Arrested apoptosis of nurse cells during Hydra oogenesis and embryogenesis.

During Hydra oogenesis, an aggregate of germ cells differentiates into one oocyte and thousands of nurse cells. Nurse cells display a number of features typical of apoptotic cells and are phagocytosed by the growing oocyte. Yet, these cells remain unchanged in morphology and number until hatching of the polyp, which can occur up to 12 months later. Treatments with caspase inhibitors can block oocyte development during an early phase of oogenesis, but not after nurse cell phagocytosis has taken place, indicating that initiation of nurse cell apoptosis is essential for oocyte development. The genomic DNA of the phagocytosed nurse cells in the oocyte and embryo shows large-scale fragmentation into 8- to 15-kb pieces, but there is virtually none of the internucleosomal degradation typically seen in apoptotic cells. The arrested nurse cells exhibit high levels of peroxidase activity and are prevented from entering the lysosomal pathway. After hatching of the polyp, apoptosis is resumed and the nurse cells are degraded within 3 days. During this final stage, nurse cells become TUNEL-positive and enter secondary lysosomes in a strongly degraded state. Our results suggest that nurse cell apoptosis consists of caspase-dependent and caspase-independent phases. The independent phase can be arrested at an advanced stage for several months, only to resume after the primary polyp hatches.

An Eph receptor sperm-sensing control mechanism for oocyte meiotic maturation in Caenorhabditis elegans.

During sexual reproduction in most animals, oocytes arrest in meiotic prophase and resume meiosis (meiotic maturation) in response to sperm or somatic cell signals. Despite progress in delineating mitogen-activated protein kinase (MAPK) and CDK/cyclin activation pathways involved in meiotic maturation, it is less clear how these pathways are regulated at the cell surface. The Caenorhabditis elegans major sperm protein (MSP) signals oocytes, which are arrested in meiotic prophase, to resume meiosis and ovulate. We used DNA microarray data and an in situ binding assay to identify the VAB-1 Eph receptor protein-tyrosine kinase as an MSP receptor. We show that VAB-1 and a somatic gonadal sheath cell-dependent pathway, defined by the CEH-18 POU-class homeoprotein, negatively regulate meiotic maturation and MAPK activation. MSP antagonizes these inhibitory signaling circuits, in part by binding VAB-1 on oocytes and sheath cells. Our results define a sperm-sensing control mechanism that inhibits oocyte maturation, MAPK activation, and ovulation when sperm are unavailable for fertilization. MSP-domain proteins are found in diverse animal taxa, where they may regulate contact-dependent Eph receptor signaling pathways.

Persistent hiccups associated with intravenous corticosteroid therapy.

Singulata, commonly known as hiccups, have many causes and are most often idiopathic and short-lived. Occasionally, hiccups can be persistent and becoming quite bothersome, distracting, and even disabling. Many medications have been implicated as a cause of hiccups. Although uncommon, corticosteroids and benzodiazepines are 2 classes of drugs most frequently associated with the development of hiccups. The following case series describes 5 patients with intravenous corticosteroid-associated hiccups reported to our adverse drug reaction reporting system between January 1998 and December 2000. Physicians using intravenous corticosteroids should be aware of this potential complication.

Field estimate of net trophic transfer efficiency of PCBs to Lake Michigan chinook salmon from their prey.

Chinook salmon (Oncorhynchus tshawytscha) has been the predominant piscivore in Lakes Michigan, Huron, and Ontario since the 1970s, and therefore accurate quantification of its energy budget is needed for effective management of Great Lakes fisheries. A new approach of evaluating a fish bioenergetics model in the field involves field estimation of the efficiency with which the fish retains PCBs from its food. We used diet information, PCB determinations in both chinook salmon and their prey, and bioenergetics modeling to generate a field estimate of the efficiency with which Lake Michigan chinook salmon retain PCBs from their food. Our field estimate is the most reliable field estimate to date because (a) the estimate was based on a relatively high number (N = 142) of PCB determinations for chinook salmon from Wisconsin waters of Lake Michigan in 1985, (b) a relatively long time series (1978-1988) of detailed observations on chinook salmon diet in Lake Michigan was available, and (c) the estimate incorporated new information from analyses of chinook salmon age and growth during the 1980s and 1990s in Lake Michigan. We estimated that chinook salmon from Lake Michigan retain 53% of the PCBs that are contained within their food.

Direct Raman imaging techniques for study of the subcellular distribution of a drug.

Direct Raman imaging techniques are demonstrated to study the drug distribution in living cells. The advantage of Raman imaging is that no external markers are required, which simplifies the sample preparation and minimally disturbs the drug mechanism during imaging. The major challenge in Raman imaging is the weak Raman signal. In this study, we present a Raman image model to describe the degradation of Raman signals by imaging processes. Using this model, we demonstrate special-purpose image-processing algorithms to restore the Raman images. The processing techniques are then applied to visualize the anticancer agent paclitaxel in living MDA-435 breast cancer cells. Raman images were obtained from a cancer cell before, during, and after drug treatment. The paclitaxel distribution illustrated in these images is explained by means of the binding characteristics of the paclitaxel and its molecular target-the microtubules. This result demonstrates that direct Raman imaging is a promising tool to study the distribution of a drug in living cells.

A prospective analysis of the treatment of friction blisters with 2-octylcyanoacrylate.

Early military investigations of blister treatment using tissue adhesives have shown promise, finding that these agents provide pain relief, prevent infection, and allow continuation of recruit training. A trial was conducted to compare a 2-octylcyanoacrylate treatment with military standard therapy. Patients were recruited during foot evaluation at the end of a 10-km military road march. Seventy-five patients were screened, and 51 were enrolled. Twenty-six patients received 2-octylcyanoacrylate and 25 received standard treatment. A total of 80 blisters were treated (40 in each group). The mean (SD) blister size in the 2-octylcyanoacrylate group was 12.7 (6.2) mm and in the standard group was 12.0 (5.7) mm. There were no statistically significant differences in any of the baseline variables. Baseline and repeated-measures visual analog scale scores demonstrated no statistically significant differences in initial or repeat pain scores. Both groups showed a similar change across time, with a nonsignificant trend toward improvement in the standard therapy group at 10 min (28.5 versus 24.9) and in the 2-octylcyanoacrylate group at 3 days (42.9 versus 50.1). Mean Likert scores were similar, indicating no difference in patient satisfaction. Time to resumption of normal activity was similar, with one patient in each group unable to return to activity at the time of follow-up. There was a trend toward an increased proportion of patients in the 2-octylcyanoacrylate group who were able to return to normal activity within 48 hours, but this did not reach statistical significance. 2-Octylcyanoacrylate was associated with a greater degree of procedural discomfort. No infected blisters were noted in either group.
