Antineuroblastoma monoclonal antibody (MoAb) was labeled with iodine-131 (131I) and injected into transplantable human neuroblastoma-bearing nude mice in vivo. Imaging of the tumor by gamma camera was then attempted. At the fourth day after injection of the antibody, relatively heavy labeling was observed in the tumor. Principal organs were removed and their 131I uptake was determined. A high radioactivity count was detected in the tumor, indicating efficacy of this antibody in tumor imaging of neuroblastoma. This antibody was labeled with 131I and injected into transplantable human neuroblastoma-bearing mice. Apparent inhibition of tumor growth was observed in animals treated as such compared with animals of the control group, MoAb alone group, and 131I alone group (P less than 0.05). In addition, pathohistologic study showed binding of 131I to tumor cells and necrotic changes, suggesting the possibility of application of this MoAb to the treatment of neuroblastoma.
1. The effects of a noradrenaline precursor, L-threo-3,4-dihydroxyphenylserine (L-DOPS), on spinal mono-(MSR) and polysynaptic reflexes (PSR) and decerebrate rigidity, were studied. 2. Although a low dose of L-DOPS (10 mg/kg, i.v.) did not affect MSR or PSR in C1 spinal rats, high doses of L-DOPS (50 and 100 mg/kg, i.v.) moderately enhanced the amplitudes of both MSR and PSR. 3. Clorgyline-HCl (1 mg/kg, i.v.), an MAO inhibitor, enhanced the excitatory effects of low-dose L-DOPS (10 mg/kg, i.v.) on both reflexes. 4. Benserazide-HCl (1 mg/kg, i.v.), an L-aromatic amino acid decarboxylase inhibitor, decreased the pressor effect, but not the stimulatory effects, of L-DOPS (100 mg/kg, i.v.) on MSR and PSR. 5. L-DOPS (300 mg/kg, i.p. or i.d.) did not affect the muscle tone of rigid hindlimbs caused by radio frequency lesioning of the midbrain. 6. These results suggest that the moderate enhancing effects of L-DOPS on MSR and PSR are due to conversion of L-DOPS to noradrenaline in the spinal cord.
Various 17 alpha-ethynylsteroids were prepared and derivatized as the corresponding triethylsilyl compounds 2-35, which were examined for a ratio of antifertility to estrogenic activity that would be more beneficial than that of the presently used agent. Among the triethylsilyl compounds evaluated, only 23 displayed this desired ratio, although two other compounds without the triethylsilyl moiety, 18 and 26, shared similar characteristics.
The antihypertensive activity of a new angiotensin converting enzyme (ACE) inhibitor, CV-5975, (R)-3-[(S)-1-carboxy-5-(4-piperidyl)pentyl] amino-4-oxo-2,3,4,5-tetrahydro-1,5-benzothiazepine-5-acetic acid, was examined in normotensive rats and various hypertensive animal models. In spontaneously hypertensive rats, CV-5975 (1 to 10 mg/kg, p.o.) had a dose-related, sustained antihypertensive action, which was more potent and longer than that of enalapril. The potency and duration of action of CV-5975 was intensified when it was administered repeatedly or combined with hydrochlorothiazide. CV-5975 (1 mg/kg, p.o.) inhibited the ACE activity of plasma and tissues; inhibition on the ACE activity of the aorta, kidney, and brain was marked when CV-5975 was administered repeatedly. In 2-kidney, 1 clip hypertensive rats (1 to 10 mg/kg, p.o.) and dogs (0.3 and 1 mg/kg, p.o.), CV-5975 had a marked, sustained antihypertensive action, which was more marked than that of enalapril. In normotensive rats (10 mg/kg), 1-kidney, 1 clip hypertensive rats (3 and 10 mg/kg), and hyporeninemic DOCA/salt hypertensive rats (1 to 10 mg/kg/day), CV-5975 administered orally once or repeatedly reduced blood pressure, whereas enalapril did not. These results indicate that CV-5975 is a potent and long-lasting antihypertensive agent, the action of which is mediated primarily by inhibiting ACE activity and partly by some unknown mechanisms.
CV-5975, (R)-3-[(S)-1-carboxy-5-(4-piperidyl)pentyl]amino-4-oxo- 2,3,4,5-tetrahydro-1,5-benzothiazepine-5-acetic acid, was found to inhibit rabbit lung angiotensin converting enzyme (ACE) activity with an IC50 of 3.1 x 10(-9) M and a Ki of 2.6 x 10(-9) M, inhibit the angiotensin I (A-I)-induced contraction of the guinea pig ileum with an IC50 of 1.3 x 10(-8) M, and augment the bradykinin (BK)-induced contraction of the ileum with an AC50 of 9.2 x 10(-10) M. The activity of CV-5975 was comparable to or slightly more potent than that of enalaprilat. The overall inhibition constant (Ki*), calculated from a steady-state analysis of enzyme reactions, was 4.4 x 10(-12) M for CV-5975; this indicates that the inhibition was about 5 times more potent than that of enalaprilat (2.0 x 10(-11) M). In rats, CV-5975 (0.03 and 0.3 mg/kg, i.v. and 3 and 10 mg/kg, p.o.) inhibited the A-I-induced pressor action more potently and for a longer period than did the corresponding doses of enalaprilat and enalapril. CV-5975 and enalapril (3 mg/kg, p.o.) augmented the BK-induced depressor action to a similar extent. In dogs, CV-5975 (0.3 and 1 mg/kg, p.o.) markedly inhibited the A-I-induced pressor action in a dose related manner, and the duration of this inhibitory activity was longer than with the corresponding doses of enalapril. These data provide evidence for the proposal that CV-5975 is a highly potent and long lasting ACE inhibitor.

APA is a common cause of constipation, and is the mild case of the imperforated anus. On diagnosing APA, anterior displacement of the anus and normal distribution of the external anal sphincter to the anus are essential. To determine the location of the anus in the perineum simple clinical technique was developed. In 61 normal cases, the result of measurement was almost same in each sex, but in 3 APA cases the location were anteriorly dislocated than normal cases. Distribution of the external anal sphincter was evaluated with electromyographic technique, and location map of the sphincter was made. In 3 APA cases, the anus was totally surrounded by the external anal sphincter, but in 17 ano-cutaneous fistula cases, the opening was anteriorly dislocated to the sphincter distribution map. As a conclusion, newly proposed simple clinical technique to determine anal location and electromyographic examination of the external and sphincter distribution are very useful in objective diagnosis of APA.
The rate of clearance (K value) of 99mTc tin colloid in the liver differentiates normal subjects from liver cirrhosis patients; so 99mTc tin colloid is as useful as 198Au colloid as a marker of liver function. There are several reports concerning volume estimation using liver scintigraphy. Our original method was devised to measure the effective liver volume by scintigraphy. By combining the K value with effective liver volume, a predictive index was obtained in order to predict the residual liver function before hepatic resection. The index in 24 patients with liver diseases was investigated before hepatic resection. Three of them died due to hepatic failure after hepatic resection. The indices were between 0.40 and 0.45 in two of these three patients and 0.338 in one. Among the patients without hepatic failure, the indices showed more than 0.45 in 19 patients and between 0.40 and 0.45 in two. These results indicate that the limitation of hepatic resection is between 0.40 and 0.45 of the predictive index.
Delayed primary operation has been started since 1982 in our department and 19 cases of solid tumor in childhood were treated with this method. They included 11 cases of advanced neuroblastoma, 5 cases of hepatoblastoma and 3 cases of yolk sac tumor in sacrococcygeal area. We discussed effectiveness of preoperative chemotherapy to the primary tumor and metastatic lesions, the best timing of operation after chemotherapy and the operative technique in delayed primary operation. Preoperative chemotherapy were useful in almost all cases of delayed primary operation, so the primary lesion and lymph nodes metastases were removed easily. We recommended it was the best timing of operation when 2 or 3 courses of preoperative chemotherapy were finished. CUSA (Cavitron Ultrasonic Surgical Aspirator) was the useful instrument to remove the primary tumor and lymph node metastases completely in advanced neuroblastoma preserving both kidneys.

The Antineuroblastoma monoclonal antibody CNM-5 was submitted to preclinical studies and then used for tumor imaging with following results. 1. CNM-5 is an antibody which belongs to the IgM fraction and has a molecular weight of 900 kilodaltons. Its protein concentration was found to be 17.99 mg/dl. 2. The 131I-labeled antibody was injected into nude mice transplanted with human neuroblastoma, and imaging was performed by a gamma camera. Accumulation of 131I in the tumor was relatively clearly seen. Various organs were removed and examined for 131I uptake. Radioactivity was easily detected in the tumor, suggesting that CNM-5 is effective for use in imaging diagnosis of neuroblastoma.
The interaction of cis-diamminedichloroplatinum II (c-DDP) and a course of 5 daily irradiations was investigated in the SCCVII/St tumor and normal tissues (duodenal crypt cells and lung) of the C3H mouse. Two schedules with daily doses of 2.4 mg/kg c-DDP given immediately before 4 Gy X ray on 5 consecutive days and a single 12 mg/kg c-DDP dose followed 24 hr later by the first of 5 daily 4 Gy X ray doses produced the most consistent and significant supra-additive effects on the SCCVII tumor. This supra-additive effect was also achieved with lower and much less toxic drug doses. These schedules produced high enhancement ratios (dose effect factors DEF) for mouse duodenal crypt cells, but the degree of enhancement was less than that for the SCCVII tumor. Schedules with a 72-hr interval between drug and radiation treatments, which produced low enhancement ratios for the SCCVII tumor and duodenal crypt cells, gave high enhancement ratios for the lung. It is concluded that c-DDP has the potential of enhancing the radiation effect on normal tissues, and the degree of enhancement depends upon the interval between X ray and c-DDP. The enhancement ratios for the SCCVII tumors are greater than for normal tissues, and this results in high therapeutic gain factors (TGF). Comparing the effects on tumor with those on normal tissues, it may be seen that there is clinical usefulness in simultaneous combination treatments and perhaps moreso in the administration of a single drug dose 24 hr before the first of 5 daily X ray fractions.
A number of silicon-substituted analogues of ethynylestradiol that exhibit modified and enhanced biological activities have been synthesized. Particularly noteworthy are a group of [(trialkylsilyl)ethynyl]estradiol analogues that exhibit high antifertility potency and markedly reduced estrogenic activity. The best compounds synthesized are 17 alpha-[(triethylsilyl)ethynyl]estradiol (5) and 17 alpha-[(tert-butyldimethylsilyl)ethynyl]estradiol (33), which show a separation of antifertility from estrogenic activity in the rat. The results of structure-activity studies indicate a good correlation between the observed biological activities and the calculated van der Waals volumes of the three variable silicon substituents.
Fundamental and clinical characteristics of 3 kinds of high-sensitivity immunoradiometric assay (IRMA) kits for thyroid stimulating hormone (TSH). i.e., RIA BEADS II (kit A), TSH kit Daiichi II (kit B) and Ab tube TSH 'Eiken' (kit C) and one conventional radioimmunoassay (RIA) kit, i.e., TSH kit Daiichi (kit D), were studied. In the recovery test and the reproducibility test, there was no significant difference between the 4 kits. The sensitivities of kits A, B and C were much higher than that of kit D, and those IRMA kits were sensitive enough to distinguish hyperthyroidism from normal samples. For low concentrations of TSH (less than 5 microU/ml), the data from kits D, B, C and A tended to show higher values in that order. The correlation between the data measured by kits B and D, and the tendency of kit A toward lower values agreed well with other reports.
