[Detection of RbAp46 expression in bone marrow cells of leukemia patients by real-time quantitative RT-PCR].

OBJECTIVE: To investigate retinoblastoma (Rb) associated protein 46 (RbAp46) gene expression levels in bone marrow (BM) cells of leukemia patients. METHODS: Real-time quantitative reverse polymerase chain reaction (QRT-PCR) method was used for detecting RbAp46 expression levels in BM cells of 140 patients with acute leukemia (AL), 13 with chronic myelogenous leukemia in chronic phase (CML-CP), 7 with CML in blast crisis (CML-BC) and 32 with non-leukemic disorders. RESULTS: The M-Estimators of RbAp46 were higher in 98 newly diagnosed ALs and 5 relapsed ALs than in 28 ALs in complete remission (CR) and 32 non-leukemic controls (178.23 and 213.65 vs 85.89 and 88.08, respectively). No statistic difference was found between the CR group and control group, or between the newly diagnosed group and relapsed group. The M-Estimators of RbAp46 in patients with CML-CP was 58.27, similar to that in control, but much lower than that in CML-BC (173.24). Among 98 newly diagnosed ALs, the M-Estimators of RbAp46 in M(3) and M(4) were the lowest in all of the subtypes. Furthermore, the RbAp46 expression levels were not correlated with the expression of the fusion genes of bcr/abl, PML-RARalpha, and multidrug resistant gene (mdr1), but were positively correlated with Wilms' tumor gene (WT1) expression levels and negatively with AML1/ETO fusion gene expression. CONCLUSION: RbAp46 expression levels in ALs and CML-BC were strikingly higher than that in non-leukemias and CML-CP, and might participate in leukemogenesis.

Ca2+-independent hypoxic vasorelaxation in porcine coronary artery.

To demonstrate a Ca(2+)-independent component of hypoxic vasorelaxation and to investigate its mechanism, we utilized permeabilized porcine coronary arteries, in which [Ca(2+)] could be clamped. Arteries permeabilized with beta-escin developed maximum force in response to free Ca(2+) (6.6 microm), concomitant with a parallel increase in myosin regulatory light chain phosphorylation (MRLC-P(i)), from 0.183 +/- 0.023 to 0.353 +/- 0.019 MRLC-P(i) (total light chain)(-1). Hypoxia resulted in a significant decrease in both force (-31.9 +/- 4.1% prior developed force) and MRLC-P(i) (from 0.353 to 0.280 +/- 0.023), despite constant [Ca(2+)] buffered by EGTA (4 mm). Forces developed in response to Ca(2+) (6.6 microm), Ca(2+) (0.2 microm) + GTPgammaS (1 mM), or in the absence of Ca(2+) after treatment with ATPgammaS (1 mM), were of similar magnitude. Hypoxia also relaxed GTPgammaS contractures but importantly, arteries could not be relaxed after treatment with ATPgammaS. Permeabilization with Triton X-100 for 60 min also abolished hypoxic relaxation. The blocking of hypoxic relaxation after ATPgammaS suggests that this Ca(2+)-independent mechanism(s) may operate through alteration of MRLC-P(i) or of phosphorylation of the myosin binding subunit of myosin light chain phosphatase. Treatment with the Rho kinase inhibitor Y27632 (1 microm) relaxed GTPgammaS and Ca(2+) contractures; but the latter required a higher concentration (10 microm) for consistent relaxation. Relaxations to N(2) and/or Y27632 averaged 35% and were not additive or dependent on order. Our data suggest that the GTP-mediated, Rho kinase-coupled pathway merits further investigation as a potential site of this novel, Ca(2+)-independent O(2)-sensing mechanism. Importantly, these results unambiguously show that hypoxia-induced vasorelaxation can occur in permeabilized arteries where the Ca(2+) is clamped at a constant value.

Use of ultrafast-laser-driven microexplosional for fabricating three-dimensional void-based diamond-lattice photonic crystals in a solid polymer material.

Micro-sized void spheres are successfully generated in a solid polymer by use of a tightly focused femtosecond laser beam from a high-repetition-rate laser oscillator. Confocal reflection images show that the void spheres are longitudinal rotational symmetric ellipsoids with a ratio of long to short axes of approximately 1.5. Layers of void spheres are then stacked to create three-dimensional diamond-lattice photonic crystals. Three gaps are observed in the [100] direction with a suppression rate of the second gap of up to approximately 75% for a 32-layer structure. The observed first- and second-order gaps shift to longer and shorter wavelengths, respectively, as the angle of incidence increases.

Characterization of full length and truncated type I human methionine aminopeptidases expressed from Escherichia coli.

Methionine aminopeptidase (MetAP) carries out an essential posttranslational modification of nascent proteins by removing the initiator methionine and is recognized as a potential target for developing antibacterial, antifungal, and anticancer agents. We have established an Escherichia coli expression system for human type I MetAP (HsMetAP1) and characterized the full length HsMetAP1 and its N-terminal-truncated mutants HsMetAP1(Delta1-66) and HsMetAP1(Delta1-135) for hydrolysis of several thiopeptolide and peptide substrates and inhibition by a series of nonpeptidic inhibitors. Although the N-terminal extension with zinc finger motifs in HsMetAP1 is not required for enzyme activity, it has a significant impact on the interaction of the enzyme with substrates and inhibitors. In hydrolysis of the thiopeptolide substrates, a relaxation of stringent specificity for the terminal methionine was observed in the truncated mutants. However, this relaxation of specificity was not detectable in hydrolysis of tripeptide or tetrapeptide substrates. Several nonpeptidic inhibitors showed potent inhibition of the mutant HsMetAP1(Delta1-66) but exhibited only weak or no inhibition of the full length enzyme. With the recombinant HsMetAP1 available, we have identified several MetAP inhibitors with submicromolar inhibitory potencies against E. coli MetAP (EcMetAP1) that do not affect HsMetAP1. These results have demonstrated the possibility of developing MetAP inhibitors as antibacterial agents with minimum human toxicity. In addition, micromolar inhibitors of HsMetAP1 identified in this study can serve as tools for investigating the functions of HsMetAP1 in physiological and pathological processes.

Effective Mie scattering of a spherical fractal aggregate and its application in turbid media.

An effective Mie-scattering model is developed to deal with the scattering property of a spherical fractal aggregate consisting of scattering particles. In this model the scattered field of a scattering particle is given by the classical Mie-scattering theory. On the basis of the Monte Carlo simulation method, we determine the physical parameters of a scattering aggregate, the scattering efficiency Q, and the anisotropy value g, as well as their dependence on the size and the effective mean-free-path length of a scattering aggregate. Accordingly, photon migration through a microscope objective focused into a turbid medium including scattering aggregates is simulated to understand the effect of complex tissue on image quality.

Monte Carlo modeling of optical coherence tomography imaging through turbid media.

We combine a Monte Carlo technique with Mie theory to develop a method for simulating optical coherence tomography (OCT) imaging through homogeneous turbid media. In our model the propagating light is represented by a plane wavelet; its line propagation direction and path length in the turbid medium are determined by the Monte Carlo technique, and the process of scattering by small particles is computed according to Mie theory. Incorporated into the model is the numerical phase function obtained with Mie theory. The effect of phase function on simulation is also illustrated. Based on this improved Monte Carlo technique, OCT imaging is directly simulated and phase information is recorded. Speckles, resolution, and coherence gating are discussed. The simulation results show that axial and transversal resolutions decrease as probing depth increases. Adapting a light source with a low coherence improves the resolution. The selection of an appropriate coherence length involves a trade-off between intensity and resolution.

Mutations at the S1 sites of methionine aminopeptidases from Escherichia coli and Homo sapiens reveal the residues critical for substrate specificity.

Methionine aminopeptidase (MetAP) catalyzes the removal of methionine from newly synthesized polypeptides. MetAP carries out this cleavage with high precision, and Met is the only natural amino acid residue at the N terminus that is accepted, although type I and type II MetAPs use two different sets of residues to form the hydrophobic S1 site. Characteristics of the S1 binding pocket in type I MetAP were investigated by systematic mutation of each of the seven S1 residues in Escherichia coli MetAP type I (EcMetAP1) and human MetAP type I (HsMetAP1). We found that Tyr-65 and Trp-221 in EcMetAP1, as well as the corresponding residues Phe-197 and Trp-352 in HsMetAP1, were essential for the hydrolysis of a thiopeptolide substrate, Met-S-Gly-Phe. Mutation of Phe-191 to Ala in HsMetAP1 caused inactivity in contrast to the full activity of EcMetAP1(Y62A), which may suggest a subtle difference between the two type I enzymes. The more striking finding is that mutation of Cys-70 in EcMetAP1 or Cys-202 in HsMetAP1 opens up the S1 pocket. The thiopeptolides Leu-S-Gly-Phe and Phe-S-Gly-Phe, with previously unacceptable Leu or Phe as the N-terminal residue, became efficient substrates of EcMetAP1(C70A) and HsMetAP1(C202A). The relaxed specificity shown in these S1 site mutants for the N-terminal residues was confirmed by hydrolysis of peptide substrates and inhibition by reaction products. The structural features at the enzyme active site will be useful information for designing specific MetAP inhibitors for therapeutic applications.

Scanning total internal reflection fluorescence microscopy under one-photon and two-photon excitation: image formation.

We propose a new type of total internal reflection fluorescence microscopy (TIRFM) called scanning TIRFM (STIRFM) that uses a focused ring-beam illumination and a high-numerical-aperture objective (NA = 1.65). The evanescent field produced by the STIRFM is focused laterally, producing a small excitation volume that can induce a nonlinear effect such as two-photon absorption. Experimental images of CdSe quantum dot nanocrystals and Rhodamine 6G-doped microbeads show that good lateral and axial resolutions are achieved with the current setup. The theoretical simulation of the focal spot produced in STIRFM geometry shows that the focused evanescent field is split into two peaks because of the depolarization effect of a high numerical-aperture objective lens. However, the point-spread function analysis of both one-photon and two-photon excitation cases shows that the detection of the focus-splitting effect is dependent on the detection pinhole size. The effect of pinhole size on image formation is theoretically investigated and confirmed experimentally with the nanocrystal images.

[Incisor torque control with fixed appliance].

OBJECTIVE: To evaluate the properties of orthodontic rectangular arch wires in torsion. METHODS: To measure and analyze the torsional moments and angles of 0.46 mm x 0.63 mm rectangular orthodontic arch wires in single 0.022 inch bracket of the maxillary central incisor with a torsion measurement apparatus. RESULTS: (1) The torsional stiffness of stainless steel arch wires, Chinese nitinol arch wires, 7.0 mm vertical loop arch wires, 3.0 mm and 6.0 mm horizontal loop arch wires is 4.909, 1.325, 4.137, 3.448 and 3.024 Nmm per degree, respectively. (2) The arch wires will be twisted 19.143, 36.832, 19.851, 21.812 and 22.418 degrees; respectively to obtain optimal force. (3) Vertical and horizontal loop decreased the torsional stiffness of the rectangular stainless steel arch wire. CONCLUSION: The straight rectangular stainless steel arch wire is not well suitable for the torque control of the incisor. Loop design in arch wire is capable of decreasing torsional stiffness. The horizontal loop is more effective than vertical loop.

[Cephlometric analysis of the cranial base in class II(1) malocclusions with different vertical types in adolescences].

PURPOSE: To explore the morphology of the cranial base in Class II(1) malocclusions with different vertical types in adolescence. METHODS: The sample of 60 males and 60 females were divided into Class II(1) high-angle group, average-angle group, and low-angle group and normal occlusion group. Each group had 30 cases. Cephalograms were analyzed for all subjects. Data were obtained to have analysis of variance and LSD multiple range test. RESULTS: The cranial floor base angle (Ba-SE-FMS) of three groups with class II(1) malocclusion was increased. The anterior cranial floor angle was more open in the high angle group, and the flexure of the middle cranial floor was increased in the low angle and average angle groups. The 'effective vertical growth' of cranial floor was decreased while the mandibular plane angle was increased. CONCLUSION: The cranial floor base angle(Ba-SE-FMS) was suitable to describe the flexure of the cranial base, and indicated the mechanism of the Class II(1) malocclusion with different vertical types in adolescence.

Two-photon fluorescence scanning near-field microscopy based on a focused evanescent field under total internal reflection.

We present two-photon fluorescence near-field microscopy based on an evanescent field focus produced by a ring beam under total internal reflection. The evanescent field produced by this method is focused by a high-numerical-aperture objective, producing a tightly confined volume that can effectively induce two-photon excitation. The imaging system is characterized by the two-photon-excited images of the nanocrystals, which show that the focused evanescent field is split into two lobes because of the enhancement of the longitudinal polarization component at the focus. This feature is confirmed by the theoretical prediction. Unlike other two-photon near-field probes, this method does not have the heating effect and requires no control mechanism of the distance between a sample and the probe.

Two-photon fluorescence endoscopy with a micro-optic scanning head.

A major obstacle in the race to develop two-photon fluorescence endoscopy is the use of complicated bulk optics to transmit an ultrashort-pulsed laser beam and return the emitted fluorescence signal. We describe an all-fiber two-photon fluorescence microendoscope based on a single-mode optical fiber coupler, a microprism, and a gradient-index rod lens. It is found that the new endoscope exhibits an axial resolution of 3.2 microm and is capable of imaging transverse cross sections of internal cylindrical structures as small as approximately 3.0 mm in diameter. This device demonstrates the potential for developing a real-time diagnostic tool for biomedical research without the need for surgical biopsy and may find applications in photodynamic therapy, microsurgery, and early cancer detection.

Penetration depth of single-, two-, and three-photon fluorescence microscopic imaging through human cortex structures: Monte Carlo simulation.

Penetration depth is investigated in terms of the performance of transverse image resolution and signal level in human cortex under single-, two-, and three-photon fluorescence microscopy. Simulation results show that, in a double-layer human cortex structure consisting of gray and white matter media, the signal level is strongly affected by the existence of the white matter medium under three-photon excitation. Compared with three-photon excitation, two-photon excitation keeps a better signal level and sacrifices a slight degradation in image resolution. In a thick gray matter medium, a penetration depth of 1500 microm with a near-diffraction-limited resolution is obtainable under three-photon excitation. It is also demonstrated that the numerical aperture has a slight influence on image resolution and signal level under two- and three-photon excitation because of the nonlinear nature in the excitation process.

Effect of ultrashort pulsed illumination on foci caused by a Fresnel zone plate.

The focal distribution produced by a zone plate under ultrashort pulsed laser illumination is investigated under the Fresnel approximation. A comparison of the diffraction patterns in the focal region between pulsed and continuous-wave illumination shows that the focal shape produced by a zone plate can be significantly altered when an ultrashort pulse is shorter than 100 fs. In particular, the focal width in the axial and the transverse directions is increased by approximately 5 and 85%, respectively, from continuous-wave illumination to 10-fs pulsed illumination.

Monte Carlo simulation of multiphoton fluorescence microscopic imaging through inhomogeneous tissuelike turbid media.

Image resolution and signal level in fluorescence microscopy through inhomogeneous turbid media consisting of scatterers of multiple sizes under single- (1p), two- (2p), and three-photon (3p) excitation have been investigated based on a modified Monte Carlo model. The effects of the size distribution and the concentration distribution of scattering particles are explored. Simulation results reveal that the size and the concentration distribution both have an impact on image formation in media consisting of small particles and that 3p excitation has the most significant impact. In media with scatterers of a large size, both size and concentration distributions lead to a slight effect. Image formation in a mixed medium containing small and large scattering particles is more affected by the large particles.

Three-dimensional optical-transfer-function analysis of fiber-optical two-photon fluorescence microscopy.

The three-dimensional optical transfer function is derived for analyzing the imaging performance in fiber-optical two-photon fluorescence microscopy. Two types of fiber-optical geometry are considered: The first involves a single-mode fiber for delivering a laser beam for illumination, and the second is based on the use of a single-mode fiber coupler for both illumination delivery and signal collection. It is found that in the former case the transverse and axial cutoff spatial frequencies of the three-dimensional optical transfer function are the same as those in conventional two-photon fluorescence microscopy without the use of a pinhole. However, the transverse and axial cutoff spatial frequencies in the latter case are 1.7 times as large as those in the former case. Accordingly, this feature leads to an enhanced optical sectioning effect when a fiber coupler is used, which is consistent with our recent experimental observation.

Resolution improvement in two-photon fluorescence microscopy with a single-mode fiber.

The dependence of spectral broadening of an ultrashort-pulsed laser beam on the fiber length and the illumination power is experimentally characterized in order to deliver the laser for two-photon fluorescence microscopy. It is found that not only the spectral width but also the spectral blue shift increases with the fiber length and illumination power, owing to the nonlinear response in the fiber. For an illumination power of 400 mW in a 3-m-long single-mode fiber, the spectral blue shift is as large as 15 nm. Such a spectral blue shift enhances the contribution from the short-wavelength components within the pulsed beam and leads to an improvement in resolution under two-photon excitation, whereas the efficiency of two-photon excitation is slightly reduced because of the temporal broadening of the pulsed beam. The experimental measurement of the axial response to a two-photon fluorescence polymer block confirms this feature.
