Suppression by coffee cherry of the growth of spontaneous mammary tumours in SHN mice.

We previously found that coffee cherry (CC), residue after removal of coffee beans, significantly suppressed the development of spontaneous mammary tumours of mice. In this paper, the effects of CC on the growth of the palpable size of this type of tumour was examined. Free access as drinking water of 0.5% solution of the hot water extract of CC for 10 days resulted in a marked inhibition of the tumour growth: The percent changes of tumour sizes were 53.8 +/- 11.7% and 13.8 +/- 10.9% in the control and the experimental groups, respectively. Associated with this, thymidylate synthetase activity in the mammary tumours was significantly lower in the experimental group than in the control. Normal and preneoplastic mammary gland growth, body weight change and weights and structures of endocrine organs were only slightly affected by the treatment. The findings indicate that CC is promising as an antitumour agent.

Effects of coffee cherry on the immune system in SHN mice.

Previously, we found that chronic ingestion of the extract of coffee cherry (CC), the residue after the removal of coffee beans, induced a marked suppression of the development and the growth of spontaneous mammary tumours in a high mammary tumour strain of SHN mice. As a possible step to clarify the mechanism of this effect, the immunomodulating role of CC was examined in this study. CC treatment resulted in significant weight gain in the spleen. CC treated experimental mice showed a significant decrease in the proportion of immature (CD4+8+) thymocytes and an increase in the percentages of mature cells expressing helper/inducer (CD4+8-) or cytotoxic/suppressor (CD4-8+) phenotypes. The proportion of T cells expressing CD25, a lymphocyte activation marker, in the spleen and peripheral blood tended to increase in the CC treated group. The natural killer activity of the spleen cells was not affected by CC ingestion. These results have revealed that CC can enhance the differentiation of thymocytes and the activation of peripheral T lymphocytes.

Hyperthermia combined with chemotherapy and irradiation for the prolongation of the postoperative prognosis in the patients with esophageal carcinoma invading neighbouring structures.

Preoperative hyperthermo-chemo-radiotherapy (HCR) has been performed on patients with esophageal carcinoma, especially in advanced cases with invasion to the neighbouring structures (a3 carcinoma), and is considered to have contributed to the favorable postoperative prognosis. 163 patients with operated locally advanced esophageal carcinoma with invasion to the neighbouring structures (a3 carcinoma) were analyzed. 40 cases were treated with preoperative HCR (HCR group), while 85 cases were treated with other preoperative adjuvant therapies including 35 with chemo-radiotherapy (CR group) and 42 with radiotherapy (R group), and the remaining 38 were operated on without any preoperative treatments. The effectiveness of the therapies and the prognosis were investigated. The survival rates of the HCR group were significantly better than those of the CR group (p < 0.05). The prognosis of cases with Grade 3 effectiveness was significantly better than those of others, moreover, in 5 cases with Grade 3 effectiveness in the HCR group, there was no case of early recurrence or metastasis after resection, while in addition, three patients have since survived for 4, 5, and 7 years, respectively. Our data showed that for the patients with locally advanced esophageal carcinoma with invasion to the neighbouring structures (a3 carcinoma), hyperthermia combined with chemotherapy and irradiation has demonstrated a favorable influence on the prognosis.

Further study on the effects of coffee cherry on spontaneous mammary tumourigenesis in mice: effects of methanol extract.

To examine the effective components in the antitumourigenic role of water extract of coffee cherry (CC), and the residue after removal of beans, the dry matter of coffee cherry was extracted repeatedly by 60% methanol and further divided into water-soluble and -insoluble fractions (CC-W and CC-M, respectively). Free access to CC-W (0.25% in tap water) markedly inhibited mammary tumour development in SHN virgin mice, while it had no effect on the growth of normal and preneoplastic mammary glands and established tumours. Associated with this, urinary excretion of some components and intrathymic T cell differentiation were stimulated by the treatment with the CC. CC-W had little affect on the endocrine system, including serum prolactin level, oestrous cycles and endocrine organs, but retarded ovarian degeneration in mice developing mammary tumours. The finding in CC-W were generally similar to those in CC. Fat emulsion of CC-M, which was only checked for mammary tumour growth owing to the limited amount available, showed no effect at the dose level used (0.25% in drinking water). Finally, the previous and the present findings taken together strongly suggest that the effective components of coffee cherry are in the water-soluble fraction.

Renal pelvic and ureteral carcinoma with huge hydronephrosis: US, CT, and MR findings.

A 78-year-old man presented huge hydronephrosis with a calculus in the left kidney, and underwent left nephroureterectomy under the diagnosis of renal pelvic and ureteral tumor. Histological examination disclosed renal pelvic and ureteral tumors featuring mixed histologic types of carcinoma. Magnetic resonance (MR) imaging was superior to computed tomography (CT) in its complete, detailed demonstration of the renal pelvic lesions. The imaging characteristics of CT, MR imaging, ultrasonography, and retrograde ureterography were discussed in the report.

Effects of coffee cherry on the activation of splenic lymphocytes in mice.

We previously observed that the ingestion by mice of a hot water extract (CC) and the methanol-extracted and water-soluble fraction (CC-W) of coffee cherry, the residue remaining after the removal of coffee beans from the fruit, enhanced the differentiation of thymocytes and the activation of peripheral T-lymphocytes; and the anti-mammary tumour effects of coffee cherry extract was considered to be associated with this immunomodulation. To study further these effects, mitogen response and some immune parameters were examined in a high mammary tumour strain of SHN mice. While the T-lymphocyte response to concanavalin A was not significantly changed by either CC or CC-W, the lipopolysaccharide response was significantly enhanced by both treatments. The proportion of CD45R/B220+ (B) cells in the splenic lymphocytes was significantly increased by CC, and the percentage of activated (CD25+) cells in B-lymphocytes was increased by CC and CC-W. These data indicate that coffee cherry extract can enhance B-lymphocyte response, and suggest that this immunopotentiation contributes to the antitumourigenic role of coffee cherry.

Comparison of urinary component levels in 4 strains of mice with different physiological characteristics.

Changes in urinary component levels before, during and after reproduction were examined with 1H-nuclear magnetic resonance (NMR) in four strains of female mice with different mammary tumour potential and related characteristics (SHN, SLN, GR/A and C3H/He). Regardless of the variations in these changes among the strains, the urinary component levels reflected physiological changes during reproduction and some strain-specific characteristics. The urinary excretion of citrate, 2-oxoglutarate and lactate increased from the virginal stage to pregnancy and declined during lactation in all strains, indicating a marked change in sugar metabolism during reproduction. SHN and SLN, which are from the same basal stock, showed a similar pattern and the level, of several components were lower than those of GR/A and C3H/He. The levels of some lower components were compensated for by higher urine excretion. The levels of taurine and betaine, both essential for fetal and pup growth, increased at lactation in only GR/A, reflecting the poorer pup growth observed in this strain. Finally, the 1H-NMR method was useful to estimate not only the metabolic but also the physiological changes of the animals without invasion, pain or distress and would thus contribute to animal welfare in the animal experiments.

Effects of taurine on reproduction and behaviour in GR/A mice.

The characteristics of GR/A mice are rather poor pup growth and vigorous activity associated with increased excretion of taurine during pregnancy and lactation. The relationship between taurine and these characteristics were studied. The group of GR/A mice given a 1% taurine solution in their drinking water on the first day of pregnancy (day when the vaginal plug was found) (group I) or on the day of parturition (group II) differed little from the control group given tap water in all reproductive parameters, including pup growth. The spontaneous motor activities of both the female and male 4-month-old offspring of group I were markedly lower than those of the offspring of the control and group II, and than that of group III, which was given taurine after of weaning. These findings strongly suggest that taurine, possibly by acting on the developing brain at the fetal stage, has a long-term effect on the behaviour of this strain of mice, while it has little effect on their reproduction.
