[Monostotic fibrous dysplasia in the parietal bone: a case report].

In this paper a case of monostotic fibrous dysplasia in the left parietal bone is described. This 51-year-old female was admitted to our hospital for a head injury on January 12, 1983, and a flat, painless hump (7 X 7cm) was incidentally found in the left parietal region. Plain x-ray film of the skull showed a multicystic lesion with slightly sclerotic margin in the left parietal bone, and outward bulging of the outer table without destruction of the inner table in tangential view. CT scan at the level of bone window clearly demonstrated the same abnormality and intradiploic mass separated by some bony septums. Angiography revealed no positive findings, but RI bone scan (99m Tc) showed an abnormal uptake in this lesion. On February 1, 1983, operation was performed to verify the nature of the lesion and for relief of the left-sided headache. Post operative course was uneventful. Histological findings of this mass were of inactive fibrous dysplasia with fibrous tissue replacing the normal bone and surrounded by island or scorlled edge of bones. Monostotic fibrous dysplasia in the cranial vault is rare. It is often possible to make diagnosis in this disease on the clinical course, plain x-ray film and CT scan showing cystic findings and outer table bulging, and RI scan demonstrating abnormal uptake.(ABSTRACT TRUNCATED AT 250 WORDS)

High accumulation of 2,3,4,7,8-pentachlorodibenzofuran to hepatic microsomes of rats.

The tissue and subcellular distributions of 14C-2,3,4,7,8-pentachlorodibenzofuran (PenCDF), one of the most important causal agents of yusho, were studied using rats. More than 60% of the radioactivity given orally was accumulated in the liver after 5 d and this high percentage persisted over a period of 3 weeks. Subcellular fractionation of the liver homogenate showed unusual separation by PenCDF-pretreatment, but the distribution of radioactivity was just parallel to those of cytochrome P-450 content and glucose-6-phosphatase (EC3.1.3.9) activity. Gas chromatographic analysis provided evidence that the extracts from the liver and its subcellular fractionations contained only unchanged PenCDF. Those results strongly suggest that PenCDF has some affinity to endoplasmic reticulum of rat liver.

Studies on cis-trans isomerization of nitrofuran derivatives by bacterial nitroreductases.

cis-trans Isomerization of 3-(5-nitro-2-furyl)-2-(2-furyl)-acrylamide(AF-2) using Escherichia coli B/r and its two 5-nitro-2-furaldehyde semicarbazone (nitrofurazone)-resistant mutants was investigated. The isomerizing activity was detected in all three strains and markedly increased with acquiring resistance to nitro-furazone in intact cells and cell free extracts. Two distinct isomerases, nicotinamide adenine dinucleotide phosphate(NADPH)-dependent one with high activity and NAD(P)H-dependent with low activity were separated by Sephadex G-100 and Sepharose 4B columns. Both enzyme activities agreed with the nitrofurazone-reducing activity due to O2-sensitive nitroreductase as reported previously. Another nitroreductase, O2-insensitive one, was unable to isomerize cis AF-2 to trans form. These results suggest that bacterial cis-trans isomerases are not O2-insensitive nitroreductase, but O2-sensitive ones.

Favorable effect of dimethyl sulfoxide on secondary amyloidosis in juvenile rheumatoid arthritis.

A girl with secondary amyloidosis as a complication of juvenile rheumatoid arthritis was administered dimethyl sulfoxide by topical application to the skin. Her gastrointestinal symptoms and massive proteinuria improved. Decreased left ventricular function and creatinine clearance also improved remarkably. The favorable effect of dimethyl sulfoxide in this single patient deserves further study in a controlled trial.

Structural analysis of gene loci for rat U1 small nuclear RNA.

Four phage clones which hybridize with U1 small nuclear RNA were obtained from a rat gene library. Two clones contain a presumed pseudogene. A third clone includes two gene candidates that are co-linear with the rat U1-RNA, 3.6kb apart and in the opposite orientation. The two genes are surrounded by identical sequences of 491bp upstream and 178bp downstream. The upstream sequences do not contain a TATA box, but share many block homologies with those for the human U1-RNA gene(1-3). A 101bp "identifier (ID) sequence", which was reported to be specifically expressed in rat brain (4), is inserted immediately after the shared sequence downstream of one of the genes. In the fourth clone, there are two putative pseudogenes, which have one or three nucleotide changes, 3kb apart and in the same orientation. Southern blot analysis of total rat DNA reveals about 50 U1-RNA genes/pseudogenes in the genome.
