Study of the evolution of acute phase reactants and of thromboxane and prostacyclin during calcium pyrophosphate-induced pleurisy in the rat.

By using a clearly defined model of acute inflammation in rats, CaPP-induced pleurisy, we compared the evolution of four APR levels (Hp, HpX, alpha 2M, SAP) at a local and peripheral site. At the same time we studied thromboxane and prostacyclin concentrations in pleural exudate. The levels of APR have shown a progressive increase up to 24 h. For alpha 2M, Hp, HpX the increase in serum concentration was more rapid than in exudate, whereas the evolution of SAP level was parallel in the intra and extra-vascular compartments. On the other hand the levels of albumin and transferrin which are not APR remained constant in serum and also in exudate from 6 h to 24 h. Thromboxane and prostacyclin levels reached a peak during the first two hours of the inflammatory reaction followed by a rapid decreased. These prostanoids may be considered as early and transient mediators of acute inflammation, whereas the acute phase proteins studied have a more prolonged role in the inflammatory reaction.

Effect of immunomodulating agents on leucocyte chemotaxis and cyclic nucleotides.

We have studied the effect of immunomodulating agents on polymorphonuclear leucocyte chemotaxis and their relation to the modification of cyclic nucleotide levels. The tested drugs (levamisole, tuftsin, azimexon, muramyl dipeptide, isoprinosine) inhibited the chemotaxis of 'normal' cells but restored the impaired chemotactic responsiveness of inflammatory cells. None of these drugs had any significant effect on cyclic nucleotide levels in 'normal' cells. All the drugs, except isoprinosine, produced an increase in the cGMP levels in inflammatory cells. These results suggest that immunomodulators are able to modify PMN chemotaxis. This effect cannot, however, be related to modification of the cyclic nucleotide levels.

[Generalized lymph node sarcoma during course of an actinic reticuloid (author's transl)].

A 50 year old man with actinic reticuloid developed a diffuse malignant lymphoma 7 years after the beginning of the dermatosis. Biopsy of lymph node showed a polymorphic proliferation with lymphocytes, numerous reticular cells and several Sternberg-like cells. An ultrastructural study showed an indifferentiate lymphocyte proliferation and numerous malignant reticular cells without the Sternberg cell picture. In this patient we discuss the role of immunosuppressive therapy, chronic herpes virus infection and chronic antigenic photostimulation. Up to now, this is the third case of an association between actinic reticuloid and malignant lymphoma.

[The effect of mepyramine and 48/80 on the histamine content of pleural exudates in the rat].

The influence of pretreatment with the antihistaminic, mepyramine, or with the histamine liberator 48/80 was studied on the volume and the histamine content of pleural exudates induced in rats by intrapleural injection of turpentine, silver nitrate or carrageenan. The fluorometric determination of histamine was performed in the exudates collected at different times after injection of the irritant. In control animals the histamine levels were different from those previously found by the authors using biological assays. In rats pretreated with mepyramine or compound 48/80, the effects observed varied according to the nature of the irritant used to induce pleurisy.

Mapping of the X-breakpoint involved in a balanced X;12 translocation in a female with mild mental retardation.

Balanced chromosomal abnormalities such as translocations and inversions have been identified in many genetic diseases. Cloning of the breakpoints involved in these abnormalities has led to the identification of the disease-related genes. Recent reports suggest the presence of a mental retardation locus at Xq11-12. We have identified a female patient with a balanced translocation t (X;12) (q11;q15) associated with mild mental retardation. We identified a yeast artificial chromosome spanning the X-chromosome breakpoint by using fluorescent in situ hybridization techniques. A cosmid library of this YAC has been constructed and the search for candidate genes is in progress.
