A radiological population study on the ossification of the posterior longitudinal ligament in the spine.

This paper reports the results of a radiological population study on the ossification of the posterior longitudinal ligament (OPLL) in both the cervical and the thoracic spine among Japanese. The study was carried out in the Yachiho-mura district in the central part of Japan, where 5074 people were living. X-ray examinations were made of 1058 of the people; there were 440 men and 618 women, 50 or more years of age. The roentgenograms showed 34 cases of OPLL in the cervical spine (3.2%): 19 men (4.3%) and 15 women (2.4%). The condition was most frequently observed at the level of C-4. Radiological classification showed 18 cases of the segmental type, 11 of the continuous type, and five of a mixed type. There were eight cases of OPLL in the thoracic spine (0.8%), four in men (0.9%) and four in women (0.6%). OPLL in the thoracic spine was most frequently observed at the midthoracic levels. All eight cases showed a continuous type of ossification. There were three subjects with OPLL in both the cervical and the thoracic spine. Therefore, the number of subjects with OPLL in either the cervical or the thoracic spine was 39 (3.7%) total.

Extra- and intra-cellular digestion of elastic fibers by macrophages in annular elastolytic giant cell granuloma. An ultrastructural study.

A case of annular elastolytic giant cell granuloma was studied with the electron microscope using acid orcein stain. Elastic fibers were digested by multinucleate giant cells (MGCs) in 2 steps: the first step was the extracellular digestion and the second was the intracellular digestion. In the first step, the orcein-reactive amorphous materials of the elastic fibers surrounded by MGCs at first disappeared, and then the fine filaments appeared in elastic fibers. This process advanced in extracellular spaces of MGCs. In the next step, the fine filaments were phagocytosed by MGCs and were digested in phagocytic vacuoles.

Thalamic posterior ventral neurons with bifurcating axons to the first and second somatosensory areas in the cat, demonstrated by the fluorescent retrograde double labeling technique.

The thalamic posterior ventral neurons with bifurcating axons to both the first and second somatosensory cortical areas (SI and SII) in the cat were examined by the fluorescent retrograde double labeling technique. After injection of Evans blue (EB) into the SI, and of 4',6-diamidino-2-phenylindol.2HCl (DAPI) into the SII of the same hemisphere, EB- and DAPI-labeled cells were observed predominantly in both the posterolateral ventral and the posteromedial ventral nuclei of the thalamus. Although EB single-labeled and DAPI single-labeled cells tended to occupy separate regions within the posterior ventral nuclei, a small number of cells double-labeled with both EB and DAPI were detected in the border zone between two single-labeled cell groups. These observations indicate that some cells in the posteromedial and posterolateral ventral nuclei project both to the SI and SII by bifurcating axons.

[An epidemiological study on the ossification of the anterior longitudinal ligament of the spine].

An epidemiological study was carried out in connection with the multiphasic screening examination of 1,057 people (442 men and 615 women) in the village of Yachiho. The purpose of this study was to define the etiology of OALL (ossification of the anterior longitudinal ligament) and disc narrowing. The prevalence of disc narrowing increased with age, but OALL was not correlated with aging. OALL was found more frequently in men, but disc narrowing showed no difference between each sexes. The body height and weight-height index were higher in the OALL cases. The body height shrinkage by aging was greater in the patients with disc narrowing cases. The index of thoracic kyphosis was higher in the OALL cases than in the patients with disc narrowing ones. OALL was associated with ossification of the other ligaments of the spine, but disc narrowing was not. This study suggests that despite some similarities, OALL is etiologically different from disc narrowing.

An epidemiological survey on ossification of ligaments in the cervical and thoracic spine in individuals over 50 years of age.

An epidemiological survey on ossification of the spinal ligaments was performed on a total of 1,058 subjects over the age of 50 years by means of roentgenography of the cervical and thoracic spine. Ossification of the posterior longitudinal ligament (OPLL) of the cervical spine was detected in 34 subjects (3.2%) with a predilection for men, whereas OPLL in the thoracic spine was found in 8 (0.8%). There were 325 cases (30.7%) of ossification of the anterior longitudinal ligament (OALL) of stage II or above by Forestier's classification in the region from the cervical to thoracic vertebrae, and these cases included a significantly greater number of men. Ossification of the ligamenta flava (OLF) was observed in 48 cases (4.5%). As for the coexistence of ossification of these ligaments, 364 individuals (34.4%) had at least one instance of OPLL and OALL (stage II or above) in the region from the cervical to thoracic spine, and OLF in the thoracic spine.

Pharmacological investigations of the new antiinflammatory agent 2-(10,11-dihydro-10-oxodibenzo[b,f]thiepin-2-yl]propionic acid. 4th communication: inhibitory effects on rat adjuvant arthritis.

The preventive and therapeutic effects of 2-(10,11-dihydro-10-oxodibenzo [b,f]thiepin-2-yl)propionic acid (CN-100) on local and systemic changes of rats with adjuvant arthritis being used frequently as experimental model of rheumatoid arthritis were investigated in comparison with those of reference drugs, indometacin and pranoprofen. Preventively and therapeutically CN-100 showed potent inhibitory effects on adjuvant primary inflammation and secondary lesion. CN-100 also exerted an evident preventive effect on destruction of foot bone, improved the changes in organ weight, and stimulated weight gain. These effects were dose-dependent, and the effects at 5.0 mg/kg were almost the same as those of indometacin and pranoprofen at 1.25 and 2.5 mg/kg, respectively. The mode of action of CN-100 resembled that of reference compounds. Although CN-100 improved the change in albumin/globulin ratio, which was a parameter of systemic inflammatory reactions, the effect was more remarkable in therapeutic administration than in preventive one. This suggests that CN-100 is suitable for clinical application.

Pharmacological investigations of the new antiinflammatory agent 2-(10,11-dihydro-10-oxodibenzo[b,f]thiepin-2-yl)propionic acid. 5th communication: antiplatelet effect of the drug and antiinflammatory effect of its main metabolite.

Since nonsteroidal antiinflammatory drugs (NSAID) usually have an antiplatelet effect, the inhibitory effect of 2-(10,11-dihydro-10-oxodibenzo[b,f]thiepin-2-yl) propionic acid (CN-100), which exerts a potent antiinflammatory effect, was compared with those of reference drugs, indometacin and pranoprofen, in this study. Indometacin at 10(-5) mol/l inhibited completely (100%) rat and rabbit platelet aggregation induced by collagen and arachidonic acid. Pranoprofen at 10(-5) mol/l also entirely inhibited rat platelet aggregation induced by the two aggregators, but an about 10 times higher concentration was required to produce 100% inhibition of rabbit platelet aggregation. CN-100 at 10(-5) mol/l exerted 100% inhibition of rat platelet aggregation induced by collagen, whereas more than 10(-4) mol/l was needed to exhibit 100% inhibition of aggregation induced by arachidonic acid and ADP. The inhibitory activity of CN-100 on aggregation of rat platelets ex vivo was weaker than those of reference NSAID, i.e., the antiplatelet effect of CN-100 was found to be weak. The main metabolite of CN-100 also had a weak antiplatelet effect, and its antiinflammatory effect on carrageenin edema and UV erythema was apparently weaker than that of CN-100. The inhibitory effect of the metabolite on endotoxin diarrhea was weak. The ulcerogenic effect of the metabolite on gastric mucosa was similar to that of CN-100, but the effect rarely seemed to be a clinical problem because it was basically weak.

Thalamic projections to the hippocampal formation in the cat.

The thalamo-hippocampal projections in the cat were studied by anterograde and retrograde tracing methods. Injections of horseradish peroxidase (HRP) into the hippocampus and dentate gyrus led to retrograde labeling of cells in the nucleus reuniens of the ipsilateral thalamus throughout almost its entire anteroposterior extent. The labeled cells were found mostly in the parvocellular ventral part, but a few cells were labeled in the dorsal part at anterior levels. Anterograde transport following injections of wheat germ agglutinin conjugated to HRP into the nucleus reuniens resulted in terminal labeling in the subicular area, the neighboring parts of the hippocampus (CA1) and the border zone of the dentate gyrus with the hippocampus.

Intimate structural association of amyloid and elastic fibers in systemic and cutaneous amyloidoses.

Elastic fibers were found in the amyloid islands of primary systemic amyloidosis, heredo-familial amyloid polyneuropathy, macular amyloidosis and amyloidosis cutis nodularis atrophicans. In macular amyloidosis, the elaunin fibers in amyloid islands were loosely arranged, waved and twisted. In systemic amyloidosis and amyloidosis cutis nodularis atrophicans, amyloid fibers did not deposit in homogeneous electron-lucent material of elastic fiber, but were located close to the elastic fiber where the peripheral microfibrils were present.

The organization of thalamic neurons projecting to the premotor cortex and the caudate nucleus in the cat studied by a fluorescent retrograde double labeling technique.

Thalamic neurons projecting to both the head of the caudate nucleus and the premotor cortex in the cat were studied by the retrograde fluorescent double labeling technique. After injections of Evans blue into the caudate nucleus, and diamidino-phenylindol into the premotor cortex, a small number of double labeled neurons appeared in the ventral anterior, ventral lateral, anteromedial, rhomboid, central dorsal, central lateral, central medial, paracentral and parafascicular nuclei, in addition to numerous single-labeled neurons. This indicates that some neurons in the thalamic nuclei send bifurcating axons to both the head of the caudate nucleus and the premotor cortex. The caudatal projections of these thalamic neurons are organized in a topical manner.

Cephem antibiotics and alcohol metabolism.

Mechanisms of the disulfiram-like reaction of cephem antibiotics were studied. Changes in ethanol (EtOH) and acetaldehyde (AcH) levels in the blood with EtOH loading following daily intravenous administration of cephem antibiotics were determined in rats and the following were found: The daily intravenous injection of cefazolin, cefotiam (CTM), cefsulodin, cefoxitin or ceftizoxime in no way varied the changes in the EtOH and AcH levels in the blood with EtOH loading. The daily intravenous injection of cefmetazole, cefoperazone, cefamandole, latamoxef, cefmenoxime or cefotetan caused the AcH level in the blood to be elevated significantly until at least 8 hours after the EtOH loading, but was inert on the EtOH level on the blood. The daily administration of 1-methyl-2-tetrazoline-5-thione (TZ), a compound having a partial structure similar to those of the cephem antibiotics elevating the AcH level in the blood on EtOH loading, was inert on the EtOH level in the blood but elevated the AcH level in the blood. The daily administration of 1-(2-dimethylaminoethyl)-2-tetrazoline-5-thione (MTZ), a compound having a partial structure similar to that of CTM, was inert either on the EtOH or AcH level in the blood. The cephem antibiotics elevating the AcH level in the blood all had a (1-methyl-1H-tetrazol-5-yl) thiomethyl group in the 3 position of the aminocephalosporanic acid nucleus. It was though that the disulfiram-like reaction caused by the cephem antibiotics was derived from the elevation of AcH level in the blood.(ABSTRACT TRUNCATED AT 250 WORDS)

Staining of amyloid with cotton dyes.

We have utilized three cotton dyes, Pagoda Red, RIT Scarlet No. 5, and RIT Cardinal Red No. 9, for the microscopic demonstration of amyloid in cutaneous histopathology. All three dyes stained amyloid specifically, yielding a bright orange color. The preparation of staining solutions and the staining procedure are simple, practical, and can be applied routinely in a dermatopathology laboratory.

The laminar arrangement of limbic thalamocortical neurons in the lateropulvinar nuclei of the cat thalamus.

The topical organization of limbic cortical projections of the lateropulvinar nuclei of the thalamus in the cat was studied with the horseradish peroxidase (HRP) technique. The dorsal margins of the dorsal lateral, medial pulvinar and lateral pulvinar nuclei project to the postsubicular and presubicular areas (presubiculum in a wide sense), the most dorsal parts of these nuclei projecting to the retrosplenial area, and the dorsal parts to the cingular area. These three zones of limbic thalamocortical neurons in the lateropulvinar nuclei are arranged in lamination from the surface inward, and may be called presubicular, retrosplenial and cingular zones.
