Integrative multi-omics profiling deciphers tumor microenvironment heterogeneity and immunotherapy vulnerabilities in lung neuroendocrine carcinomas.

INTRODUCTION: Lung neuroendocrine carcinomas (Lu-NECs) are rare, highly aggressive lung tumors with poor prognosis and limited therapeutic options. Understanding the tumor immune microenvironment (TIME) is crucial towards personalized therapeutic strategies. OBJECTIVES: This study aims to systematically characterize the heterogeneity and complexity of the TIME in Lu-NECs by integrating proteomic, transcriptomic, and genomic data. METHODS: We performed comprehensive immune-proteomic profiling of 76 Lu-NECs across diverse histopathological subtypes to elucidate intra-tumoral TIME heterogeneity at the proteomic level. Validation was conducted in multiple independent cohorts, including 112 Lu-NECs using immunohistochemistry, 147 Lu-NECs, and 17 small cell lung carcinoma samples using transcriptomics. We integrated proteomic, transcriptomic, genomic, and clinical data to assess molecular, immunological, and clinical features, as well as therapeutic vulnerabilities across different immune subtypes. RESULTS: We delineated the immuno-proteomic landscape of Lu-NECs and identified two major immuno-proteomic clusters with distinct immunological, molecular, and clinical characteristics. IPC1 was characterized by high immune cell infiltration, while IPC2 exhibited sparse immune cell presence. Genomic analysis revealed distinct mutational patterns, with IPC1 showing a higher incidence of APOBEC-associated mutation signatures and IPC2 being enriched for mutations associated with defective DNA mismatch repair and tobacco-related mutagens. Functional analyses indicated that IPC1 was related to immune and oncogenic signaling activity, whereas IPC2 was associated with cancer stemness and proliferation-related features. Furthermore, IPC1 and IPC2 demonstrated histological subtype-specific clinical benefits from postoperative chemotherapy. Finally, we developed a machine learning model (iPROM) to predict Lu-NECs immune classification and improve risk stratification, which was validated across multiple independent cohorts. CONCLUSIONS: This study advances the understanding of the tumor immune microenvironment in Lu-NECs through multi-omics characterization and highlights potential personalized therapeutic vulnerabilities tailored to the specific immune landscapes of Lu-NECs.

A test of the generality of leaf trait relationships on the Tibetan Plateau.

Leaf mass per area (LMA), nitrogen concentration (on mass and area bases, N(mass) and N(area), respectively), photosynthetic capacity (A(mass) and A(area)) and photosynthetic nitrogen use efficiency (PNUE) are key foliar traits, but few data are available from cold, high-altitude environments. Here, we systematically measured these leaf traits in 74 species at 49 research sites on the Tibetan Plateau to examine how these traits, measured near the extremes of plant tolerance, compare with global patterns. Overall, Tibetan species had higher leaf nitrogen concentrations and photosynthetic capacities compared with a global dataset, but they had a slightly lower A(mass) at a given N(mass). These leaf trait relationships were consistent with those reported from the global dataset, with slopes of the standardized major axes A(mass)-LMA, N(mass)-LMA and A(mass)-N(mass) identical to those from the global dataset. Climate only weakly modulated leaf traits. Our data indicate that covarying sets of leaf traits are consistent across environments and biogeographic regions. Our results demonstrate functional convergence of leaf trait relationships in an extreme environment.

[Therapeutic efficacy of lixue xiaoyin decoction in treating psoriasis and its effect on plasma edothelin].

OBJECTIVE: To study the therapeutic efficacy of Lixue Xiaoyin Decoction (LXD) in treating psoriasis and its effect on plasma endothelin (ET). METHODS: Two hundred and twenty patients were divided into two groups. LXD was taken orally by the 118 patients in the treated group, while Compound Qingdai Capsule (CQC) taken orally by the 102 patients in the control group. The therapeutic efficacy was evaluated after 8 weeks of treatment. The ET content was determined by radioimmunoassay before and after treatment. RESULTS: The total effective rate was 73.72% in the treated group and 54.90% in the control group with significant difference (chi2 = 8.52, P < 0.01). The plasm ET level in patients was significantly higher than that in the healthy subjects. ET in both groups was all lowered after treatment, but the decrement was more obvious in the treated group, showing significant difference when compared with that in the control group (P < 0.01). CONCLUSION: LXD shows good therapeutic efficacy in the treatment of psoriasis. It can improve the microcirculation, inhibit the division of epithelial cells, promote the epidermic cell differentiation, and shows regulative effect on plasma ET.

A genetic algorithm for detecting target geometric figures.

A genetic algorithm is proposed to detect target geometric figures in an given image. Float-point encoding was adopted to process the parameters of a geometric figures to be detected. On the basis of classical Hough transform, a fitness function was obtained for each individual task, and the individuals with the highest fitness function were identified and copied into the cohort of the next generation. For the rest of the individuals, operation with multi-point crossover or uniform mutation was performed to form new individuals in the next generation. When the termination conditions for this genetic algorithm were met, the best-fitted individual was decoded and output as the parameters of the detected geometric figures. This algorithm can eliminate noise interference with good convergence and accurate results, and may save time and storage space during relatively easily programmed computation in comparison with classical Hough transform.

Enantioseparation of vesamicol in human serum by capillary electrophoresis with solid phase extraction and sulfated-beta-cyclodextrin.

An enantioseparation of racemic vesamicol in human serum by capillary electrophoresis with solid phase extraction and sulfated B-cyclodextrin (S-B-CD) is presented The separation was achieved on an uncoated 72 cm x 50 microm id fused silica capillary maintained at 30 degrees C and + 15 kV applied voltage using a run buffer of 128 micro-B-CD in 50 mM phosphate buffer at pH 5. The detection wavelength was 260 nm. Bond Elut C18 solid phase extraction cartridges were used in the sample preparation of the vesamicol samples from serum. Among the CDs studied, the migration order of the enantiomers was reversed in CM-B-CD compared to S-B-CD. Increases in migration time and differences in time between enantiomers was observed with increasing concentrations of S-B-CD. Baseline separation was achieved in the 2-20 microg/ml range of enantiomer concentration (r > .996). A sample stacking technique was used to improve peak shape and LOD. LODs were 0.5 microg/ml for each enantiomer. Studies of various factors and CE conditions showed the effect of CD type, CD concentration, buffer type, buffer concentration and pH on stability and resolution.

A median filter algorithm importing ISODATA dynamic clustering for medical imaging.

To improve conventional median filter algorithm employed in medical imaging, we proposed a new median filter algorithm importing ISODATA dynamic clustering for pattern recognition. The result of clustering was set as the parameters to decide if median filter was necessary and when it was, how the process was to be carried out. As shown in our test, this algorithm was capable of eliminating serious impulse noises and retain thorough image details, therefore enhanced signal to noise ratio and quality of the images in contrast with the conventional median filter algorithm.

A simple analysis model simulating the correlation of cervical traction force with the pressure in the cervical nucleus pulposus.

OBJECTIVE: To design a simple model for analyzing the correlation between the changes in cervical traction force and that of the pressure in the cervical nucleus pulposus. METHODS: Multiple non-linear mathematical models were established by means of regression analysis to calculate the correlation coefficient and variance, and the capacity of each model to predict the changes on the test points was evaluated. RESULTS: Logarithm model was apparently not adequate for the purpose and quadratic model and power model showed poor capacity for predicting the changes of the experimental data. Exponential model and cubic model, in contrast, well illustrated the trend of the changes in the data, therefore best met the demands. CONCLUSIONS: Exponential model best describes the trends of the changes of the experimental data and can procure continuous values of the pressure reduction in the cervical nucleus pulposus in association with varied cervical traction forces, thus offering useful information for relevant theoretical analysis.

[Correlation of the (rotation) traction force with the pressure in the nucleus pulposus in multiple segments of the cervical vertebra].

OBJECTIVE: To study the correlation between cervical (rotation) traction force and the pressure in the cervical nucleus pulposus based on an examination of multiple segments of the cervical vertebra. METHODS: The data obtained from our observation were processed by the exponential model, and the changes in the pressure reduction (P) in the cervical nucleus pulposus were analyzed. RESULTS: When stretching followed by rotation was imposed on the cervical vertebra, an increase of P was first observed due to the stretching before a reduction in P caused by the rotation. The P of C(3-4) segments was constantly greater than that of C(5-6) segments, while the changes in P of C(4-5) followed different patterns. CONCLUSION: Changes of P in each segment of the cervical vertebra are similar in response to the load force, and there is no correlation of monotonic increase or decrease between the changes of P and the location of the cervical vertebra.
