[Identification of leaf rust resistance genes in wheat (Triticum aestivum L.) cultivars using molecular markers].

A collection of 68 cultivars of common wheat has been screened for leaf rust resistance genes with the use of molecular markers. Markers of genes Lr1, Lr9, Lr10, Lr19, Lr20, Lr21, Lr24, and Lr26 have been used. It has been suggested that allele Xgwm295 be used as a marker for identifying the Lr34 gene. The genes originating from Triticum aestivum L., as well as the Lr26 gene contained in rye translocation 1RS, are the most frequent. Genes originating from wild wheats were rarer in the cultivars studied.

[Modulation of the cat gene expression in E. coli cells by simple (AC)20 repeats].

Probable relationship between modulation of gene expression by simple repeating sequences and competition capacity of the host was studied. (AC)20 repeat can both stimulate and inhibit the expression of chloramphenicol acetyl transferase gene expression in E. coli cells depending on this gene's position. Modulation of the gene expression by simple (AC)20 repeat and competitive capacity of bacteria containing this plasmid are closely related.

[ret/PtC1 and ret/PTC3r1 rearrangement in thyroid cancer cells, arising in residents of Belorus in the period after the accident at the Chernobyl nuclear power plant].

After the accident at the Chernobyl nuclear power plant, a considerable increase in the incidence of thyroid cancer among children in Belarus was observed. In the present study, the frequency of the c-ret protooncogene rearrangements in samples of thyroid carcinomas resected and diagnosed in 1998 from individuals in Belarus was investigated. The ret/PTC1 oncogene was detected in 19% of the samples, and the ret/PTC3r1 oncogene, in 14%. The number of ret/PTC1 rearrangements observed in tumor cells from the patients whose age at the time of the accident was from 1 to 10 years, was greater compared to those whose age at the time was from 10 to 20 years, irrespective of the year of surgery (1996 or 1998). The majority of the patients with ret/PTC3r1 rearrangements lived in Gomel oblast, which was contaminated by the Chernobyl meltdown.

[Effect of simple repeats (AC)n and (AT)n on chloramphenicol acetyltransferase gene expression in Escherichia coli].

According to some published reports, simple repeats can modulate gene transcription. The ability of (AC)n and (AT)n sequences to modulate gene expression in E. coli was investigated. These repeats were inserted in the leader sequence, in its 3' uncoding region, and in both these sites. When simple sequences were inserted in the leader sequence, gene expression was the highest for constructions containing 40 bp insertion. An increase in the insertion length led to a decrease in the gene expression. The (AC)20 sequence insertions in a 3' uncoding site of cat gene did not notably change its expression. Gene expression was much more modified by inserting the simple sequences on both sites of cat gene. Cat gene expression depended on the ratio of simple sequence lengths in its 5' and 3' uncoding sites. It was the highest when (AC)n sequence in the gene leader sequence or its 3' uncoding region was 40 bp. Gene expression modulation by simple sequence is assumed to be associated with the DNA lability increase.

[Genetic transformation of plants, recombination processes, and gene expression regulation in transgenic plants].

Recent studies in molecular biology and gene engineering were conducive to accumulation of numerous facts which help better understand the processes of plant genome functioning and follow up the effects of recombination processes in purposefully modified genome of a transformed plant on its expression. This review analyzes the major approaches to studies and results attained in plant gene engineering and plant genome imprinting.

[Deletional and insertional analysis of the antitumorigenicity of the plasmid R388 in Agrobacterium tumefaciens].

The study of the plasmid R388 deletional derivatives has shown the antitumorigenicity determinant of the plasmid to be contained by the tra-operon. Transposon Tn5 has been used for selection of a number of insertional mutants having lost the antitumorigenic properties. The difference in the locations of Tn5 inserts was found, all of the latter being localized within the tra-operon. The data obtained suggest the locus of the plasmid R388 responsible for its antitumorigenic properties to be located in the region of tra-genes distally to the rep-region.

[CD44 gene expression in cancerous thyroid cells].

The peculiarities of alternative CD44 mRNA splicing in thyroid cancer tissue of children from radiocontaminated areas was investigated. CD44 gene expression in thyroid cancer tissues of children exposed to radiation resembled that in spontaneously emerged cancers. It was concluded that CD44 gene expression is not the primary target of radioactive irradiation. Probably, the CD44 mRNA splicing deregulation is the consequence of cancer.

[The molecular genetic mapping of cereal crops].

The application of modern methods of genetic mapping using RFLP and PCR technologies allowed to advance essentially in construction of rye genome genetic maps and mapping of some morphological and breeding-valuable genes. Genetic mapping of cereal genomes, such as rye, wheat, maize and rice using common set of DNA-probes permitted to reveal considerable evolutionary conservation in gene organization and localization. This allows to use more effectively method of comparative mapping for fast localization and tagging of genes in genomes of less investigated species.

[Comparative molecular-genetic mapping of genomes of rye (Secale cereale L.) and other cereals].

The genetic map of rye consisting of 149 RFLP, 20 isozyme and 12 microsatellite markers was developed. Using the collection of cross-hybridizing probes, the presence of multiple translocations in rye genome with respect to wheat and barley genomes was shown. However, within large regions of genome a strict collinearity of marker order was observed that allow us to use the method of comparative mapping for an introduction of new genes. In the developed genetic map 18 morphological and breeding-valuable genes mapped in different rye populations were integrated. The comparative analysis of homeological loci in genomes of Triticeae species as well as in genomes of rice and maize was carried out. The genes controlling a number of morphological traits, plant height, photoperiodic response and winter/spring growth habit were shown to be conserve among cereals and to form clear homoeologous rows.

[Chromosome aberrations in different tissues of the murine rodents and amphibians from radionuclide-contaminated regions of Belarus].

Cytogenetic investigations of amphibian and rodent populations from the contaminated by radionuclides regions of Belarus was conducted. A significant increase in the levels of cytogenetic anomalies in bone marrow cells and intestinal epithelium of amphibians and rodents, the levels of alveolar macrophages of rodents, and micronuclei in peripheral blood erythrocytes of amphibians was revealed. Presence of chromosome type aberrations indicates that radiation as a mutagenic factor was involved. No strict relationship between the levels of contamination of the area and levels of cytogenetic anomalies was found.

[A structural analysis of the BRS1 repeat from the genomic DNA of barley (Hordeum vulgare L.)].

The paper presents the results of analysis of the primary sequence of repetitive barley DNA called BRS1 (index number in Genbank BRS-1 U72261). The sequence encodes the open reading frame for 337 amino acids which has homology to retrotransposon-like element of Arabidopsis. There is a block consisting of 11 direct and 7 inverted repeats outside the open reading frame, the sequence ATACTAATGGCGCACC being the base of the repeats.
