Synthesis of anilino-monoindolylmaleimides as potent and selective PKCbeta inhibitors.

We report herein synthesis of PKCbeta-selective inhibitors possessing the novel pharmacophore of anilino-monoindolylmaleimide. Several compounds of this series exhibited IC50's as low as 50 nM against human PKCbeta2. One of the most potent compounds, 6l, inhibited PKCbeta1 and PKCbeta2 with IC50 of 21 and 5 nM, respectively, and exhibited selectivity of more than 60-fold in favor of PKCbeta2 relative to other PKC isozymes (PKCalpha, PKCgamma, and PKCepsilon).

Skeletal unloading alleviates the anabolic action of intermittent PTH(1-34) in mouse tibia in association with inhibition of PTH-induced increase in c-fos mRNA in bone marrow cells.

UNLABELLED: We analyzed the effect of unloading by tail suspension on the anabolic action of intermittent PTH in the tibia of growing mice. Unloading alleviated the PTH-induced increase of bone formation and accelerated bone resorption, consequently reducing bone mass. Reduction of the PTH-induced anabolic actions on bone was associated with unloading, which was apparently related to suppression of c-fos mRNA expression in bone marrow. INTRODUCTION: The effects of intermittent parathyroid hormone (PTH) administration on unloading bone have not been well elucidated at the cellular and molecular levels. We tested the effects of PTH on unloaded tibias of tail-suspended mice. MATERIALS AND METHODS: Eighty male C57BL/6J mice, 8 weeks of age, were divided into four groups with loading or unloading and administration of PTH (40 microg/kg body weight) or vehicle five times per week. Mice were killed at 8 or 15 days, and both tibias were obtained. Bone histomorphometry of the trabecular bone in the proximal tibia, development of osteogenic cells, and mRNA expression of osteogenic molecules in bone marrow cells were assessed. RESULTS AND CONCLUSIONS: At 15 days of unloading, bone volume decreased in PTH-treated mice. The increase in the bone formation rate by PTH was depressed, and the osteoclast surface was thoroughly increased. The increase in alkaline phosphatase-positive colony-forming units-fibroblastic (CFU-f) colonies induced by PTH was maintained and that of TRACP+ multinucleated cells enhanced. The PTH-induced increase in c-fos mRNA was depressed, but the increases in Osterix and RANKL mRNA were maintained. Unloading promoted the PTH-associated osteoclastogenesis and seemed to delay the progression of osteogenic differentiation in association with reduction of the PTH-dependent increase of c-fos mRNA in bone marrow cells.

Analysis and numerical computation of diffraction of an optical field by a subwavelength-size aperture in a thick metallic screen by use of a volume integral equation.

Diffraction of an optical field by an aperture in a thick metallic screen is analyzed numerically by use of a three-dimensional volume integral equation together with a generalized conjugate residual method and fast Fourier transformation. Numerical results were validated by reciprocity and the independence of the results of the truncated discretized volume size used in numerical calculations. Near and far fields of square, circular, and triangular apertures in a thick screen are obtained numerically. Some of the numerical results obtained in the present study agree with previously reported experimental results. The surface plasmon polaritons excited on the sidewalls of the aperture can explain the basic characteristics of near-field distribution of apertures. The Bethe-Bouwkamp theory was found to be insufficient to explain the basic characteristics of the near field around the subwavelength aperture in a practical metallic screen.

Immediate early genes expressed in chlorovirus infections.

Twenty-three chlorovirus genes expressed in host cells as early as 5-10 min postinfection (p.i.), or immediate early, were isolated and characterized. Some showed significant homology with those for transcriptional factors and mRNA-processing proteins including TFIIB, helicases, mRNA capping enzyme, nucleolin, and bean transcription factor. Others code for (i) factors influencing translation such as aminoacyl tRNA synthetases and ribosomal protein, and (ii) unknown proteins. Enzymes involved in polysaccharide synthesis were also found. All transcripts of these genes had a poly(A) tail, which decreased in size after 20 min p.i., possibly caused by the shortening by an exonuclease. Often, due to readthrough either from an upstream ORF or into a downstream ORF, a few extra transcripts for each gene appeared after 40 min p.i., suggesting a change in promoter selection and termination accuracy at this point. A typical TATA-box and a common element 5'-ATGACAA were in the promoter region of almost all of the immediate early genes, which may be recognized by host RNA polymerase and transcription factors.

Prostaglandin E2 receptor (EP4) selective agonist (ONO-4819.CD) accelerates bone repair of femoral cortex after drill-hole injury associated with local upregulation of bone turnover in mature rats.

Prostaglandin E(2) (PGE(2)) is essential for fracture healing. Systemic administration of EP4 ligands such as PGE(2) and other synthetic EP4 agonists appears to transduce anabolic signals by binding to receptor EP4. Therefore, the present study was designed to test whether administration of EP4 agonist accelerates the healing of drill-hole injury in the femoral diaphysis. After surgery, a total of 128 Wistar rats, at the age of 12 weeks, were assigned to basal control (n = 8), and three groups with respective doses of 0 (vehicle control), 10 (low-dose), and 30 (high-dose) microg/kg body weight of the agent were subcutaneously injected twice a day. Femoral specimens were obtained at 0, 5, 7, 14, 21, and 28 days. In EP4 agonist-treated groups, the total bone volume of the regenerating bone in the defect did not significantly differ, but the regenerated cortical bone volume measured by histomorphometry and cortical bone mineral content (Ct. BMC) by pQCT dose-dependently increased at 14 and 21 days compared to the control. In the high-dose group, the value of osteoclast surface significantly increased compared with that in the control at 14 days. Expression levels of osteocalcin and TRAP mRNAs in the injured tissue increased at 14 days. Expression levels of EP4, BMP-2, and RANKL mRNAs increased at 7 days in the high-dose group. The bone mineral values of the lumbar bone at 28 days, measured by DXA, did not differ in the three groups. These data indicated that systemic administration of EP4 agonist ONO-4819.CD accelerated cortical bone healing after drill-hole injury by upregulating the local turnover of the regenerating bone.

Simulation of confined and enhanced optical near fields for a long narrow aperture in a pyramidal structure on a thick metallic screen.

A new type of long narrow aperture in a pyramidal structure on a thick metallic screen is proposed, and optical wave scattering by this structure is simulated. This aperture structure provides high emission intensity and small spot size simultaneously through excitation of the surface plasmon polaritons on the sidewalls of the pyramidal structure. Scattering of optical waves by this structure in the thick metallic screen is solved numerically with a volume integral equation by generalized conjugate residual iteration and fast Fourier transformation. The basic characteristics of the near-field intensities of the aperture are investigated in detail.

Trends in pertussis among infants in the United States, 1980-1999.

CONTEXT: Reported cases of pertussis among adolescents and adults have increased since the 1980s, despite increasingly high rates of vaccination among infants and children. However, severe pertussis morbidity and mortality occur primarily among infants. OBJECTIVE: To describe the trends and characteristics of reported cases of pertussis among infants younger than 12 months in the United States from 1980 to 1999. DESIGN, SETTING, AND PARTICIPANTS: Cases of pertussis in infants younger than 12 months in the United States reported to the National Notifiable Disease Surveillance System of the Centers for Disease Control and Prevention between 1980 and 1999, and detailed case data from the Supplementary Pertussis Surveillance System. MAIN OUTCOME MEASURES: Incidence and demographic and clinical characteristics of cases. RESULTS: The incidence of reported cases of pertussis among infants increased 49% in the 1990s compared with the incidence in the 1980s (19 798 vs 12 550 cases reported; 51.1 cases vs 34.2 cases per 100 000 infant population, respectively). Increases in the incidence of cases and the number of deaths among infants during the 1990s primarily were among those aged 4 months or younger, contrasting with a stable incidence of cases among infants aged 5 months or older. The proportion of cases confirmed by bacterial culture was higher in the 1990s than in the 1980s (50% and 33%, respectively); the proportion of hospitalized cases was unchanged (67% vs 68%, respectively). Receipt of fewer doses of vaccine was associated with hospitalization, when cases were stratified by age in months. CONCLUSIONS: The incidence of reported cases of pertussis among infants increased in the 1990s compared with the 1980s. The limited age group affected, the increased rate of bacteriologic confirmation, and the unchanged severity of illness suggest that an increase in infant pertussis has occurred apart from any change in reporting. Strategies are needed to prevent the morbidity and mortality from pertussis among infants too young to be fully vaccinated, according to the current recommended schedules of vaccination in the United States.

Vascular endothelial growth factor is expressed along with its receptors during the healing process of bone and bone marrow after drill-hole injury in rats.

In this study, we investigated the expression of vascular endothelial growth factor (VEGF) mRNA along with its receptors in the healing process following rat femoral drill-hole injury. The cellular events involved in the differential expression of VEGF were studied by reverse transcription-polymerase chain reaction, immunocytochemistry, and in situ hybridization. Abundant alkaline phosphatase-positive osteoprogenitor cells were present in the bone marrow cavity surrounding the wound region at day 3. Some of the cells were immunoreactive for Flk-1, a marker of angioblasts. At day 5, osteoblasts expressing osteocalcin mRNA actively participated in bone formation. After day 11, medullary bone gradually decreased and hematopoietic cells covered the wound region. The expressions of the VEGF splice variants VEGF120 and VEGF164 were detected at days 1 and 3, and VEGF188 mRNA began to appear from day 5. The expressions of the three VEGF splice variants gradually decreased after day 11. VEGF immunoreactivity and mRNA expression were strongly detected in angioblasts, osteoprogenitor cells, and osteoblasts between days 3 and 7, but gradually decreased after day 11. Immunoreactivity for Flt-1 was also detected in endothelial cells, osteoprogenitor cells, and osteoblasts between days 3 and 7. However, immunoreactivity for Flk-1 was not detected on osteoblasts but rather on endothelial cells. These findings indicate that the differential expression of VEGF splicing isoforms along with its receptors may play an important role in the healing process after rat femoral drill-hole injury.

Accurate analysis of power coupling between two arbitrarily ended dielectric slab waveguides by the boundary-element method.

The boundary integral equations that are called guided-mode extracted integral equations are applied to the investigation of the power-coupling-properties between two arbitrarily ended dielectric slab waveguides. The integral equations derived in this paper can be solved by the conventional boundary-element method. The reflection and coupling coefficients of the guided wave, as well as the scattering power, are calculated numerically for the case of incident TE guided-mode waves. The results presented are checked by the energy conservation law and the reciprocity theorem. Numerical results are presented for several geometries of coupling, including systems with three-layered symmetrical and asymmetrical slab waveguides.

Chitin synthesis in chlorovirus CVK2-infected chlorella cells.

Hyaluronan synthesis in chlorovirus PBCV-1-infected Chlorella cells was previously reported (DeAngelis et al., 1997). In contrast, we report here on the detection, characterization, and expression of a gene for chitin synthase (chs) encoded by chlorovirus CVK2 isolated in Kyoto, Japan. The CVK2 chs gene encoding an open reading frame of 516 aa was expressed as early as 10 min postinfection (p.i.), peaked at 20-40 min p.i., and disappeared at 120-180 min p.i. The chitin polysaccharide began to accumulate as chitinase-sensitive, hair-like fibers on the outside of the virus-infected Chlorella cell wall by 30 min p.i. All chloroviruses without the gene for hyaluronan synthase (has) alternatively contained the chs gene, suggesting the importance of polysaccharide production in the course of virus infection. A few chloroviruses possessed both the chs and has genes and produced chitin and hyaluronan simultaneously. Polysaccharide accumulation on the algal surface may protect virus-infected algae from uptake by other organisms, such as protozoa. Since CVK2 was reported to encode two chitinases and one chitosanase, CVK2 is a very peculiar virus that encodes enzymes required for both the synthesis and the degradation of chitin materials.

Electroencephalographic changes during intravenous olfactory stimulation in humans.

The intravenous olfaction test with thiamin propyl disulfide (TPD) is a simple procedure widely used in Japan. An olfactory stimulus is provided by intravenous injection of TPD (2 ml) over the course of 20 sec. The subject smells n-propyl mercaptan (a decomposition product of TPD discharged from the blood into alveoli) in expired air after treatment. In this preliminary study we recorded electroencephalograms (EEGs) in normal subjects during three stages: 1) eyes-closed rest (prestimulus), 2) olfactory sensation after TPD injection, and 3) disappearance of sensation. In each of these stages, we calculated and compared EEG powers according to the band components of each electrode position. This study was designed a) to evaluate by frequency analysis EEG changes during olfactory sensation after TPD injection, and b) to identify the most significant changes in EEG power according to frequency band and electrode location. During the intravenous olfactory stimulation, alpha 2 and beta 2 waves were activated over the frontal and temporal regions. After disappearance of olfactory sensation, these waves decreased in the same regions. EEG powers returned to prestimulus levels.

Numerical simulation of an arbitrarily ended asymmetrical slab waveguide by guided-mode extracted integral equations.

The scattering phenomenon from an arbitrary-shaped end of a asymmetrical slab waveguide for the cases of TE and TM guided modes is simulated by means of boundary integral equations that are called guided-mode extracted integral equations. The integral equations that we derive can be solved by the conventional boundary-element method. Numerical results are presented for problems of three-layer asymmetrical waveguides with tilted ends. The reflection coefficient, reflected and scattered powers, and radiation patterns are calculated numerically for the cases of incident TE and TM guided modes.

[Chemoradiotherapy for lung, esophageal, and uterine cervix cancer].

Some early cancer is completely cured by radiation therapy alone. In many advanced cases, however, local control rates are low with radiation therapy. Chemoradiotherapy has recently become a standard treatment to improve therapeutic-results. The objective of chemoradiotherapy is to improve local tumor control, to control potential systemic metastases, or both. In chemoradiotherapy, drugs effective against primary tumors are selected, platinum compounds often being key drugs. Regarding the timing of radiation and chemotherapy, three patterns are possible: sequential, concurrent, and alternating. In the sequential pattern, the severity of acute toxicity decreases; however, an extended total therapeutic period can lower the antitumor effect. In the concurrent pattern, the maximum antitumor effect is expected because of the simultaneous action of radiation and chemotherapy, but the severity of acute toxicity is intensified to the extent that treatment withdrawal or dose reduction may be required. In the alternating pattern, radiation therapy is quit transiently during chemotherapy. As a result, the irradiation period is extended so that the antitumor effect might be reduced. To explore the methods and results of chemoradiotherapy in each disease, this paper also reports on the results of meta-analysis and randomized trials. Although chemoradiotherapy is not recommended for all patients, it is considered to be more appropriate than radiation therapy alone in patients who have good performance status and adequate organ function.

[Genital tuberculosis occurring in the spermatic cord: a case report].

Genital tuberculosis occurring in the spermatic cord is a rare disease. A 70-year-old man presented with a mass on the left side of the scrotum which had been painless and had gradually enlarged over the previous 4 months. Surgical excision was performed. The tumorous mass was located in the spermatic cord but did not connect with the testis or epididymis. The removed specimen was 15 x 20 x 15 mm in size and weighed 6 g. Histopathological diagnosis was tuberculosis. At present, 27 months after surgery, recurrence has not been found.
