[Management and prognosis of advanced ovarian cancer associated with bowel metastasis and obstruction].

OBJECTIVE: To investigate the high risk factors and prognostic factors of bowel metastasis and obstruction in the patients with late stage ovarian cancer by analyzing the clinical characteristics and to find out appropriate methods to solve this difficult problem. METHODS: 151 cases with ovarian cancer in stages I and IV who received the treatment from January 1, 1988 to December 31, 1992 were analyzed retrospectively. RESULTS: Operative residues, diffuse bowel metastasis, small intestinal involvement and papillary serous cystadenocarcinoma were found to be high risk factors for bowel obstruction. Age, duration between operation and obstruction, level of CA125 at occurrence of obstruction, numbers of operation and the histologic grade were disclosed as prognostic factors. The prognosis was much worse if the score is more than 6. CONCLUSION: Due attention paid to the high risk factors and the prognostic factors should be emphasized to improve the quality of life and the survival rate.

[Gene expression of HGF and its receptor in experimental hepatomas].

OBJECTIVE: To understand the regularity of changes of gene expression about HGF and its receptor (HGFR) in experimental hepatomas model of Wistar rats and the relationship between this change and tumor biological behaviour. METHODS: The gene expression of HGF and HGFR in the canceration course of rats was determined by using experimental hepatomas model of Wistar rats established by diethylnitrosamine (DENA), with digoxigenin-labled probe. RESULTS: The positive expression of HGF and HGFR was found in rats liver tissues, but it was different in distinct stages. The positive expression of HGF was the highest in precirrhosis stage, and next in the cirrhosis stage. The canceration stage was the same as in the normal group. The positive expression of HGFR was gradually increased following the canceration course and the canceration stage was the highest, but we didn't find the expression of HGFR in normal liver tissues. CONCLUSIONS: There are close relations between the gene expression of HGF and HGFR and evolution course during the canceration of rats liver. HGF and HGFR systems play an important role in regulating tumor growth and metastasis.

A study on detection of serum fasting total bile acid and choloyglycin in neonate for cholestasis.

Enzyme-linked colorimetric analysis and radioimmunoassay were employed to detect serum fasting total bile acids (FBA) and choloyglycin (CG) respectively in cases of 32 neonatal hepatitis, 33 cases of neonatal unconjugated hyperbilirubinemia and 34 cases of breast milk jaundice (BMJ). FBA and CG in acute period of neonatal hepatitis were obviously elevated and decreased gradually in convalescent and recovered period. The difference was significant for each stage as compared with controls. Acute FBA correlated strongly with the course. Both neonatal unconjugated hyperbilirubinemia and BMJ differed significantly from controls in FBA and CG. The results suggested that serum FBA and CG were helpful in judging the course and state of neonatal hepatitis, and cholestasis might existed in neonatal unconjugated hyperbilirubinemia.

[Genetic polymorphism of antithrombin III in five Chinese populations].

The genetic polymorphisms of antithrombin III (AT III) in 5 Chinese populations were studied by isoelectric focusing on polyacrylamide gels followed by immunoblotting. The products of two alleles at the antithrombin III locus together with 1 AT III anodic variant and 3 cathode variants were observed. The results revealed that the predominant allele of AT III blood group system in Chinese populations was AT III *1, with frequencies between 0.9773-0.9850. Frequencies of AT III *2 ranged from 0.0091 to 0.0169. These results indicate that the polymorphic information content of AT III blood group system in Chinese populations is limited.

[Subcellular location of blood group substances ABH using transmission electron microscopical immunocytochemistry technique].

The subcellular location of blood group substances ABH was studied by using transmission electron microscopical immunocytochemistry. The cells were the epithelial cells of both mucous membranes and glands of normal human stomach, duodenum and transverse colon. Both cellular ultrastructure and antigens should be well preserved in transmission electron microscopical immunocytochemistry. In this paper, various fixatives, embedding media and immunostaining methods were investigated and compared. The results indicate that the solution containing 2% paraformaldehyde and 0.2% glutaraldehyde (2PG) is the best fixative and Epon 812 medium is the best embedding medium for preserving ABH antigens and protecting cellular ultrastructure. Post-embedding immunogold staining technique is superior to pre-embedding immunoperoxidase staining.

[An experimental study on the anticancerous effects of CD3-TIL derived from human primary hepatic carcinoma].

In this study, CD3-TIL produced by in vito induction with CD3 and IL-2 (CD3-TIL) was compared with TIL stimulated by only IL-2 (TIL) in their in vitro proliferation, and both in vitro and in vivo anticancerous effects. It was found that CD3 at the concentration of 100 ng/ml was the most suitable stimulus of in vitro TIL proliferation (CD3-TIL) which was significantly greater than that by simple IL-2 stimulation. Also in vitro anticancerous activity of CD3-TIL was greater than TIL. CD3-TIL significantly inhibited tumor growth, causing shrinkage of tumor mass, and prolonging the life span of the nude mouse with tumor burden.

[Surgical treatment of primary liver cancer].

The mortality rate of patients with primary liver cancer (PLC) was 20.40 per 100,000 Chinese per year, that takes the second place among all forms of cancer. Surgical resection remains the modality of primary choice in the treatment of PLC. 2051 patients with PLC were treated by liver resection from 1960 to 1993. Of those, 94.1% were hepatocellular carcinoma (HCC) and the incidence of cirrhosis and chronic hepatitis was found in 86.5%. Small PLC (< or = 5 cm in diameter) was found in 25.1% of all cases. In the small PLC group, 176 cases were found the tumor < or = 3 cm in diameter. All 2051 patients received liver resection and 44.0% of them had local radical resection. The overall mortality rate within one month after operation was 1.1%. The overall 5-year survival rate was 36.1%. Improved surgical result was obtained in 515 patients with small PLC (< or = 5 cm in diameter). No operative death occurred in this group and the 5-year survival rate was 79.8%. Moreover, the patients with very small PLC (< or = 3 cm in diameter) prolonged their survival. The 5-year survival rate was 85.3%. Early diagnosis and early resection of PLC are one of the key points for improving the long-term results. Improvements in operative techniques play an important role in reducing post-operative complications, lowering operative mortality and obtaining better operative results. Rehepatectomy for recurrent liver cancer can improve the surgical results. 95 patients underwent rehepatectomy with a 5-year survival rate of 53.2% and 25.0% after the first and second operation respectively. One patient survived 18 years. For unresectable large tumors, two-stage resection can be used. We collected 659 cases of large unresectable tumors from 1974 to 1994. Among them, 73 cases were resected in two-stage operation. The resection rate was 11.1%, and the 5-year survival rate was 61.5%. Postoperative comprehensive treatment is also important for upregulating cellular immunological function, preventing tumor recurrence, and improving surgical results.

[Anti-human AFP variant McAb in radioimmunodetection for primary hepatocellular carcinoma].

We investigated the affinity and special combination of anti-human AFP variant monoclonal antibody (AFP-R-LCA McAb) for cells of AFP positive hepatocellular carcinoma (HCC). AFP-R-LCA McAb was labeled by 131I radioisotope (131I-AFP-R-LCA McAb injected into the peripheral veins of patients with HCC or liver cirrhosis after hepatitis B. 131I-AFP-R-LCA McAb was gathered in tumor of HCC in 6 AFP positive patients, but there was no positive gathering in HCC in 6 AFP negative or 4 liver cirrhosis patients. AFP-R-LCA McAb has strong affinity and special combination to AFP positive cells of HCC and can be recognized as a carrier for radioimmunodetection and radiommunotherapy.

[Effects of different processed products of radix Angelica sinensis on clearing out oxygen free radicals and anti-lipid peroxidation].

Different processed products of Radix Angelica Sinensis could clear out superoxide radical (O2.-) generated through hypoxanthinexanthine oxidase system and hydroxyl radical (.OH) generated through Fenton action, and inhibit lipid peroxidation of supernatant hepati homogenate in mice induced by free radical generation system. There exist significant differences among the different processed products.

A single base-pair change (ATG-->ATC) nullifies the activity of cytosolic fumarase in Saccharomyces cerevisiae.

A respiratory defective pet mutant (W303 delta FUM1) of Saccharomyces cerevisiae deficient in fumarase was transformed with the plasmid construct pG5/ST7. This plasmid contains the entire FUM1 gene with only a single base pair change (ATG-->ATC) confined to the putative second inframe translation initiation codon. While transformation of the fumarase deficient mutant with pG5/ST7 resulted in an elevation of fumarase activity in the mitochondria of the transformed strain, fumarase activity in the cytosol remained negligible his result indicated that the cytosolic fumarase isoenzyme is synthesized exclusively from the second translation initiation codon of FUM1. Results of RACE-PCR of the 5' ends of FUM1 transcripts confirmed that the FUM1 gene synthesizes two distinct species of transcripts. These data provide strong evidence for the synthesis and targeting of two fumarase isomers.

Mesolimbic dopamine terminals and locomotor activity induced from the subiculum.

The role of the mesolimbic dopamine terminals in the nucleus accumbens in the initiation of locomotion in rats was studied. Locomotor activity was initiated by activation of the excitatory input from the ventral subiculum to the nucleus accumbens with NMDA. Measurements of locomotor activity, induced by unilateral administration of NMDA into the ventral subiculum, were compared before and after destruction of the mesolimbic dopamine terminals in the nucleus accumbens. The dopamine terminals were destroyed by injection of 6-OHDA into the ventral tegmental area which projects to the nucleus accumbens. Injection of NMDA into the ventral subiculum caused an almost four-fold increase in locomotor activity. However, this increase was abolished after the destruction of the mesolimbic dopamine terminals in the nucleus accumbens. The results suggest that the mesolimbic dopamine terminals are essential in transmitting subicular signals to the output neurones within the nucleus accumbens.

Folate receptor type gamma is primarily a secretory protein due to lack of an efficient signal for glycosylphosphatidylinositol modification: protein characterization and cell type specificity.

A novel isoform of the human folate receptor (FR, type gamma) was recently identified in hematopoietic tissues [Shen et al. (1994) Biochemistry 33, 1209-1215]. In that report, Cos-1 cells, transiently transfected with the cDNA for FR-gamma, produced relatively poor expression of the receptor on the cell surface. In this study, several recombinant Chinese hamster ovary (CHO) cell lines were produced by stable transfection with the cDNA for FR-gamma followed by amplification. Similar recombinant CHO cell lines were produced that expressed the glycosylphosphatidylinositol-(GPI-) anchored FR type beta and a truncated form of FR type beta (FR-beta delta), in which the normal carboxyl-terminal signal for GPI anchor attachment was deleted. Both FR-gamma-and FR-beta delta-expressing CHO cells produced a [3H]folic acid binding protein in the medium with a similar time course over a 24-h period; in contrast to intact FR-beta, relatively insignificant amounts of either FR-gamma or FR-beta delta were associated with the CHO cell surface and this was unaltered by the absence of serum in the medium. The FR-gamma- and FR-beta delta-producing CHO cells did not differ significantly in intracellular FR levels. Furthermore, the mRNA level for FR-gamma did not exceed that for FR-beta delta. When deglycosylated with hydrogen fluoride, both FR-gamma and FR-beta delta showed similar apparent molecular weights on Western blots as predicted for the intact polypeptides.(ABSTRACT TRUNCATED AT 250 WORDS)

Polyadenylation of c-mos mRNA as a control point in Xenopus meiotic maturation.

c-mos protein, encoded by a proto-oncogene, is essential for the meiotic maturation of frog oocytes. Polyadenylation of c-mos messenger RNA is shown here to be a pivotal regulatory step in meiotic maturation. Maturation is prevented by selective amputation of polyadenylation signals from c-mos mRNA. Injection of a prosthetic RNA, which restores c-mos polyadenylation signals by base pairing to the amputated mRNA, rescues maturation and can stimulate translation in trans. Prosthetic RNAs may provide a general strategy by which to alter patterns of mRNA expression in vivo.

A periodic table of symmetric tandem mismatches in RNA.

The stabilities and structures of a series of RNA octamers containing symmetric tandem mismatches were studied by UV melting and imino proton NMR. The free energy increments for tandem mismatch formation are found to depend upon both mismatch sequence and adjacent base pairs. The observed sequence dependence of tandem mismatch stability is UGGU > GUUG > GAAG > or = AGGA > UUUU > CAAC > or = CUUC approximately UCCU approximately CCCC approximately ACCA approximately AAAA, and the closing base pair dependence is 5'G3'C > 5'C3'G > 5'U3'A approximately 5'A3'U. These results differ from expectations based on models used in RNA folding algorithms and from the sequence dependence observed for folding of RNA hairpins. Imino proton NMR results indicate the sequence dependence is partially due to hydrogen bonding within mismatches.

Role of Rel-related factors in control of c-myc gene transcription in receptor-mediated apoptosis of the murine B cell WEHI 231 line.

Treatment of immature murine B lymphocytes with an antiserum against their surface immunoglobulin (sIg)M results in cell death via apoptosis. The WEHI 231 B cell line (IgM, kappa) has been used extensively as a model for this anti-Ig receptor-mediated apoptosis. Anti-sIg treatment of WEHI 231 cells causes an early, transient increase in the levels of c-myc messenger RNA and gene transcription, followed by a rapid decline below control values. Given the evidence for a role of the c-myc gene in promoting apoptosis, we have characterized the nature and kinetics of changes in the binding of Rel-related factors, which modulate c-myc promoter activity. In exponentially growing WEHI 231 cells, multiple Rel-related binding activities were detectable. The major binding species was identified as p50/c-Rel heterodimers; only minor amounts of nuclear factor kappa B (NF-kappa B) (p50/p65) were detectable. Cotransfection of an inhibitor of NF-kappa B (I kappa B)-alpha expression vector reduced c-myc-promoter/upstream/exon1-CAT reporter construct activity, indicating the role of Rel factor binding in c-myc basal expression in these cells. Treatment with anti-sIg resulted in a rapid transient increase in the rate of c-myc gene transcription and in the binding of Rel factors. At later times, formation of p50 homodimer complexes occurred. In cotransfection analysis, p65 and c-Rel expression potently and modestly transactivated the c-myc promoter, respectively, whereas, overexpression of the p50 subunit caused a significant drop in its activity. The role of activation of Rel-family binding was demonstrated directly upon addition of the antioxidant pyrrolidinedithiocarbamate, which inhibited the anti-sIg-mediated activation of the endogenous c-myc gene. Similarly, induction after anti-sIg treatment of a transfected c-myc promoter was abrogated upon cotransfection of an I kappa B-alpha expression vector. These results implicate the Rel-family in Ig receptor-mediated signals controlling the activation of c-myc gene transcription in WEHI 231 cells, and suggest a role for this family in apoptosis of this line, which is mediated through a c-myc signaling pathway.

Utilization of [14C]phenylalanine derived from arylphorin or free amino acid in Manduca sexta pharate adults.

The role of arylphorin as a storage protein was studied using 14C-arylphorin. 14C-arylphorin was produced optimally by incubating one-half fat body from Manduca sexta fifth instar larvae at 22 degrees C for 24 h, in 1 ml of medium containing amino acids at 25% of their physiological concentration with [U-14C]-phenylalanine (phe) provided initially without nonlabeled phenylalanine. Nonlabeled phe was provided after 1 h at 16% of its physiological concentration. The specific activity of 14C-arylphorin produced in vitro was 30 times greater than that generated in vivo. Injection of 14C-arylphorin into pharate adults was used to study the distribution of 14C-phe derived from this protein into 14CO2 and tissues for comparison with injection of free 14C-phe during the middle (days 6 to 12 pharate adult) and late (days 12 to 17 pharate adult) stages of adult development. Appearance of 14CO2 from 14C-arylphorin as compared to 14C-phenylalanine showed a slower time course during both the middle and late stages of development, in keeping with the time needed for degradation of the protein. In accord with faster phe turnover near the end of adult development, total 14CO2 production was greater and the retention of 14C in hemolymph and fat body was less compared to the middle stage of development regardless of whether 14C-arylphorin or 14C-phe was injected. In the middle stage of development, the appearance of 14C in the cuticle and head parts was greater, whereas incorporation into abdomen and thorax was less than during the late stage of development. Since the pattern of 14C distribution from 14C-arylphorin and 14C-phe was similar, one major function of arylphorin must be as a storage protein replenishing the supply of free amino acids used for synthesis of adult tissues. These results also suggest a limited contribution of M. sexta arylphorin to formation of the cuticle subsequent to day-6 pharate adult.
