[Clinical and diagnostic significance of microalbuminuria and activity of tubular enzyme N-acetyl-beta-D-glucosaminidase (NAG) determination in patients with hyperuricosuria].

AIM: Development of the approaches to detection of hyperuricosuric stage of purin metabolism derangement and specification of methods for early diagnosis of urate damage to the kidney. MATERIALS AND METHODS: The study included 115 young subjects whose parents suffer from gout with renal involvement or isolated urate nephropathy. Each patient was examined clinically with evaluation of family history for gout risk factors. Three times for 9 months measurements were made of uricemia and uricosuria, microalbuminuria and activity of tubular enzyme NAG. RESULTS: 45 (39.1%) patients had neither disturbances of purin metabolism nor renal affection. 70 (60.9%) patients had hyperuricosuria. In 23 (32.9%) of them microalbuminuria increased to > 20 mg/day, NAG activity to 5 u/l. Chronic tubulointerstitial nephritis was diagnosed in 17 (73.9%) patients. Six patients (26.1%) developed asymptomatic affection of the kidneys. The rest 47 patients had normal levels of microalbuminuria and NAG. There were 3 cases of hyperuricemia with microalbuminuria rising to 160-200 mg/day and further development of urinary syndrome. Hyperuricemia in them was registered at microalbuminuria higher that 160 mg/day. CONCLUSION: Microalbuminuria higher than 20 mg/day and NAG activity higher than 5 u/day are important diagnostic indicators of renal affection in hyperuricosuria including asymptomatic one. Microalbuminuria above 160 mg/day gives grounds to discuss the role of morphological changes prior to proteinuria.

[Lytic ability, in relation to gram-positive microorganisms, of a preparation isolated from a Pseudomonas lytica culture].

A lytic enzyme isolated from P. lytica was studied with respect to its effect on pathogenic grampositive and gramnegative organisms. All the grampositive organisms were lyzed by the enzyme to this or that extent. The cells of staphylococci were the most sensitive. The gramnegative organisms were resistant. It was suggested that the lytic enzyme could be used in preparing drugs for treating certain skin diseases caused by pathogenic staphylococci.

[Use of an experimental model of mixed chlamydial-gonococcal infection in ovo for evaluating the etiotropic action of chemopreparations].

The detection of the differentiated chemotherapeutic activity of tetracyclin and penicillin has been used as an example for demonstrating the possibility of using the experimental in ovo model of mixed chlamydial and gonococcal infection for the detection and primary selection of effective etiotropic preparations, simultaneously affecting Chlamydia trachomatis and Neisseria gonorrhoeae.

[Modelling of an association of Chlamydia trachomatis and Neisseria gonorrhoeae in in ovo experiments].

In experiments in ovo mixed chlamydial and gonococcal infection has been obtained by the successive infection of developing chick embryos with C. trachomatis and N. gonorrhoeae into the yolk sack. The competitive interrelations between the associated microorganisms with respect to their pathogenicity characteristics for chick embryos have not been established. This simulator is intended for use in the primary selection of etiotropic chemical preparations capable of producing combined effect on C. trachomatis and N. gonorrhoeae.

[Cultivation of Neisseria gonorrhoeae in an L-929 cell culture].

N. gonorrhoeae strain b has been found to be capable of retaining its viability in medium 199 with 10% of inactivated cattle serum added and in monolayer cell culture L-929 in the above medium. The characteristics obtained in the present investigation permit simulating the mixed association of gonococci and chlamydiae in the culture system used in this work.

[Some peculiarities of ATPase in tumor mitochondria].

The oxidative phosphorylation and ATPase activity (initial and stimulated by DNP and Mg2+) in tumor mitochondria were investigated. The intact mitochondria of Zajdela hepatoma, in contrast to liver mitochondria, exhibit the ATPase activity which is slightly stimulated by 2,4-dinitrophenol and is markedly activated by Mg2+. The mitochondria from transplantable solid tumors (adenocarcinoma 755, Iensen sarcoma, sarcoma 45) despite satisfactory morphological integrity under electron microscopy are biochemically less intact than the mitochondria of hepatoma. ATPase of these mitochondria is also slightly stimulated by 2,4-dinitrophenol and significantly by Mg2+. The ATPase activity of thymus mitochondria, the normal tissue with sufficiently high proliferative activity, corresponds to that of tumor mitochondria. The total amount of enzyme in mitochondria of tumors investigated and thymus is not lowered, since the ATPase activity in the presence of both DNP and Mg2+ corresponds to the ATPase activity of liver mitochondria. The Mg2+ ATPase activity of tumor mitochondria is not sensitive or is only partly sensitive to oligomycin. The data obtained are indicative of a high lability of the phosphorylating system in tumor and thymus mitochondria. A possibility of reorganization of the energy mechanism of tumor mitochondria and some normal tissues in connection with increased metabolism requiring high energy consumption, is discussed.
