B cell suppressing and CD8+ T cell enhancing effects of photosensitive dye platonin in humans.

Platonin is one of the photosensitive dyes of trithiazole pentamethine cyanine. It is used as an effective medicine for rheumatoid arthritis. In our study, platonin suppressed the immunoglobulin (Ig) production of human peripheral blood lymphocytes (PBL) stimulated by Staphylococcus aureus Cowan I (SAC) or pokeweed mitogen (PWM). It also suppressed the PWM induced Ig production of B cells when T cells and/or B cells were pretreated with platonin, respectively. The percentage of CD8+ T cells was increased by platonin. Our results suggest that platonin suppresses Ig production through suppressing B cells and enhancing CD8+ (suppressor/cytotoxic) T cells.

Comparative evaluation of chewing function with removable partial dentures and fixed prostheses supported by the single-crystal sapphire implant in the Kennedy Class II partially edentulous mandible.

Differential chewing function with removable partial dentures (RPDs) and fixed prostheses supported by the single-crystal sapphire implant was evaluated in five subjects with Kennedy Class II partially edentulous mandibles by means of electromyography. Rehabilitation with the single-crystal sapphire implant resulted in regular chewing patterns with a low variation coefficient and higher activity of chewing-side masticatory muscles compared to RPD rehabilitation. This difference in chewing function between the two rehabilitation modalities could be the result of differences in stability of occlusion and neurophysiologic feedback systems.

IL-2 receptor(p55)/Tac-inducing factor. Purification and characterization of adult T cell leukemia-derived factor.

Many human T cell lymphotropic virus-I (HTLV-I) transformed T cells from adult T cell leukemia (ATL) patients continuously produce a humoral factor called ATL-derived factor (ADF) which induces IL-2R/Tac expression on T and NK cells. Using gel filtration, procion red Sepharose, DEAE, and reverse phase chromatography, we have purified ADF protein to homogeneity from 15 liters of serum-free culture supernatant of an HTLV-I(+) T cell line ATL-2. Purified ADF protein had the m.w. of 14,000 by SDS-PAGE and gel filtration, and its isoelectric point is around 5.0. ADF did not react with heteroantibodies against IL-1 alpha and IL-1 beta, which have also IL-2R/Tac-inducing activity on suitable target cells. Partial N-terminal amino acid sequence of ADF is different from other cytokines such as, IFN, BSF-2, and various IL whose cDNA has been cloned. Western blot analysis using rabbit antibodies against N-terminal 10mer synthetic peptide of ADF showed that IL-1 alpha and ADF are different proteins. ADF had its IL-2R/Tac-inducing activity not only on human NK-like cell line YT, but also on HTLV-I(+) T cells, such as ED. In contrast, macrophage-derived IL-1 lacked IL-2R/Tac-inducing activity on ED cells despite their IL-2R/Tac induction on YT, indicating that ADF and IL-1 have their effect via different receptors.

A soluble 'anchorminus' interleukin 2 receptor suppresses in vitro interleukin 2-mediated immune responses.

The immunosuppressive effects of a recombinant soluble IL-2 receptor L chain (s-IL-2R) were analyzed. S-IL-2R protein was obtained from the conditioned medium of L cells transfected with a mutant cDNA clone encoding the extracytoplasmic portion of the IL-2 receptor (IL-2R) and was purified to homogeneity by an IL-2-coupled sepharose column, following by reverse phase chromatography (HPLC). Soluble IL-2R protein thus prepared retained the ability to bind IL-2 specifically and suppressed the in vitro IL-2-mediated immune responses, including proliferation of IL-2-dependent cell line (CTLL-2), induction of secondary cytotoxic T lymphocytes (CTL) and the mixed lymphocyte reaction (MLR), but did not suppress the growth of IL-3-dependent cell line. Kinetic studies revealed that s-IL-2R exhibited the suppressive effects on the proliferative responses of alloantigen stimulated human tonsillar cells, only when added at an early stage, namely 0-48 h after culture onset, whereas cyclosporin A (CsA) exhibited an inhibitory effect only when added at between 0 and 24 h. This implies that s-IL-2R exerts its effect on an early stage of lymphocyte activation. The observed immunosuppressive effects of s-IL-2R suggest the possibility that s-IL-2R might be useful for the protection of rejection crisis in organ transplantation.

Electromechanical effects of bPTH-(1-34) on rabbit sinus node cells and guinea pig papillary muscles.

The effects of bPTH-(1-34), a synthetic preparation of bovine parathyroid hormone containing the first 34 amino acids, on electromechanical activity of isolated rabbit sinus node cells and guinea pig papillary muscles were examined by microelectrode techniques. In sinus node cells, bPTH-(1-34) (10(-7) M) decreased the cycle length of spontaneous firing (SPCL) accompanied by an increase in the maximum upstroke velocity at phase 0 (dV/dtmax) without affecting the amplitude of the action potential (APA) or the maximum diastolic potential (MDP). All the effects of bPTH-(1-34) on sinus node cells were abolished by verapamil (5 X 10(-7) M), but not by pindolol (2 X 10(-7) M). In constantly driven guinea pig papillary muscles, bPTH-(1-34) caused a positive inotropic effect (+31%). The parameters of the action potential were not significantly affected. This inotropic effect of bPTH-(1-34) was inhibited by pretreatment with verapamil or a low calcium medium (0.12 mM), but was not affected by pindolol. In contrast, ryanodine (2 X 10(-6) M), an inhibitor of internal Ca2+ release, which decreased the contraction with a prolongation of the action potential duration augmented the inotropic effect of bPTH-(1-34). In papillary muscles depolarized by 26 mM [K+]O, bPTH-(1-34) enhanced the slow action potential. In voltage clamp experiments using a single sucrose-gap method, bPTH-(1-34) caused an increase in the peak amplitude of the slow inward current, while it did not affect the outward current.(ABSTRACT TRUNCATED AT 250 WORDS)

Expression of proto-oncogenes in normal and tumor tissues of human skin.

The expression of c-fos, c-myc, Ha-ras, N-ras, EGF-receptor, and cardiac actin genes was examined in 7 normal epidermis, 3 cellular nevi, and 8 skin tumors including 6 malignant and 2 benign tumors of human origin. These genes were transcribed in most normal and tumor tissues, though no tumor-specific expression of proto-oncogenes (c-fos, c-myc, Ha-ras, and N-ras) could be detected. However, there was a characteristic parallelism between the expression of c-fos and c-myc in normal epidermis, while the parallelism was not always definite in skin tumors. The ratio of c-fos/c-myc transcripts in normal epidermis was constant compared with the expression of other genes examined. These data suggest that c-fos and c-myc are expressed in all normal skin tissues, and that maintenance of a constant ratio of c-fos/c-myc is closely related to ordered cell growth of the tissues.

Comparison between effects of caffeine and ryanodine on electromechanical coupling in myocardium of hibernating chipmunks: role of internal Ca stores.

1. To clarify the cause of uncoupling of Ca influx through Ca channels and the contractility of the myocardium in hibernating chipmunks, the electromechanical effects of two different internal Ca store inhibitors, caffeine and ryanodine, and a cardiotonic agent, isoprenaline, were investigated in papillary muscles of hibernating animals. 2. Ryanodine (10(-6) M), an inhibitor of internal Ca release, abolished the contraction with a marked inhibition of the action potential plateau (APp). In such preparations, an increase in Ca influx induced by isoprenaline (5 x 10(-8) M) failed to augment the contraction, indicating uncoupling of Ca influx and contraction. 3. In ryanodine pretreated preparations, 10 mM caffeine produced an early phase of APp, but did not affect the contraction abolished by ryanodine, while a higher concentration of caffeine (25 mM) markedly increased the contraction with an augmentation of the electrical response. 4. In the absence of ryanodine, caffeine (5 mM) almost abolished the contraction with a greater inhibition of APp. In such preparations, isoprenaline greatly increased the contraction with an augmentation of the early phase of APp. 5. These effects were not significantly affected by additional application of ryanodine, but were inhibited by nifedipine, a Ca channel blocker. 6. These observations suggest that in cardiac muscles of hibernating animals, lack of the positive inotropic effect of isoprenaline may be attributed to a rapid and effective sequestration of increased cytoplasmic Ca through Ca influx by internal stores, probably by enhancement of their ability to take up Ca.

Successful resection of alpha-fetoprotein-producing lung cancer.

We present a case of successfully resected alpha-fetoprotein (AFP)-producing primary lung cancer. AFP in the serum decreased from 289 ng/ml to an undetectable level after a curative operation. The resected tumor was stained positively by the immunohistochemical study for AFP. The histologic appearance of the tumor was well-differentiated adenocarcinoma similar to that of fetal gut epithelium and apparently different from extragonadal germ cell tumor. This case provides evidence that lung cancer cells possess the potential to produce AFP.

[Treatment of nonseminomatous testicular tumor with liver metastases].

Therapeutic course of 2 cases of nonseminomatous testicular tumor with liver metastasis is reported. One case had mixed tumors including embryonal carcinoma, choriocarcinoma and yolk sac carcinoma, and was positive pulmonary metastasis already at the initial examination. In the other case having mixed tumors of embryonal carcinoma and choriocarcinoma, metastasis to the supraclavicular lymph node was detected at the initial examination. In both cases liver metastasis occurred after complete response could be obtained by treatment chiefly consisting of PVB therapy. For liver metastasis four-drug combination treatment using cisplatin, vinblastine, adriamycin and actinomycin D was performed with partial response. However, this patient eventually died. The other case received VAB-6 therapy with complete response for liver metastasis. It is advisable to consider other modalities therapy in addition to conventional chemotherapy in cases of testicular tumor with liver metastasis since the prognosis is poor in these cases.

Identification of a pre-hibernating state in myocardium from nonhibernating chipmunks.

During the hibernating season, the amplitude of the cardiac action potential plateau of nonhibernating chipmunks was reduced. Replacing external Ca by Sr inhibited the electromechanical responses of these preparations. Similar properties were observed in hibernating animal preparations, suggesting that changes in cardiac function are already triggered before hibernation begins.

Rat lymphoid cell lines with HTLV-I production. III. Transmission of HTLV-I into rats and analysis of cell surface antigens associated with HTLV-I.

Newborn WKA rats given a single intraperitoneal injection of MMC treated TARS-1, a rat T cell line producing HTLV-I, were shown to accumulate high titred antibodies specific for HTLV-I. Adult WKA rats rejected TARS-1 transplant with transient appearance of anti-HTLV-I antibodies. However, rats maintained under daily administration of Bredinin, an immunosuppressive drug after TARS-1 transplant showed continuous production of antibodies specific for HTLV-I by aging. Type-C virus particles similar to HTLV-I were demonstrated by electronmicroscopy in the short-term cultured spleen cells of these rats. The evidence indicates that HTLV-I can be transmitted into newborn and immunosuppressed adult rats and they may provide a suitable animal model of ATL and related conditions in man, especially for elucidating the virus-host interactions involved in the leukemogenesis of HTLV-I. By using monoclonal antibodies, cell surface antigens associated with HTLV-I were also analysed.

Can strontium replace calcium as an activator of internal calcium release in cardiac muscles? Study on a dual action of A23187.

The positive inotropic effect of A23187 has previously been indicated to be attributed to both the enhancement of the slow Ca inward current and the facilitation of Ca release from internal stores. The electromechanical effects of replacing external Ca by Sr on this dual action of A23187 were examined on guinea-pig papillary muscles driven at 0.2 Hz. Replacing most of external Ca by Sr (Sr 2.5 mM plus Ca 0.2 mM) (Sr-medium) increased the developed tension. This procedure simultaneously prolonged the time to peak developed tension (tPT) and the action potential duration at all repolarization phases (APD 10, APD 30 and ADP 80). In such preparations, A23187 (10(-6) M) caused a marked positive inotropic effect (938 +/- 180% of control) accompanied by a prolongation of APD at an early repolarization phase (APD 10), but did not affect tPT. Thereafter, returning superfusate to normal Ca medium (Ca 1.2 mM) in the presence of A23187 also caused a positive inotropic effect, but markedly shortened tPT. The electrical and mechanical responses to A23187 in Sr-medium were completely blocked by nifedipine (2 X 10(-6) M), Ca-channel-blocker, while the positive inotropic effect in normal Ca medium was not blocked by nifedipine. Ryanodine, an inhibitor of internal Ca release, did not affect the positive inotropic effect of A23187 in Sr-medium. These results suggest that in Sr-medium, an internal Ca pool of the myocardium, most likely sarcoplasmic reticulum, does not play a role in the activation of contraction.

Expression of murine interleukin-2 cDNA in E. coli and biological activities of recombinant murine interleukin-2.

Murine interleukin-2 (MIL-2) cDNA was inserted into an expression vector carrying an Escherichia coli tryptophan promoter and was expressed in E. coli. Recombinant MIL-2 produced by E. coli supported the growth of murine CTLL-2 cells, but not that of human T-cell blasts. Recombinant MIL-2 strongly inhibited the binding of recombinant human IL-2 (HIL-2) to murine responder cells, but only very weakly inhibited the binding to human responder cells. Moreover, recombinant MIL-2 induced secondary alloantigen specific cytotoxic T lymphocytes (2 degrees CTL) from memory CTL and activated natural killer (NK) cells in murine systems in the same manner as recombinant HIL-2. The results suggest that the species hierarchy (that MIL-2 derived from native cell culture does not act on human T-cells) is due to the protein moiety, not the sugar moiety, and is to be ascribed to the difference in binding affinity of MIL-2 and HIL-2 to murine and human responder cells respectively, and that recombinant MIL-2 shares identical biological and immunological activities with recombinant HIL-2. Thus, MIL-2 might be a convenient tool for extensive studies of the pharmacological and physiological activities of IL-2 in murine models.
