A capillary diffusion method for determining hemolytic complement activity in undiluted serum .

A method is presented for measuring the hemolytic activity of undiluted guinea pig serum. Undiluted serum is placed in contact with a column of sensitized sheep red blood cells in an agar matrix in a capillary tube. After incubation, the length of the zone of lysis is measured. Comparison with a standard curve prepared from dilutions of a pool of normal serum allows relative quantitation. Because the serum is not diluted, this method allows detection of reversible inhibitors of the complement system.

New method for rapid characterization of molecular shapes: applications in drug design.

We present a method for the rapid quantitative shape match between two molecules or a molecule and a template, using atom triplets as descriptors. This technique can be used either as a rapid screen preceding the computationally expensive shape-based docking method developed by Kuntz and co-workers or as a stand-alone method to rank compounds in a large database for their fit to a shape template. The merits and limitations of this method are discussed in detail with examples.

Effect of a pretest listening exposure on the ascending-descending gap at most comfortable loudness levels.

The effect of approach mode (ascending versus descending, A-D) on comfortable loudness was determined for a 1000 Hz pure tone using Békésy audiometry. Two groups (experimental and control) of 10 normal-hearing young adult subjects measured their most comfortable loudness (MCL) levels pre- and post-treatment. The experimental group received a treatment of a listening exposure to a 70 dB intensity range before the test. The results demonstrated a statistically significant change of the A-D gap in the experimental group while the A-D gap for the control group remained essentially unchanged. It is proposed that MCL is underestimated in the ascending approach and overestimated on the descending testing, which results in the A-D gap. In addition, the results demonstrate the importance of the procedures used in research and clinic of the MCL.

Studies on type II collagen induced polyarthritis in rats. Effect of complement depletion.

The effect of cobra venom factor on the developing and the established lesion of collagen-induced rat polyarthritis has been examined. In the developing lesion, decomplementation by cobra venom factor results in a delay in the onset of inflammation and a decrease in the radiological parameters of joint destruction. Under the conditions of the decomplementation, antibody titers to collagen are not decreased. In the established lesion, treatment with cobra venom factor has no effect, on either the inflammatory lesion or the various radiological parameters of joint destruction.
