In vitro inhibition of Giardia lamblia and Trichomonas vaginalis growth by bithionol, dichlorophene, and hexachlorophene.

Bithionol, dichlorophene, and hexachlorophene, which are used in treating some helminthic infections, killed trophozoites of Giardia lamblia and Trichomonas vaginalis in modified BI-S-33 and Asami media, respectively. Virtually all G. lamblia and T. vaginalis cells were killed within 24 h with a 0.42 mM concentration of these compounds, except that 0.93 mM dichlorophene was required for sterilizing T. vaginalis in the same period. In modified BI-S-33 and Asami media from which bovine and human sera were omitted, respectively, the inhibitory actions of the compounds against in vitro growth of these protozoa were significantly enhanced. Trophozoites of G. lamblia and T. vaginalis could be killed in shorter than 10 min with 0.074 mM dichlorophene and 0.0025 mM hexachlorophene, respectively, in serum-free media. G. lamblia, which was incubated in the complete medium containing dichlorophene, showed a characteristic swelling of the ventral side which led to disruption of the parasite, whereas bithionol caused a thin crack in the cytoplasm of T. vaginalis incubated in Asami medium. The crack appeared to enlarge and result in vacuolization of T. vaginalis. These observations suggest that bithionol, dichlorophene, and hexachlorophene merit further evaluation to ascertain whether they are useful for treatment of giardiasis and trichomoniasis.

Evaluation of a new bronchodilator, Formoterol, using biochemical parameters.

The authors administered orally 40, 60 and 80 micrograms of Formoterol which was developed as a new bronchodilator and 4 mg of Salbutamol which is considered to be a beta 2-selective drug, to healthy adults. Pulse rate, blood pressure and blood levels of cyclic GMP, free fatty acid, cyclic AMP, glucose and lactic acid were measured in order to evaluate efficacy and dosage of Formoterol and to compare efficacy of this drug with that of Salbutamol. As a result, it is considered that Formoterol has a strong dose dependent beta 2-effect with 40 micrograms of Formoterol almost as high as 4 mg of Salbutamol in beta 2-effect. In addition, Formoterol has almost no alpha-effect and its beta 1-effect can be considered negligible in the doses of 60 micrograms and 80 micrograms.

[Fundamental and clinical studies of a sustained release preparation of cefaclor in the surgical field].

Fundamental and clinical studies of S6472 (sustained release preparation of cefaclor (CCL] were conducted in the surgical field and it was confirmed that the preparation is a useful drug. The following is the summary of the results from the fundamental and clinical studies: In vitro antibacterial activity. CCL showed MICs of 0.78 to 6.25 micrograms/ml against almost strains of S. aureus, E. coli and Klebsiella isolated from surgical wound regions, and the antibacterial activities were stronger than those of cephalexin (CEX). Clinical efficacy. S6472 was orally administered to 33 patients with skin and soft tissue infections in 2 divided doses. As a result, excellent clinical response was observed in 13 patients, good response observed in 14 patients, fair in 4 and poor in 1. The clinical efficacy in 1 of the 33 patients was unknown. Overall clinical effective rate was 84.4%. Adverse reaction. In 2 patients, mild gastrointestinal symptoms were observed.

Lacrimal gland accumulation of 67Ga-citrate in patients with Sjögren's syndrome.

The extent of 67Ga accumulation in the two lacrimal glands in patients with keratoconjunctivitis sicca (KCS) of Sjögren's syndrome was studied. Of the two main groups one consisted of 69 subjects without ophthalmic complaints (control group), the other consisted of 26 patients with KCS of Sjögren's syndrome. Of the 26 patients with KCS, 7 had been diagnosed as probable KCS (probable sub-group) and the other 19 had been diagnosed definite KCS (definite sub-group). About 3 mCi (111 MBq) 67Ga-citrate was injected IV into each subject and this was followed by scintigraphy at 24, 48, and 72 h after the injection of 67Ga. A positive finding in the lacrimal gland was noted in 64 of 69 subjects (92.7%) in the control group and in 7 of 7 patients (100%) with probable KCS. Three of 19 patients with definite KCS (15.7%) showed positive findings under scintigraphy. When the scintigraphic finding in the lacrimal gland is not positive in patients with suspected KCS of Sjögren's syndrome, they can then be diagnosed with little risk as definite KCS cases. Shirmer's test was performed on subjects in the probable and definite groups. There was statistical significance between the positive and equivocal or negative scintigraphic finding and Schirmer's values. These results suggest a correlation between gallium accumulation in the lacrimal gland and the tear production.

Harmonic-intensity analysis of normal and hoarse voices.

Objective evaluation of normal and hoarse voices is performed considering the characteristic that hoarse voices show a prominent fundamental frequency intensity compared with harmonics in the voice spectrum. The relative harmonic intensity Hr, obtained from a stable portion of the sustained vowel/a/, is defined as the intensity of the second and higher harmonics expressed as a percentage of the total voice intensity. Ninety-five percent of the normal voices examined have Hr larger than the critical value of 67.2%, whereas 90% of the hoarse voices have Hr smaller than the critical value. The harmonic-intensity analysis thus provides good discrimination between normal and hoarse voices.

[An evaluation of radioactive colloid for measuring the volume of effective liver].

A methods for liver scintigraphy for finding the effective liver volume were developed. Matrix ROI (region of interest) was set on the anterior liver scintigram with 99mTc tin colloid and 198Au colloid, respectively. The various depths of liver under matrix ROI were calculated from the count rate noting the absorption coefficient. The effective liver volume was the sum total of the areas of matrix ROI to the various depths. In order to check the accuracy of this method, the effective liver volume was calculated using the liver phantom and the patient's liver scintigraphy. The volume obtained for the liver phantom was nearly equal to the real volume. The liver volume in 19 patients using 99mTc tin colloid and 198Au colloid showed good correlation with the volume as measured by computed tomography (CT) scan (r = 0.841, r = 0.749).

Health status of Indochinese refugees in Japan: statistical analyses on anemia, eosinophilia and serum alkaline phosphatase.

Detailed statistical analyses were attempted on anemia, eosinophilia and elevation in serum alkaline phosphatase in the Indochinese refugees in Japan. A high statistical correlation was found between hemoglobin concentration and hematocrit for most of the refugees except the Cambodian females. Refugees with parasitic infections, particularly hookworm infection had significantly lower values for hemoglobin concentration and hematocrit. Significant eosinophilia was observed in the refugees infected with hookworms, Strongyloides stercoralis, Rhabditis sp., Endolimax nana and/or Hymenolepis nana. However, these parasites were found in the same subpopulations more frequently than expected by chance. Elevation in serum alkaline phosphatase was observed in young Cambodian immigrants. Analyses showed that the variations of this enzyme could be well predictable by those of enzymes such as serum glutamic oxaloacetic transaminase, which are good markers for liver functions, in Cambodian immigrants, 13-24 of age, whereas no such correlation was observed in non-Cambodians of the same age group.

Cerebral circulatory and cardiac effects of vinpocetine and its metabolite, apovincaminic acid, in anesthetized dogs.

Cerebral circulatory and cardiac actions of vinpocetine and its metabolite, apovincaminic acid (AVA), were studied in anesthetized dogs. Injections of vinpocetine (0.03 to 1.0 mg/dog) into the vertebral or coronary artery produced dose-dependent increases in the blood flow of each artery. The action was more prominent in the vertebral than in the coronary artery. Intravenous bolus injections of vinpocetine (0.3 and 1.0 mg/kg) transiently increased vertebral flow and superior sagittal sinus flow velocity in a dose-dependent manner. Femoral flow was slightly increased with a fall in diastolic pressure. AVA (1.0 to 5.0 mg/kg) did not significantly increase vertebral, but increased femoral flow, which was accompanied by systemic hypotension. A similar trend of actions was noted after i.v. infusion of both agents (0.15 and 0.5 mg/kg/min for 30 min). An i.d. administration of vinpocetine (10 mg/kg) increased vertebral and regional cerebral cortical flows, accompanying with a fall in diastolic blood pressure. Femoral flow was not affected. An i.v. bolus injection of vinpocetine (1, 3 and 10 mg/kg) transiently decreased systemic blood pressure, left ventricular maximum dp/dt and heart rate in a dose-dependent manner. Cardiac output tended to be increased, and total peripheral resistance decreased. Coronary flow was transiently increased at 1 and 3 mg/kg, but tended to be decreased at 10 mg/kg. AVA, in the same dose range, decreased systemic blood pressure and total peripheral resistance, and increased cardiac output and coronary flow in a dose-dependent manner. An i.v. infusion of vinpocetine (0.5 mg/kg/min for 30 min) decreased systemic blood pressure, left ventricular maximum dp/dt, total peripheral resistance and heart rate.(ABSTRACT TRUNCATED AT 250 WORDS)
