High chromosome numbers of seminomata and malignant teratomata of the testis: a review of data on 103 tumours.

Cytogenetic data on 103 seminomata and malignant teratomata of the testis from the literature and (partly in the form of DNA measurements) from this laboratory show that modal chromosome numbers are generally 50 or more. The only exceptions were 2 seminomata in which diploid and pseudodiploid karyotypes respectively were found, but the dividing cells may not have been tumour cells. Malignant tumours of the testis thus differ from those of all other sites (including the ovary) that have been studied sufficiently, where hypodiploid tumours are common. The reason for this difference is unknown. Mechanisms whereby high chromosome numbers, particularly the near-triploid numbers commonly found in testicular tumours, may be achieved are discussed briefly.

Histological cell type and DNA value in the prognosis of squamous cell cancer of uterine cervix.

Based on the evaluation of 362 cases of squamous cell carcinoma of the uterine cervix, the distribution of the tumours in relation to their modified Broders' grade, histological cell type as proposed by Wentz and Reagan, and the clinical stage of disease was evaluated. The morphological characteristics of the 3 cell types-large cell non-keratinizing, keratinizing, and small cell cancers-were described. The 5 year survival in relation to Broders' grade, cell type, extent and DNA values of the malignant cells were evaluated and compared. Broders' grading system was not useful in predicting the biological behaviour of cervical squamous cancer. The histological cell type and extent of the tumour were important factors in prognosis. The 5 year survival for large cell cancer was 51·8%, keratinizing cancer 34·7% and small cell cancer 10·0%. The 5 year survival was 63·3% for stage I neoplasms, 52·9% for stage II neoplasms, 30·7% for stage III neoplasms and 15·0% for stage IV neoplasms. When the DNA values of neoplastic cells were considered in relation to cell type and extent of disease, the biological behaviour of cervical squamous cell cancers was determined more accurately. The 5 year survival of women with cervical cancer in which the DNA values of the neoplastic cells exceeded 155 was more favourable than those with DNA values of less than 155. This difference in 5 year survival was evident for comparable cell type and clinical stage of disease.

Y bodies and similar fluorescent chromocentres in human tumours including teratomata.

The presence of Y bodies and similar fluorescent chromocentres in the interphase cells of 73 benign and malignant neoplastic lesions of males, and 69 of females, has been assessed in preparations stained with quinacrine dihydrochloride. In the male, Y bodies were usually present, although none was seen in 16 of the 65 malignant tumours. Sometimes the Y body was present in duplicate, for example, in some regions of a benign polyp of the colon and generally in 10 of the malignant tumours. The series includes 5 seminomata and 12 malignant testicular teratomata, all of which were positive for Y bodies. The tumours of females failed to show similar bodies, with 5 exceptions: one of 13 carcinomata of the ovary showed a body resembling the Y body in about half the cells (however, a similar body was seen in lymphocytes from this case) while a further carcinoma of probable ovarian origin, and 3 of 13 ovarian dermoids, showed a similar body though in less than 20% of the cells.Although quinacrine fluorescence studies on interphase tumour cells may be of value in suggesting the presence or absence of Y chromosomes, it is desirable that these studies be supplemented by the investigation of the fluorescence pattern of the metaphase chromosomes.

Variable sex chromatin pattern in an early carcinoma of the bladder.

Sex chromatin studies on squash preparations of a well differentiated transitional cell carcinoma of the bladder without evidence of invasion from a female aged 63 revealed a single body in some regions, but two to four bodies in others. All regions were near diploid according to DNA estimations. Previous observations on a variety of invasive tumours of females showed that the presence of more than one sex chromatin body is generally associated with a high chromosome number. The pattern of two or more bodies in near-diploid cells seen in this non-invasive tumour may therefore characterize an intermediate stage of clonal evolution, eventually resulting in malignancy, when the cell line has not yet achieved the ability to invade or metastasize.

Modal deoxyribonucleic acid value and survival in carcinoma of the breast.

On the basis of their modal deoxyribonucleic acid values, 67 carcinomas of the breast fell into two discrete groups. The lower (near-diploid) group showed a significantly better eight-year survival rate than the higher (triploid-tetraploid) group. The four-year survival rates of 40 further cases classified in the same way showed similar trends.
