Factors influencing the release of renin in patients under chronic dialysis treatment.

Plasma renin activities (resting PRA, post-dialysis delta PRA) were studied in 61 patients under chronic dialysis treatment. Removed sodium and removed water were estimated at each dialysis. Delta PRA/removed-sodium and delta PRA/removed-water were calculated as indices in response to the removal of sodium and water during the dialysis. 1)Resting PRA (pre-dialysis PRA) was positively correlated to delta PRA/removed-sodium, delta PRA/removed-water, serum osmolality, and diastolic blood pressure, but negatively to serum sodium concentration, age, and pulse pressure/diastolic blood pressure. Statistically significant factors controlling the resting PRA were delta PRA/removed-sodium, delta PRA//removed-water, and serum sodium concentration. Resting PRA was slightly correlated to diastolic blood pressure and age. 2)Post-dialysis PRA was significantly correlated to the resting PRA, delta PRA/removed-sodium, delta PRA/removed-water, serum sodium concentration, and age, but not to the blood pressure indices.

2-Amino-5-methyl-5-hexenoic acid, a methionine analog produced by Streptomyces sp. MF374-C4.

2-Amino-5-methyl-5-hexenoic acid (AMHA), a new methionine analog, was isolated from a fermentation broth of Streptomyces sp. MF374-C4 based on its reversal of the effect of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in a test system that determines the size of growth zones of revertants (His+) of Salmonella typhimurium TA1535. AMHA also inhibited growth of the tester strain in a synthetic medium. These AMHA activities were abolished by methionine. The incidence of spontaneous streptomycin-resistant mutations of Escherichia coli K12 was not decreased by AMHA at concentrations where cell growth was partially inhibited. AMHA inhibited protein synthesis but not DNA or RNA synthesis in S. typhimurium TA1535 and E. coli K-12. The analog inhibited formation of methionyl-tRNA but not of valyl-tRNA in a cell-free system of E. coli, and supported ATP-PPi exchange in the cell-free system. At concentrations where it inhibited cell growth, AMHA decreased the number of foci, induced by ROUS sarcoma virus, on cultured sheets of chick-embryo fibroblasts. The effects of AMHA on focus formation and on the cell growth were overcome by methionine.

SCE-963, a new broad-spectrum cephalosporin: in vitro and in vivo antibacterial activities.

SCE-963 {7beta-[2-(2-aminothiazol-4-yl)acetamido]-3-[({1-(2-dimethylaminoethyl)- 1H-tetrazol-5-yl}thio)methyl]-ceph-3-em-4-carboxylic acid}, a new semisynthetic cephalosporin, showed excellent antibacterial activity against gram-positive and gram-negative bacteria, including Haemophilus influenzae, indole-positive Proteus, Enterobacter species, and Citrobacter freundii. The minimum inhibitory concentrations of SCE-963 against most strains of clinically isolated Escherichia coli, Klebsiella pneumoniae, H. influenzae, and Proteus mirabilis were within the range of 0.2 to 0.78 mug/ml. These activities were about 10 times more potent than those of cefazolin, cephaloridine, and cephalothin. Variations in pH, addition of horse serum, and type of growth medium had no significant effect on the activity of the cephalosporin, but the inoculum size elicited a considerable effect on the activity of beta-lactamase-producing strains of bacteria. SCE-963 exerted bactericidal and bacteriolytic effects on Staphylococcus aureus and E. coli. The pronounced in vitro activity was reflected in the remarkable protection in mice infected with a wide range of gram-negative bacteria, such as E. coli, K. pneumoniae, P. mirabilis, Proteus vulgaris, Proteus morganii, and Proteus rettgeri. The protective effects of SCE-963 in mice infected with E. coli, K. pneumoniae, and P. vulgaris varied according to the challenge dose. The activity of SCE-963 was far more potent when the drug was administered parenterally rather than orally.

Absorption, distribution and excretion of SCE-963, a new broad-spectrum cephalosporin, in mice, rats, rabbits and dogs.

A single dose of 20 mg/kg of SCE-963 [7beta-]2-(aminothiazol-4-yl)acetamido]-3-[[[1-(2-dimethylaminoethyl)-1H-tetrazol-5-yl]thio]methyl]ceph-3-em-4-carboxylic acid] was administered subcutaneously to mice, intramuscularly to rats, rabbits and dogs. Plasma and tissue levels of SCE-963 reached a peak in 15 approximately 30 minutes after administration. In mice, rats and dogs, SCE-963 was distributed at high concentration in the descending order in the kidney, liver, plasma, lung and spleen, and in rabbits, in the kidney, plasma, lung, liver and spleen. The SCE-963 levels in the liver of mice, rats and dogs were higher than those of cefazolin, cephaloridine and cephalothin. The plasma and tissue levels of SCE-963 in mice and rats diminished rapidly, but those in rabbits and dogs declined gradually. SCE-963 was mainly excreted in the urine. The rate of excretion of SCE-963 in the bile was two to three times higher than that of cefazolin.

[Viscosity changes of zinc phosphate cements in setting process (author's transl)].

A better understanding of setting process of zinc phosphate cements is important to obtain optimum conditions of manipulation. Viscosity changes of zinc phosphate cements in setting process were investigated using a rotational viscometer in various conditions. The results obtained were as follows. 1) The viscosity of the cement mixture increased exponentially as a function of time. 2) The setting process of the cements was characterized by two parameters, the coefficient of setting speed and the setting time. The coefficient of setting speed was defined as the slope of the linear portion of log viscosity versus time curve, and the setting as the time required to reach a certain level of viscosity after the start of the mixing. 3) Shear rate was observed to affect on the time-related changes of viscosity. As the shear rate increased, the coefficient decreased and the setting time increased. These changes were more remarkable in the lower range of shear rate. 4) The coefficient of setting speed was found to be dependent on environmental temperature in accordance with Arrhenius' low. 5) Apparent activation energy calculated 16.41 kcal/mol by Arrhenius' equation. The coefficient increased by a factor of 2.6, when the temperature was raised from 20 degrees to 30 degrees C. 6) A decrease of the P/L ratio caused the logarithm of the coefficient to increase linearly and the setting time to decrease. 7) All the four commerical zinc phosphate cements tested showed different values of both the coefficient and the setting time.
