The lipoxygenase product hydroperoxyeicosatetraenoic acid (HPETE) has immunosuppressive properties in vitro and in vivo. It was observed that 15-HPETE inhibit the sheep red blood cell rosette formation and the concanavalin A-induced blast transformation of human lymphocytes. This inhibition was HLA-linked. HLA-B12 subjects were less sensitive than non-B12 subjects. It is likely that HPETE acids are macrophage mediators which inhibit some lymphocyte functions.

In vitro generation of helper T cells and suppressor T cells that regulate the cytolytic T lymphocyte response to trinitrophenyl-modified syngeneic cells.
Helper T cells and suppressor T cells have been generated in vitro that regulate the cytolytic T lymphocyte (CTL) response to trinitrophenyl (TNP)-modified syngeneic cells. B6D2F1 helper cells generated to TNP-modified parental (P1) cells augment the CTL response to those P1-TNP-modified antigens but not to P2-TNP-modified antigens. The generation of these helper T cells requires the presence of splenic adherent cells and these helper T cells are radioresistant. A soluble factor can be obtained from the helper T cell cultures that can also augment the CTL response. The suppressor T cells generated in culture do not demonstrate the specificity observed with the helper T cells; however, they are antigen-dependent in their induction. Whether helper or suppressor activity is obtained depends upon the length of time cells are cultured in vitro.
[Inhibition of the lymphocyte response by metabolites released by the lipoxygenase of mouse macrophages].
Arachidonic acid can be transformed into a series of metabolites by the lipoxygenase enzyme activity of Mouse peritoneal macrophages. The resulting metabolites inhibit tritiated thymidine uptake by Mouse splenocytes stimulated by ConA or PHA. They suppress the development of killer cells. When mice are injected with 15-hydroperoxide, their splenocytes show a decreased H3-thymidine uptake after lectin stimulation.

Metabolism of polyunsaturated fatty acids by mouse peritoneal macrophages: the lipoxygenase metabolic pathway.
When resident macrophages from mice are incubated with exogenous polyunsaturated fatty acids, they produce lipoxygenic metabolites. To delineate this metabolic chart we used high pressure liquid chromatography and gas chromatography prior to mass spectrometry-computer system. The lipoxygenic activity of these cells leads to many compounds. Among them we describe the monohydroxylated metabolites and vicinal hydroxyepoxyenes. In the mechanism of formation of the latter unstable cyclic precursors might occur as intermediates between hydroperoxides and them. Dihydroxy compounds could arise from hydrolysis of unstable epoxide precursor which could be the second substrate of the glutathione transferase system and could lead to thioaminolipids.

Cryoglobulinemic neuropathy: a pathological study.
A 53-year-old woman developed symmetrical polyneuropathy of the lower limbs a few months after she was found to have myeloma with cryoglobulinemia. In musculocutaneous nerve biopsy material, electron microscopy showed both axonal degeneration and demyelination. The most striking finding was the presence, in the endoneurial space, of numerous masses of closely packed tubular structures. These masses also were found in the walls of all the vasa nervorum and within the lumen of some vessels. The morphological features and dimensions of the deposits within nerve were identical to those of cryoprecipitates extracted from serum and examined with the electron microscope. An example of myeloma neuropathy with cryoglobulin deposits within the endoneurial space has not been reported previously.

In vitro inhibition of human lymphocyte transformation by natural or synthetic prostaglandins.
We have examined the role of 45 natural or synthetic prostaglandins (PGs) in the in vitro responsiveness of human lymphocytes toward two different varieties of mitogenic stimuli: the mixed lymphocyte culture (MLC) and the tuberculinic test (LTT). Addition of PGs to culture of lymphocytes decreases the uptake of 3H thymidin.
HLA phenotypes and cadmium blood level.
The distribution of HLA phenotypes was studied and the amount of cadmium blood level was determined in a group of 100 healthy subjects. The HLA-A3 phenotype seems associated with high cadmium levels in the blood. The lymphocytes transformation test with PHA and different concentrations of cadmium indicates that A3 cells are more sensitive than others to inhibition by cadmium.
