Coronary endothelium-protective effects of defibrotide in ischaemia and reperfusion.

Defibrotide is known to enhance prostacyclin (PGI2) release from the vascular endothelium. We investigated the vasoactive effects of defibrotide in isolated rat hearts perfused at constant flow subjected to ischaemia and reperfusion. Defibrotide at 10 or 100 micrograms/ml did not exert any direct vasoactive effect on normal rats hearts. However, ischaemia and reperfusion resulted in an impaired vasodilation to acetylcholine, an endothelium-dependent vasodilator. In contrast, the vasodilator response to the endothelium-independent dilator, nitroglycerin, was unaffected. Defibrotide, at 10 or 100 micrograms/ml, markedly restored the vasodilation to acetylcholine 10 nmol/l to 1 mumol/l (P less than 0.01) without influencing the vasodilator response to nitroglycerin (2 to 200 micrograms/l). Haemoglobin (150 nmol/l) inhibited the dilation to acetylcholine in response to defibrotide. However, no evidence of PGI2 release was observed with acetylcholine-induced vasodilation in the presence or absence of defibrotide. Additionally, 10-100 micrograms/ml of defibrotide did not significantly decrease superoxide radicals generated by a xanthine-xanthine oxidase synthetic system under conditions in which superoxide dismutase was effective. Thus, defibrotide appears to exert an endothelium-protective effect preserving endothelium-derived relaxing factor (EDRF) without directly scavenging free signals.

Slit ventricle syndrome after cyst-peritoneal shunting for the treatment of intracranial arachnoid cyst.

Serious complications following installation of a cyst-peritoneal shunt in an 8-year-old boy with asymptomatic arachnoid cyst in the middle cranial fossa are reported. Highly elevated intracranial pressure without association of ventriculomegaly seen in this patient indicates slit ventricle syndrome caused by malfunction of the cyst-peritoneal shunt. This phenomenon is worth recognizing as a possible consequence of cyst-peritoneal shunting for the treatment of intracranial arachnoid cyst.

Intraoperative subdural hematoma in a patient with arachnoid cyst in the middle cranial fossa.

A case of arachnoid cyst in the middle cranial fossa complicated by intraoperative subdural hemorrhage is presented. Accumulation of cerebrospinal fluid (CSF) outside the outer wall of the arachnoid cyst was found to induce the rupture of well-developed, fragile leptomeningeal vessels at the base of the middle cranial fossa. This phenomenon is noteworthy as a pitfall in surgery for intracranial arachnoid cyst and suggests the bleeding source of subdural hematoma occasionally associated with this lesion.

A survey of the natural occurrence of Fusarium mycotoxins, deoxynivalenol, nivalenol and zearalenone, in cereals harvested in the Netherlands.

A survey for the natural occurrence of Fusarium mycotoxins, deoxynivalenol (DON), nivalenol (NIV) and zearalenone (ZEN), in Dutch cereals (totaling 29 samples) harvested in 1984/1985, showed that 90%, 79% and 62% of samples were contaminated with DON, NIV and ZEN, respectively. Average contents (ng/g) in the total of positive samples were 221 (DON), 123 (NIV) and 61 (ZEN). Among the cereals examined, the highest concentrations (ng/g) was 3198 (DON), 1875 (NIV) and 677 (ZEN) in a yellow corn sample for animal feed. The results of this survey show that Dutch cereals were relatively significantly contaminated with Fusarium mycotoxins.

Symptomatic subacute subdural haematoma following spontaneous acute subdural haematoma.

Two patients with non-traumatic acute subdural haematoma were observed, initially without surgical intervention. Eleven days after the onset, each patient developed hemiparesis and an increase in severity of headache. Serial computed tomography scans demonstrated that the initial hyperdense haematomas became hypodense with a definite increase in volume. The term "symptomatic subacute subdural haematoma" was proposed to properly define this pathological process, which necessitated removal of the haematoma in the subacute stage. The mechanism of increase in the volume of the haematoma is discussed.

Acute subdural haematoma with rapid resolution.

Two adult patients with rapid spontaneous resolution of an acute subdural haematoma are reported. Neurological recovery occurred at the same time, followed by a benign clinical course without surgical intervention. The author discusses the possible influence of this phenomenon on the treatment of acute subdural haematomas.

Epidural haematoma in the newborn infants: therapeutic consequences from the correlation between haematoma content and computed tomography features. A review.

Eight reported cases of neonatal epidural haematoma are reviewed. Special attention is directed at the correlation between the haematoma content and computed tomography (CT) features. Seven cases had a high density haematoma on CT, of which four showed liquid, one semi-liquid, one liquid and solid, and one solid haematoma. The low density haematoma in one case was liquid. These results suggest that evacuation of the epidural haematoma via percutaneous epidural tapping is warranted as the first line of treatment irrespective of high density on CT.

Direct positive inotropic and vasoconstrictor effects of endothelin.

Endothelin, a newly discovered vasoconstrictor peptide, when added to isolated cat papillary muscles, induced a direct positive inotropic effect that was slow in onset but of long duration. The magnitude of the developed force was concentration dependent. Endothelin exerted a marked concentration-dependent vasoconstriction in isolated cat carotid arteries and rabbit aortic rings. In both the carotid arteries and the aortic rings, endothelin induced a similar vasoconstrictor effect in the presence or absence of an intact endothelium. Addition of propyl gallate, a 5-lipoxygenase inhibitor, ibuprofen, a cyclooxygenase inhibitor, or SKF-525A, a cytochrome P450 inhibitor, at 2 microM did not significantly attenuate the ability of endothelin to vasoconstrict aortic rings in the presence or absence of an intact endothelium. These results demonstrate that the vasoconstrictor activity of endothelin operates independently of all three pathways of arachidonic acid metabolism (i.e., lipoxygenase, cyclooxygenase, or cytochrome P450 pathways) and is not dependent upon other endothelium-derived mediators (e.g., endothelium-derived relaxing factor, or eicosanoids) in these preparations. Moreover, endothelin exerts a direct positive inotropic effect in isolated cat ventricular myocardial tissue.

Direct cardiovascular actions of two metabolites of linoleic acid.

Two newly discovered oxidation products of linoleic acid (i.e., 9,10-epoxy-12-octadecenoate termed Leukotoxin A, and 12,13-epoxy-9-octadecenoate termed Leukotoxin B) are produced by neutrophils in a variety of species. These substances appear to combat bacterial infection although they also have detrimental effects on normal organ function. Administration of Leukotoxin A or B to isolated cat papillary muscles decreased developed force, an index of myocardial contractility, in a concentration-dependent manner. Leukotoxin B was more active in decreasing the developed force than Leukotoxin A at high concentrations. Leukotoxin A or B, when added to isolated perfused cat carotid arteries, produced a significant vasoconstriction which in vivo would result in an increased vascular resistance. Thus, leukotoxins exert significant direct effects on the cardiovascular system in cats. Leukotoxins A and B are both cardiodepressant and vasoactive independent of release of other blood borne mediators.

Avulsion of choroid plexus during revision of ventricular shunting: its high incidence and predictive value on computed tomography scan.

Over a 2-year period, 5 of 10 patients with revision of ventricular shunting were complicated by intraventricular and/or intracerebral hemorrhage due to avulsion of the choroid plexus. Retrospective analysis of computed tomography scans prior to the revision disclosed the presence of an intraventricular or periventricular mass adjoining the ventricular catheter in four of the five patients. Histologic findings of the soft tissue at the tip of the removed catheter were compatible with the choroid plexus accompanied by granulation tissue. The risk factors useful for detecting the possible occurrence of this complication are discussed.

Comparative investigation of vitamins and their analogues on terminal differentiation, from preadipocytes to adipocytes, of 3T3-L1 cells.

1. The effects of 20 kinds of vitamins or their analogues on the growth rate of preadipocytes and the terminal differentiation of preadipocytes to adipocytes was systematically compared in 3T3-L1 cells. 2. The addition of vitamin C markedly increased the growth rate of preadipocytes at over 50 microM. 3. The addition of vitamin K3 slowed down the growth rate at over 0.1 microM. 4. In water soluble vitamins and their analogues tested, the vitamin B6 group and vitamin C significantly stimulated the differentiation, and consequently increased the glycerophosphate dehydrogenase activity and triglyceride accumulation, to a concentration of over 10 microM. 5. Many fat soluble vitamins and their analogues (the vitamin A group, including beta-carotene, the vitamin D group, vitamin E and the vitamin K group) strongly inhibited the adipose conversion of 3T3-L1 cells at microM level.

Proliferation of 3T3-L1 preadipocytes in a completely defined serum-free medium.

We have developed a completely defined serum-free medium that supports the growth of Swiss 3T3-L1 fibroblasts to nearly the same extent as Dulbecco's modified Eagle's medium supplemented with 10% fetal calf serum. With ASF301 medium [former name, RITC 80-7; Yamane et al. Exp. Cell Res. 134, 470 (1981)], most of the 3T3-L1 cells survived for at least 10 days, but did not grow. ASF301 medium contains insulin and mouse-epidermal growth factor as growth factors, which are termed "progression factors". So we examined the effects of "competent factors" on the proliferation of 3T3-L1 preadipocytes. Growth in the medium supplemented with competent factors reached a confluent monolayer in 6-7 days. Furthermore, it was confirmed that 3T3-L1 cells grown in the serum-free medium retained the properties of differentiation into adipocytes. Our serum-free medium should be a useful tool for research on the growth and differentiation of 3T3-L1 preadipocytes.

Clinical and immunohistochemical studies of subependymal giant cell astrocytomas associated with tuberous sclerosis.

Two cases of TS associated with brain tumors had severe psychomotor retardation and early onset of long-term intractable convulsions, compared with cases without tumors. In one case, the tumor was partially cystic and progressed rapidly. Immunohistochemical studies of neuron specific enolase, glial fibrillary acidic protein and myelin basic protein revealed differences in positivity between cell types and between cases. These results suggested that the origin of the tumor cells could be variably differentiated cells.

Enzymic method for the spectrophotometric determination of aspartame in beverages.

A sensitive spectrophotometric method for the determination of aspartame in beverages is described. The method involves the enzymic conversion of aspartame into formaldehyde by the alpha-chymotrypsin-alcohol oxidase system, followed by the formation of a chromophore with 4-aminopent-3-en-2-one. The calibration graph was linear in the range 2.0-30.0 micrograms ml-1 of aspartame. Many common ingredients of beverages do not interfere with the proposed method. The method was applied to the determination of the aspartame content of various real samples, and the results obtained were compared with those given by high-performance liquid chromatography.

Up-regulation of thrombomodulin in human umbilical vein endothelial cells in vitro.

Previous studies have shown that thrombomodulin (TM) on endothelial cells is down-regulated by endotoxin, interleukin-1 beta (IL-1 beta), and tumor necrosis factor (TNF). This loss of anti-coagulant potential is thought to be related to the hypercoagulable state in sepsis, inflammation, and cancer. The current studies describe up-regulation of TM in human umbilical vein endothelial cells (HUVECs) by several compounds as judged by increased surface cofactor activity, surface TM antigen, and TM mRNA levels. Surface TM activity was increased by active phorbol esters (10(-8) M, 24-48 h), analogs of cAMP (1-10 mM, 4 h), and forskolin (10(-5) M, 24-48 h). Up-regulation of TM in HUVECs by 4 beta-phorbol 12-myristate 13-acetate (PMA) and dibutyryl cAMP (dBcAMP) was due to de novo synthesis of TM protein resulting from increased TM mRNA levels. The results suggest that protein kinase C and protein kinase A may be involved in cellular regulatory mechanisms for TM expression. In addition, PMA effects on surface TM activity are biphasic, with an initial reduction followed by a significant enhancement. Hence, we propose that compounds capable of increasing intracellular cAMP concentrations in HUVECs may be useful in preventing thrombosis by increasing the anti-thrombotic properties of endothelial cells.
