[Two aspects of masculinity-femininity. II].

This study was designed to explore two aspects (mother-father; woman-man) of Masculinity-Femininity in personality. Sixty-five middle school students, 150 high school students and 219 university students were asked to rate the desirability for them, their fathers and mothers, and society of each of 45 items comprising a questionnaire which described valid characteristics of these two aspects. Analysis of the results revealed that male subjects in all age groups seemed to feel that their fathers desired them to be more masculine (i.e., father and man) than they, themselves, desired. Those of high school and university age also seemed to feel that their mothers and fathers and society desired more motherliness of them than they, themselves, desired. Female high school and university subjects seemed to feel that their fathers and mothers desired much more femininity (mother and woman) and much less masculinity (father and man) than they, themselves, desired.

Low responsiveness to the anti Leu 4 antibody by T cells from patients with active systemic lupus erythematosus.

It has been widely accepted that murine monoclonal antibodies against the CD 3 antigen have mitogenic effects on human T cells which mimic antigen binding via the T cell receptors. We report that T cells from patients with active systemic lupus erythematosus cannot respond to anti Leu 4 antibody, an anti CD 3 murine monoclonal antibody of the subclass IgG1. It was observed that T cells from patients with the active disease expressed as much of the Leu 4 and OKT 3 epitopes as compared to high responders, and sera from active patients could not change the expression of the Leu 4 epitope. Similarly, the low responsiveness could not be attributed to suppressor T cells, poor production of IL 2 or less expression of the IL 2 receptor. The accessory nonrosetting mononuclear leucocytes from high responders could convert the low responsiveness of these patients to high, whereas the accessory cells from active patients failed to help the responses of T cells from high responders. Likewise, exogenous IL 1 and anti Leu 4 antibody bound to Sepharose beads independently or cooperatively failed to propagate T cells from low responders. We suggest that the low response to anti Leu 4 antibody as demonstrated in the T cells of patients with active SLE may be due in part to the impairment of the secondary signals, distinct from IL 1, between helping accessory cells and responding T cells, or to the dysfunction of intracellular transmission of stimulation, and may play significant roles in the pathogenesis of SLE.

Immunoelectron microscopy of Proteus vulgaris by the plasma polymerization metal-extraction replica method: differential staining of flagellar (H) and somatic (O) antigens by colloidal golds.

Flagellar (H) and somatic (O) antigens of Proteus vulgaris were differentially stained with antibodies coupled to different sizes of colloidal gold particles, and the distribution of these antigens was visualized by the plasma polymerization metal-extraction replica (PMR) method. The H antigen, labeled with 5 nm colloidal gold, was almost exclusively located on the flagella, whereas the O antigen, labeled with 10 nm colloidal gold, was almost exclusively located on the bacterial body. The marker gold particles were clearly observed as electron-dense particles on the relatively low contrast background of three-dimensional replica image of the flagellated bacteria. Thus, the PMR method may prove to be a useful tool for studying the localization of multiple substances on the cell surface, at a high resolution and in three dimensions. The diameter of the flagella measured by the replica method was about 15 nm, close to the value obtained by negative staining (16 nm). When treated with anti-flagellar (H) factor serum and protein A-gold, the diameter of flagella was significantly increased to about 35 nm. This increase in diameter was presumably caused by binding of immunoglobulins to H antigens of flagella.

[Investigated reports of tooth fractures cases from November, 1986 to September, 1988].

Researching the actual condition of tooth fractures, clinical records and X-ray photographs of 2717 patients, visited P.D.I. Dental Clinic from November, 1986 to September, 1988 were investigated statistically. 174 cases were tooth fractures in 2717 patients. The frequency rate of tooth fractures at an early age was low, increased from the latter half of the 30s and was highest at the latter half of the 50s accounted for over 10%, and decreased from the 60s. In the upper anterior teeth, the crown fractures caused by traumatic factors, at an early age showed a high proportion, and in the lower anterior the tooth fractures were rare. In the posterior the root fractures of non-vital teeth attributed to dental treatment at the middle and advanced age indicated a high proportion. In the case of tooth restored with metalcore and bridge, the proportion of the root fractures were high, the metalcore, was used in 63%, and in the bridge in 27% of the root fractures.

[Comparisons of bioavailability of various calcium salts. Utilization incisor dentin in parathyroidectomized rats].

In the present study, we compared the bioavailability of various calcium salts in the diet such as calcium lactate, calcium carbonate and calcium phosphate by examining the effects of these calcium salts on serum calcium concentration and on the mineralization of incisor dentin in parathyroidectomized (PTXed) rats. Each rat was given daily 12 g of a low calcium (0.1% Ca) diet beginning from 3 days before the operation and continuing for 11 days after. Then the diet was changed to those containing various calcium salts (1.5% Ca). The serum calcium and phosphorous concentrations were examined every three days, just before feeding time and 6 hours after. The degree and site of the mineralization of incisor dentin were examined histologically by a time marking method using lead acetate. In the PTXed rats maintained on a low calcium diet, the serum calcium concentration decreased to about 4.5 mg/dl and the mineralization of incisor dentin was inhibited. On the 11th day, just before feeding on diets containing various calcium salts, the serum calcium concentrations were very similar for each group, but at 6 hours after feeding, concentration was 7.6 mg/dl in the calcium lactate group, 6.2 mg/dl in the calcium carbonate group and 4.8 mg/dl in the calcium phosphate group. On the 17th day following administration of the high calcium diets, the serum calcium concentrations were 9.8 mg/dl in the calcium lactate group, 8.4 mg/dl in the calcium carbonate group and 4.4 mg/dl in the calcium phosphate group. Mineralization of the incisor dentin was best in the calcium lactate group, moderate in the calcium carbonate group and poor in the calcium phosphate group. We also examined the effects of dietary phosphorus contents on serum calcium concentrations and on the mineralization of incisor dentin using PTXed rats. On the 17th day following the administration of diets (1.5% Ca, with calcium lactate) containing various amounts of phosphorus, the serum calcium concentrations in the calcium lactate group were 8.8 mg/dl (0.4% P), 6.9 mg/dl (0.8% P) and 4.2 mg/dl (1.6% P) respectively. Mineralization of incisor dentin was also inhibited in rats fed a high phosphorus diet. These results suggest that in PTXed rats, absorption of calcium from the intestines and mineralization of the incisor dentin is best by the administration of calcium lactate, moderate by calcium carbonate and poor by calcium phosphate, and that phosphorus in the diet inhibits calcium absorption from the intestines.

[von Recklinghausen's disease associated with malignant lymphoma].

A 59-year-old man was admitted to the Saga Medical School Hospital in November 1986 because of unconsciousness and lymph node swelling. He had café au leit spots and multiple neurofibromas since his thirties. The leukocyte count was increased. The level of serum LDH and Ca were extremely elevated. His bone marrow aspiration showed the invasion of lymphoma cells. Lymph node biopsy from his left neck and the study of surface markers were performed. And then, his illness was diagnosed as non-Hodgkin's lymphoma (diffuse, medium sized cell type, B cell type, stage IV B.) following von Recklinghausen's disease. After the admission, his general condition improved by chemotherapy, but he had a relapse 6 months later. And he died in November 1987. The case of von Recklinghausen's disease associated with malignant lymphoma in rarely reported. We reviewed the literature and discussed von Recklinghausen's disease associated with malignant lymphoma including this case.

Small-square (SS) net exists in trout muscle.

The small-square-net form (with the large-square-net form) was clearly observed in brown trout muscle in cross-square-net-section. Fresh adductor mandibularis and dorsal muscle strips, isometrically clamped to prevent contraction, were prefixed in 3% glutaraldehyde, 0.1 M cacodylate, pH 7.4, and post-fixed in 1% osmium tetroxide in veronal acetate buffer at 2 degrees C. The samples were dehydrated, embedded in epon-araldite, cross-sectioned, and stained with uranyl acetate and lead citrate for electron microscopic observation. The sections provided the first clear evidence of the small-square-net form, coexisting with the basket-weave pattern, in fish muscle. In previous experiments, we could not demonstrate the small-square-net pattern convincingly with guppy tissue (Yamaguchi et al., 1985a) which may be more susceptible to chemical or physical stress than is trout tissue. The existence of the diverse structural patterns in cross-sections of some, if not all, vertebrate striated muscle must be derived from a basic unit which although it may vary in size among species is structurally identical. The findings agreed with our unified model of narrow and wide Z-line structure (Yamaguchi et al., 1980; Yamaguchi et al., 1985a; Yamaguchi et al., 1985b).

A new analysis of natural killing activity and the role of monocytes in normal subjects.

A new analysis of natural killing activity in peripheral blood is proposed. In this new analysis, we did not use a fixed E/T ratio, which is practised in the conventional analysis. We employed the individual effector/target cell ratio (E/T ratio) according to the number of effector cells in the peripheral blood: the individual E/T ratio of the person was set precisely at the point when the effectors in 1 ml of the person's peripheral blood encountered 2 x 10(4) of target cells. Asymptomatic healthy persons were divided into high activity and low activity groups. Comparison between mononuclear and non-adherent cells in terms of natural killing activity indicated that monocytes possessed a suppressive effect on the activity. In the asymptomatic low activity group, monocytes were more strongly suppressive as the natural killing activity of non-adherent cells became higher. The degree of this suppression was controlled by the change in the ratio of monocytes over large granular lymphocytes (Mo/LGL ratio). The asymptomatic high activity group showed the same suppressive pattern as patients with common cold syndrome, who were found to be in an activated state of natural killing activity. In an activated state, the monocyte suppression was to a lesser degree although the natural killing activity was higher than that in a non-activated state. Also in an activated state, the degree of the suppression was controlled by the Mo/LGL ratio. In addition, the degree of monocyte suppressive function as determined by one Mo/LGL ratio was almost identical both in an activated state and in a non-activated state of natural killing activity.(ABSTRACT TRUNCATED AT 250 WORDS)

Different effect of prostaglandin E2 on B-cell activation by two distinct B-cell differentiation factors, B151-TRF1/IL-5 and B151-TRF2: selective inhibition of B151-TRF2-induced antibody response through increases in intracellular cyclic AMP levels.

Effects of prostaglandin E2 (PGE2) on murine B-cell activation induced by two distinct B-cell differentiation factors, B151-TRF1/IL-5 and B151-TRF2, were examined. A final differentiation of unprimed B cells into IgM-producing cells induced by B151-TRF2 was markedly inhibited by PGE2 at physiological concentrations (around 10(-8) M), whereas B151-TRF1/IL-5- induced antibody responses of unprimed as well as activated B cells were not affected by PGE2, even at 10(-6) M. B-cell responses induced by B151-TRF2-like factors from autoimmune-prone MRL/lpr mice were also inhibited by PGE2. Biphasic increases in intracellular cyclic AMP (cAMP) levels were induced by culturing B cells with 10(-6) or 10(-8) M PGE2: rapid increases within 8 min and delayed increases around 16 hr. The direct addition of dibutyryl cAMP to cultures of B cells resulted in marked inhibition of antibody responses when stimulated with B151-TRF2 but not with B151-TRF1/IL-5. The B151-TRF2-induced antibody responses were also inhibited by cAMP-elevating reagents such as forskolin, cholera toxin and theophylline. Furthermore, 2'.5'-dideoxyadenosine, which is an inhibitor of adenylate cyclase, prevented the PGE2-mediated cAMP accumulation in unprimed B cells as well as the PGE2-mediated inhibition of B151-TRF2-induced B-cell responses when added at the initiation of culture. These results suggest that PGE2 inhibits B151-TRF2-induced antibody responses through the activation of adenylate cyclase and subsequent accumulation of intracellular cAMP, whereas B151-TRF1/IL-5-responsive B cells are resistant to the inhibitory effect of PGE2 and cAMP.

[P-fimbriated Escherichia coli and serum antibody responses to P-fimbriae in pregnant women with urinary tract infections and their children].

Escherichia coli (E. coli) is still a major pathogen in urinary tract infections (UTI). It was found that 15 out of 20 cases (75%) of E. coli related UTI were caused by P-fimbriated E. coli, compared to the mere 15% of E. coli isolated from urine and 22% from the stool of healthy controls that were P-fimbriated. All patients studied were pregnant women and their delivered children. Antibody responses to P-fimbriae in the sera of these patients were detected with enzyme-linked immunosorbent assay (ELISA) using purified P-fimbriae. Positive antibody responses were observed at titers of 800-6400 in 8 out of 9 cases of UTI of pregnant women caused by P-fimbriated E. coli. The high level of antibody titers persisted for one month on average and then decreased. These antibodies to P-fimbriae were essentially IgG and transmitted to delivered children from UTI mothers. Therefore, the protective role of these antibodies from P-fimbriated E. coli infections in new born children has been suggested.

[Acute biphenotypic leukemia followed by two color flow cytometry].

In this paper is reported a case of acute biphenotypic leukemia who was treated by chemotherapy and pursued its effect by two color flow cytometry. A 33-year-old male patient was admitted due to fever and general fatigue and diagnosed as acute leukemia by hematological findings. Surface markers were investigated to find positive reaction of Leu 12 (CD19), J 5 (CD10), My 7 (CD13) and My 9 (CD33), in which Leu 12 and My 9 were simultaneously expressed on the same blast cells by two color flow cytometry. He was treated with daunorubicin, enocitabine, mercaptopurine, vincristine, and prednisolone to obtain partial remission. Then, he was administered L-asparaginase, doxorubicin, vincristine and prednisolone to reach complete remission. The effect of chemotherapy was investigated by not only bone marrow puncture, but also by two color flow cytometry. From the findings in this case, the two flow cytometry was proved to be a useful tool for not only diagnosis of acute mixed leukemia, aut also the judgement of the effect of treatment.

Epidemiological study on sarcoidosis in Japan. Recent trends in incidence and prevalence rates and changes in epidemiological features.

This paper presents the results of an investigation made on the recent trends in the estimated prevalence and incidence rates of sarcoidosis in Japan and the changes in its epidemiological characteristics based on the results of nation-wide surveys conducted in 1972 and 1984. Age-adjusted mortality trends based on vital statistics were also examined. The estimated prevalence rate in males increased from 3.0 per 100,000 population in 1972 to 3.8 in 1984 and in females from 3.3 to 5.6, respectively (p less than 0.01), while the estimated incidence rate remained unchanged (1.2 per 100,000 in males and 1.4 in females). The age-adjusted mortality rate showed a gradually decreasing tendency from 0.2 to 0.1 per million. The estimated age-sex specific prevalence and incidence rates showed a pronounced increase in the middle-aged and elderly age groups particularly in females but a marked decrease in those 19 years and under. The increase in prevalence rate may be attributable to the prolonged duration of the disease. Noteworthy changes in the pattern of the disorder included a marked increase in the proportion of patients with eye lesions (males: from 31.3% to 44.4% and females: 41.6% to 58.7%) indicative of poor prognosis and statistically insignificant increase in pulmonary parenchymal lesions (males: from 38.1% to 43.4% and females: from 33.5% to 36.4%), the majority of which showed pulmonary mottling not accompanied with severe fibrosis often seen in Europe and America. The standardized prevalence ratios were observed in hot, warm and cold areas.(ABSTRACT TRUNCATED AT 250 WORDS)

[Surgical repair of scimitar syndrome by direct anastomosis between right pulmonary vein and left atrium].

A surgical case of 8-year-old boy with scimitar syndrome is presented. The patient was admitted to the hospital because of exertional dyspnea and underdevelopment. Cardiac catheterization revealed a large amount of left to right shunt and O2 step up at the level of the inferior vena cava below the diaphragm. Angiography showed that the pulmonary vein draining the right lower lobe pierced the diaphragm and emptied into IVC. At operation the right pleural cavity was entered and a large anomalous pulmonary vein trunk was found which headed downward and medially and eventually entered in the diaphragm. A direct anastomosis between the scimitar vein and left atrium was made. To avoid kinking or stenosis of this vein, the parenchyma of the right lung (S7) was divided to create a passway of the vein. Anastomosis was performed without excessive tension or kinking on it. We conclude that the method applied in this case might be a procedure of choice for the repair of scimitar syndrome especially when there is no associating atrial septal defect or scimitar vein drains into IVC at the level lower than hepatic vein.

[Genetic markers and thrombin reaction in a family of Bernard-Soulier syndrome].

A family with Bernard-Soulier syndrome (BSS) was investigated with reference to the genetic markers and thrombin reactions. The proband was a 24-year-old man with a life-long history of epistaxis and gingival bleeding. His parents were first cousins; furthermore, his father was born to parents of second cousins. His father also had bleeding tendency and was also diagnosed as having BSS. However, his mother and elder brother were normal. Genetic marker analysis among the family members suggested that the 16th chromosome was associated with the development of BSS, because only the haptoglobin genotype coded on the 16th chromosome was the marker in both the proband and his father. In addition, they both exhibited decreased thrombin-induced platelet aggregation at a low dose, but an almost normal reaction at a high dose of thrombin.
