OBJECTIVE: To evaluate the effects of cellular immune function and cytokines on the pathogenesis of aplastic anemia (AA) and its clinical significance. METHODS: T lymphocyte subsets and HLA-DR antigen expression in the peripheral blood cells were assayed, and the levels of G-CSF, IL-6, TNF alpha, IFN alpha and IL-8 in the PBMNC culture supernatants were determined in 38 AA patients and 20 normal control with APAAP and ELISA methods. RESULTS: CD4+ cells, CD4+/CD8+ cells and G-CSF level were lower, and CD8+ cells, HLA-DR+ cells and IL-6, TNF alpha, IFN alpha and IL-8 levels were higher in AA patients than in normal controls. The level of G-CSF was positively correlated with CD4+ cells and CD4+/CD8+ cells and negatively with IFN alpha level. IL-6 level was negatively correlated with WBC count and CD4+ cells. TNF alpha level was positively correlated with CD8+ cells and negatively with CD4+/CD8+ cells. IL-8 level was positively correlated with CD8+ cells and HLA-DR+ cells. CONCLUSION: The cellular immune dysfunction and cytokine aberration participate in the pathogenesis of AA.
OBJECTIVE: To dissect the molecular mechanism of toxic neuropathy induced by allyl chloride (AC). METHODS: Fluorescence molecular probe (Fura-2/AM), electron probe X-ray microprobe analysis (EPMA) and biochemical methods were used to determine the concentrations of cytosolic free Ca2+, the contents of intracellular Ca2+ percentage, Ca(2+)-free calmodulin (CaM), the activity of Ca2+/CaM-dependent protein kinase II (Ca2+/CaM-PK II), and cytoskeletal protein synthesis in chicken embryo brain cells induced by AC. RESULTS: The contents of Ca2+ percentage, the concentrations of cytosolic free Ca2+, and the activities of Ca2+/CaM-PK II in the cells were increased significantly as AC was added (P < 0.01). However, the content of Ca(2+)-free CaM and the synthesis of cytoskeletal proteins were markedly decreased (P < 0.01). CONCLUSION: The results suggest that one of the mechanism of AC-induced cytoskeletal injury in vitro might be related to the elevation of intracellular Ca2+, activated CaM and Ca2+/CaM-PK II.
Thirty-two rabbits were used to investigate the effect of nitric oxide (NO) on the myoelectric activity of the sphincter of Oddi (SO) and arterial blood pressure. Intravenous infusion of NG-nitro-L-arginine (L-NNA), an inhibitor of nitric oxide synthase (NOS), caused increase in SO phasic wave amplitude and arterial blood pressure. The increase in SO phasic wave amplitude by infusion of L-NNA was reversed by L-arginine, but arterial blood pressure remained at a high level. It was observed that SO phasic wave amplitude, frequency and arterial blood pressure were significantly decreased by intravenously infusion of sodium nitroprusside (SNP), a substance producing NO. These observations suggest that (1) NO inhibits the resting activity of SO and modulates relaxation of vascular smooth muscle; (2) different tissues show different responses to L-NNA and L-arginine.
OBJECTIVE: The benefit of glucocorticoid for the treatment of acute optic neuritis remains controversial. The efficacy of oral prednisone in 12 patients (14 eyes) with acute optic neuritis was reviewed. METHODS: The time of visual symptoms before treatment was 3-15 days. The visual acuities were worse than 0.1 in all patients, with positive relative afferent pupillary defect. Visual field defect and delay latency of P100 in pattern VEP were also found. The regimen of glucocorticoid therapy was oral prednisone, starting with 160 mg daily followed by reducing the dosage by 20 mg every three days until 40 mg per day was attained and then tapering the dosage at 70 mg every other day until stoppage of the drug. The treatment was 3-11 months and 9 patients were followed up more than one year. RESULTS: The visual acuities improved rapidly and stable. After 4 days, the visual acuity was 0.2 or better in 10 eyes. It was better than 0.6 in 12 eyes at 15 days. At 6 months, all had the visual acuity better than 0.7, with 85.7% equal or better than 1.0. CONCLUSIONS: The regimen of oral prednisone beginning with 160 mg followed by tapering for three months would be feasible. Pattern VEP was a sensitive and credible sign for evaluating the extent of demyelination.
With the use of high performance capillary electrophoretic system, the authors developed a micellar electrokinetic chromatographic method with direct sample injection to separate and assay serum or bile fluid bilirubin subspecies. The four bilirubins, alpha, beta, gamma and delta subspecies, in serum or in bile fluid were successfully separated within 20 min. Under the concentration of combined bilirubin (bilirubin ditaurite), there was a good relationship between the concentration and the peak area. The detection limit reached 1.5 mumol/L. Being compared with the results of the first run with a concentration of 255 mumol/L and migration time of 16 min, the results of the 50th run were 238 mumol/L and 16.7 min respectively.

Two chiral drugs of EMD-56431 and chlorpheniramine are separated by cyclodextrin-modified micellar electrokinetic capillary chromatography using beta-cyclodextrin(beta-CD) and sodium deoxycholate(SDC) as selector in the paper. All the electrolytes in experiments contain 0.1 mol/L borate. The length of capillary is 45.0 cm as total and 30.0 cm as effective. The running and loading voltages are all 7.8 kV. The effects of pH and concentrations of SDC and beta-CD are studied, in which the best chiral separation conditions for EMD-56431 are pH 10.4, [beta-CD] = 50 mmol/L, [SDC] = 150 mmol/L, and those for chlorpheniramine are pH 9.0, [beta-CD] = 50 mmol/L, [SDC] = 100 mmol/L. The mechanism of chiral separation for the buffer system is initially believed as: the micellar monomer exists almost all as inclusion body with beta-CD, some CD-SDC inclusion complex may exist in micellar because the SDC micellar's gather number is only 4 and the SDC molecular is so big that it can only partly enter beta-CD. Then, the good separation ability of the system is supplied while the ratio of concentration between SDC and beta-CD is in 4:1-4:3; but there will be a optimized total concentration for SDC and beta-CD. The complex interaction among sample, SDC and beta-CD makes intricate change for migration along with the selector's concentration, and the same complex results are also made in pH experiments because of electroosmosis and the acidity of SDC and components. The phenomenon of increasing beta-CD solubility is showed. The beta-CD's solubility with 100 mmol/L SDC can be increased above 150 mmol/L.

OBJECTIVE: The purpose of this article is to improve the biological activity of the implant pretreated by use of cold plasma technic and to investigate the biological effects after implanted into bone. METHODS: First, HA granular was set in the electric field with high voltage and vacuum, then the gaseous mixture of ammonia, hydrogen and nitrogen was led to. By coupled capacity glow discharge, the plasma was produced and the pretreatment was completed. In this way, the surface of HA granular may obtain active amino structure. Sequently, the material was implanted into bone of the animal and the dynamic changes at the material/bone interface were histologically observed. RESULTS: The active group was definitely formed on the surface of the material after pretreatment and there were no adverse effects on the morphology and properties of the base material. Furthermore, the early formation of new bone at the interface of the material/bone was detected, and obvious effects on facilitating bone growth and osseointegration were obtained. CONCLUSION: The findings indicated that cold plasma pretreatment technic was an effective method to improve the biological activity of implant material.
OBJECTIVE: To detect tumor micrometastases in axillary lymph nodes of breast carcinoma so as to obtain more reliable information about nodes status and its clinical significance. METHOD: The expression of CK19 mRNA was measured in 61 axillary lymph nodes (LNs) of 15 breast carcinomas by using reverse transcriptase-polymerase chain reaction (RT-PCR). RESULT: CK19 mRNA expression was detected in all 15 breast carcinomas, but not in 5 control LNs. Of 61 axillary LNs analysed by HE staining and CK19 RT-PCR, 7 LNs were found to have metastases by histological staining and CK19 RT-PCR. Of the remaining 54 axillary nodes, 12 were found to have metastases by using CK19 RT-PCR method. CONCLUSION: The CK19 RT-PCR method is more sensitive than histological study for the detection of micrometastases in LNs (P < 0.01). This new method is of value in differentiating breast cancer patients at high risk for relapse from those with negative lymph node.
OBJECTIVE: To confirm the effect of auxiliary partial heterotopic liver transplantation (APHLT) in the treatment of fulminant hepatic failure (FHF). METHOD: On 30 rat models of FHF, 62% of partial liver grafts were implanted below the host residual liver with 28 successful operations. The survival rate, biochemical test for liver function, (99m)Tc-HIDA liver scintigraphy and pathological changes of the grafts and host livers were studied. RESULT: The survival rate in 48 hours was 71.4% compared with 0% in 15 FHF animals. 14 days after the operation, the host liver regenerated obviously and the liver function recovered while the graft shrank gradually. 30 days later, the graft became fibrosis completely. CONCLUSION: Depending on the temporary function support of the graft, the native residual liver could regenerate and restore its function while the graft tended to atrophy.
An HPLC method for the determination of lactitol in mice serum was developed. A liquid chromatograph equipped with a refractive index detector and a Waters Sugar-Pakl column was used. The column temperature was maintained at 90 degrees C and the mobile phase was redistilled water. One milliliter of the serum sample was treated with 3.0 mL methanol. The precipitate was centrifuged for 15 min at 10,000 r/min. Two milliliters of the supernatant was dried on a water bath at 100 degrees C. The residue was dissolved with 1.0 mL water and 10 microL of which was injected. The average recovery was 98.6%. The detection limit was 0.1 microgram (S/N = 3.75). The linear range was 0-600 mg/L (r = 0.9992). Both the RSD's within day and between days were below 1.0% (n = 5).
By low stringency PCR amplification of genomic DNA using the primers designed based on the conservation of zinc finger motif, we got 8 gradient eletrophoretic bands. After recovery of the second and third bands, the DNA fragments in them were cloned and sequenced. Compared to the GenBank database, among these 60 segments containing zinc finger motif, 23 segments were novel zinc finger genes' genomic segments. Then the human brain tissue cDNA library was screened, using these segments as probes, and 44 positive clones were obtained. Rescreening 28 of them, we got 20 rescreened clones. All of them were sequenced and sent to the GenBank DNA database for sequence analysis, the results showed that 16 were novel C2H2 type zinc finger protein cDNA segments. The cDNA segments encoding the novel C2H2 type zinc finger proteins provide the basic materials for cloning of full length cDNA of valuable novel zinc finger protein genes.
To study the mechanism of cinnarizine in preventing motion sickness, TXB2, 6-Keto-PGF1 alpha in rats' blood plasma and Na(+)-K(+)-ATPase activity in the endothelial cells of their cerebellar capillary were measured and analysed by a radioactive immunity analyser and a computer image system. The results showed that TXB2 and 6-Keto-PGF1 alpha in rats' blood plasma in the cinnarizine preventing group (CPG) decreased remarkably, compared with those in the motion sickness group(MSG) (p < 0.05). The activity of Na(+)-K(+)-ATPase in the endothelial cells of rats' cerebellar capillary in CPG was higher than that in MSG (p < 0.01). The authors suggest that the lower concentration of TXB2 and 6-Keto-PGF1 alpha in rats' blood plasma in CPG is closely related to cinnarizine which prevents Ca2+ from entering into the platelets and into the endothelial cells of blood vessels. The higher activity of Na(+)-K(+)-ATPase in the cerebellum may be caused by cinnarizene which dilates the blood vessels in the brain, increases the blood flow therein, and hinders Ca2+ from getting into the cerebellum cells. These change are believed to be the important mechanism of how cinnarizine prevents motion sickness.
To study the changes of inorganic and organic substance around cementum-dentin junction in the tissues of human natural tooth, the method of electron probe microanalysis was used in this research project. Determinations of weight percentage (wt%) were made for Ca, P and S elements around cementum-dentin junction. The results showed that the changes in quantities of Ca, P and S elements around cementum-dentin junction were not significant(P > 0.1). These suggest that the quantities of inorganic and organic substance have not obviously varied with the anatomic structure of natural tooth.
Hydroxyapatite was extensively used to repair osteoshortage recently. To improve its bondibility and X-ray obstructibility, we added ytterbium oxide of different proportions in the synthetic process of hydroxyapatite. Ytterbium oxide is characterized by high activity and X-ray obstructibility. A cheap biomaterial with good biocompatibility and high X-ray obstructibility has been got in this way.
