Induction and degradation of copper-induced metallothioneins in rat liver as studied at isometallothionein levels.

Cupric chloride was injected i.p. into female rats in an endeavour to clarify the detailed induction and degradation pathways of hepatic copperbinding protein at isoprotein levels. Injected copper (Cu) in serum decreased to a control level within 6 h, the depletion rate being an intermediate between cadmium (Cd) (within 1 h) and zinc (Zn) (within 1 day). Cu in the liver increased with time after injection and was found mainly in the metallothionein fraction: here it was separated mainly into two isoproteins which corresponded to the two isometallothioneins and were accompainied by Zn. The relative peak height of the two isoproteins changed with time. The overall changes were of an intermediate pattern for those of Cd and Zn loadings.

Rat kidney metallothionein induced by injection of Cd-thionein: Changes of chromatographic properties with time and their relation to copper content and kidney dysfunction.

Cadmium (Cd)-thionein was injected i.p. into adult rats to try to establish the relation between copper (Cu) content in the kidneys and its effect on the properties of metallothionein induced in the kidneys. Elution profiles of the re-synthesized kidney metallothionein changed with time after injection both on a Sephadex and on a gel permeation column (TSK GEL SW3000). The changes were correlated with Cu content in the kidneys caused by kidney dysfunction upon injection of Cd-thionein.

Relationship between HLA-D and in vitro and in vivo responsiveness to Candida allergen.

In vitro reaction to Candida allergen was studied in 100 normal healthy Japanese and related to HLA. A significant association was found between the low responder group (< 7,000 c.p.m.) and HLA-B15 and the high responder group (> 7,000 c.p.m.) and HLA-B7, as well as Dwl. Four HLA-D homozygote cells of types HLA-Dwl and DHO, were tested; these fitted in the high responder group. HLA-DYT and DEn fitted into the low responder group. Responsiveness to Candida allergen corresponded to skin test (r = 0.884, P < 0.01). From these results, in vitro lymphocyte reaction to Candida allergen and skin tests showed close correlation and an association with HLA antigens, HLA-D in particular.

A radiometric assay for the combined measurement of phagocytosis and intracellular killing of Candida albicans.

A radiometric assay for combined measurement of phagocytosis and intracellular killing of Candida albicans by human polymorphonuclear leucocytes (PMNs) is presented. The assay, based upon the incorporation of 3H-uridine into the micro-organisms, makes it possible to measure phagocytosis and intracellular killing simultaneously but independently in a single sample. Thus it is possible to determine in a single assay whether increased survival of the micro-organism is due to reduced ingestion or reduced ability of the PMN to kill. The assay is objective, quantitative and convenient for clinical application. It is also suitable for analysing the effects of various agents, including serum factors and drugs, on PMN function.

Gel and anion exchange chromatographic properties of copper-containing metallothioneins.

Chromatographic properties of copper-binding proteins (copper-containing metallothioneins) induced in rat liver and kidney by the injection of either copper in the liver and kidney or cadmium in the kidney were compared with those of metallothioneins, with low or no copper content, induced by the injection of cadmium or zinc in the liver both on a Sephadex G-75 column and on a DEAE Sephadex A-25 column. The copper-containing metallothioneins were eluted at a slightly slower rate on a Sephadex G-75 column and at a higher buffer concentration on an anion exchange column than the metallothioneins with low or no copper content regardless of the inducing metals, copper or cadmium. The different chromatographic properties of the metallothioneins with high and low copper contents may be explained by the more compact conformation of the proteins with high copper contents than those with low or no copper contents and by the changes of electrostatic charge with coordination by copper.

Inhibition of 7,12-dimethylbenz[alpha]anthracene-induced mammary tumorigenesis in rats by a synthetic protease inhibitor, N,N-dimethylamino-(p-(p'-guanidinobenzoyloxy)) benzilcarbonyloxyglycolate.

Oral administration of N,N-dimethylamino-(p(p'-guanidinobenzoyloxy))-benzilcarbonyloxy glycolate depressed induction of breast tumors in rats by a single intravenous injection of 7,12-dimethylbenz[alpha]anthracene. The latent period was delayed in the group fed on a diet supplemented with N,N-dimethylamino-(p(p'-guanidinobenzoyloxy))benzilcarbonyloxy glycolate. Administration of this compound for 40 days after 7,12-dimethylbenz[alpha]anthracene also significantly reduced the average weight of tumors per rat. These findings suggest that protease may play an important role in the process of mammary tumorigenesis.
