[Analysis of proteins with altered expression in human esophageal squamous cell carcinomas].

BACKGROUND & OBJECTIVE: Esophageal squamous cell carcinoma is characterized by high incidence and high mortality. This study was designed to identify the proteins dysregulated in ECSS by. METHODS: Microdissection of routinely unstained frozen sections was used to procure cancer cells from seven esophageal squamous cell carcinomas, and esophageal epithelial cells from normal tissues adjacent to the tumors. By two-dimensional electrophoresis, protein profiles of normal and cancerous tissues were obtained. Selected proteins with altered expression in tumors were identified through MALDI-TOF-MS. RESULTS: Compared the protein profiles of tumors with those of normal epithelia, 11 proteins were found as dysregulated in tumors. Among them, HSP27 was observed up-expressed and three isoforms of ANNEXIN I were seen down-expressed in cancerous tissues. CONCLUSION: Two-dimensional electrophoresis coupled with mass spectrometry can contribute to rapidly screen dysregulated proteins in human maligancies. Altered expressions of HSP27 and ANNEXIN I were the frequent events in Chinese esophageal squamous cell carcinomas.

[Lung cancer risk associated with genetic polymorphism in myeloperoxidase (-463 G/A) in a Chinese population].

OBJECTIVE: To examine the relationship between susceptibility to lung cancer among Chinese and genetic polymorphism at nucleotide -463 (G/A) in myeloperoxidase (MPO), an enzyme found in lysosomes of phagocytes and involved in the formation of hydroxyl radicals and activation of various smoking-related carcinogens. METHODS: The association of this polymorphism with lung cancer in a hospital-based case-control study of 314 patients and 320 age- and sex-matched controls was tested. The MPO genotypes were determined by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP). RESULTS: The allele frequency for MPO-463A was found to be 11.0% for controls, compared with 15.0% for patients. Multivariate analysis showed an increased risk for overall lung cancer in subjects having GG genotype (OR, 1.7; 95% CI, 1.2 - 2.5), however, the elevated risk was limited to squamous cell carcinoma (OR, 2.4; 95% CI, 1.4 - 3.9; n = 177) but not to adenocarcinoma (OR, 1.3; 95% CI, 0.8 - 2.1; n = 137). In addition, the risky effect of the GG genotype on squamous cell carcinoma of the lung was evident only in the smokers and those who smoked >/= 26 pack-years (OR, 20.5; 95% CI, 5.6 - 75.3) as compared with GA and AA genotypes (OR, 6.2; 95% CI, 1.7 - 22.5) but not in the nonsmokers or those who smoked < 26 pack-years. CONCLUSION: Our data support the hypothesis that -463A polymorphism in the MPO gene may reduce the susceptibility to lung cancer in the Chinese.

[Research of the anti-tumor effect of human endostatin mediated by recombinant adeno-associated virus].

BACKGROUND & OBJECTIVE: It was reported that endostatin could inhibit tumor angiogenesis, then inhibit the growth and metastasis of tumor. However, there was few report about the treatment usage of endostatin. This study was designed to explore the effect of endostatin mediated with recombinant adeno-associated virus(rAAV) on tumor growth and metastasis. METHODS: To obtain the endostatin gene complete cDNA by RT-PCR, and clone it into the plasmid pSNAV and package the recombinant rAAV-SS-endostatin; to analyze its anti-tumor effect through animal experiments. RESULTS: In the B16F10 melanoma tumor model, C57BL/6 mice were inoculated with 10(11)TU rAAV-SS-endostatin i.m. injection, which the inhibition rate was 57.1%; In the lung metastatic model, the inhibition rate of metastases was 70.7%. CONCLUSIONS: rAAV-SS-endostatin can effectively inhibit tumor growth and metastasis.

[The method and implementation of measuring the peak of bioelectrical signals].

It is always very difficult to process the bioelectrical signals, sampling, amplifying and quantifying because of the different sources it comes from and the mechanism it depends on. This paper mainly introduces a method of measuring the peak value of bioelectrical signals, expatiates on the structure, testing result, analysis for error and measure taken for improvement of the circuit designed by this method. At last, the paper also points out that the method is wonderful, simple, easy to carry out, and can be used in many medical systems.

Expression of cell cycle/growth regulator genes in human hepatocellular carcinoma and adjacent normal liver tissues.

We investigated the gene expression of the cell cycle/growth regulators in hepatocellular carcinoma (HCC) through the usage of Atlas human cancer array membranes, semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and Northern blot. Hybridization of cDNA array membrane was performed with alpha-32P-labeled cDNA probes synthesized from RNA, isolated from HCC and adjacent non-cirrhotic normal liver. RT-PCR of 24 paired specimens and Northern blot of 4 paired specimens were used to confirm the expression patterns of the cell cycle/growth regulator genes identified by Atlas array hybridization. Among 79 genes related to cell cycle/growth regulators, transcription factor DP2 (TFDP-2) and E2F-3 were up-regulated, whereas dual-specificity mitogen-activated protein kinase kinase 1 (MAPKK1) and cell division protein kinase 3 (CDK3) were down-regulated in HCC. RT-PCR of TFDP-2 gave result consistent with Atlas human cancer cDNA array findings. Northern blot analysis of TFDP-2 and E2F-3 of 4 paired specimens all showed up-regulation in HCC compared to normal liver tissues. The results obtained from Atlas microarray provided for the first time a liver cancer-specific expression profile, which identified the gene expressions comprehensively and systematically. The findings may lead to better understanding of the mechanism of onset and progression of HCC. The rapid and high-throughout method of profiling gene expression by cDNA array provides an overview of the key factors that may be involved in HCC. Some genes are reported for the first time in HCC.

Gene expression profiles of hepatoma cell line BEL-7402.

BACKGROUND/AIMS: To investigate the gene expression of cancer related genes in hepatoma cell line BEL-7402 through the usage of Atlas Human Cancer Array with 588 well-characterized human genes related with cancer biology. METHODOLOGY: Hybridization of cDNA membrane was performed with 32P-labeled cDNA probes synthesized from RNA isolated from Human hepatoma cell line BEL-7402 and non-cirrhotic normal liver which was liver transplantation donor. Bioinformatics was used to analyze the result. The reverse transcription polymerase chain reaction of 24 pairs of specimens and northern blot of 4 pairs of specimens were used to confirm the expression pattern of some genes identified by Atlas arrays hybridization. RESULTS: The differential expression cell cycle/growth regulator in hepatoma cell line BEL-7402 showed a stronger tendency toward cell proliferation with up-regulation of E2F-3 and TFDP-2. The anti-apoptotic factors such as Akt-1 were up-regulated. Whereas the promotive genes of apoptosis were down-regulated, such as BAK and caspase-3. Besides this, some genes were up-regulated, such as Integrin beta 8, DNA-PK, CSPCP, cyclin C etc. A number of genes were down-regulated, which included LAR, ABL2, SKY, TDGF1 etc. In general, expression of the cancer progression genes were up-regulated, while expression of anti-cancer progression genes were down-regulated. The results of reverse transcription polymerase chain reaction of 24 pairs of specimens and Northern Blot of 4 pairs of specimens were consistent with the expression pattern of some genes identified by Atals arrays hybridization. CONCLUSIONS: Investigation of gene expression profile of hepatoma cell line BEL-7402 should help to disclose the molecular mechanism of the onset, progression and prognosis of hepatocellular carcinoma. The quick and high-throughout method of profiling gene expression by cDNA array provides us with an overview of key factors that may be involved in hepatocellular carcinoma, and may find the clue to the study of hepatocellular carcinoma carcinogenesis and molecular targets of diagnosis and therapy.
