Studies on peptides. CXI. Synthesis of chicken gastrin-releasing peptide.

The heptacosapeptide amide corresponding to the entire amino acid sequence of chicken gastrin-releasing peptide (cGRP) was synthesized similarly to the synthesis of porcine GRP by assembling six peptide fragments followed by deprotection with 1 M trifluoromethanesulfonic acid-thioanisole in TFA. A new carboxyl-activating reagent, thiazolidine-2-thione, was preferentially adopted for preparation of necessary fragments. The synthetic cGRP, purified by ion-exchange chromatography, followed by partition chromatography, was active as the synthetic porcine GRP, when plasma immunoreactive gastrin level was examined in rats. No obvious difference was observed when synthetic and natural cGRP preparations were compared by HPLC, immunochemical property and biological activity in dogs.

Rigid spine syndrome: clinical and histological problems.

A case is described with the characteristic clinical features of rigid spine syndrome: childhood onset in a male, very slowly progressive weakness and muscle atrophy, limitation of neck and trunk flexion, joint contractures, normal intelligence, no clear family history, increased serum CPK levels and myopathic patterns in EMG and muscle biopsy specimens. Myopathic patterns with various histological changes in muscle biopsy specimens were seen in all eight reported cases of the rigid spine syndrome, but there are no characteristic histological findings in the rigid spine syndrome.

Isolation and characterization of a proteodermatan sulfate from calf skin.

A proteodermatan sulfate was extracted from calf skin with 3 M MgCl2 at 4 degrees C in the presence of protease inhibitors and purified by DEAE-cellulose chromatography followed by CsCl density gradient centrifugation. The molecular weight of the proteoglycan was estimated to be approximately 115,000, based on the results of sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The proteoglycan contains neutral sugars such as galactose, xylose, mannose, fucose, and glucose, but its amino acid composition is similar to that of proteodermatan sulfates from bovine heart valve, tendon, sclera, and pig and rat skin. The core protein is a glycoprotein with a molecular weight of 56,000 and seems to be monodisperse. However, the dermatan sulfate component, which is the only glycosaminoglycan constituent composed of 95% 4-sulfated disaccharide unit and 5% 6-sulfated disaccharide unit, seems to be polydisperse with a number average molecular weight of 17,000, as judged from both gel chromatography and chemical analysis. Analysis by beta-elimination and reduction of the dermatan sulfate-peptide(s) indicated that the glycan components are linked to the core protein through an O-glycosidic linkage between xylose and serine residues. From these results, we propose a model for the structure of calf skin proteodermatan sulfate that is composed of 3--4 chains of dermatan sulfate components which are covalently linked to a core (glyco-) protein via O-glycosidic linkages.

Studies on peptides. CII. Synthesis of a heptacosapeptide amide corresponding to the entire amino acid sequence of gastrin-releasing peptide.

The heptacosapeptide amide corresponding to the entire amino acid sequence of gastrin-releasing peptide (GRP) was synthesized by assembling seven peptide fragments followed by deprotection with a new reagent system, IM trifluoromethanesulfonic acid-thioanisole in TFA. The deprotected peptide was purified by ion-exchange chromatography on CM-cellulose followed by partition chromatography on Sephadex G-25. The latter was found to remove effectively the Met (O) derivative of GRP. The highly purified synthetic GRP was active as synthetic bombesin on the molar basis. A new carboxyl-activating reagent, thiazoline-2-thione, was employed for preparation of the necessary fragments.

[Studies on establishment and maintenance of persistent infection with measles virus in NC-37 cell. II. Effect of DNA-synthesis-suppressing factor (DSF) on the production of interferon in NC-37 cell infected with measles virus (author's transl)].

The attenuated strain of measles viruses, Schwarz, CAM-70, or subacute sclerosing panencephalitis (SSPE) virus of measles viruses, Halle, Mantooth was good interferon inducer, but the virulent strain, Edmonston, Toyoshima induced few amount of interferon or not. Furthermore, an inverse relationship seemed to exist between the ability to induce interferon and DNA synthesis suppressing factor (DSF). The attenuated strain or SSPE virus which had not a capacity to produce DSF could induce interferon and establish persistent infection in human lymphoid cell NC-37, while virulent strain did not induce interferon but DSF. Maximum yield of progeny virus was precede the detection of interferon in culture fluids. A persistently infected cells also produced continuously interferon and progeny virus. It is demonstrated that DSF suppressed interferon production by infection of measles virus and that DSF had not an activity like interferon.

[Studies on establishment and maintenance of persistent infection with measles virus in NC-37 cell. III. Role of interferon in establishment and maintenance of persistent infection (author's transl)].

Human lymphoblastoid cell, NC-37, persistently infected with measles virus could always produce interferon in culture medium. Persistently infected cells were sensitive to interferon and acquired antiviral state, but normally yielded infectious progeny virus in culture fluid. Sensitivity of measles virus to interferon was studied because measles virus continued to replicate in these persistently infected cell. Attenuated and SSPE viruses which established persistent infection were relatively resistant to antiviral effect of interferon while wild strain had a sensitivity. Interferon produced endogenously in these persistently infected cells did not play an important role in maintenance of persistent infection caused by the suppression of virus replication. As it was found that persistently infected cell treated with interferon as high as 10(4) units/ml was significantly decreased virus yield in parallel with hemadsorption, it is suggested that endogenous measles virus which has been replicate in persistently infected cell might be also sensitive to high titer of interferon. NC-37 cell pretreated with interferon (10(3) units/ml) was infected with wild strain of measles virus. The treated cell led to decrease approximately 30% of virus yield and hemadsorption at the time when maximum yield was obtained in control cell, and that cell death reduced one week, but persistent infection could not be established.

[Studies on establishment and maintenance of persistent infection with measles virus in NC-37 cell. 1. Production of DNA-synthesis-suppressing factor (DSF) in NC-37 cell infected with measles virus].

Wild strains of measles virus, such as Edmonston, Toyoshima and freshly isolated KM strain which had not a capacity to establish a persistent infection in NC-37 cell inhibited 80-90 percent of host cellular DNA synthesis, while attenuated or subacute sclerosing panencephalitis (SSPE) viruses, such as Schwarz, Halle and Mantooth which had a capacity to establish persistent infection inhibited about 50 percent of host cellular DNA synthesis 6 or 7 days after infection of virus. However, inhibition of DNA synthesis in cells infected with attenuated or SSPE virus recovered gradually to the same level of control cell within two weeks after infection of virus, and persistently infected cells were not suppressed cellular DNA synthesis in synchronized cell culture. NC-37 cells infected with wild strain produced host cellular DNA-synthesis-suppressing factor (DSF) into culture fluid, but it could detect no such factor in culture fluid of cells infected with attenuated, SSPE virus, or carried virus and persistently infected cell. Furthermore, persistently infected cell treated wtih DSF caused the suppression of DNA synthesis, and so these cells died of metabolic inhibition within one week after inoculation of DSF. NC-37 cell infected with Mantooth strain was inhibited the establishment of persistent infection by the treatment of cell with DSF.
