Comparative study of the work load between one-man buses and two-man buses.

The differences in physiological and safety conditions of one-man buses and two-man buses were examined from the view point of occupational fatigue. This survey consisted of a work load study which included a time study, study of subsidiary behavior, auditory task, memory test, Galvanic Skin Response (GSR) and physiological function tests and a self-administered questionnaire which involved items concerning safety and subjective fatigue complaints. The visual and postural restrictions in the one-man bus were greater than in the two-man bus. The mental capacity of the one-man bus drivers was found to be less. Greater mental fatigue and stress were observed in the one-man bus. More subjective fatigue complaints were observed in the one-man bus. More cases of near accidents were observed in the one-man bus. From these results it was concluded that the one-man bus caused bus drivers a greater mental and physical work load.

Effects of 7-ethoxycarbonyl-4-hydroxymethyl-6,8-dimethyl-1(2H)-phthalazinone (EG 626), a new phosphodiesterase inhibitor, on the cardiovascular system of the dog.

7-Ethoxycarbonyl-4-hydroxymethyl-6,8-dimethyl-1(2H)-phthalazinone (EG 626) (0.3-3 mg/kg i.v.), a new cyclic AMP phosphodiesterase inhibitor, increased femoral, renal, coronary, carotid, vertebral and sagittal blood flows in anesthetized dogs. Systolic tension in left ventricular wall, heart rate and cardiac output also increased. These properties of EG 626 resembled those of papaverine (0.1-1 mg/kg i.v.). However, the heart rate increasing activity of EG 626 was less than that of papaverine at equipotent does on the vasodilator actions. In the isolated right atria of guinea pigs, EG 626 was more selective for increasing contractility than for increasing the sinus rate. The cardiovascular actions of EG 626 were also examined in perfused vascular beds and papillary muscle preparations of dogs. An i.v. infusion of EG 626 or papaverine (0.3 mg/kg/min) enhanced the increasing action of isoprenaline on left ventricular systolic pressure and coronary sinus outflow. The vasodilator effect of EG 626 was no suppressed by atropine, propranolol nor clemastine, and the inotropic action was not modified by pindolol. These results indicate that EG 626 may be a useful vasodilator with a cardiotonic activity but with less potency to cause tachycardia than papaverine.

Significance of a gastric mass screening survey.

We analyzed the rate of occurrence of gastric cancer in a population of 39,250 by mass screening survey. In 0.123% of the total subjects gastric cancer was detected, 0.064% was at early stage and 0.059% at an advanced stage. The incidence of gastric cancer in the newly examined persons was 0.17%, 0.08% was at early stage and 0.09% at advanced stage. Of the frequently examined persons 0.07% was found to have gastric cancer, 0.043% at early stage and 0.027% at an advanced stage. The rate of occurrence of early gastric cancer was significantly higher than that of advanced gastric cancer within a 1 1/2 year interval of screening. From these results, gastric mass screening survey is considered to be effective in detecting asymptomatic gastric cancer in the "newly examined persons," and also to be beneficial in detecting "early" gastric cancer at intervals of 1 1/2 years of screening.

[The metastatic patterns of gastric cancer from its histopathological characteristics in the primary lesion--particularly in relation to peritoneal dissemination and liver metastases].

The purpose of this study was to catagorize the metastatic patterns of advanced gastric cancer into liver and peritoneal metastases from the stromal behavior of the primary lesion in 278 patients. According to our data, peritoneal disseminations were encountered 5.0% of the medullary type, 30.0% of the intermediate type and 38.0% of the scirrhous type, while the corresponding figures for liver metastases were 28.8%, 6.0% and 1.9%, respectively. The differences among the three types were significant with both patterns of metastasis. From the present data, we assume that the histological pattern in the stroma of primary gastric cancer shows the biological behavior of cancer growth and metastatic patterns, particularly in close relation to peritoneal dissemination and liver metastases.

Phospholipid vesicle formation using nonionic detergents with low monomer solubility. Kinetic factors determine vesicle size and permeability.

The method developed previously for formation of unilamellar vesicles from mixed micelles of egg lecithin and octyl glucoside [Mimms, L. T., Zampighi, G., Nozaki, Y., Tanford, C., & Reynolds, J. A. (1981) Biochemistry 20, 833-840] has been extended to allow for (1) use of nonionic detergents with much lower critical micelle concentrations and (2) variation in the time course of detergent removal. The results demonstrate the importance of kinetic factors, especially in the determination of vesicle size: initially formed vesicles are small, but the size increases slowly thereafter if detergent is not removed too quickly. Vesicle size remains fixed when the molar detergent/lipid ratio falls below about 1/1, and detergent removal becomes increasingly difficult thereafter, presumably because flip-flop of detergent from the inner to the outer leaflet of the bilayer membrane is very slow. Residual detergent (to about 25 mol %) has surprisingly little effect on anion permeability but increases cation permeability to the point where the normal discrimination between anions and cations (in pure lipid vesicles) is lost. Detergent added to initially detergent-free vesicles readily partitions into vesicular membranes (presumably only into the outer leaflet) and has a qualitatively similar effect on permeability. Vesicles produced by this method, regardless of residual detergent level, were found to be predominantly unilamellar: no multilamellar liposomes or other lipid aggregates could be detected within the accuracy of the methods employed.

Calcium transport mechanism in crayfish gastrolith epithelium correlated with the molting cycle. II. Cytochemical demonstration of Ca2+-ATPase and Mg2+-ATPase.

Periodical changes in Ca2+-ATPase and Mg2+-ATPase activity were observed cytochemically in the crayfish gastrolith epithelium during the molting cycle in relation to the calcium transport mechanism. The ATPase activity was demonstrated by a new one-step lead citrate method. The reaction products were mainly restricted to the matrix of type II cell mitochondria. The Ca2+-ATPase activity was intensely observed in two calcium moving stages, the small gastrolith period which indicates the beginning of gastrolith formation, and the aftermolt , when the calcified gastrolith has been dissolved in the stomach and then reabsorbed from the stomach epithelium into the newly formed soft exoskeleton through the blood. Although the intensity of reaction products of Mg2+-ATPase varied in each stage, the enzymatic activity was observed throughout all molting stages. Reaction products were observed in all mitochondria, basement membranes, apical cytoplasmic membranes, and in some lysosomes. In conclusion, periodical changes in the two types of ATPase activity were seen in the mitochondria of gastrolith epithelium during the molting cycle, but Ca2+-ATPase activity seemed to be more prominently synchronized to the calcium movement in the gastrolith epithelium than Mg2+-ATPase activity. There results provide the strong evidence that Ca2+-ATPase may act strongly in the calcium transport system of crayfish molting.

The renin-angiotensin-aldosterone system and circadian rhythm of urine variables in normotensive and hypertensive subjects.

Forty-two middle-age male subjects, aged from 47 to 54 years old, were divided into three groups; 14 normotensives (NT), 16 borderline hypertensives (BHT) and 12 hypertensives (HT). The environmental effects were neutralized as much as possible by keeping their activities synchronized and their plasma and urine variables simultaneously investigated using the same protocol as under normal physiologic conditions. The circadian rhythms of urinary excretions of Na, K, Cl, creatinine, aldosterone, adrenaline and noradrenaline were analyzed by a multivariate method (cosinor method). Both systolic and diastolic blood pressure were statistically significantly different among the three groups and body weight was greater in HT than in NT. Other plasma and urine variables examined, however, were not significantly different among the three groups. A statistically significant circadian rhythm was detected in the urine variables. The rhythm characteristics, such as the mesor, amplitude and acrophase, of the urine variables except for the acrophase of urinary Na excretion were not significantly different among the three groups; the circadian acrophase of urinary Na in HT and BHT appeared significantly earlier than in NT. The pathophysiological and clinical significance of the earlier appearance of the circadian acrophase of urinary Na in hypertensive subjects still remains a unique characteristic requiring further investigation.

Effects of consecutive night-shift work on health conditions among car-cleaners of super-express trains.

The Shinkansen Super-Express trains of the Japanese National Railways, the so-called bullet trains, run from early morning until late at night. Accordingly the car-cleaners of the Shinkansen must do late night shift work. A self-administered questionnaire survey including items about health and work conditions was given to 246 workers at Osaka Station, who were then divided into 3 groups: Group A consisted of 102 night workers working 5 successive days with 2 days off, Group B consisted of 124 alternate-day 24-hour workers working 3 days a week, while Group C included 20 day-workers working 6 days a week. A subjective fatigue inventory was administered before and after work for a week to 20 workers selected from each of three groups, group A, B above group D whose shift was 2 times 2 successive night shifts with one day off, and one holiday a week. The results of the questionnaire survey revealed that the rates of complaints such as gastroenteric disorders and general fatigue were the highest in group A. The results of the subjective fatigue inventory show a tendency for the number of complaints in group A and B to increase significantly after the last work shift of the week, but not in the case of group D. The authors demonstrated that there was a close relationship between patterns of night-shift work and various kinds of health problems, and concluded that to insert a day off on the third day of the group D shift was effective in reducing the work load.

[AFP producing stomach cancer--a study of transplanted tumor in nude mice].

AFP producing cancer of the stomach was divided into 2 groups; Group A demonstrated a marked elevation of serum AFP levels before gastrectomy and was positive for AFP by PAP staining; group B showed delayed elevation of AFP levels at recurrence or in the terminal stage. Experimentally, we could establish three cell lines from group A and two lines from group B from transplanted tumor in nude mice. Tumor growth, changes of serum AFP levels and PAP staining investigated in each group. Group A and B were not only thought to exhibit different biological behavior, but also different mechanisms in AFP production. Group A cells may have the ability to secrete high levels of AFP; this may be a property of these tumor cells. On the other hand, group B tumor cells initially have little ability to secrete AFP. However, they may transform into AFP producing cells as a phenomenon of cell differentiation.

[Experimental studies on oral administration of nitrosourea anti-tumor agent, MCNU].

Experimental studies with orally administered MCNU, a water-soluble nitrosourea, yielded the following results. MCNU produced a significant increase in life span, and 60-day survivors were observed by various schedules in L1210 leukemia. The therapeutic ratios of MCNU were almost similar to those of CCNU. With Lewis lung carcinoma and Ehrlich ascites carcinoma implanted into the stomach wall, its antitumor activity by oral administration was slightly more effective than by intravenous route. In Beagle dogs, hematologic toxicity and gastrointestinal toxicity (vomiting, diarrhea) were noted by oral administration, similar to intravenous administration, but its toxicity was mild. The maximum blood level of MCNU was noted at 30 minutes after oral administration in Beagle dogs. The half life (23.7 min) by oral administration was similar to that by intravenous route. From these results, the oral administration of MCNU deserves the consideration as a form of treatment now given other MCNU routes.
