Selective ablation of antennular sensilla on the Caribbean spiny lobster Panulirus argus suggests that dual antennular chemosensory pathways mediate odorant activation of searching and localization of food.

In spiny lobsters and other decapod crustaceans, odorant-mediated searching behavior patterns are driven primarily by chemosensory neurons in the antennules. Two groups of antennular chemosensory neurons can be distinguished on the basis of the sensilla that they innervate and their central projections: those that innervate the aesthetasc sensilla on the lateral flagella and project into the glomerularly organized olfactory lobes, and those that innervate other (i.e. non-aesthetasc) sensilla on both lateral and medial flagella and project into the stratified and non-glomerularly organized lateral antennular neuropils. By ablating different groups of antennular sensory neurons or sensilla, we examined the role of aesthetasc and non-aesthetasc chemosensory neurons in regulating local searching behavior of Caribbean spiny lobsters, Panulirus argus, for food (squid) in a low-flow environment. The results show that odorant-mediated activation of searching and localization of food under these conditions requires only a subset of functional antennular chemosensory neurons, since neither aesthetasc chemosensory neurons nor non-aesthetasc chemosensory neurons are by themselves necessary for these types of behavior. However, ablation of aesthetasc chemosensory neurons together with subsets of non-aesthetasc chemosensory neurons from either the medial or lateral flagella impairs the ability of lobsters to locate the food. This reveals a large degree of functional redundancy but also some complementary functions between aesthetasc and non-aesthetasc chemosensory neurons, and hence between these dual antennular chemosensory pathways, in odorant-mediated searching behavior of lobsters under these conditions.

Prevalence and location of peripheral nerve found on prostate needle biopsy.

We examined 238 prostate biopsy cores from 40 sextant biopsies by routine H&E stain to determine the presence of prostatic adenocarcinoma (PCa), perineural invasion (PNI), and nerves. Step sections were immunostained for S-100 protein to confirm the presence of nerves. Nerves were distributed evenly in specimens from apex, mid gland, and base. No significant difference was observed in the number of nerves in these areas. Significantly more nerves were found using the S-100 stain than the H&E stain. There was no significant difference between cancerous and benign specimens. However, there was a significant reduction in the nerve density in cancerous compared with benign specimens. PNI was identified in 3 of 11 PCas. All foci of PNI were identified on H&E stain. The S-100 immunostain detected no additional foci of PNI. Nerves are distributed evenly in standard sextant biopsy specimens from prostatic apex, mid gland, and base. In addition, the specimens with PCa have nerve distribution similar to that of benign specimens, suggesting that finding no PNI may represent true absence of PNI in the specimens. Careful examination of routine H&E-stained specimens is sufficient to detect all important PNI.

Pharmacokinetics of cocaine in maternal and fetal rhesus monkeys at mid-gestation.

We compared pharmacokinetics of cocaine and its metabolite, benzoylecgonine, in pregnant rhesus monkeys and their fetuses at mid-gestation: 1) after a single intravenous dose of cocaine, 2) after a single oral dose of cocaine, 3) after the last oral cocaine administration of a 50-day-long chronic cocaine treatment, and 4) on the last day of a 50-day-long chronic treatment with five daily intravenous cocaine injections. We found that intravenous administrations of cocaine produced maximal maternal levels of benzoylecgonine below the plasma levels for cocaine. In contrast, oral administrations resulted in the maximal maternal plasma levels of this metabolite significantly above those of cocaine. The bioavailability of the orally administered cocaine was calculated as 25%. Cocaine was detectable in the fetal plasma at maximal levels of approximately 1/5 of peak maternal levels for both single intravenous and single oral administrations. The maximal plasma levels of benzoylecgonine for the fetuses of the intravenously treated mothers were close to those of cocaine, whereas peak levels of this metabolite in the plasma of the fetuses of the mothers receiving the oral treatments were above those of cocaine. The chronic treatments resulted in significantly higher maximal levels of cocaine in the fetal circulation compared with those produced by single drug administrations.

Molecular markers for renal cell carcinoma: impact on diagnosis and treatment.

Molecular tumor markers are used to aid the differential diagnosis of renal cell carcinoma and to monitor disease progression and recurrence. They may also provide prognostic information and help in the design of therapeutic regimens. So far, no satisfactory markers are available for the diagnosis of renal cell carcinoma. Many markers have been evaluated for their potential use as prognostic factors. Some are promising, yet need vigorous clinical trials to validate. Genetic changes detected by molecular and cytogenetic methods provide markers specific for renal cell carcinoma. High-throughput DNA and tissue microarray techniques promise to revolutionize the discovery and validation of novel molecular markers.

[Cloning and sequencing of prolactin gene cDNA in three chicken breeds].

The total RNA was extracted from the pituitary of two Chinese native chicken breeds, Yuehuang and Taihe Silkies, and one layer Isa, using RNeasy Kit (QIAGEN, Germany). The total RNA was used to synthesize a specific fragment with RT-PCR, in which the primers were designed based on the sequence of broiler prolactin gene cDNA. The specific fragment was ligated to a linear plasmid pBSSK and cloned with XL1-Blue. The sequencing of prolactin cDNA was carried out with ABI PRISH 377XL DNA Sequencer after cloning. The cDNA sequences and deduced amino acid sequences of prolactin gene of two Chinese native chicken breeds and one layer were compared with that of broiler, dwarf chicken and turkey. There was 93.97%-99.89% cDNA sequence homology among Chinese native, layer, broiler and dwarf chickens, in which there was the highest (99.87%) between those of Taihe and dwarf chickens. There was 98.25%-100% deduced amino acid sequence homology among Chinese native, layer, broiler and dwarf chickens, in which there was the highest (100%) between those of Taihe Silkies and dwarf chickens. It was found that Yuehuang and Taihe Silkies had the same signal peptide cleavage site Leu-Pro-Ile-Cys among amino acids sequence deduced from pre-prolactin cDNA as broiler, dwarf and turkey, while layer Isa had a different cleavage site Pro-Pro-Ile-Cys. Such difference might cause a different translation processing of pre-prolactin, which could make layer Isa non-broody. There were different amino acids in the positions 71, 141, 150 and 175 of deduced prolactin amino acid sequences among Yuehuang, Taihe Silkies, layer, broiler and dwarf chickens. There was a heparin-binding site in positions 175-181 (L-R-R-D-S-H-K) among prolactin amino acid sequences of broiler and Taihe Silkies.

Correlation between the expression of cyclin A protein and p53 activity in oral squamous cell carcinomas.

Cyclins and wild-type p53 protein are prime cell cycle regulators and may be involved in tumorigenesis. Cyclin A is a late S cyclin and its abnormalities have been reported in several cancers, including oral squamous cell carcinomas. To explore whether aberrant G1/S in p53 mutant tumours leads to increased cyclin A protein in oral squamous cell carcinomas (OSCC), a total of 39 samples were evaluated for the expression of cyclin A and p53 protein by an immunohistochemical method using a labelled polymer assay. These samples comprised two hyperkeratotic and three oral premalignant lesions (two moderate and one severe dysplastic lesions), and 27 OSCC, together with seven healthy controls. The results demonstrated that the cyclin A protein was localized and highly expressed in the nuclei of the tumour cells. Although there was no correlation between cyclin A detection and the local lymph node involvement, a positive correlation was noted between the positivity of cyclin A and p53 protein (p <0.05). The results suggested that cyclin A may contribute to the progression of oral cancer and correlated to some degree with that of the p53 gene activity.

[Chromosomal aberration analyzed by comparative genomic hybridization in nasopharyngeal carcinoma].

OBJECTIVE: To identify genetic alteration in primary sporadic nasopharyngeal carcinoma(NPC) in Hunan. METHODS: Comparative genomic hybridization was applied to investigate the genomic imbalance in a series of 20 cases of primary nasopharyngeal carcinomas. RESULTS: The common chromosomal alterations included gains of chromosomes 1q, 2, 3q, 7q,8q,12 as well as losses of chromosomes 3p, 9p, 11q, 16q. CONCLUSION: There are multiple regions of chromosomes with copy number changes in NPC. The tumor suppressor genes and oncogenes on these regions may be involved in the development and progress of nasopharyngeal carcinoma.

Lack of association of prostate carcinoma nuclear grading with prostate specific antigen recurrence after radical prostatectomy.

PURPOSE: Grading prostate cancer using the Gleason system relies only on architectural tumor growth, in contrast to other systems, such as the WHO system, which grade prostate carcinoma based on nuclear features as well as architectural patterns. The prognostic significance of nuclear grading remains controversial since most studies were performed before prostate specific antigen (PSA) screening became widely available. We evaluated the significance of nuclear grade for predicting PSA recurrence in a contemporary cohort of patients treated with radical prostatectomy for clinically localized prostate carcinoma. MATERIALS AND METHODS: Nuclear grades 1 to 3 were determined in 141 consecutive radical prostatectomies in 1995. Predominant and worst nuclear grade was determined by a consensus of 3 pathologists. Statistical analysis compared nuclear grade with Gleason score using the chi-square test. The Cox proportional hazards analysis was performed to calculate the ability of nuclear grade, Gleason score and other variables to predict PSA recurrence. RESULTS: We identified a significant association of Gleason score with worst nuclear grade (p = 0.007). All 6 cases with a Gleason score of 8 or greater had a worst nuclear grade of 3, in contrast to 36 of 60 (60%) with a score 6 or less, in which the worst nuclear grade was 3. Of the 141 patients 31 (21.9%) had PSA recurrence at a median followup of 3.7 years. The univariate Cox model revealed significant associations of PSA recurrence with Gleason score 8 or greater (hazards ratio 5.5, p = 0.005), extraprostatic extension (hazards ratio 3.4, p = 0.001), positive surgical margin (hazards ratio 2.6, p = 0.009), seminal vesicle involvement (hazards ratio 7.3, p <0.001), preoperative serum PSA (hazards ratio 1.03, p = 0.007), tumor stage (hazards ratio 3.6, p = 0.001) and maximal tumor dimension (hazards ratio 2.4, p <0.001). However, overall and worst nuclear grade did not predict PSA recurrence (p = 0.89 and 0.13, respectively). Nuclear grade did not fit any multivariate model tested, which otherwise included Gleason score, log(PSA), surgical margin status, extraprostatic extension, seminal vesicle status, tumor size and pathological stage. By varying sample fixation time we also showed that benign prostate tissue in the same section as prostate carcinoma had grade 2 or 3 nuclear changes, that is moderate to marked anaplasia. CONCLUSIONS: High nuclear grade is associated with high Gleason score. However, prostate carcinoma with a Gleason score of 6 or less shows extreme variability. Nuclear grade determined by light microscopy failed to predict PSA recurrence in a contemporary series of men with clinically localized prostate cancer treated with radical prostatectomy. Nuclear morphology is subject to tissue fixation and processing artifact. Any nuclear morphometric study must consider this artifact.

[Phospholipase activities of Candida albicans isolates from oral pre-malignant lesions and oral cancers].

OBJECTIVE: The aim of this study is to compare activities of phospholipase of Candida albicans isolates from oral cavities of two groups of patients, including oral pre-malignant lesions and oral cancers. METHODS: A total of 55 isolates, including oral pre-malignant lesions (21), oral cancers (15), and normal health control (19), were studied using the precipitate zone assay on the egg yolk agar. RESULTS: No significant difference was found in phospholipase activities between these isolates from normal controls and oral pre-malignant lesions, while the activities of phospholipase of isolates from oral cancers were higher than those of the normal controls. CONCLUSION: Candida albicans may play different roles in different manners in these two closely related diseases.

[The examination of DNA repair capacity in the patients with oral precancerous lesion].

OBJECTIVE: The occurrence of oral precancerous lesion is related with not only carcinogenic environmental factors but also some self-regulating mechanisms. The genetic heredity factors determine individual's DNA repair capacity. The study is attempted to investigate the change of DNA repair capacity of the patients with the precancerous lesion. METHODS: The study is conducted in 16 pairs of oral precancerous lesion and normal controls by using host cell reactivation assay, while some dangerous factors such as smoking, alcohol intaking and family history of cancers are also recorded. RESULTS: The host cell reactivation in the patients with precancerous lesion is lower than that of normal controls (P < 0.05) and is shown to be an independent risk factor in the occurrence of oral precancerous lesion (OR: 3.25 95% CI = 1.01-10.49). CONCLUSION: The host cell reactivation is likely to be a helpful indicator for predict prognosis of cancer for the individuals with highly genetic sensitivity to cancer.

[Effects of isocarbophos on the integrity of epidermic cell membrane of human skin].

OBJECTIVE: To investigate the effects of isocarbophos, a new and highly effective organophosphorus pesticide on the membranes of keratinocytes. METHODS: The epidermic cells of human skin cultured in the medium were adopted as a biological material. The activity of lactate dehydrogenase (LDH) was used as an indicator. We tested the activities of LDH and alkaline phosphatase (ALP) in the medium in different isocarbophos dosage conditions. RESULTS: It was found that the activity of LDH in the medium decreased with the increase of isocarbophos dosage. The LDH activities of the dosage groups were statistically different from those of the positive and negative groups. The ALP activities were of no significant difference between the groups. CONCLUSION: The above findings reveal that isocarbophos does not damage the membranes of keratinocytes directly, however it inhibits the LDH of human skin keratinocytes; thus it is suggested that isocarbophos like most organophosphorus pesticides used widely can pass through the intact human skin and poison the body before the skin is damaged. Therefore, protection during work hours is of paramount importance.

[Rapid construction of directional cDNA library from human nasopharynx].

OBJECTIVE: To construct a directional cDNA library from human adult nasopharynx by SMART (switching mechanism at 5' end of RNA transcript) technique. METHODS: The total RNA was separated from human adult nasopharynx epithelial tissue and the first-strand cDNA was synthesized through reverse transcription by a modified oligo(dT) primer(contained sfi IB site) while the SMART oligonucleotide(contained sfi IA site) was utilized as a template so that the first-strand cDNA could be extended over the 5' end of mRNA. The double-strand cDNA was amplified by LD-PCR(long-distance PCR) with the above two primers and then digested by sfi I (IA & IB) restriction enzyme. After cDNA size fractionation through CHROMA SPIN column, the double-strand cDNA was ligated into the sfi I-digested lambda TripIEx2 vector and then the recombinant DNA was packaged in vitro. RESULTS: The unamplified human adult nasopharynx cDNA library consists of 1.5 x 10(6) independent clones in which the percentage of recombinant clones is about 100%. The titer of the amplified cDNA library is 3.8 x 10(9) pfu/ml and the average exogenous inserts of the recombinants is 1.5 kb. CONCLUSION: These results shows that the human adult nasopharynx cDNA library has an excellent quality and lays solid foundation for screening and cloning new tumor suppressor genes of nasopharyngeal carcinoma(NPC) and tissue-specific genes of human nasopharynx.

[Concentration of interleukin-5 and granulocyte-macrophage colony stimulation factor in nasal polyps tissue and its significance].

OBJECTIVE: To study the relationship between the concentration of interleukin-5 (IL-5) and granulocyte-macrophage colony-stimulation factor (GM-CSF) in nasal polyps and its significance. METHODS: The concentration of IL-5, GM-CSF was measured by enzyme linked immunosorbent assay (ELISA) in a total of 38 cases, and the number of eosinophils were counted in the same tissues. RESULTS: The statistical analysis showed that the concentrations of IL-5 and GM-CSF have significant difference (P < 0.05) between the nasal polyps and the control group. There is a positive correlation between the quality of IL-5/GM-CSF and the extent of eosinophils (r = 0.75/0.71). CONCLUSION: This study demonstrated that there are inductive compartments, namely the microenvironment, in which immune-effector and inflammatory cell function is modulated in nasal polyps. A special discussion has been made of eosinophils and IL-5/GM-CSF in the formation of nasal polyps, and the effect of the IL-5/GM-CSF in the accumulation of eosinophils.

Freshly isolated astrocyte (FIA) preparations: a useful single cell system for studying astrocyte properties.

Astrocytes are cell constituents of the mammalian CNS whose intricate relationships with neurons, blood vessels and meninges in situ are well documented. These relationships and their complex morphologies imply numerous functions. Over the past quarter century or so, however, the main experimental basis for determining which roles are likely have been derived from studies on primary astrocyte cultures, usually prepared from neonatal rodent brains. We list a number of examples where these cultures have shown quantitative and qualitative differences from the properties exhibited by astrocytes in situ. The absence of an adequate reliable database makes proposals of likely hypotheses of astrocyte function difficult to formulate. In this article we describe representative studies from our laboratory showing that freshly isolated astrocytes (FIAs), can be used to determine the properties of astrocytes that seem more in concordance with the properties exhibited in situ. Although the cells are most easily isolated from < or =15 day old rat hippocampi they can be isolated from up to 30 day old rats. The examples we describe are that several different types of K(+) currents can be determined by patch clamp electrophysiology, of all the mGluRs only mGluR3 and 5 were detected by single cell RT-PCR, and that single cell Ca(2+) imaging shows that the mGluR5 receptor is functional. It was found that the frequency of cells expressing mGluR5 declines with the age of the animal with the mGluR5b type splice variant replacing the mGluR5a type, as occurs in the intact brain. It is concluded that FIAs can be used to determine the individual characteristics of astrocytes and their properties without the problems of indirect effects inherent in a heterogeneous system such as the slice, and without the problem of cultures unpredictably reflecting the in situ state. The FIAs obviously cannot be used to study interactions of astrocytes with the other CNS components but we propose that they will provide a good database on which hypotheses regarding such interactions can be tested in slices. FIAs can also be isolated from brain slices or intact brain after various pharmacological or electrophysiological perturbations to determine the changes in astrocyte properties that correlate with the perturbations.

Magnet used for protein crystallization: novel attempts to improve the crystal quality.

The accuracy of the structures of biological macromolecules determined by X-ray crystallography is of fundamental importance, both for the understanding of life processes and for medical applications. The resolution of the structure is thus critical, and is largely determined by the quality of single crystals. Here we report the results of applying a magnetic field and a magnetization force during growth of the snake muscle fructose-1,6-bisphosphatase and human estrogenic 17beta-hydroxysteroid dehydrogenase crystals. For both enzyme proteins, the quality of the crystals improved with repeated assay, and their data sets were collected at significantly higher resolutions. These results coincide with a mechanism involving the reduction of convection, due to both the hydrodynamics within a magnet and the partially reduced gravity induced by a magnetization force. The density difference between the crystal and solution becomes less significant, and the sedimentation speed of the crystals is also reduced in the presence of the magnetization force.

Effect of Interfacial Alcohol Concentrations on Oil Solubilization by Sodium Dodecyl Sulfate Micelles.

Alcohol partitioning and its effect on oil solubilization in Winsor Type I microemulsion systems was investigated. The microemulsion systems consisted of sodium dodecyl sulfate (SDS), pentanol, isopropanol (IPA), and dodecane, with either deionized water or an aqueous solution of 50 mM CaCl(2). Alcohol partitioning between aqueous, oil, and interfacial phases of the microemulsion was described using a pseudophase model in which the alcohol was assumed to self-associate in the oil phase. Partitioning in these miroemulsions was consistent with pentanol self-association in the oil phase. IPA did not self-associate but co-associated with pentanol in the oil phase. IPA concentrations as high as 20 g/kg of water had no effect on pentanol partitioning. The partition coefficient for pentanol between aqueous and interfacial phases was about 220 on a mole fraction basis. However, pentanol saturated the interfacial phase at a mole ratio of 3 : 1 pentanol to SDS. Addition of pentanol beyond that sufficient to saturate the interface resulted in large amounts of pentanol partitioning into the oil, reaching concentrations in excess of 25 g dL(-1) of oil phase. Dodecane solubilization increased linearly with pentanol mole fraction in the interface up to the 3 : 1 pentanol-to-SDS saturation level. The fact that dodecane solubilization was unaffected by pentanol at concentrations beyond those necessary for interfacial saturation suggests that pentanol behaves as a cosurfactant and not a cosolvent in these microemulsion systems. Inhibition of insulin-like growth factor-I-mediated cell signaling by the von Hippel-Lindau gene product in renal cancer.

Insulin-like growth factor-I (IGF-I)-mediated signaling is thought to be involved in the regulation of multiple cellular functions in different tumors including renal cell carcinoma (RCC). Blocking IGF-I signaling by any of the several strategies abolishes or delays the progression of a variety of tumors in animal models. Herein, we demonstrate that in RCC cell lines, IGF-I-mediated signaling is found to be inhibited in the presence of wild type von Hippel-Lindau (VHL) tumor suppresser gene. Moreover, molecular modeling and biochemical approaches have revealed that beta-domain of the VHL gene product by interacting directly with protein kinase Cdelta inhibits its association with IGF-IR for downstream signaling. We also demonstrated that RCC has IGF-I-mediated invasive activity where protein kinase Cdelta is an important downstream molecule, and this invasiveness can be blocked by wild type VHL. These experiments thus elucidate a novel tumor suppresser function of VHL with its unique kinase inhibitory domain.

Structure of the cytoplasmic beta subunit-T1 assembly of voltage-dependent K+ channels.

The structure of the cytoplasmic assembly of voltage-dependent K+ channels was solved by x-ray crystallography at 2.1 angstrom resolution. The assembly includes the cytoplasmic (T1) domain of the integral membrane alpha subunit together with the oxidoreductase beta subunit in a fourfold symmetric T1(4)beta4 complex. An electrophysiological assay showed that this complex is oriented with four T1 domains facing the transmembrane pore and four beta subunits facing the cytoplasm. The transmembrane pore communicates with the cytoplasm through lateral, negatively charged openings above the T1(4)beta4 complex. The inactivation peptides of voltage-dependent K(+) channels reach their site of action by entering these openings.
