Transient spectra of intermediates in the photolytic sequence of octopus rhodopsin.

The intermediate photolytic sequence of octopus rhodopsin was studied at different temperatures and different pH values by means of a flash photolysis-rapid scan spectrophotometry near physiological temperature. The first photoproduct in the photolysis of rhodopsin was lumirhodopsin. Transformation of lumirhodopsin leads to mesorhodopsin took place independently of the pH of the solution. Mesorhodopsin was transformed to acid metarhodopsin in acid solution. In alkaline solution, mesorhodopsin was transformed to transient acid metarhodopsin whose absorption spectrum was similar to acid metarhodopsin. Transient acid metarhodopsin was then transformed to alkaline metarhodopsin reaching a tautomeric equilibrium which was determined by the pH of the solution.

Synthesis, properties and reactions of 3 beta-benzoyloxy-7 alpha-15 beta-dichloro-5 alpha-cholest-8(14)-ene.

Treatment of 3 beta-benzoyloxy-14 alpha,15 alpha-epoxy-5 alpha-cholest-7-ene (I) with gaseous HCl in chloroform at -40 degrees C gave, in 87% yield, 3 beta-benzoyloxy-7 alpha,15 beta-dichloro-5 alpha cholest-8(14)-ene (III). Reduction of the latter compound with lithium aluminum hydride in ether at room temperature for 20 min gave, in 86% yield, 7 alpha-15 beta-dichloro-5 alpha-cholest-8(14)-en-3 beta-ol (IV). The latter compound was fully characterized and assignments of the individual carbon peaks in the 13C nuclear magnetic resonance spectra of this sterol have been completed. Reduction of III with excess lithium aluminum hydride in refluxing ether for 4 days gave, in 74% yield, 5 alpha-cholesta-7,14-dien-3 beta-ol (VI). Reduction of the dichloro-steryl benzoate III with lithium triethylborohydride in tetrahydrofuran gave, in 88% yield, 5 alpha-cholest-8(14)-en-3 beta-ol (VII). A similar reduction using lithium triethylborodeuteride led to the formation of [7 beta, 15 xi-2 H2]-VIIa. Treatment of III with concentrated HCl in a mixture of chloroform and methanol gave, in 79% yield, 3 beta-benzoyloxy-5 alpha-cholest-8(14)-en-15-one (II) which was characterized as such and as the corresponding free sterol.

Assumed initiation site of fibroin gene transcription.

To map the initiation site of fibroin gene transcription on cloned genomic fibroin DNA (pFb29), we looked for precursors of fibroin mRNA, first by applying the hybridization mapping method to the RNA from the posterior silk gland of Bombyx mori. The 6.2-kilobase (kb) DNA fragment used for the assay, from 5.7 kb upstream to 0.5 kb downstream from the cap site, was end-labeled at the Xho I site (position +514) within the intervening sequence of the fibroin gene. By analyzing the labeled DNA protected by the RNA from nuclease S1 treatment, we have shown that the intervening sequence is transcribed together with the surrounding gene sequences in the posterior silk gland (producer) but not in the middle silk gland (nonproducer). Large transcripts that could include entire intervening (1 kb) and fibroin mRNA (16 kb) sequences were found. The 5' end of such transcripts was located around or near the cap locus. Synthesis and analysis of the DNA complementary to the transcripts primed within the intervening sequence region also confirmed that the 5' end of the transcripts is near the cap locus. These results strengthen the possibility that the cap site is the transcription initiation site of fibroin gene.

Purification and characterization of a lectin from rice bran.

A rice bran lectin was purified to homogeneity by precipitation with ammonium sulfate and chromatography on ovomucoid-Sepharose and CM-cellulose. The molecular weight of the dimer lectin was estimated to be around 37,000 by ultracentrifugation studies. The sedimentation coefficient was 3.8S. On Sepharose 6B gel filtration in the presence of 6 M guanidine-HCl, the lectin showed a molecular weight of 19,000. On reduction and carboxymethylation, the lectin further dissociated into two nonidentical subunits, with molecular weights of about 11,000 and 8,000. These subunits did not show hemagglutinating activity. Equilibrium dialysis experiments using N-acetyl-[1-14C]glucosamine indicated that about 1.8 mol of the sugar was bound to 19,000 g of the lectin. The lectin was mitogenic against mouse splenic lymphocytes and human peripheral lymphocytes. The lectin enhanced the rate of glucose oxidation and inhibited epinephrine-stimulated lipolysis in mouse adipocytes. Some characteristics of the lectin are compared with those of wheat germ agglutinin.

Purification and characterization of a lectin from the mushroom, Flammulina veltipes.

A lectin was purified to homogeneity from the mushroom, Flammulina veltipes, by zinc acetate treatment and CM-cellulose column chromatography. Its molecular weight was estimated to be 20,000 by gel filtration and polyacrylamide gel electrophoresis. The lectin does not contain carbohydrate, half-cystine, methionine, or histidine. On gel filtration sith Sepharose 6B in the presence of 6M guanidine-HCl, the purified lectin dissociated into two nonidentical subunits, FVA-L (molecular weight, 12,000) and FVA-S (8,000). The hemagglutinating activity was retained only in the FVA-L subunit. The lectin is mitogenic with respect to mouse spleen lymphocytes.

Echocardiographic and angiocardiographic features of supravalvular aortic stenosis in children.

Fourteen patients with SVAS served as subjects of echocardiographical and angiocardiographical studies and the data obtained were compared. There were significant decreases in aortic diameter at the level of ascending aorta compared to those of the aortic leaflets and significant differences of the percentage change of the aorta between the patients with SVAS and the normals. Echocardiography is, therefore, a useful noninvasive method for evaluating SVAS. If the values of percentage change of the aortic diameter obtained by echocardiogram are below -30%, the values of peak systolic pressure gradients across the supravalvular stenotic region should be higher than 20 mmHg. The value of LV M/V under 1.0 suggests that the pressure gradient is lower than 20 mmHg.

Effect of dietary proteins on the turnover of rat liver argininosuccinate synthetase.

The rates of synthesis and degradation of arginosuccinate synthetase in rat liver under various dietary conditions were determined. The relative rate of the enzyme synthesis in the livers of rats fed on 70% casein diet was 4.0 times greater than that for rats fed on 5% casein diet. The rate constants of degradation (Kd of argininosuccinate synthetase were estimated to be 0.15 and 0.16 day-1 under 70% and 5% casein feeding, respectively. When the dietary conditions were changed acutely from 70% to 5% casein diet or vice versa, the rates of the enzyme synthesis decreased or increased, respectively, and the rates of enzyme degradation were also affected. The change from 5% to 70% casein diet caused a transient decrease in the rate of degradation. After the enzyme activity had achieved a new steady-state level, the enzyme degradation proceeded at the normal steady rate. On the other hand, the change from 70% to 5% casein diet caused a transient increase in the rate of degradation. Thus, the only factor regulating the amount of enzyme in rat liver is the rate of enzyme synthesis under the steady-state conditions. However, the rates of both enzyme synthesis and degradation are involved in the regulation of the amount of enzyme during dietary transition.

Rat liver glutamine 5-phosphoribosyl-1-pyrophosphate amidotransferase [EC 2.4.2.14]. Purification and properties.

Glutamine 5-phosphoribosyl-1-pyrophosphate (PRPP) amidotransferase (amidophosphoribosyltransferase), [EC 2.4.2.14] was purified 1,600-fold from rat liver. The preparation gave two protein bands on acrylamide gel electrophoresis, of which only the main band showed enzyme activity. The molecular weight of the enzyme was estimated to be 215,000, 200,000, and 195,000 by Sephadex G-150 gel filtration, polyacrylamide gel electrophoresis, and sucrose density grandient ultracentrifugation, respectively. The apparent Km values for glutamine and PRPP were 1.24 mM and 0.57 mM, respectively. The concentration-activity curve for PRPP changed from a hyperbolic to a sigmoidal form on addition of AMP or GMP, and this inhibition by AMP was prevented by increasing the PRPP concentration. In the presence of high concentrations of inorganic phosphate, the catalytic activity was decreased and the sensitivity to AMP inhibition was slightly increased. The molecular size of liver amidotransferase was not changed by the addition of PRPP, AMP, or 2-mercaptoethanol. The purified rat liver enzyme has a broad pH-range of activity between 6.5 and 8.5.

Inhibition of sterol biosynthesis in animal cells by 14 alpha-alkyl-substituted 15-oxygenated sterols.

Reported herein are the results of investigations of the effects of a number of 14 alpha-alkyl-substituted 15-oxygenated sterols, prepared by chemical synthesis, on sterol biosynthesis and the levels of 3-hydroxy-3-methylglutaryl CoA reductase activity in L cells and in primary cultures of fetal mouse liver cells grown in serum-free media. Several of the compounds, most notably 14 alpha-ethyl-5 alpha-cholest-7-en-3 beta, 15 alpha-diol and 14 alpha-ethyl-5 alpha-cholest-7-en-15 alpha-ol-3-one, were found to be extraordinarily potent inhibitors of sterol synthesis in these cells. For example, the latter compound caused a 50% inhibition of the incorporation of labeled acetate into digitonin-precipitable sterols in L cells in culture at a concentration of 6 X 10(-9) M.

Reconstitution of hybrid toxin from Fragment A of diphtheria toxin and a subunit of Wistaria floribunda lectin.

A hybrid protein was prepared in which Fragment A of diphtheria toxin was linked to a monovalent subunit of the lectin from Wistaria floribunda seeds by a disulfide bridge. The hybrid molecule was reconstituted effectively from a reduced mixture of Fragment A and the lectin subunit by oxidation with o-phenanthroline and CuSO4. The monovalent hybrid protein was partially purified and found to be toxic to L cells. The toxicity of the hybrid protein on the cells was blocked by the addition of either N-acetyl-D-galactosamine, which specifically binds to the lectin, or by anti-Fragment A antibody.

Kinetic study of photoregeneration process of digitonin-solubilized squid rhodopsin.

In the photoregeneration process of squid rhodopsin, an intermediate has been found at neutral pH values (phosphate buffer) with a flash light (lambda greater than 540 nm). An intermediate R430, with the 11-cis retinal as chromophore, is produced from metarhodopsin in light and is converted to rhodopsin through the processes R430 leads to P380 and P380 leads to rhodopsin. The pH dependence of the velocity of the conversions suggests that processes R430 leads to P380 and P380 leads to rhodopsin involve a protolytic reaction and that the ionized group is a histidine residue of opsin. Kinetic parameters show that the largest conformational change in opsin occurs in the conversion of R430 leads to P380.

Induced synthesis of immunoglobulin messenger RNA accompanies induction of immunoglobulin production in cultured mouse spleen cells.

Immunoglobulin kappa type light chain mRNA (Lkappa mRNA) accumulated in parallel with secretion of immunoglobulin M in cultured mouse spleen cells activated by lipopolysaccharide. Actinomycin D suppressed the accumulation of kappa chain mRNA completely without affecting the degradation rate of kappa chain mRNA. The half life of kappa chain mRNA was about 9 h. Available evidence indicates that lipopolysaccharide stimulates de novo synthesis of kappa chain mRNA. The accumulation of kappa chain mRNA was markedly suppressed by inhibitors of DNA or protein synthesis such as hydroxyurea, cytosine arabinoside and cycloheximide.

Accumulation of immunoglobulin messenger ribonucleic acid in immunized mouse spleen.

We have measured the concentration of mRNAs coding for immunoglobulins, k and lambda type light chains and gamma 1 type heavy chain, in mouse spleen cells activated by bacterial lipopolysaccharide or sheep red blood cells. These mRNAs were quantitated by hybridization to radioactive DNA complementary to highly purified immunoglobulin mRNAs from mouse myelomas. In the lipopolysaccharide-stimulated spleen cells, only light chain mRNA accumulated, whereas gamma 1 type heavy chain mRNA remained unvaried. The light chain mRNA concentration also increased in purified bone-marrow-derived lymphocytes. The lipopolysaccharide-induced light chain mRNA was similar to light chain mRNAs purified from myelomas. The accumulation and disappearance of light chain mRNA in bone-marrow-derived lymphocytes coincide with the kinetics of synthesis of immunoglobulin M which is the major species induced by lipopolysaccharide. In sheep red blood cell stimulated spleen, the specific accumulation of k type light chain and gamma 1 type heavy chain mRNAs parallels immunoglobulin G synthesis. These results seem to indicate that the increment of immunoglobulin mRNA concentration in bone-marrow-derived lymphocytes is important for induction of immunoglobulin synthesis.
