Indomethacin attenuates post-suspension hypotension in Sprague-Dawley rats.

Orthostatic hypotension is a serious condition that is sometimes manifested in astronauts during standing postflight. These observations may be related to impairment of autonomic function and/or excessive production of endothelium-dependent relaxing factors. To evaluate the role of the cyclooxygenase inhibitor indomethacin as a countermeasure against the post-suspension reduction in mean arterial pressure (MAP), we examined the cardiovascular responses to 7-day 30 degrees tail-suspension and a subsequent 6-hr post-suspension period in conscious male Sprague-Dawley rats. Indomethacin (2 mg/kg) or saline were administered intravenously prior to release from suspension and at 2 and 4 hrs post-suspension. Direct MAP and heart rate were determined prior to suspension, daily and every 2 hrs post-suspension. During suspension, MAP did not change, in contrast, during post-suspension; it decreased compared to parallel non-suspended, untreated animals. There were no significant changes in heart rate. The reduction in MAP post-suspension was associated with significant increases in plasma prostacyclin. Indomethacin attenuated the observed post-suspension reduction in MAP and reduced plasma prostacyclin levels. Also, the baroreflex sensitivity for heart rate was modified by indomethacin--the MAP threshold for baroreflex activation was raised and the effective MAP range expanded. Thus, the post suspension reduction in mean arterial pressure may be due to overproduction of vasodilatory prostaglandins and/or other endothelium-dependent relaxing factors and alteration in baroreflex activity.

[Ultrasonic measurement of the elastic properties of goat condyles].

OBJECTIVE: The purpose of this experiment is to investigate the elastic properties of goat mandibular condyles and to provide basic constants for biomechanical study of temporomandibular joint. METHODS: A fresh goat mandible was adopted and the standard condyle specimen of 4 mm x 6 mm x 10 mm was obtained from it. The structure density of the condyle was determined according to Archimede's principle. The transmitting velocities of ultrasonic wave in different directions of goat condyles were measured by using the ultrasonic system, which was composed of a transmitting transducer of 1.25 MHz, a receiving transducer of 2.5 MHz, an ultrasonic analyzer (5052UA) and a digital storage oscilloscope (HP54601A). The elastic coefficients and technical constants could be calculated by elastic equations. RESULTS: The structural density of the sample and the wave velocities in different directions of the condyle were both obtained by direct measurement. And then, nine elastic coefficients and all of the technical constants of orthotropic condyles were calculated. CONCLUSION: 1. It is practicable to measure elastic properties of the condyle by using ultrasonic technique. 2. Goat condyles of the mandible are a type of anisotropic and linearly elastic material. 3. The direction perpendicular to the transverse ridge of the goat condyle has the highest Young's modulus. As to shear modulus, the direction along the transverse ridge shows the highest value.

[Detection of telomerase activity in laryngeal carcinoma and the association between telomerase activity and lymphatic metastasis].

OBJECTIVE: To investigate the telomerase activity in human laryngeal squamous cell carcinomas (LSCC) and the association between telomerase activity and lymphatic metastasis. METHOD: Telomerase activity was determined in 47 specimens of LSCC and 10 specimens of laryngeal inflammatory polyps using PCR-ELISA. RESULT: Telomerase activity was detected in 39 of 47 (83%) LSCC, but was not detected in all 10 laryngeal inflammatory polyps. There were significant differences between them (P < 0.01). Telomerase activation in LSCC with positive cervical lymph node involvement appeared to be higher (13 out of 13 cases, 100%) than in LSCC with negative cervical lymph node involvement (26 out of 34, 76.47%), (P < 0.05). CONCLUSION: Telomerase is activated frequently in LSCC and may become a biomarker of LSCC. The detection of telomerase activity in LSCC may have some value for predicting metastasis potential and prognosis and can guide the clinical practices applying selective neck dissection.

[Immunity against Schistosoma japonicum induced by phage display peptides mimicking antigenic epitopes of Trichinella spiralis].

OBJECTIVE: To obtain the short peptides mimicking antigenic epitopes of Trichinella spiralis (T. s.), and explore their cross protective immunity against Schistosoma japonicum (S.j.) in mice. METHODS: IgG antibodies were purified from sera of mice infected with T.s.. The purified IgG was used to immunoscreen a phage random peptide library of 7 amino-acid residues displayed as a fusion to protein of filamentous phage. Positive clones were obtained by affinity selection, the reactivity of each clone binding to specific IgG was detected by ELISA. Kunming mice were immunized subcutaneously three times with mixed phage clones. The mice were sacrificed 45 days after challenge. The worms and the liver eggs were counted. RESULTS: After three rounds of panning, the relevant phages had been enriched approximately 150 times in production as compared to those from the first round. Of 24 phage clones randomly selected from the third round biopanning, 21 clones were shown to actually bind to the specific IgG. As compared with the control group, the worm and the liver egg reduction rates in vaccination group were 42.8% and 66.3% (P < 0.001), respectively. CONCLUSION: The above results demonstrate that antigenic epitopes of T. s. can be prepared by immunoscreening phage random peptide library and a significant protective immunity against S. j. can be induced by these epitopes in mice.
