Hyaluronuria in a case of progeria. (Hutchinson-Gilford syndrome).

A classic case of progeria (Hutchinson-Gilford syndrome) in a 9-year-old Japanese boy is presented. The characteristic clinical features in this patient were similar to those reported in the literature. The total amount of acid glycosaminoglycans excreted in the urine was within the normal range, but there was an increase in hyaluronic acid excretion. The hyaluronuria was a novel finding in progeria, providing a common linkage with the hyaluronuria found in Werner's syndrome.

Epithelial dysplasia in cancerous and noncancerous esophagi.

Subserial examinations of the noncancerous esophagus from 86 autopsy materials (male 47, female 39) in Kagoshima Prefecture revealed epithelial dysplasia in 54 instances (63%); 32 males (68%) and 22 females (57%). Higher grades of dysplasia were shown in 17 males (36%) and 3 females (8%). Histologically a formation of the rete ridge-like elongation of basal epithelium was characteristically conspicuous in these materials. Tabacco and alcohol intakes were regarded as promoting factors of dysplasia. Distribution and severity of dysplasia around carcinoma were histologically examined on semiserial blocks in 100 surgical materials under the consideration of preoperative treatment. A coexistence of carcinoma with extensive dysplasia was found in 24 instances and that with multifocal dysplasia in 46. Multicentric in situ carcinoma was found in 19 patients. These in situ carcinomas were frequently surrounded by severe dysplasia and occasionally showed gradual transition to the latter. The lesion of invasive carcinoma sometimes did not show sharp demarcation from the surrounding mucosa, especially in cases of well differentiated squamous cell carcinoma. The results indicate that dysplasia plays a significant role on carcinogenesis of the esophagus as a precursor lesion.

Increased excretion of urinary glycosaminoglycans in a case of Rothmund Thomson syndrome.

The exercretion of urinary total glycosaminoglycans (GAG) in a case of Rothmund Thomson syndrome associated with osteosarcoma was increased about 2--3 times that of normal control. Since the excretion of urinary total GAG in two cases of osteosarcoma was within normal range, the increased excretion of total GAG might be on account of the metabolic disorder of GAG in Rothmund Thomson syndrome. The prominent fractions obtained by Dowex 1 column chromatography from this syndrome were 0.75 M Fr and 1.0 M Fr, in which the major GAG were indicated to be partially degraded forms of heparan sulfate and chondroitin sulfate, respectively.

Hemangiosarcoma of the spleen and liver metastases: angiographic manifestations.

The arteriographic findings in two cases of hemangiosarcoma of the spleen with liver metastases are described. These are the second and third reported cases of angiographically demonstrated malignant splenic tumors of vascular origin. Despite the absence of tumor vessels, there were multiple vascular takes in the arterial through the venous phase, mimicking benign cavernous hemangioma. Multiple metastases in the liver were the only clue to malignancy. Arteriography combined with liver-spleen scintigraphy not only provides information which is valuable in the preoperative diagnosis but also aids in management of the patient.

Separation and properties of the NAD-linked and NADP-linked isozymes of succinic semialdehyde dehydrogenase in Euglena gracilis z.

Euglena gracilis z contained two succinic semialdehyde dehydrogenases (EC 1.2.1.16), one requiring NAD and the other NADP, and these isozymes were separated from each other and partially purified. The NAD-linked isozyme was relatively stable on storage at 5 degrees C whereas the NADP-linked one was extremely unstable unless 30% glycerol or ethyleneglycol was added. The optimum pH was 8.7 and optimum temperature 35-45 degrees C for both isozymes. They were inhibited by Zn2+ and activated, particularly the NAD-linked enzyme, by K+. Sulfhydryl reagents activated both isozymes. The Km values for succinic semialdehyde were 1.66 - 10(-4) M with the NAD-linked isozyme and 1.06 - 10(-3) M with the NADP-linked one. The NADP-linked isozyme was induced by glutamate while the NAD-linked one was not. Probable roles of these isozymes in the physiology of Euglena gracilis are discussed.

Postmortem study of a case of Werner's syndrome.

The postmortem study of a classic case of Werner's syndrome is presented. The subject was a Japanese man who died at the age of 39. Major findings included general microsplanchnia, extreme atrophy of the testes and skin, calcified aortic atherosclerosis, an increase of basophils in the pituitary, aspiration pneumonia, chronic pyelonephritis and a meningioma in the occipital area of the brain. Histologically, no osteoporosis was evident in the lumbar spine and iliac bone. The findings suggest that in Werner's syndrome the dominant pathologic factor may be found in connective tissue other than bone.

[Angiographic findings of tentorial branches of the internal carotid artery in gliomas infiltrating the tentorium (author's transl)].

The tentorial branches, originating from the cavernous portion of the internal carotid artery, showed pathological findings in two cases of brain tumors infiltrating the tentorium: a glioblastoma multiforme of the temporooccipital and basal regions and a medulloblastoma diffusely involving the cerebellar hemisphere, vermis brachium pontis and pons. The value and importance of the tentorial branches are emphasized in the diagnosis of glioma infiltrating the tentorium. The percentage of the visualization and the measurements of the visualized segment of these branches were described using selective internal carotid angiograms by the transfemoral catheter technic on 50 presumably normal adults. The percentage of the visualization of the tentorial branches was 24 percents and the average visualized segment measured 15 mm with a range of 5 to 30 mm on conventional angiograms.
