[Synthesis cloning and expressions in E coli of human insulin A and B chain genes].

Human insulin A and B chain genes were designed and synthesized by using a rapid and simple method. The synthesized A and B chain genes were cloned separately. The expression (plasmids) pWR 590-HIA and pWR 590-HIB were constructed, and the two plasmids can direct the synthesis of the approximately 590 amino acid-long truncated beta-galactosidases fused to human insulin A or B chains. The fused A or B chain proteins were isolated from the fermented cells and cleaved with BrCN. The resulting mixtures were sulfonated and the sulfonated A and B chains were purified. Human insulin was obtained by using an A and B chain combination method.

Pharmacokinetic studies of norethisterone-3-oxime and norethisterone-3-oxime acetate in rhesus monkey.

After [6,7-3H]-labelled norethisterone-3-oxime (NETO) and norethisterone-3-oxime acetate (NETO-AC) were given intravenously or orally through a nasal tube with 1 mg of respective unlabelled steroid to Rhesus monkey, serum samples were collected at various periods, and radioactivity was counted with or without reverse-phase HPLC separation in advance. Pharmacokinetics of NETO and NETO-AC were compared with those of norethisterone (NET) and norethisterone acetate (NET-AC) respectively which were studied in a similar experimental design. The results indicated that the serum concentration-time curve of NETO and NET could be adequately described by a two-compartment model. Average t 1/2 ka, t 1/2 alpha and t 1/2 beta with standard deviation for oral administration were 0.21 +/- 0.08 (h), 1.28 +/- 0.31 (h) and 10.01 +/- 4.59 (h) for NET and 0.37 +/- 0.81 (h), 0.90 +/- 0.26 (h) and 8.55 +/- 2.21 (h) for NETO respectively. NETO metabolized to NET which had a similar serum profile with its precursor. NET-AC also metabolized to NET, but more rapidly. It disappeared from blood 8-12 h after nasal feeding. NETO-AC was non-detectable at all when given orally because it metabolized immediately and extensively in the animal body. Its major metabolites, NETO, NET and NET-AC already appeared in the first blood sample drawn 15 min after administration. NETO-AC, when injected intravenously, declined abruptly and could not be detected 4 h later. Among the metabolites, only the deacetylated products (NET and NETO) reached relatively higher levels and sustained longer in blood.(ABSTRACT TRUNCATED AT 250 WORDS)

[Distribution of bromo[methylene-3H]acetamide in Oncomelania snails, carps, and mice].

Bromoacetamide show a high molluscicidal effect, low toxicity to fish and good solubility in water. It is a promising molluscicide. The present paper reported the distribution of bromo[methylene-3H]acetamide in Oncomelania snails, carps, and mice. Radioactivity in animals was measured by liquid scintillation counting. In Oncomelania snails the radioactivity gradually increased with exposure time and drug concentration. The highest level was found in the head-foot, reaching 12.5 x 10(3) dpm.mg-1 after exposure to the drug for 24 h, whereas the levels in liver and other tissue were 3.0 x 10(3) and 3.8 x 10(3) dpm.mg-1 respectively. Carps absorbed bromo[methylene-3H] acetamide similarly as in Oncomelania snails, but the levels in tissues were less than those in Oncomelania snails. Bromo[methylene-3H]acetamide 0.5 ml (1.4 mumol.L-1) was given ig to mice. Blood samples of 3 mice were collected at 0.5, 1, 3, 6, 12, 24 and 96 h after ig. The peak levels was found within 12 h and then declined gradually.

[Pharmacokinetics of norethindrone-3-oxime and norethindrone in rhesus monkeys].

After intravenous administration or nasal feeding of (6, 7-3H)-labelled norethindrone-3-oxime (NETO) or norethindrone (NET) to rhesus monkeys, the serum concentrations were determined by measuring the radioactivity after separation with HPLC. The serum concentrations of total extractable radioactivity were also measured without HPLC separation. In cases of nasal feeding, NETO and NET were quickly absorbed, and almost all of them were eliminated within 24 hours. NETO, when given via both routes, was partly metabolized to NET and partly remained in original form. The blood concentration--time curves for NETO and NET were adequately fitted to two compartment models. No significant difference in pharmacokinetic parameters between the two drugs was observed. The absolute bioavailability for NETO and NET were found to be 64.46 +/- 34.60% and 35.02 +/- 26.49% respectively.

[Immuno-electron microscopical studies on the retinal basement membrane of chick embryo].

Retinal basement membrane (RBM), also called inner limiting membrane of retina, is constituted by extracellular matrix. It was reported that neurite outgrowth of a neuron was closely related to extracellular matrix, particularly the laminin. In this laboratory RBM was used as the optimal substrate for retinal cells in culture. We have studied the surface of RBM and its relation to neurite outgrowth by scanning electronmicroscopy and immunogold transmission electronmicroscopy. RBM could be separated by mechanical disruption of the retina mounted between 2 adhesive substrata (membrane filter and poly-L-lysine coated glass). The surface of RBM studied was the side of RBM facing the optic fiber layer and ganglion cell layer. Small particles densely distributed on surface of RBM (Plate I, Fig. 1 and 2) were shown to be chrysanthemum-like structures with radiative arms under the scanning electronmicroscopy (Plate I, Fig. 3 and 4). The radiative arms of RBM of 12-day old chick embryo (E 12) were more in number and longer in length than that of the 6-day old chick embryo (E 6). The axons of ganglion cell from E 6 retinal strip extended out very well on RBM (Plate I, Fig. 5). Growth cone was active with filopodia. The chrysanthemum-like structures changed to ball-particles when the RBM was cultured for 24 hr. Some of ball-particles lay over the growth cone, and some beside it. Over and beside the nerve fiber could also be seen some ball-particles. When many neurites grew on RBM, a lot of ball-particles were shown to be displaced and piled up (Plate I, Fig. 6). The whole amount RBM labeled by indirect immunogold staining of Müller glial cell could be observed by transmission electronmicroscopy. The gold particles wer located at the chrysanthemum-like structure of E 6 RBM (Plate II, Fig. 7) and E 12 RBM (Plate II, Fig. 8). It was suggested that those structures were the end foot of Müller glial cells. Staining of PBS control or mouse serum control was negative (Plate II, Fig. 9 and 10).(ABSTRACT TRUNCATED AT 250 WORDS)

[Pharmacokinetics of norethindrone-oxime and norethindrone in rabbits after oral administration].

After oral administration of [3H]-labelled norethindrone-oxime (NETO) or norethindrone (NET) to rabbits, the blood concentrations of NETO and NET were determined by measuring the radioactivity after separation with HPLC. The pharmacokinetic parameters of the compounds were also compared. Experimental results indicate that both NETO and NET were quickly absorbed. Their elimination from blood revealed a fast and a slow phase. One portion of NETO was metabolized to NET, and the remainder was excreted in unchanged form. The amount of NETO and its metabolite, NET, in serum were about equal within 24 hours. The serum concentration-time curves of NETO and NET adequately fitted to a two-compartment model. There was significant difference between NETO and NET in Tmax (P less than 0.05). But no significant differences in other parameters (P greater than 0.05) were observed.
