Cytotoxic effects of 24R,25-dihydroxycholecalciferol on the proliferation of various tumor cell lines in vitro and its antitumor effects in vivo.

24R,25(OH)2D3, one of the endogenous active metabolites of vitamin D3, showed suppressive effects on the proliferation of various tumor cells in vitro, and showed a suppressive effect on the growth of human osteosarcoma transplanted in nude mice. 24R,25(OH)2D3 increased bone strength lowered by the tumor burden which causes abnormal calcium metabolism and results in hypercalcemia.

Therapeutic benefits and safety of carvedilol in the treatment of renal hypertension. An open, short term study. Carvedilol Renal Hypertension Study Group in Japan.

Carvedilol, a new beta-blocker with vasodilating activity, was given orally to 9 hypertensive inpatients with impaired renal function in a dosage regimen of 5 to 20mg once daily to evaluate its clinical efficacy and safety. Treatment with carvedilol produced a significant decrease in blood pressure from 172/101 to 150/87mm Hg (p less than 0.01), but it did not cause orthostatic hypotension. Heart rate was decreased from 74.3 to 72.8 beats/min, but the decrease was not statistically significant. Serum creatinine and BUN levels were unchanged and other laboratory parameters were within normal limits. There were no side effects in any of the patients during the trial. These results suggest that carvedilol is a useful and safe drug for the treatment of renal hypertension.

Temporal bone findings in cloverleaf skull syndrome.

Cloverleaf skull syndrome was first reported in 1960 by Holtermüller & Wiedemann as Kleeblattschädel-Syndrome. The case in the present report was that of a male infant 26 days old at the time of death. Besides a trilobed skull and abnormalities of the extremities, he had low-set ears, stenosis of both external auditory canals, a beak-like nose, microphthalmia, retentio testis and congenital dermal sinus-like anomaly at the coccygeal region. A chromosome study revealed a normal male karyotype. The right temporal bone showed some anomalies through the external and middle ears. The bony portion of the external auditory canal was narrowed. The middle ear cavity was filled by mesenchymal tissue devoid of cell components. The incus was almost attached to the lateral tympanic wall. In addition, the stapes was thickened and bent anteriorly. The sensory and neutral elements of the inner ear were normal.

[Primary malignant fibrous histiocytoma of the gallbladder: a case report].

A 75-year-old female patient was admitted to our department because of fever and right hypochondraligia. A giant tumor of the gallbladder was confirmed by CT scanning, ultrasonography and PTC. She underwent complete removal of the main tumor and right hemicolectomy. The extirpated tumor was solid, but partially occupied by coagulated and necrotic tissue, 13 X 11 X 9cm in size, and 900g in weight. On the basis of the histologic features of the tumor, MFH (pleomorphic type) originated from the gallbladder was clearly diagnosed. Her postoperative course was uneventful, but she died of cachexia due to recurrence on 79 days after operation. Malignant fibrous histiocytoma (MFH) is a soft tissue sarcoma, usually developing in the extremities, less commonly in the retroperitoneal and the abdominal cavity. MFH originated from the gallbladder is very rare, and only 3 cases were reported in Japan so far. This is the fourth reported case in Japan.

[A modified Tanabe-Chiba medium for detection of Entamoeba histolytica].

Tanabe-Chiba's (T-C) medium, which has been routinely used for detection of Entamoeba histolytica in feces in Japan, was modified by adding some commercially available enrichments to the liquid part and by eliminating the agar slant. This is tentatively called M medium. M medium and T-C medium were compared for their efficiency for detecting E. histolytica in the feces of monkeys. No significant difference in the detection rate was found between the two media. However the rate of multiplication of the amoebae in M medium was significantly higher than that in T-C medium within the limited time of 24-48 h of cultivation.

Pathological basis for dementia in elderly patients with idiopathic Parkinson's disease.

We have examined the pathological basis for dementia in elderly patients with Lewy-type Parkinson's disease (PD). 37 (66.1%) of 56 examined cases (mean age of 77.9 years) had evidence of dementia. According to the distribution pattern of Lewy bodies and senile changes, as well as abiotrophic changes of the nucleus basalis of Meynert, nucleus paranigralis and locus ceruleus, demented PD could be subdivided into three groups: (1) demented PD without cerebral alzheimerization (21.6%, 8 cases); (2) demented PD with cerebral alzheimerization (64.9%, 24 cases), and (3) demented PD with combined senile-vascular changes (13.5%). Based on neuropathological findings, damage not to a single, but to multiple neuronal networks including the innominatocortical cholinergic, ceruleocortical noradrenergic as well as mesocortical dopaminergic systems could play a role in the development of dementia in PD. We also discussed the nosological situation of 'diffuse Lewy body disease', regarding it as one distinct disease entity.

Noradrenaline induces rhythmic bursting in sympathetic preganglionic neurons.

In the in vitro slice preparation of upper thoracic cord of the cat, noradrenaline, at concentrations of 10-50 microM, induced rhythmic bursting in 30% of sympathetic preganglionic neurons of the intermediolateral nucleus. The bursting frequencies were between 0.2 and 1.0 Hz at membrane potentials between -45 and -65 mV. The bursting rhythm could be reset by short intracellular current pulses. In the presence of tetrodotoxin noradrenaline produced a rhythmic oscillation in membrane potential in the same frequency range as the bursting. The frequency of oscillation was voltage dependent. Neuronal input resistance decreased at the oscillation peak and the oscillation was abolished by cobalt or low calcium.
