Increased interleukin-8 expression by cigarette smoke extract in endothelial cells.

It has been reported that cigarette smoking worsens alcohol-induced gastric lesions through neutrophil infiltration. We hypothesize that IL-8, a potent chemotactic factor for neutrophil is likely to be involved in this ulcerogenic process. To evaluate this phenomenon, the ability of cigarette smoke extract (CSE) to induce endothelial cell expression of IL-8 was examined. Two different fractions (ethanol or chloroform soluble extracts) of CSE with their chemical types identified showed a time- and dose-dependent increase on IL-8 secretion from ECV304 cell line. Protein kinase C (PKC) inhibitor GF109203X had no effect on IL-8 response in basal secretion and also to these stimuli. Protein tyrosine kinase (PTK) inhibitor genistein and protein kinase A (PKA) inhibitor H8 at respective concentrations significantly reduced chloroform and ethanol soluble extract-induced IL-8 expression by about 34 and 35% respectively at 8 h after incubation. It is concluded that CSE increases IL-8 release from human endothelial cells through PTK and PKA activation.

Improvement of phase I drug metabolism with Schisandra chinensis against CCl4 hepatotoxicity in a rat model.

The seed extract of Schisandra chinensis was investigated in the rat for its restorative or therapeutic effect on Phase I hepatic drug metabolism following intoxication by carbon tetrachloride (CCl4). Male Sprague Dawley rats (220-250 g) were divided into two sets, one included rats with or without CCl4 intoxication, the other included CCl4 intoxicated rats with or without treatment of Schisandra extract. With the treatment regimen, rats received four oral doses of Schisandra (160 mg/kg) or the same volume of water at 8, 24, 32 and 48 h after CCl4 intoxication. A single oral dose (80 mg/kg) of antipyrine, a conventional probe for oxidative drug metabolism, was then administered. The levels of liver serum transaminases and cytochrome P450 were measured and the pharmacokinetics of antipyrine were assessed using a non-compartmental approach via WinNonlin. In comparison to the rats without CCl4 intoxication (t1/2: 2.2 +/- 0.9 h; Cl/F: 0.30 +/- 0.01 L/h/Kg; P450: 0.611 +/- 0.190 nmol/mg protein), CCl4 administration significantly decreased elimination (t1/2: 12.0 +/- 3.9 h) and oral clearance (Cl/F: 0.049 +/- 0.018 L/h/kg) of antipyrine, and markedly reduced the content of P450 (0.075 +/- 0.011 nmol/mg protein). Data obtained from intoxicated animals treated by Schisandra extract, compared to those without treatment, showed significant (p < 0.05) improvement in the t1/2 (4.45 +/- 1.7 h) and Cl/F (0.096 +/- 0.018 ml/h) estimates of antipyrine and a 2-3 fold increase in P450 level (0.190 +/- 0.072 nmol/mg protein). Findings in this study suggest that the seed extract of Schisandra appeared to be a promising agent for the improvement of Phase I oxidative metabolism in the liver damaged by CCl4.

Oral absorption and bioavailability of tea catechins.

The absorption characteristics and oral bioavailability of three tea catechins, namely (-)-epicatechin (EC), (-)-epicatechin gallate (ECG), and (-)-epigallocatechin gallate (EGCG), were assessed in this study. Male Sprague Dawley rats (210-230 g) received either an intravenous (i.v. 50 mg/kg) or oral (5000 mg/kg) dose of decaffeinated catechin-fraction containing EC (5%), EGCG (50%), and ECG (13%). Concentrations of the compounds in plasma, urine, and feces were measured using HPLC. A non-compartmental approach was employed for pharmacokinetic analysis. Results indicated that maximum plasma concentrations for the catechins (15-112 micrograms/ml) were achieved at 2 h post-oral dosing and the apparent volume of distribution (Vd/F) ranged from 30 to 63 l/kg. Absolute bioavailability (F) of EC, EGCG, and ECG was assessed to be 0.39, 0.14, and 0.06, respectively. Estimates of terminal elimination half-life (t1/2, lambda z) of the catechins after oral dosing were 451-479 min and were 1.4-10 fold longer than those observed for the i.v. dosing. The discrepancy in terminal elimination and low rate and extent of absorption indicated the possibility of flip-flop kinetics. Respective urinary recoveries were 0.17-4.72% and 2.11-14.2% after oral and i.v. dosing. In conclusion, the low systemic availability of tea catechins observed could be a result of slow absorption, high first pass effect, and wide tissue distribution.

Angiotensin II increases neuronal delayed rectifier K(+) current: role of 12-lipoxygenase metabolites of arachidonic acid.

Angiotensin II (Ang II) elicits an Ang II type 2 (AT(2)) receptor-mediated increase in voltage-dependent delayed rectifier K(+) current (I(KV)) in neurons cultured from newborn rat hypothalamus and brain stem. In previous studies, we have determined that this effect of Ang II is mediated via a Gi protein, activation of phospholipase A(2) (PLA(2)), and generation of arachidonic acid (AA). AA is rapidly metabolized within cells via lipoxygenases (LO), cyclooxygenase (COX) or p450 monooxygenase enzymes, and the metabolic products are known regulators of K(+) currents and channels. Thus in the present study, we have investigated whether the AT(2) receptor-mediated effects of Ang II on neuronal I(KV) require AA metabolism and if so, which metabolic pathways are involved. The data presented here indicate that the stimulatory actions of Ang II and AA on neuronal I(KV) are attenuated by selective blockade of 12-LO enzymes. However, the effects of Ang II are not altered by blockade of 5-LO or p450 monooxygenase enzymes. Furthermore, the actions of Ang II are mimicked by a 12-LO metabolite of AA, but 5-LO metabolites such as leukotriene B(4) and C(4) do not alter neuronal I(KV). These data indicate that the AT(2) receptor-mediated stimulation of neuronal I(KV) is partially mediated through 12-LO metabolites of AA.

Pharmacokinetic and pharmacodynamic interactions between intravenous ciprofloxacin and oral ferrous sulfate.

Changes in oral bioavailability and in vitro antimicrobial activity have been the focus of many previous interaction studies for metal cations and quinolones. This study is the first to examine the possibility of an interaction in the systemic circulation using ciprofloxacin and ferrous sulfate as representative interactants in a rat model, and to determine the changes, if any, in the pharmacokinetics and pharmacodynamics of the antibiotic. To minimize direct physical interaction in the gastrointestinal (GI) tract, the current study design required the male Sprague Dawley rats (220-240 g) to be dosed with 100 mg/kg of oral ferrous sulfate and 5 mg/kg of intravenous ciprofloxacin. Control animals received only intravenous ciprofloxacin. Blood and urine samples were collected over time for quantitation of ciprofloxacin independently by both HPLC (H) and microbiological (M) assays. Results showed that the disposition of ciprofloxacin in control animals was biexponential with a mean (+/-SD) terminal elimination half-life (t(1/2,lambda z)) of 0.93+/-0.30 h. A large apparent volume of distribution (V(d,lambda z): 6.96+/-1.56 L/kg) was observed. In addition, concentration vs. time profiles generated by both assays were similar. When the antibiotic was dosed with oral iron, parameter estimates generated by HPLC appeared to show a wider distribution and a longer elimination of ciprofloxacin; mean V(d,lambda z) and t(1/2,lambda z) estimates increased by 2- and 4-fold, respectively. Relative to controls, antibiotic exposure (AUC(0-infinity) was also significantly higher (p<0.05) in the presence of iron (1.89+/-0.15 vs. 1.00+/-0.39 mg/h/L). A strong assay dependency was observed for ciprofloxacin concentrations observed post-distribution; the respective M/H ratios for AUC(0-infinity) and urinary recovery were 1.1 and 0.9 for controls and 0.7 and 0.5 for animals receiving oral iron. This iron related reduction in antimicrobial activity was in clear contrast to the higher exposure and longer t(1/2,lambda z) of the antibiotic. In conclusion, concomitant oral iron dosing induced significant changes in the pharmacokinetics/pharmacodynamics of intravenous ciprofloxacin.

[Functional and structural analysis of a prokaryotic enhancer-like element in Escherichia coli MC1061 strain].

Cat and lacZ genes were used as reporter gene and three prokaryotic enhancer-like element (MC2, MC8 and MC9) were identified in the genomic DNA of MC1061 strain. All three fragments can improve the expression of lacZ gene by 2-5 times with the orientation independence. The results of in vivo transcription and Dot blot hybridization assays suggested that MC8 regulated the expression of lacZ at transcription level. Stepwise deletion expreriments showed the functional domain of MC8 located at 450-950 bp, and in regions 450-600 bp and 840-950 bp contain at least one functional loci. Sequence data indicated three are 3 A + T rich sections in MC8, 2 of them are in the functional loci.

Functional analyses of patient-derived IgG monoclonal anticardiolipin antibodies using in vivo thrombosis and in vivo microcirculation models.

Antiphospholipid antibodies (aPL) have been associated with thrombosis and pregnancy losses in patients diagnosed with antiphospholipid syndrome (APS) and enhance thrombus formation in vivo in mice, but the mechanism of thrombosis by aPL is not completely understood. It has been proposed that aPL may affect endothelial cell (EC) function and/or induce their activation, transforming their anticoagulant surface into procoagulant, thus predisposing to thrombosis. This study proposes to test the hypotheses that some IgG anticardiolipins (IgG aCL) with thrombogenic properties in mice, exert their effects through activation of endothelium. We studied seven patient-derived monoclonal aCL for their thrombogenic properties in an in vivo pinch-induced thrombosis model, and their functional activities in activating EC by analyzing in vivo leukocyte adhesion to endothelium in microcirculation in venules in exposed murine cremaster muscle and in vitro adhesion molecule expression in cultured EC. The binding of the monoclonal aCL to EC was also tested. In addition to the previous identified thrombogenic IS2, four of the five new more IgG monoclonal aCL (from two patients) were found to be thrombogenic. Of these five thrombogenic aCL, three caused more in vivo leukocyte adhesion to EC in microcirculation, as compared to that induced by the H2 control human monoclonal IgG, and enhanced expression of adhesion molecules (particularly VCAM-1) on cultured EC. These data show that about 2/3 patient-derived IgG monoclonal aCL are thrombogenic and suggest that some thrombogenic IgG aCL exert their effects through activating EC.

[Epidemiological aspects of the genotype of hepatitis C virus].

OBJECTIVE: To research the epidemiological characteristics of genotype of hepatitis C virus. METHODS: 107 HCV isolates from 7 cities in southern, northern and northeast of China were analyzed using PCR and second generation line probe assay (INNO-LiPA). RESULTS: The analysis of HCV genotypes showed that: 1. HCV l b was the common genotype (83 17%), however the frequency of genotype l b in northeast (72.22%) was lower than northern (87.65%, P <0.01), and the frequency of the genotype 2 was in low level (6.86%). The rate of coinfection with l b and other genotype was 10.29%. 2. The rate of infection with genotype l b in female patients was higher (93.61%) than that in male (75.0%) showing statistical significance (P <0.01). 3. In three age groups (< 30, 30 - 49 and > 50 years old) the rates of infection with genotype l b were 77.77%, 83.92% and 90.90%, respectively. 4. The detection rates of genotype l b in patients with infection period > 10 years and <10 years were 100% and 80.43% respectively (P <0.01). CONCLUSIONS: There are some relationship between HCV genotype and HCV geographic distribution, sex, age and infection period of patients.

99mTc-HL91 "hot spot" imaging of mice bearing human carcinoma by gamma camera and the effects of tumor necrosis on imaging.

OBJECTIVE: To evaluate the hypoxia-avid agent 99mTc-HL91 (99mTc labeled 4, 9-diaza-3, 3, 10, 10-tetramethyldodecan-2, 11-dione dioxime) as the tracer of tumor "hot spot" imaging and the influence of tumor necrosis on the image. METHODS: After injection of 99mTc-HL91, 6 nude mice bearing human breast cancer MCF-7 and 18 nude mice bearing human pancreatic adenocarcinoma were subjected to gamma camera imaging, postmortem analysis, and autoradiography and imaging of tumor sections. RESULTS: The image of tumor was identified 1 hour after injection of 99mTc-HL91. Images demonstrated gradually increased 99mTc-HL91 uptake in the tumor 1-12 hours after injection (P < 0.05-0.001). Six hours after injection, the radioactivity ratios of tumor to thorax and tumor to head were higher than 2.1. Six hours after injection, the radioactivity ratios of tumor to brain, muscle, blood, heart, lung and kidney in pancreatic adenocarcinoma bearing nude mice were 101.0 +/- 114.7, 30.0 +/- 30.3, 19.9 +/- 21.9, 14.4 +/- 15.1, 3.71 +/- 2.41 and 0.46 +/- 0.26, respectively, and the radioactivity ratios in breast cancer MCF-7 bearing nude mice were close to these figures. The radioactivity of non-necrotic tumor was 3.77 times that of necrotic tumor. However, the radioactivity ratios of tumor to liver, intestine and stomach were lower than 1.3. Autoradiographs and images of tumor sections showed that the radioactivity was higher in the region of solid tumor than in the necrotic region. CONCLUSION: 99mTc-HL91 via gamma camera positively identifies regional tumor in nude mice bearing human cancer. 99mTc-HL91 retention is lower in necrotic tumor than in non-necrotic tumor. The low radioactivity ratio of tumor to abdominal organs limits the application of 99mTc-HL91 in detecting abdominal tumors.

[In vitro studies on dendritic cell derived from cord blood and its role in antitumor immunity].

OBJECTIVE: To investigate the way of inducing dendritic cell (DC) from precursor in cord blood and its role in antitumor immunity. METHODS: Cord blood was collected under sterile condition and the mononuclear cells (MNCs) were separated by centrifugal in density gradient (Ficoll-Hypaque). The adhesion cells, collected among the MNCs, were cultured with interleukin-4 (IL-4, 50 ng), granulocyte-macrophage colony-stumilating factor (GM-CSF, 100 ng) and tumor necrosis factor alpha (TNF-alpha, 1-2 ng) in dulbecco's modified eagles medium (DMEM) medium. Cell phenotype was analyzed on 10th day with CD1a, CD16, CD14, CD19, CD40, CD56, CD80, CD83, and human leukocyte antigens-D-related (HLA-DR) antibody by using indirect immunofluorescence assays. After cultured with tumor cytolysis antigen for 2 days, the cells were incubated with auto T cell separated by E-rosette formation. Methyl thiazolyl tetrazolium (MTT) assay was employed to test inhibition rate of tumor specific cytotoxic T lymphocyte (CTL) on HO9810 tumor cell line. RESULTS: Our results indicated that DC precursors in cord blood can be induced to differentiate and mature in the medium containing IL-4, GM-CSF, and TNF-alpha. Some DC specialized antigens expressions were seen on 10th day after cultured with cytokines. After incubation with tumor cytolysis antigen, the DC can activate the CTL to become tumor specialized CTL, which had shown significantly inhibition on growth of HO9810 tumor cell line. CONCLUSIONS: Primary in vitro experiment has shown that the precursors in cord blood can be induced to functional DC which activate T lymphocyte to become tumor specialized CTL. Our result indicate that it is an efficient and convenient way in antitumor immunity by inducing DC precursors in cord blood to functional DC and may have a great implication on clinical application.

[Detecting mycobacteria and their L-forms in peripheral blood from pulmonary tuberculosis patients by cultivation with hemolyzed-centrifugated blood in liquid medium].

OBJECTIVE: To establish a new method of detecting mycobacteria and their L-forms in blood. METHODS: 65 samples of peripheral blood from patients with pulmonary tuberculosis were cultivated directly or cultivated with hemolyzed-centrifugated blood sediments in 92-3TB and 92-3TBL liquid medium, and the cultures were staining with immunoenzyme technique (ABC method). RESULTS: In 65 specimens, the positive rates of mycobacteria and their L-forms were 15%, 26% respectively with total isolation rate of 32%. In cultivating with hemolyzed-centrifugated blood the positive rate was much higher than direct cultural method (P < 0.05). In sputum the positive rates of acid-fast bacilli and their L-forms were 38% and 20% respectively, with total isolation rate of 52%, higher than that in peripheral blood (P < 0.05), and the total positive rate of combinative detection of blood and sputum salmples was 65%. CONCLUSIONS: Mycobacteria and their L-forms exist in peripheral blood of patients with pulmonary tuberculosis, and mainly in L-forms. Cultivation with hemolyzed-centrifugated blood in liquid medium and staining with immunoenzyme technique are valuable in routine detection of mycobacteria and their L-forms in peripheral blood. The positive rate of bacterial culture of pulmonary tuberculosis could be increased in combination with sputum mycobacteria and their L-forms examination.

[Study of platelet membrane glycoproteins and its fibrinogen binding reaction in patients with cerebral infarction and diabetes mellitus].

OBJECTIVE: To investigate the change of platelet membrane glycoproteins and its fibrinogen binding reaction in patients with cerebral infarction and type II diabetes mellitus (DM), and assess their value in the study of thrombosis and pro-thrombosis state. METHODS: By flow cytometry, the platelet glycoprotein (GP) I b, GP II b, GP III a, GP II b- III a complex and P-selectin expression as well as fibrinogen binding reaction were analyzed in 85 patients with type II DM and 28 with cerebral infarction, and compared with that in 30 normal subjects. RESULTS: The platelet membrane GP II b- III a complex, P-selectin expression and fibrinogen binding reaction were higher, while the platelet surface GP I b was lower in the patients with cerebral infarction and type II DM with vascular disease than those in normal group. The GP II b and GP III a were not significantly changed. There was no difference of these parameters between the type II DM without vascular disease patients and normal group. CONCLUSION: The platelet fibrinogen binding reaction, P-selectin and GP II b- III a complex expression were increased while the GP I b decreased in patients with cerebral infarction and type II DM with vascular disease.

[Effects of different cryoprotectants on biological properties of hematopoietic cells derived from cord blood].

OBJECTIVE: To search for effective cryoprotectants that can be used in cord blood bank to cryopreserve hematopoietic cells. METHODS: Four kinds of combined reagents were used to cryopreserve cord blood cells. The effects of these cryoprotectants on biological properties of hematopoietic cells were monitored and compared after the cells were refrigerated for 1 to 4 months. The detected biological properties included the number of total nucleated cells (TNC), CD(34)(+) cells and CFCs. RESULTS: The cryoprotectants of Dextran-40 + 10%DMSO resulted in the highest recovery rates, TNC, CD(34)(+) cells and CFCs were 77.8%, 78.5% and 87.2%, respectively. The combination of 10%DMSO + 6% AB serum + 5%HES led to the lowest recovery rates. CONCLUSION: Dextran-40 + 10%DMSO is an ideal cryoprotectant combination for cryopreserving hematopoietic cells in the cord blood bank.

[Effects of maxillary growth of Wistar rats with bilateral artificial cleft palate after premaxillary orthopedic treatment on expression of proliferating cell nuclear antigen(PCNA)].

OBJECTIVE: Being based on the principle of Latham's appliance, this experiment is designed to detect activity of osteoblasts in the maxillary sutures of Wistar rats with bilateral artificial cleft palate by immunohistochemistry technique, and evaluate the effects of this appliance on the growth and development of the maxilla. METHODS: The animal models of male infant Wistar rats with bilateral cleft palate were made by removing some palatal bones, splitting a "V" gap of 1.5 cm wide at the line between premaxilla and segments of maxilla. A sort of appliances, which could be fixed in the mouth of Wistar rats with bilateral cleft palate for correcting protrusion premaxilla was constructed basing on the principle of Latham's appliance. Then the diferent pathological changes of osteoblast proliferation between the experimental group and the two controlled groups were examined. The jugomaxillary sutrues, temporomalar sutures and sphenoipalatine suture were harvested 7, 14 days after premaxillar orthopedic treatment, followed by 4% paraformaldehyde fixing about 1 hour, demineralization with 15% EDTA and 0.5% paraformaldehyde for 48 hours, distilled water washing for a night, dimethylbenzene transparant dealing, and paraffin wax embeding. Proliferating Osteoblasts in all these sutures were investigated using immunohiostochemical technique with monoclonal antibodies of proliferating cell nuclear antigen (PCNA). RESULTS: Seven days after orthopedic treatment, no significant difference was observed between the experimental group and the two controlled groups. While after fourteen days, obvious PCNA-positive expression were observed in cells of all these sutures of the experimental group. CONCLUSION: The distribution of proliferating cells and the degree of cell proliferation change after premaxillary orthopedic treatment. And significant cell proliferation is observed in the experimental group, but there are no significant differences between the two controlled groups.

[Cephalometrical analysis of effects of premaxillary orthopedic treatment on maxillary growth of Wistar rats with bilateral artificial cleft palate].

OBJECTIVE: To measure the differences between the experimental group and the two controlled groups by using the cephalometric radiography technique to study the effects of premaxillary orthopedic treatments on the growth and development of maxilla. METHODS: To make an animal model with bilateral cleft palate by surgery operation: General anesthesia was carried on by injecting 2.5% pentobarbital (25 mg/kg) into abdominal cavity, then almost the same bone of rats' palate were removed from bilateral palate. An appliance, based on the principle of Latham's appliance was made and designed for premaxillary orthopedic treatment. It was fixed in rats' palate using pins under the condition of general anesthesia. After 20 days of orthopedic treatment, the rats were anesthetized by ether, and cephalometric films were taken under self-designed cephalometer and analysed aided by computer. RESULTS: The distances that represented the maxillary position in the posterior-anterior (PA) direction significantly increased (P < 0.05), while the angles that represented the direction of premaxillary growth significantly decreased (P < 0.05) between the experimental group and the two controlled groups. There were no significant difference between the two controlled groups. CONCLUSION: Premaxilla of rats can be effectively moved backward and rotated downward and backward on the PA direction by appliance. The appliance can move the segment palate forward during the premaxillary backword. The appliance can stimulate maxillary growth.

[Determination of the saflor yellow-A in Carthamin tinctorius].

The content of the saflor yellow-A in Carthamin tinctorius medicinal materials was determined by the HPLC method. The C18-ODS(150 x 4.5 mm) column and methanol-acetonitrile-0.7% H3PO4(26:2:72) as a mobile phase were used. The flow rate was 1.0 ml/min and the detective wavelength was 403 nm. The average recovery was 100.72% and the relative standard deviation (RSD) was 1.73%.

[Impact of glutamine of gut permeability and clinical prognosis on the aging patients undergoing gastric-intestinal operation].

OBJECTIVE: To evaluate the impact of parenteral nutrition supplemented glutamine on aging patients undergoing gastric-intestinal operation. METHODS: 30 patients above 60 years old undergoing gastric-intestinal operation, a randomized double-blind protocol was designed, divided into two groups, received impact isocaloric parenteral nutrition. The study group received alanyl-glutamine [0.5 g/(kg.d)]. To observe plasma amino acids profile, nitrogen balance, intestinal permeability and clinical prognosis, examine clinical chemistry variables and observe the adverse reactions in order to find out its safety. RESULTS: The patients in both groups were comparable prior to the operation. The plasma glutamine level of study group is higher than the control group, it's cumulative nitrogen balance values were prior to the control group, L/M ratio was lower than the control group. The complications related to infection was observed more in the control group. No adverse reaction was observed in both groups. CONCLUSIONS: Ala-Gln-supplemented PN improved nitrogen balance and maintained intestinal permeability, reduced complications.

[Preventive and therapeutic effect of Chinese herbs naofeikang on experimental emphysema and pulmonary artery hypertension in hamsters].

OBJECTIVE: To observe preventive and therapeutic effect of Chinese herbs Naofeikang on hypoxic pulmonary artery hypertension of elastase-induced emphysema hamsters and investigate its mechanism. METHODS: Hamsters for 30 days after intratracheally-instilled elastase, were kept in hypoxia environment under normal atmospheric pressure for 15 days, 50 hamsters were divided into prevention group (Prv), treatment group (T), emphysema + hypoxia (EH), and control group(N). Mean pulmonary artery pressure (MPAP) was measured before hamsters were killed. And then right ventricle hypertrophy index (RVHI) was measured, as well as circulating endothelial cells (CEC) and cells recoveries from bronchoalveolar lavage fluid (BALF) were counted. Meanwhile, pulmonary tissue changes were studied under light microscope with morphometric analysis. RESULTS: Compared with EH group, MPAP, CEC, and cells recoveries of BALF of Prv and T groups were significantly decreased (P < 0.05 or P < 0.01); Mean linear intercept and percentage of arterial media area of Prv and T groups had totally a significant difference (P < 0.01) in comparison with EH group. CONCLUSIONS: Chinese herbs Naofeikang could lower pulmonary hypertension, preserve vessel endothelial cells and lessen the inflammatory reaction in pulmonary tissue. Thereby, it could hinder the further development of emphysema and inhibit the remodeling of pulmonary small artery.
