An investigation of the role of true hypha production in the pathogenesis of experimental oral candidosis.

From 427 cases of human oral candidosis 2135 yeast clones were screened for the presence of germ tube-negative C. albicans and variants that formed only pseudohyphae in serum; one strain of each was found. The pathogenic potential of the serum-pseudohyphal and germ tube-negative C. albicans variants was investigated in the oral cavity of the rat; both variants failed to induce palatal candidosis, in contrast to a germ tube-positive C. albicans control strain. The pathogenic potential of C. albicans strains appears to be dependent on the formation of true germ tubes.

Relative effectiveness of various yeasts, Candida spp. and Torulopsis glabrata, for inducing palatal infection in the Wistar rat.

Candida albicans serotypes A and B, Candida tropicalis and Torulopsis glabrata were tested for their ability to produce palatal candidosis in the rat. C. albicans serotype A consistently produced palatal candidosis whereas serotype B did so in only two of five animals tested, indicating a difference in pathogenicity of the two serotypes. C. tropicalis and T. glabrata did not induce changes under the conditions tested and their colonization of palatal mucosa in commensal form was dependent on the presence of an acrylic plate.

An in vitro and in vivo study of the effect of incorporation of chlorhexidine into autopolymerizing acrylic resin plates upon the growth of Candida albicans.

Chlorhexidine acetate was incorporated into autopolymerizing acrylic resin and, after studying its ability to diffuse out in vitro, an investigation was made into the potential of the mixture to treat palatal candidosis in the rat. Chlorhexidine was found to diffuse out of acrylic in fungicidal concentrations for up to three weeks when mixed with the acrylic powder in the proportion of 7.5% (w/w). At this concentration it was found that palatal candidosis as produced by the technique of Shakir et al. was cured or prevented. However, rats fitted with chlorhexidine supplemented plates were found not to take sufficient food during the experimental period to maintain their body weight.

The oral yeast flora of 10-year-old schoolchildren.

The yeast flora of the hard palate mucosa and pooled saliva of 122 schoolchildren (average age 9 years 10 months, 63 males, 59 females) has been studied in detail. Of the population sampled, 71.3% carried oral yeasts. The carriage was approximately 3 times greater in females than in males. The yeast isolates were Candida albicans 71% Saccharomyces spp. 19.7% and Candida tropicalis 8.6%. No isolates of Torulopsis spp. or Rhodotorula spp. were recovered. 71.5% of the Candida albicans isolates were serotype A and 26.3% were serotype B.

Purification and characterization of liver microsomal cytochromes p-450: electrophoretic, spectral, catalytic, and immunochemical properties and inducibility of eight isozymes isolated from rats treated with phenobarbital or beta-naphthoflavone.

Eight different forms of cytochrome P-450 (P-450) were purified to electrophoretic homogeneity by a common procedure from liver microsomes of rats treated with phenobarbital or beta-naphthoflavone. Antibodies were prepared to seven of these forms in rabbits. The eight P-450s were distinguished by spectral properties of the ferric, ferrous, and ferrous carbonyl forms, apparent monomeric molecular weights, peptide mapping, immunological reactivity as discerned by double-diffusion immunoprecipitin analysis and crossed immunoelectrophoresis, and catalytic activities toward the substrates acetanilide, aminopyrine, aniline, benzo[a]-pyrene, d-benzphetamine, N,N-dimethylnitrosamine, 7-ethoxycoumarin, 7-ethoxyresorufin, ethylmorphine, p-nitroanisole, testosterone, and (R)- and (S)-warfarin. Crossed sodium dodecyl sulfate-polyacrylamide gel immunoelectrophoresis was used to estimate the levels of each of the eight forms of P-450 present in the liver microsomes of untreated rats and rats treated with phenobarbital, 5,6-benzoflavone, pregnenolone-16 alpha-carbonitrile, isosafrole, or the polychlorinated biphenyl mixture Aroclor 1254. In each situation, the sum of the levels of these eight P-450s was at least as high as the spectrally determined P-450 content. The results clearly demonstrate that individual forms of P-450 can be induced by different compounds and that a single compound can lower the level of one form of P-450 while inducing one or more other forms of P-450. Catalytic activities toward each of the substrates observed with microsomal preparations are compared to rates predicted on the basis of the content of each of the eight P-450s. These studies provide a basis for further studies on the regulation of individual P-450s, the physical properties of the different P-450s, and the metabolic consequences of changes in the forms of P-450 in rat liver models.

Nystatin-resistance of candida albicans isolates from two cases of oral candidiasis.

Two cases of oral candidiasis are described which failed to respond to nystatin therapy when used in combination with triamcinolone acetonide. The isolates of C. albicans obtained from the patients after treatment showed high in vitro resistance to nystatin when tested in combination with triamcinolone acetonide. Triamcinolone acetonide was detected in the saliva of both patients after treatment. Addition of this saliva to the isolates of C. albicans obtained after treatment was found to confer nystatin resistance. Both patients were treated with miconazole nitrate and a mycological and clinical cure was obtained in one of the cases.

Frequency of Candida albicans serotypes in patients with denture-induced stomatitis and in normal denture wearers.

A comparison has been made between the serotypes of oral Candida albicans taken from patients with denture-induced stomatitis and from a group of age and sex-matched controls. C albicans isolates were obtained from the fitting surface of the denture, the palatal mucosa supporting the denture, and the buccal mucosa. Twenty-nine of the 30 patients samples in the group affected by denture-induced stomatitis had only C albicans serotype A on their palatal mucosa and denture surface, while mixtures of serotypes were obtained from the buccal mucosa of these patients. Only one patient had serotype B isolates from the palatal mucosa and denture surface. In contrast, the control group had mixture of A and B serotypes from both the denture surface and the palatal mucosa, as well as from the buccal sites sampled. It would appear from these results that denture-induced stomatitis is associated with the proliferation of a single C albicans serotype, usually serotype A.

Purification and characterization of microsomal cytochrome P-450s.

1. Eight different forms of cytochrome P-450 have been purified to electrophoretic homogeneity. Electrophoretic, spectral and catalytic properties of these cytochrome P-450s are presented and comparison is made with preparations presented elsewhere in the literature. 2. The levels of these forms of cytochrome P-450 present in liver microsomes of rats treated with various compounds have now been quantified. Several forms of cytochrome P-450 are induced, in a more or less coordinate manner, while levels of other cytochrome P-450s are lowered, during administration of commonly used inducing agents. 3. The role of cytochrome P-450 purification and characterization studies in the understanding of the total field is discussed, along with directions in which future research is needed.

A microbiologic and ultrastructural investigation of germ-tube formation by oral strains of Candida tropicalis.

Presumptive germ tube-positive and -negative Candida tropicalis strains have been obtained from both normal children and patients who have candidiasis. Examination of these strains by conventional microbiologic tests confirmed them to be C. tropicalis. Ultrastructural examination of the filaments produced by the positive strains revealed two types: true germ tubes and those with basally constricted cytoplasm. A fluctuation analysis of the positive strains showed two subgroups: germ tube-positive and -negative. It is suggested that C. tropicalis strains may be divided into germ tube-positive and -negative strains, and that germ-tube formation must therefore not be used as a sole criterion for identification of C. albicans.

Yeast flora of the mouth and skin during and after irradiation for oral and laryngeal cancer.

The quantitative and qualitative changes occurring the fungal flora of 22 patients with oral and 9 with laryngeal carcinoma were studied during and after radiation therapy. Each patient received 6000 rad of externally applied radiation in divided doses for 5 weeks. The fungal flora was isolated from the patients' oral cavity and irradiated skin sites during irradiation and 2 weeks and 4-6 months afterwards. The number and types of fungi increased in both groups of patients after the start of irradiation and persisted at high levels for at least 4-6 months after treatment. Candida albicans and C. tropicalis were the principal yeasts isolated throughout the period studied but seven other species were also identified. All the yeast isolates were sensitive in vitro to miconazole, ketoconazole, amphotericin B and nystatin. Any of these antifungal agents should be appropriate for therapy.

An investigation into the origin and nature of 'amyloid' in a calcifying epithelial odontogenic tumour.

Fresh and fixed tissue from a resection specimen of a calcifying epithelial odontogenic tumour (CEOT) was prepared for histological, histochemical, immunological and biochemical investigation in order to study the nature of the amyloid-like material. The fixed tissue gave positive reactions with Congo-red, Thioflavin T and the dimethylamino benzene (DMAB)-method for tryptophan. The diazotization-coupling (DC) method for tyrosine was negative. The major protein purified from the unfixed tissue by sequential gel filtration had a molecular weight of 9,800. The amino acid analysis of this protein had similarities with tuft enamel protein, immune amyloid and the variable light chain component (VK). From the data obtained in this study, it is not possible to determine the precise nature of the amyloid-like material in this CEOT. However, the results do support the concept that 'amyloid' should be considered as a term describing a broad group of related proteins.
