Troglitazone increases system A amino acid transport in 3T3-L1 cells.

System A is one of the most highly regulated transport systems for transport of neutral amino acids into mammalian cells. Stimulation of uptake of alpha-[3H]methylaminoisobutyric acid (MeAIB), a nonmetabolizable system A substrate, by a novel insulin-sensitizing agent, troglitazone, in 3T3-L1 adipocytes was investigated. Treating adipocytes with troglitazone alone resulted in a time- and dose-dependent increase in the uptake of MeAIB. The peak stimulation appeared about 24 h after troglitazone addition. Both troglitazone- and insulin-stimulated transport activities increased markedly after the induction of differentiation of preadipocytes into adipocytes, and declined to a steady state level in adipocytes. The stimulated MeAIB uptake exhibited substrate specificity typical of system A and was mediated by a single component as determined by Eadie-Hofstee plots. The stimulation by troglitazone and that by insulin were similarly sensitive to actinomycin D and cycloheximide, suggesting that both agents may induce de novo synthesis of the same type of system A transport. Apart from the insulin-independent effect, troglitazone also showed an insulin-dependent action characterized by enhanced sensitivity to insulin. The synergistic stimulation of MeAIB uptake by coadministration of insulin and troglitazone was most prominent at the early stages of adipocyte differentiation. Pretreating cells with troglitazone during the differentiation attenuated the sensitivity of insulin to inhibition by actinomycin D, suggesting that troglitazone may enhance the insulin action by stabilizing messenger RNA involved in system A function.

Transformation of cortisone acetate using immobilized Arthrobacter simplex cells.

A new cell immobilization method was developed in which cells of Arthrobacter simplex BY-2-3-51 were immobilized by entrapment with a polyvinyl alcohol-alginate complex carrier. The immobilized cells preparation contains spherical beads with high mechanical strength and delta 1-dehydrogenase activity. The activation conditions and enzymatic properties of the immobilized cells were studied. When the initial substrate concentration was 20 mg/mL, the conversion of cortisone acetate to prednisone acetate by immobilized cells in the shaken culture was 98% at 18 hours. The stability of immobilized cells was maintained by activation at proper times.

Detection of p53 gene mutations in human leukemia by PCR-SSCP analysis and direct nucleotide sequencing.

OBJECTIVE: To look for mutations of the p53 gene in leukemic patients and study the relationship between abnormalities in p53 gene and leukemogenesis. METHODS: The peripheral blood and Bone Marrow Samples were collected from 36 patients with various leukemia types including 14 cases of lymphocytic leukemia [8 cases of acute lymphocytic leukemia (ALL), 4 cases of chronic lymphocytic leukemia (CLL), 2 cases of hairy cell leukemia (HCL)] and 22 cases of myelocytic leukemia [11 cases of acute nonlymphocytic leukemia (ANLL), 11 cases of chronic myelocytic leukemia (CML)]. DNA structures of exon 5-8 of the p53 gene were scanned by PCR-SSCP (single strand conformation polymorphism analysis of polymerase chain reaction products). The appropriate DNA fragments were amplified, Purified and sequenced directly. RESULTS: By PCR-SSCP analysis, shifts in electrophoretic mobility of the p53 gene were detected in 3 of 14 patients with lymphocytic leukemia (2 ALL and 1 CLL), but none in 22 patients with myelocytic leukemia including one in blastic crisis. Direct nucleotide sequencing in one patient with ALL showed transition of CTG to CAG at codon 257 of exon 7, resulting in a change of its encoded amino acids from aspartic acid to valine. To our knowledge, the mutation at this codon has not been previously reported hitherto. CONCLUSIONS: The p53 gene mutations are specifically associated with lymphocytic leukemia. Alternations of the p53 gene may play a certain role in leukemogenesis in some cases of lymphocytic leukemia.

[The treatment of fracture of patella by internal fixation with tension band from musculofascial tendon].

In the study of the efficacy of internal fixation with tension band of musculofascial tendon in the treatment of fracture of patella, 52 cases were reported. After a following-up of 6 months to 13 months the bone healing was observed, in 7.5 weeks in average, and the function of the knee joint had recovered to normal or almost normal. It was concluded that the treatment of fracture of patella by internal fixation with tension band from musculofascial tendon was a ideal and practicable method.

A clinical investigation on massage for prevention and treatment of recurrent respiratory tract infection in children.

The TCM therapeutic principle of "eliminating the pathogenic factors by supporting the healthy energy" was adopted in massage to treat and prevent recurrent respiratory tract infection of children. Susceptible and healthy children of the same age were used as controls. As a result, the therapeutic effect of the treatment group is significantly better than that of the controls, with all of the immunologic indexes being approximately normal when the patients were reexamined 3 and 6 months after the massage. Statistically, the difference was significant (P < 0.01). Massage has been proved very helpful in improving the general constitution, enhancing the immune functions, preventing and treating the condition, and in health care as well.

[Study on the relationship between occupational factors and work ability of middle aged workers].

Based on the assessment of work ability index (WAI) of middle aged workers, associations between occupational factors and work ability decline of middle aged workers were examined in a case-control study. 1037 workers (age ranged from 40 to 60) were included in the study. The study group comprised 180 workers of mean age 47.71 years representing those who's work ability decreased. The control group comprised 857 workers of mean age 47.22 years representing those who's work ability was on a normal level. Results showed that some common occupation hazards such as dusts, noise, vibration, humidity, and high temperature were more frequently occurred in work environment of the study group than that of the control group. High risks were found in workers with these hazards. Another significant risk factor of work ability decline was heavy physical load. Significantly elevated risks were observed for physical load perception(OR = 2.52), repetitive work (OR = 1.36), poor work postures(OR = 1.48), and carrying heavy loads(OR = 1.59). As compared to physical loads, mental loads were not in relation to work ability decline. The information about occupational factors affecting work ability will be helpful for redesiging the work for the middle aged workers.

[Establishment of two human esophageal carcinoma cell lines and their cytogenetic analysis].

OBJECTIVE: To explore the relation between chromosomal aberration and carcinogenesis of the esophagus by molecular cytogenetic analysis of two esophageal carcinoma cell lines. METHODS: Small tissue blocks taken from resected specimens of patients with esophageal cancer (EC) were cultured in medium 199 added with 1.5% fetal bovine serun, and two esophageal carcinoma cell lines, EC8712 and EC8733, were established in 1987. They grew in monolayers. Molecular cytogenetic studies were performed on the two cell lines using chromosome G-banding, fluorescence in situ hybridization (FISH) and comparative gene hybridization (CGH) techniques. RESULTS: Both cell lines were aneuploid. Loss of chromosome Y, partial deletion of 1p, translocation of 2q and amplification of 5p, were observed in the 2 cell lines. Amplification of 8q and 13q in EC8733 and deletion of 17p in EC8712 were detected. CONCLUSION: The presene of the reported chromosomal aberrations may be related to the carcinogenesis and cancer development of the esophagus.

[Immuno-epidemiological investigation of recurrent spontaneous abortion].

OBJECTIVE: To probe into immune pathogeny of recurrent spontaneous abortion (RSA). METHODS: Immunological analysis was made in ABO group antibodies by indirect hemagglutination, anti-zona pellucida antibodies by BA-ELISA, anti-phospholipid antibodies by ELISA, blocking effect (MLR-BE) by blocking test one-way mixed lymphocyte reaction, cytotoxic index (CI) by complement-dependent cytotoxic test, anti-idiotypic antibodies to blocking antibodies by MLR-BE inhibiting test, and effects of blocking antibodies on CD antigens by flow cytometry. In contrast to the healthy fertile women, 129 cases of primary abortion and 16 cases of secondary abortion were retrospectively analyzed for immunoetiological compositions. RESULTS: Among causes of RSA, lack of blocking antibodies accounted for 31.7%, anti-zona pellucida antibodies for 20.4%, anti-phospholipid antibodies for 12.5%, and ABO group antibodies for 11.5%. Among causes of primary abortion, lack of blocking antibodies accounted for 31.4%, anti-zona pellucida antibodies for 20.4%, anti-phospholipid antibodies for 8.5%, and ABO group antibodies for 8.4%. Among causes of secondary abortion, imbalance of idiotypic-antiidiotypic network of blocking antibody accounted for 39.4%, anti-phospholipid antibodies for 31.4%, and ABO group antibodies for 22.4%. CONCLUSION: According to the above evaluation of immuno-epidemiology of RSA, RSA may be etiologically classified into lack of materno-fetal immuno-recognition, materno-fetal inappropriate immuno-recognition, and disorder of materno-fetal immuno-recognition.

[Circulating ICAM-1 in sera of renal allograft recipients in monitoring of acute rejection].

OBJECTIVE: To study the significance of ICAM-1 in sera of renal allograft recipients in mornitoring of acute rejection. METHODS: The levels of circulating intercellular adhesion molecule-1 (cICAM-1) in sera of 87 renal transplantation recipients, including 29 patients with acute rejection and 8 patients with cyclosprine nephrotoxicity were measured by sandwich-ELISA. RESULTS: The levels of cICAM-1 were elevated transiently after transplantation. They decreased and reached normal levels within 2 weeks. In the acute rejection episode, the levels of cICAM-1 were significantly higher than those of patients with stable graft function and cyclosporine nephrotoxicity (P < 0.001). The increase of cICAM-1 was seen 1-3 days early than the diagnosis was made. On successful antirejection therapy, the levels of cICAM-1 decreased to normal, but in patients with steroid-resistant rejection, it did not decrease. CONCLUSION: The cICAM-1 in sera can be used to estimate the graft function, and it is also useful in prediction and differential diagnosis of acute rejection and prognosis.

[Chemical constituents in the stem and leaf of Isodon oresbius].

Five triterpenoids were isolated from Isodon oresbius. Their structures were identified as ursolic acid, 2 alpha, 19 alpha-dihydroxy ursolic acid, 2 alpha, 3 alpha, 19 alpha-trihydroxy-12-ursen-28-oic acid, 2 alpha-hydroxy oleanolic acid and 2 alpha, 23-dihydroxy oleanolic acid by physico-chemical constants and spectral analysis.

[The role of mast cells in myocardial fibrosis in SHRsp].

OBJECTIVE: To study the role of mast cells in hypertensive myocardial fibrosis. METHODS: The myocardial collagen and the degree of myocardial fibrosis were assessed using Masson's trichrome staining and image analysis plus toluidine blue as well as proliferating cell nuclear antigen (PCNA) staining and transmission electronic microscopy for SHRsp, aged 34 weeks (S group, n = 6). Besides, age and sex-matched WKY (W group, n = 5) were used as the controls. RESULTS: MC count increased 3.9 folds and PCNA immunostaining positive cells (PCNAP) increased 3.9 fold (both P < 0.0001) in S vs W group. MC were positively correlated with the interstitial collagen volume fraction (ICVF), perivascular collagen area/vascular lumenal cross area (PVCA/VLCA), and PCNAP (r = 0.47, P < 0.05; 0.59, P < 0.001; 0.68, P < 0.001 respectively). Electromicroscopy showed specific piecemeal degranulation of the mast cells, locating between the praliferative interstitial collagen fibers. CONCLUSION: The findings suggested that MCs play a role in certain degree to the myocardial fibrosis present in SHRsp.

Induction of anti-DNA antibodies by immunization with activated lymphocytes and active chromatin.

OBJECTIVE: To investigate the primary autoantigens which contribute to the production of anti-DNA antibodies. These antibodies are serological hallmark and pathogenic factor of systemic lupus erythematosus (SLE). METHODS: Nonautoimmune predisposed BALB/c mice were immunized with concanavalin A (Con A) activated, lipopolysaccharide (LPS) activated and nonactivated syngeneic spleen cells. Nuclei and chromatin from activated/nonactivated lymphocytes were isolated and syngeneic mice were immunized. Sera were taken after the third immunization. IgG anti-dsDNA antibody was determined by ELISA (calf thymus DNA treated with S1 nuclease was used as the coated antigen). The glomerular IgG deposition was observed by immunofluorescence one month after the third immunization. RESULTS: Con A activated T cells and LPS activated B cells induced anti-double stranded (ds) DNA antibody in syngeneic nonautoimmune BALB/c mice and formed the glomerular IgG deposition. Further studies showed that active chromatin isolated from activated lymphocytes induced anti-ds DNA antibody, but not resting chromatin isolated from nonactivated lymphocytes. CONCLUSIONS: Activated lymphocytes and their active chromatin could be the autoimmunogen(s) driving the anti-dsDNA antibodies. The change of chromatin's antigenicity by environmental factors and genetic background may be the common pathway to SLE pathogenesis.

[Role of basolateral amygdaloid nuclei in sleep and wakeful state regulation].

The role of basolateral amygdaloid nuclei (BLN) in sleep and wakeful state (W) was investigated with polysomnography. Electrical lesion of bilateral BLN increased slow wave sleep (SWS) and paradoxical sleep (PS), but decreased W. Bilateral injection of kainic acid (KA), which selectively destroyed the soma of neurons, resulted in biphasic effects. At d 1 after KA, insomnia occurred. At d 4, SWS increased while W decreased, but PS did not change. Unilateral injection of NO donor sodium nitroferricyanide (SNP) caused an enhancement of W and a decrease of SWS. Injection of nitric oxide synthase inhibitor N-nitro-L-arginine (L-NNA) led to opposite effects. L-arginine, a precursor of NO, had no direct effect on sleep and W, but blocked the effects of L-NNA. The results suggest that BLN plays an important role in regulating sleep and wakeful state. Destruction of BLN increases SWS, and NO in BLN has W-promoting and SWS-inhibiting effects, which may be mediated by activation of neurons in BLN.

[The protective effect of captopril during myocardial ischemia and reperfusion and its mechanism].

OBJECTIVE: To investigate the myocardial protective effect of captopril (Cpl) during ischemia and reperfusion injury. METHODS: Sixteen mongrel dogs were randomly divided into the control and Cpl groups. In the control group, only modified St. Thomas cardioplegia was used. In the Cpl group, St. Thomas cardioplegia and reperfusional blood containing captopril 4.6 mumol/L were used. The plasma renin activity (RA), contents of angiotensin II (AT-II) and endothelin-1 (ET-1) in arterial blood were determined before cardiopulmonary bypass (CPB) and 5, 35, 60 min after reperfusion. The myocardial RA, contents of AT-II, ET-1 and malondyldialdehyde (MDA) were also measured. Parameters of cardiac function were monitored simultaneously. RESULTS: In the control group the plasma RA, AT-II and ET-1 contents were increased after reperfusion and significantly higher than those before CPB (P < 0.01). However, the content of AT-II in the Cpl group was not increased after reperfusion. The content of ET-1 in the Cpl group was increased but significantly lower than that in the control group (P < 0.01). The myocardial AT-II, ET-1 and MDA contents in the Cpl group were significantly lower than those in the control group. The cardiac index (CI) and coronary blood flow (CBF) in the Cpl group regained normal after reperfusion at 35 min. CONCLUSIONS: Cardioplegia and reperfusion blood containing 4.6 mumol/L captopril could ameliorate the cardiac performances by inhibiting myocardial and systemic RAS and decreasing the ET-1 production.
