A series of 3-substituted 5-methoxycarbonyl-5-methoxycarbonylmethyl-1,4,2-dioxazoles was prepared by addition of hydroxamic acids to acetylene esters. Eleven of these previously unknown compounds were submitted to general pharmacological screening, and several displayed modest CNS depressant activity.
Tissue distribution of 99mTc-1-thioglucose was studied in inbred albino Fisher rats. High levels of radioactivity were observed in the kidney with concurrent low pancreatic levels and pancreas/liver ratios of 0.4 following intravenous administration of the agent. High pancreatic concentration were observed with pancreas/liver ratios of 4.0 after intraperitoneal administrations. Future studies on pancreatic radiopharmaceuticals should avoid production of artificially high pancreatic values by intraperitoneal injection.

Six non-nitrosated and three nitrosated analogs of the antitumor and diabetogenic agent, streptozotocin, have been found to produce hyperglycemia in fasted rats. The hyperglycemia produced by the non-nitrostaed analogs was of early onset and did not result in the production of permanent diabetes. The data emphasizes the important role of the nitroso group of streptozotocin in the production of the hypoglycemic and permanent diabetic phases while the sugar moiety may have a direct influence on production of the early hyperglycemia phase.
Four new tolbutamide analogs were prepared and evaluated as hypoglycemics. Hypoglycemic action was observed in two members of the class, and 99m-Tc-chelates were tested as potential radiopharmaceutical imaging agents.
Seven new 3-amino-2-imino-1,3-benzothiazin-4-ones, prepared from the condensation of cyanogen and 2-mercaptobenzhydrazides, were evaluated in a neuropharmacological mouse profile. CNS depression was observed in several members of the class.
