Conductive staining of biological specimens for scanning electron microscopy with special reference to ligand-mediated osmium impregnation.

Biological specimens for scanning electron microscopy are made electron-conductive and non-charging by intense osmication with tannic acid, thiocarbohydrazide, hydrazine hydrate or other bifunctional ligands (or mordants). Such osmication also minimizes tissue shrinkage during dehydration and drying, reduces tissue damage during metal coating and scanning observations, and circumvents the use of a thick metal coating which hinders high resolution. Intense osmication when repeated eliminates the need for a metal coating and produces a well-contrasted scanning image with good resolution. Moreover, such osmication reduces tissue damage during ion implanting, enhances the depth of imaging, allows x-ray microanalysis without coated metals, and permits resin-embedded observations, as well as continuous or repeated microdissections in the scanning electron microscope without charging. Intense osmication forms a surface coat or thickens the cell membrane. This surface coat consists of electron dense deposits of 4-20 nm thickness. The hydrazine hydrate-osmium method can be used in the vapor phase. The vapor phase application has the advantage of staining even plastic samples for non-metal-coated scanning electron microscopy.

Analytical studies on neurocirculatory asthenia and related diseases using psychological and cardiovascular parameters.

In order to analyze the nature of the cardiovascular complaints in neurocirculatory asthenia (NCA) and related diseases, 83 cases were studied on the basis of the results of psychological tests (Cornell Medical Index and Yatabe-Guilford test) and cardiovascular and respiratory parameters. Almost all of the patients with normal psychological test results had abnormalities in one or more of the following: blood pressure, pulse rate, cardiac index, TII in the standing position, and cardiothoracic ratio. The data suggest that the cardiovascular complaints of the normal psychological test group can be attributed to somatic factors, especially to a small size of the heart. On the other hand, the group with psychological abnormalities had hearts of normal sizes, a less marked lowering of TII in the standing position, but markedly abnormal respiratory patterns. This indicates that the respiratory abnormalities seen in this group are likely to be the symptoms of cardiac psychoneurosis.

A new microwave device for rapid thermal fixation of the murine brain.

A microwave applicator was developed to provide rapid thermal fixation of enzymes in the murine brain. Although the power output of the device is only 1.3 kW, whole-brain levels of acetyicholine after microwave fixation are comparable to those observed at a power output of 5 kW, and are markedly higher than those obtained by a conventional method (freezing) of fixation.

Cellular immunity in rheumatic heart disease.

Cellular immunity to non-specific antigens (PPD, Candida albicans, and streptococcal antigens) and mitogen (PHA) was investigated in patients with valvular heart disease of rheumatic origin. The result disclosed that lowered response in cellular immunity exists in these patients. The possible significance of this finding in occurrence of rheumatic activity was discussed.
