, serine, threonine and tyrosine were in the cerebrospinal fluid of the younger boys statistical significant higher as compared to the older boys and that the concentration of serine in the cerebrospinal fluid of the younger girls was statistical significant higher than in the group of older girls.
