[A study of the genetic diversity of common wild rice (O. rufipogon Griff.) and cultivated rice (O. sativa L.) by RFLP analysis].

The genetic diversity of cultivated rice and common wild rice from 10 countries in Asia were evaluated by RFLP analysis using 44 probes of single copy. We used the parameters such as proportion of polymorphic loci (P), average number of alleles per locus (A), average number genotype per locus (Ng), degree of heterozygate per locus (Ho) and average gene diversity (Hs) showing genetic diversity. It is indicated that the common wild rice from China has the largest genetic diversity in the 10 countries, and the secondary is the common wild rice from India. The results also showed that the genetic diversity in Chinese common wild rice is larger than the genetic diversity in common wild rice of South and Southeast Asia. The average gene diversity (Hs) of South Asian wild rice is higher than Southeast Asian wild rice, while the parameters value of proportion of polymorphic loci (P), average number of alleles per locus (A) and average number genotype per locus (Ng), Southeast Asian wild rice is higher than South Asian wild rice. It is found that the genetic diversity in cultivated rice is obviously lower than in common wild rice. In the detected 44 loci, the number of polymorphic loci on O. sativa is as the same as 3/4 in O. rufipogon, the number of alleles in O. sativa is as the same as 60% in O. rufipogon, the number of genotype in O. sativa is as same as 1/2 in O. rufipogon. In O. sativa, the genetic diversity of indica is larger than that of japonica. O. rufipogon from China have highest value in degree of hetrozygate per locus (Ho), and Ho of wild rice is as two times as cultivated rice. It is showed that in the proceeding of wild rice evolved into cultivated rice, Ho and number of alleles was reduced, and the genetic diversity was decreased.

Factor structure of satisfaction in daily life of elderly residents in Kitakyushu.

The purpose of this study was to investigate what items elderly persons living at home consider important for satisfaction in daily life and what factor structure is obtained by a principal components analysis. Two percent of the residents 60 years old or over of Yahatanishi Ward, Kitakyushu, were randomly sampled as subjects. A questionnaire asking residents to select items considered important in daily life was sent by mail, and 722 responses (72.5%) were eligible for analysis. "Health", "social security and pension" and "self-care" were considered important in daily life with the highest selection rates. A five-factor solution for 25 items with a selection rate greater than 4.5% was obtained by an oblique rotated principal components analysis: "health and disability", "social environment and activity", "socio-economic condition", "emotional stability in marital and family life" and "social intercourse and companionship". These factors appeared to be the fundamental framework of the subjective domain of health-related quality of life of elderly persons living at home.

Selective genotyping for the role of 5-HT2A, 5-HT2C, and GABA alpha 6 receptors and the serotonin transporter in the level of response to alcohol: a pilot study.

BACKGROUND: The vulnerability to alcohol dependence appears to be genetically influenced through a variety of mechanisms. One potentially genetically mediated channel may be a low level of response (LR) to alcohol, which has been seen in children of alcoholics and noted to predict future alcohol abuse and dependence. This pilot study uses a case and control genetic association approach to evaluate the possible role of five genotypes in both LR and alcoholism in informative subgroups of men with high and low LR scores documented 15 years earlier. METHODS: As part of a larger study, 41 men, about 39 years old, were selected from among the first 113, completed 15-year follow-ups in a prospective study. The 17 subjects whose LRs at age 20 were in the lower third were compared on five polymorphisms of four genes with 24 men whose reactions to alcohol had been above the median. RESULTS: The 14 men with the LL genotype of the serotonin transporter (5-HTT) polymorphism and the seven with the Pro/Ser genotype of the GABAA alpha 6 polymorphism had demonstrated lower LR scores at about age 20, and had significantly higher proportions of alcoholics than the other genotypes for those loci. All four subjects with combined LL and Pro/Ser genotypes had developed alcoholism and demonstrated the lowest LR scores overall. There was no evidence that two polymorphisms of the 5-HT2A receptor gene and one of the 5-HT2C receptor gene were related to LR or alcoholism in this sample. CONCLUSIONS: These results are consistent with animal and human studies suggesting a possible role for genetic variation in the GABAA alpha 6 and the serotonin transporter in the reaction to alcohol and the alcoholism risk.

Gender-related differences in scores of the Barthel Index and Frenchay activities index in randomly sampled elderly persons living at home in Japan.

The purpose of this study was to examine for gender-related differences in activities of daily living (ADL) and lifestyle of elderly persons living at home, and to support our hypothesis that the gender-related difference in lifestyle of stroke patients derives from their lifestyle prior to the stroke. Participants were randomly sampled elderly persons living at home. Questionnaire sheets including subject profile, Self-Rating Barthel Index (disability index), and Self-Rating Frenchay Activities Index (activity index) were mailed and collected, and the data were analyzed with the t-test and General Linear Model (factorial model with interaction). A total of 752 subjects were recruited, and their average age was 67.1 years. No significant gender-related differences were evident in the disability index including self-care and mobility domains (t-test, P > 0.05). In contrast gender-related differences in the activity index were significant (t-test, P < 0.05) for three factors; gender, age group, and living conditions, and in a covariate disability index (GLM, P < 0.05). Because randomly selected elderly persons in this study exhibited a prominent gender-related difference in lifestyle, we believe the lifestyle difference in stroke patients that we have previously described derives primarily from their premorbid attitude to daily life.

Relationship between a GABAA alpha 6 Pro385Ser substitution and benzodiazepine sensitivity.

OBJECTIVE: In humans, interindividual variation in sensitivity to benzodiazepine drugs may correlate with behavioral variation, including vulnerability to disease states such as alcoholism. In the rat, variation in alcohol and benzodiazepine sensitivity has been correlated with an inherited variant of the GABAA alpha 6 receptor. The authors detected a Pro385Ser [1236C > T] amino acid substitution in the human GABAA alpha 6 that may influence alcohol sensitivity. In this pilot study, they evaluated the contribution of this polymorphism to benzodiazepine sensitivity. METHOD: Sensitivity to diazepam was assessed in 51 children of alcoholics by using two eye movement measures: peak saccadic velocity and average smooth pursuit gain. Association analysis was performed with saccadic velocity and smooth pursuit gain as dependent variables and comparing Pro385/Ser385 heterozygotes and Pro385/Pro385 homozygotes. RESULTS: The Pro385Ser genotype was associated with less diazepam-induced impairment of saccadic velocity but not with smooth pursuit gain. CONCLUSIONS: The Pro385Ser genotype may play a role in benzodiazepine sensitivity and conditions, such as alcoholism, that may be correlated with this trait.

Reliability of the State-Trait Anxiety Inventory, Form Y in Japanese samples.

The internal consistency of the State-Trait Anxiety Inventory, Form Y was examined using data collected from Japanese participants by five diverse surveys, in which one included American university students. Cronbach coefficient alpha was calculated separately for state and trait items as well as for anxiety-present and absent items. The internal consistency was higher for the anxiety-absent items than those of the state and trait anxiety items, but this tendency was not clear for the anxiety-present items. The trait anxiety items showed the lowest internal consistency for all Japanese groups, whereas the anxiety-present items showed the lowest alpha for American university students. It can be considered that this difference might induce the difference in two--factor structure between Japanese and people in Western countries.

Effects of mailed advice on stress reduction among employees in Japan: a randomized controlled trial.

We conducted a randomized controlled trial (RCT) to examine the effects of mailed advice on reducing psychological distress, blood pressure, serum lipids, and sick leave of workers employed in a manufacturing plant in Japan. Those who indicated higher psychological distress (defined as having GHQ scores of three or greater) in the baseline questionnaire survey (n = 226) were randomly assigned to an intervention group or a control group. Individualized letters were sent to the subjects of the intervention group, informing them of their stress levels and recommending an improvement in daily habits and other behaviors to reduce stress. Eighty-one and 77 subjects in the intervention and control groups, respectively, responded to the one-year follow-up survey. No significant intervention effect was observed for the GHQ scores, blood pressure, serum lipids, or sick leave (p > 0.05). The intervention effect was marginally significant for changes in regular breakfasts and daily alcohol consumption (p = 0.09). The intervention effect was marginally significant for the GHQ scores among those who initially did not eat breakfast regularly (p = 0.06). The study suggests that only sending mailed advice is not an effective measure for worksite stress reduction. Mailed advice which focuses on a particular subgroup (e.g., those who do not eat breakfast regularly) may be more effective.

Job stressor-mental health associations in a sample of Japanese working adults: artifacts of positive and negative questions?

To examine whether positive and negative components of mental health were differently related to job stressors and life events, correlational analyses were conducted using data for the GHQ-12 and some scales of the NIOSH Generic Job Stress Questionnaire collected from 765 workers in Japan. Six positive items and six negative items of the GHQ-12 were summed up for positive and negative components of mental health (GHQ-POS, GHQ-NEG). The GHQ-POS was significantly correlated with only positively-oriented job stressors. The GHQ-NEG was significantly correlated with only negatively-oriented job stressors. Most correlations were significantly different between GHQ-POS and GHQ-NEG. This correlation pattern resulted in smaller, but significant, correlations between job stressors and the GHQ-12. These results may reflect "measuring similarity" due to item-wording. Detailed inspection suggested that GHQ-POS and GHQ-NEG, as well as positively- and negatively-oriented job stressors, were not attributable only to positive affectively or negative affectivity, respectively. Work-related events showed higher correlations with all mental health variables than their job stressors' counterparts. For females, mental health variables were seldom correlated with job stressors, but significantly correlated with life events. These results might indicate the superiority of "event-type stress measure" as compared to perceived rating scale in assessing job stressors. Further direction was discussed.

Cold milk accelerates oro-cecal transit time during the luteal phase but not the follicular phase in women.

The effect of the menstrual cycle on oro-cecal transit time (OCTT) has been controversial. Since poor reproducibility of OCTT measurements by lactulose might be responsible for this controversy, we measured OCTT with either milk or a solid test meal during the luteal and follicular phases of the menstrual cycle. Nine healthy young women (21.9 +/- 0.42 years old) with regular menstruation were studied for 4 consecutive menstrual cycles. Control (37 degrees C) or cold (10 degrees C) milk was used as a liquid meal, and the OCTT measurements were taken 3 times at each milk temperature during each of the 2 phases for 3 consecutive menstrual cycles. OCTT after a solid test meal (cooked rice, miso soup, a boiled egg, and cooked soybeans with mixed vegetables) was studied twice in 1 menstrual cycle. Breath hydrogen was determined every 15 min for 6 h. OCTT was defined as the time when breath hydrogen showed a sustained rise of 3 ppm or more from baseline. OCTT was not different between the luteal and follicular phases when the test meal was control milk or the solid meal. When cold milk was used as the test meal, OCTT was significantly shorter during the luteal phase (134 +/- 15 min) than during the follicular phase (165 +/- 21 min). In conclusion, cold milk accelerates OCTT during the luteal phase but not the follicular phase of the menstrual cycle in women.

[Central nervous system relapse with multiple brain masses in an acute promyelocytic leukemia patient treated with all-trans retinoic acid].

A 22-year-old woman with fever and bleeding tendency was given a diagnosis of acute promyelocytic leukemia (APL) on the basis of laboratory findings including a WBC count of 106 x 10(3)/microliter (90% blasts) and a platelet count of 1.6 x 10(4)/microliter. Induction therapy was started with all-trans retinoic acid (ATRA) and cytotoxic chemotherapy. After the patient achieved complete remission, ATRA was discontinued and consolidation chemotherapy was started. However, 4 months after onset, leukemic blasts were detected in cerebrospinal fluid. Temporal central nervous system remission was induced by intrathecal chemotherapy only. However, 2 months later, multiple focal mass lesions had developed in the brain. ATRA (45 mg/m2) was restarted together with multiple intrathecal injections of anticancer drugs, and a third remission was achieved. It is conceivable that the incorporation of ATRA in induction chemotherapy is related to the development of this rather rare complication of APL. The outcome in this case suggested orally administered ATRA may be effective in treating brain metastasis of APL.

[Recurrent bleeding tendency due to factor VIII inhibitor during therapy for bullous pemphigoid].

A 67-year-old woman with bullous pemphigoid (who had no history of hemorrhagic disease or blood transfusion) experienced extensive ecchymosis of the trunk and extremities and marked submucosal bleeding of the pharynx and larynx with risk of obstruction and suffocation. This bleeding tendency was the manifestation of a coagulation disorder due to factor VIII inhibitor. Immunosuppressive therapy, steroid pulse therapy, prednisolone (PSL), and cyclophosphamide (CPA) alleviated the bleeding and yielded negative assays for factor VIII inhibitor. However, because the patient stopped treatment, the bleeding recurred and was especially severe from a large hematoma with ruptured skin on the right hand. The bleeding was mitigated by transfusion of factor VIII concentrate combined with steroid pulse therapy. This was followed by CPA pulse therapy and oral PSL and CPA, resulting in the disappearance of factor VIII inhibitor again. We reported this case because factor VIII inhibitor complication of bullous pemphigoid is very rare and immunosuppressive therapy consisting of PSL, CPA, and pulse therapy was effective.

Identification of a naturally occurring Pro15-Ser15 substitution in the serotonin5A receptor gene in alcoholics and healthy volunteers.

We screened the serotonin5A receptor gene coding region in 186 unrelated alcoholic patients and 187 controls. A relatively abundant amino acid substitution and two synonymous DNA substitutions were detected. Two synonymous variants, A12T and C789T, had rarer-allele frequencies of 23% and 1%, respectively. The Pro15Ser substitution is located in the amino terminal, extracellular domain of the receptor adjacent to a putative phosphorylation site. Pro15Ser had rarer-allele frequencies of 8.1% and 5.9% in Finnish alcoholic patients and controls, respectively (p=n.s.).

Characterization of temperature rise of the brain and the rectum following intracerebroventricular administration of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate and kainate in rats.

Intracerebroventricular administration of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) or kainate caused a rise of the temperature of the brain and the rectum in urethane-anesthetized rats. An AMPA-kainate receptor antagonist, 6,7-dinitroquinoxaline-2,3-dione (DNQX), significantly suppressed the AMPA- and kainate-induced rises of brain and rectal temperatures. An N-methyl-d-aspartate receptor antagonist, MK-801, also suppressed the rises of the brain and rectal temperatures induced by AMPA or kainate, but the profiles of the suppressive effects of MK-801 were different between rats treated with AMPA and kainate. An antipyretic agent, indomethacin, completely suppressed the AMPA-induced rises of brain and rectal temperatures. Although indomethacin completely suppressed the kainate-induced rise of the rectal temperature as well, the brain temperature was still raised. These findings suggest that distinct mechanisms may be involved in the temperature rise of the brain and the rectum mediated through AMPA and kainate receptor stimulation.

Molecular chaperone GRP94 binds to the Fanconi anemia group C protein and regulates its intracellular expression.

The FAC protein encoded by the gene defective in Fanconi anemia (FA) complementation group C binds to at least three ubiquitous cytoplasmic proteins in vitro. We used here the complete coding sequence of FAC in a yeast two-hybrid screen to identify interacting proteins. The molecular chaperone GRP94 was isolated twice from a B-lymphocyte cDNA library. Binding was confirmed by coimmunoprecipitation of FAC and GRP94 from cytosolic, but not nuclear, lysates of transfected COS-1 cells, as well as from mouse liver cytoplasmic extracts. Deletion mutants of FAC showed that residues 103-308 were required for interaction with GRP94, and a natural splicing mutation within the IVS-4 of FAC that removes residues 111-148 failed to bind GRP94. Ribozyme-mediated inactivation of GRP94 in the rat NRK cell line led to significantly reduced levels of immunoreactive FAC and concomitant hypersensitivity to mitomycin C, similar to the cellular phenotype of FA. Our results demonstrate that GRP94 interacts with FAC both in vitro and in vivo and regulates its intracellular level in a cell culture model. In addition, the pathogenicity of the IVS-4 splicing mutation in the FAC gene may be mediated in part by its inability to bind to GRP94.

Expression of Xa1, a bacterial blight-resistance gene in rice, is induced by bacterial inoculation.

The Xa1 gene in rice confers resistance to Japanese race 1 of Xanthomonas oryzae pv. oryzae, the causal pathogen of bacterial blight (BB). We isolated the Xa1 gene by a map-based cloning strategy. The deduced amino acid sequence of the Xa1 gene product contains nucleotide binding sites (NBS) and a new type of leucine-rich repeats (LRR); thus, Xa1 is a member of the NBS-LRR class of plant disease-resistance genes, but quite different from Xa21, another BB-resistance gene isolated from rice. Interestingly, Xa1 gene expression was induced on inoculation with a bacterial pathogen and wound, unlike other isolated resistance genes in plants, which show constitutive expression. The induced expression may be involved in enhancement of resistance against the pathogen.

Can positive affect items be used to assess depressive disorders in the Japanese population?

BACKGROUND: The purpose of the present study was to examine the measurement properties of positive affect items among the Japanese population. METHODS: Responses to the Japanese version of the Center for Epidemiologic Studies Depression Scale and four additional negatively revised items of the original positive affect items were compared for 85 Japanese psychiatric out-patients with dysphoric-mood-related symptoms and 255 demographically matched controls. RESULTS: Responses to positive affect items were generally comparable between the two groups, whereas responses to negative symptom items were markedly different (P < 0.002 for all comparisons). The group difference was most marked for symptom persistence. Responses to the four negatively revised items of positive affect revealed a similar picture to that of the negative symptom items. The internal consistency of the scale significantly improved when the original positive affect items were replaced by the negatively revised items (P < 0.001 for both). CONCLUSIONS: Positive affect items with positive wording cannot be used to assess depressive disorders in the Japanese population adequately, but this can be done with the corresponding negatively revised items.

Antimicrobial activity of a compound isolated from an oil-macerated garlic extract.

A compound showing antimicrobial activity was isolated from an oil-macerated garlic extract by silica gel column chromatography and preparative TLC. On basis of the results of NMR and MS analyses, it was identified as Z-4,5,9-trithiadeca-1,6-diene-9-oxide (Z-10-devinylajoene; Z-10-DA). Z-10-DA exhibited a broad spectrum of antimicrobial activity against such microorganisms as gram-positive and gram-negative bacteria and yeasts. The antimicrobial activity of Z-10-DA was comparable to that of Z-ajoene, but was superior to that of E-ajoene. Z-10-DA and Z-ajoene are different in respect of substitution of the allyl group by the methyl group flanking a sulfinyl group. This result suggests that substitution by the methyl group would also be effective for the inhibition of microbial growth.

A convenient and efficient synthesis of sialyl Lewis X.

A convenient synthesis of the sialyl Lewis X (sLe(x)) tetrasaccharide, NeuAc alpha 2-3Gal beta 1-4(Fuc alpha 1-3)GlcNAc (8), as a carbohydrate ligand for selectins is described. The key step is the reaction between NeuAc alpha 2-3GalSMe (5) as a glycosyl donor and the suitably protected Fuc alpha 1-3GlcNAc derivative (4) as the glycosyl acceptor by using dimethyl(methylthio)sulfonium triflate (DMTST) as the promoter.
