Expression in Escherichia coli of the extrinsic 18-kDa protein of photosystem II of spinach.

A cDNA encoding the precursor for the 18-kDa protein of PSII of spinach was expressed in Escherichia coli. When the cell lysate was incubated at 7 degrees C, the precursor was degraded by proteases of E. coli to a polypeptide of 18 kDa (P18) that consisted of the mature protein moiety plus the last four residues of the transit peptide. P18 was able to reconstitute the water-oxidizing complex of NaCl-treated PSII membranes supplemented with the 23-kDa protein. Moreover, P18 was cleaved by the prolyl endoproteinase of spinach specifically at the Pro-12-Leu-13 bond, as was the authentic 18-kDa protein. These properties of P18 indicate that the present expression system is potentially useful for studies of the substrate specificity of the endoproteinase, as well as of the structure-function relationships of the 18-kDa protein.

Taxonomic study of bacteria isolated from plants: proposal of Sphingomonas rosa sp. nov., Sphingomonas pruni sp. nov., Sphingomonas asaccharolytica sp. nov., and Sphingomonas mali sp. nov.

The taxonomic positions of 10 strains of 3-ketolactose-forming bacteria which were isolated from the roots of plants (Rosa sp., Psychotria nairobiensis, Ardisia crispa, Prunus persica, and apple trees) were investigated. The DNA base compositions of these strains ranged from 64.0 to 65.7 mol%, the isoprenoid quinone of each strain was ubiquinone 10, 3-hydroxy fatty acids were lacking in the cellular fatty acids of these organisms, and all of the strains contained a sphingolipid with the long-chain base dihydrosphingosin. These are characteristics of the genus Sphingomonas. On the basis of morphological, physiological, and chemotaxonomic characteristics, together with DNA-DNA hybridization and 16S ribosomal DNA sequence comparison data, we propose the following four new species of the genus Sphingomonas: Sphingomonas rosa (type strain, IFO 15208) for the strains isolated from rose plants and formerly named [Agrobacterium rhizogenes]; Sphingomonas pruni (type strain, IFO 15498) for the strains isolated from Prunus persica; and Sphingomonas asaccharolytica (type strain, IFO 15499) and Sphingomonas mali (type strain, IFO 15500) for the strains isolated from apple trees. Two strains which were isolated from Psychotria nairobiensis and formerly named [Chromobacterium lividum] were identified as Sphingomonas yanoikuyae strains.

Bacillus curdlanolyticus sp. nov. and Bacillus kobensis sp. nov., which hydrolyze resistant curdlan.

Taxonomic characteristics of seven bacterial strains which were isolated from soil and hydrolyze resistant curdlan were studied. These bacteria were aerobic, spore-forming rods, contained menaquinone 7 as a major quinone, contained anteiso-C15:0 and iso-C16:0 as major cellular fatty acids, had guanine-plus-cytosine contents of 50 to 52 mol%, and could be divided into two groups on the basis of physiological and chemotaxonomic characteristics and DNA-DNA hybridization data. We propose the following two new species: Bacillus curdlanolyticus for strains YK9, YK121, YK161, YK201, and YK203, with type strain YK9 (= IFO 15724); and Bacillus kobensis for strains YK205 and YK207, with type strain YK205 (= IFO 15729).

Complete nucleotide sequence of a skin element excised by DNA rearrangement during sporulation in Bacillus subtilis.

As part of the Bacillus subtilis genome sequencing project, we have determined the complete nucleotide sequence of a skin element which is located between spoIVCB and spoIIIC. The entire sequence of this element is 48,032 bp in length, and contains 57 ORFs with putative ribosome-binding sites. Two of them correspond to previously sequenced and characterized genes, cwIA and spoIVCA. Furthermore, seven ORF products identified in this element show interesting similarities with known proteins present in data banks, including the phi 105 immunity repressor, the phi 105 Cro-like protein and the SPP1 terminase. These results indicate the possibility that the skin element is a cryptic remnant of an ancestral temperate phage.

Necrotizing fasciitis in association with hyperimmunoglobulin E syndrome.

A case of necrotizing fasciitis in association with hyperimmunoglobulin E (HIE) syndrome is reported. The patient was a 17-year-old Japanese boy with a clinical history of recurrent skin and pulmonary infections and eczematoid dermatitis, markedly elevated serum levels of IgE, and coarse facies. He had a gangrenous swelling on the lower abdominal wall, and his general condition was poor with high fever. The involved site was accompanied by subcutaneous gas; the culture of the pus of the lesion grew anaerobes without mixed growth of Staphylococcus aureus. Exhaustive debridement of necrotic fascia, which extended much farther than the gangrenous area, and administration of antibiotics had a curative effect on the gangrenous soft tissue infection. To the best of the authors' knowledge, this is the first published case of necrotizing fasciitis in association with HIE syndrome.

Accelerated ventricular rhythm in the newborn.

Accelerated ventricular rhythm was observed in two newborn infants. Neither of them had any causative clinical symptoms for the ventricular arrhythmia. The arrhythmia disappeared when the infants were 18 days and 45 days old, respectively. Arrhythmia was noted in the fetal period, especially in case 1.

Laparoscopic mesenterioadhesiotomy and Tenckhoff catheter placement in patients with predisposing abdominal surgery.

Peritoneoscopic surgery has been performed widely for a variety of abdominal surgical diseases. We describe here a safe and reliable technique of laparoscopic-assisted mesenterioadhesiotomy and peritoneal Tenckhoff catheter placement in patients who have previously undergone abdominal surgery. Five patients suffering from end-stage renal failure previously underwent single and/or polyabdominal surgery. The surgical procedures included hysterectomy, ovarian resection, appendectomy, and transabdominal right nephrectomy. Under general endotrachial anesthesia, a laparoscope was placed down through a direct cut made using a trocar. After CO2 gas insufflation, another one or two trocars were put in place for surgical procedures. To avoid intestinal injury, mesenterioadhesiotomy was performed carefully using a high-frequency hook electrode, forceps, and scissors forceps, and the Tenckhoff catheter was subsequently inserted with forceps directly into Douglas' fossa. Peritoneal equilibration tests performed 30-70 days after the initiation of continuous ambulatory peritoneal dialysis (CAPD) treatment revealed moderate to good peritoneal effectiveness. This procedure permits the surgeon to perform safe and exact catheter placement into Douglas' fossa even when there is a possibility that peritoneal and mesenterial adhesion are present. We believe that this technique of catheter placement may extend the indication for CAPD treatment in patients with predisposing lower abdominal surgery.

Specific determination of histamine in fish by high-performance liquid chromatography after diazo coupling.

A new approach to the pre-column derivatization and analysis of histamine by HPLC is described, the method being based upon the formation of a diazo-coupled derivative of histamine. This derivatization method is simple, efficient, sensitive and specific for histamine and other imidazole compounds without a preliminary clean-up step. The liquid chromatographic system allows for rapid, bonded-phase separation with visible-light detection of these compounds within 20 min at a 10-pmol sensitivity. By using this method, the production of a large amount of histamine was confirmed in the muscle of mackerel Scomber japonicus incubated with the intestinal contents of the mackerel.

Effect of hypoglycemic stress on the pars intermedia of the mouse pituitary gland: an ultrastructural analysis.

The present study was made to clarify the relationship between functions of the pars intermedia of the mouse pituitary gland and hypoglycemic stress. Morphometrical analysis of the ultrastructures of the pars intermedia cells showed (1) a rise in the percentage volume of rough endoplasmic reticulum (r-ER) indicative of an increase in protein synthesis, (2) an increase in the number of Golgi granules per unit Golgi area showing an induction of granule-forming activity and (3) a decrease in the numerical density of secretory granules reflecting a release of the secretory granules. These findings suggest that hypoglycemic stress induced by daily treatment with insulin or restriction of food intake was able to elicit heightened secretory activity of the pars intermedia cells of the mouse pituitary gland. However, acute hypoglycemic stress induced by food deprivation did not cytologically affect the pars intermedia. These observations suggest that repeated hypoglycemic stress, rather than acute hypoglycemic stress, may be a natural physiological stimulus of the pars intermedia of the mouse pituitary gland.

Collaborative work to determine an optimal administration period and optimal parameters for detection of effects on male fertility in rats--male reproductive toxicity study of haloperidol.

Haloperidol, a neuroleptic, was orally administered at 0, 3, 10, 30 and 60 mg/kg/day in a 4-week dosing study, and 0, 3, 10 and 30 mg/kg/day in a 9-week dosing study, to Sprague-Dawley male rats which were then sacrificed for histopathological examination or mated with untreated females. The males in the mating groups were continuously treated during the mating period. Sperm positive females were sacrificed on day 20 of gestation. The males in the histopathology groups were sacrificed after 4- or 9-week dosing and their testes were fixed in Bouin's fluid and sections stained with HE or PAS. The mated males were sacrificed and reproductive organ weights were determined. At 3 mg/kg or more, reduced spontaneous motor activity was observed and body weights were lowered. The absolute weight of testes was decreased with 60 mg/kg in the 4-week dosing study, and was decreased with 10 and 30 mg/kg in the 9-week dosing study. However, the relative weight was increased or showed a tendency to increase. In the 4-week dosing study, decrease in the fertility index with 60 mg/kg and increase in pre-implantation loss with 30 mg/kg or more were noted. However, there were no adverse effects on the copulation index in any of the treated groups. The males given 60 mg/kg showed slight changes in the testes (necrosis of pachytene spermatocytes in seminiferous tubules of stage VII, exfoliation of round spermatids in the lumina and atrophy of Leydig cells) and seminal vesicles (atrophy of epithelial cells). Atrophy of Leydig cells was also observed at 30 mg/kg in the 4-week dosing study. In the 9-week dosing study, neither male reproductive ability nor histopathological parameters were affected by haloperidol up to 30 mg/kg. From the results detailed above, it may be concluded that 4-weeks dosing before mating is suitable for detection of effects of haloperidol on male reproductive ability, and that histopathological changes together provide an optimal parameter for predicting male reproductive disorders.

Televised violence: a Japanese, Spanish, and American comparison.

In Japan, Spain, and the USA during one week acts of violence (verbal and nonverbal) were rated on the Index of Television Violence by 3 raters in each country. In all three countries televised violence was more likely depicted in interpersonal conflict than against property. Fewer scenes of physical or fatal injury appeared in Japan than in the other two countries. Other observations were made. Replication is urged.

Effects of excess doses of alpha-tocopherol on the lipids and function of rainbow trout liver.

Rainbow trout (Oncorhynchus mykiss) were kept on diets containing three different alpha-tocopherol levels (0, 10, and 1,000 mg/100 g diet) to investigate the effects of excess doses of alpha-tocopherol on the liver lipids and liver functions. Both the content of the lipids except alpha-tocopherol in the liver and the triglyceride ratio in total lipids were significantly increased by the diets containing 1,000 mg of alpha-tocopherol per 100 g of diet. Many lipid droplets, 5-20 microns in the diameter, were observed in the liver. The TBARS per milligrams of liver lipids was decreased by 1,000-mg doses of alpha-tocopherol. GOT and GPT activity in serum were in the normal range regardless of the dose of alpha-tocopherol. From these data, it is considered that excess alpha-tocopherol promotes lipid accumulation enough to form droplets in the liver tissue, although liver function is not impaired.

Polyacrylamide gel electrophoretic patterns of chicken serum in acute inflammation induced by intramuscular injection of turpentine.

To develop a method to detect hidden inflammation using serum protein in chickens, changes in serum proteins with acute inflammation were analyzed using a turpentine-induced inflammation model. Inflammation in the pectoral muscle of a 14-wk-old White Leghorn became apparent 3 h after the injection of turpentine and became more severe thereafter. Coincident with the development of inflammation, changes in serum proteins were analyzed by electrophoresis on polyacrylamide gradient gels. The electrophoretic patterns were divided into 21 segments. Two of these segments increased remarkably. These were located near the center of the electrophoretic pattern and were identified as transferrin due to iron staining, correlation of movement against a commercial transferrin sample in SDS-PAGE, and immunoblotting. These results suggest that transferrin may serve as a marker for inflammation in chicken.

[Clean intermittent catheterization in neurogenic bladder patients with vesicoureteral reflux].

BACKGROUND: For neurogenic bladder (NGB) patients with vesicoureteral reflux (VUR), renal deterioration constitutes a primary threat to survival. Although anti-reflux surgery for those patients was reported to be effective, the efficacy of clean intermittent catheterization (CIC) in such patients still remains to be clarified. METHODS: Sixteen neurogenic bladder patients with VUR were treated with CIC. Eight of them had spina bifida and 3 had radical hysterectomy for uterine cancer. Other 5 patients had spinal cord injury, spondyrocace, measles encephalopathy, and 2 unknown diseases, respectively. Hyperactive bladder was noted in 11 patients, whereas hypoactive bladder was noted in 3. Three patients were not evaluated. RESULTS: VUR was improved in only 3 patients (3 ureters), whose VUR grades were not more than III. Nine patients (13 ureters) had to have antireflux surgery. Although 3 of them needed bladder augmentation afterwards, the success rate of the antireflux surgery was 84.6%. CONCLUSION: We concluded that CIC alone was not effective to control VUR in neurogenic bladder patients. Nevertheless, CIC remained a good treatment option when VUR was managed surgically.

[Effects of castration on apoptosis and transforming growth factor-beta 1 mRNA in the neonatal mouse seminal vesicles].

BACKGROUND: In the adult male rat prostate, castration induces apoptosis of epithelial cells concomitant with the increase in transforming growth factor-beta 1 (TGF-beta 1). In the present study, we investigated the effects of castration on apoptosis and TGF-beta 1 mRNA in neonatal mouse seminal vesicles. METHODS: 5-day-old BALB/c mice were castrated by Pfeifer's method. We estimated the weight, 3H-thymidine uptake by whole seminal vesicles, the amount of TGF-beta 1 mRNA by RT-PCR method, and the apoptotic index of both epithelium and mesenchyme. RESULTS: The castration of 5-day-old neonatal mice resulted in much less weight of seminal vesicles and DNA synthesis estimated by 3H-thymidine uptake by whole seminal vesicles compared to intact neonatal mice, indicating that the growth of neonatal mouse seminal vesicles depends on androgens secreted by the testis. The amount of TGF-beta 1 mRNA estimated by RT-PCR method increased 4 days after castration at 5 days of age. However, the castration did not induce apoptosis in the seminal vesicles. CONCLUSION: The present study indicates that castration of neonatal mice does not induce apoptosis in the seminal vesicles, although it does a transient increase in TGF-beta 1 mRNA in the seminal vesicles.

[Protective effect of alpha-tocopherol in retinal light damage of pigmented rabbits--evaluation by nitro blue tetrazolium staining].

Free radicals are thought to be one of the causes of retinal light damage. Experiments were performed to identify the production of free radicals in light exposed retinas of rabbits using nitro blue tetrazolium (NBT) staining. In addition, a free radical scavenger, alpha-tocopherol, was administered systemically before light exposure to evaluate the protective effect against the retinal light damage. Pigmented rabbits were exposed to xenon light without long waves for 60 minutes. The eyes were enucleated immediately after light exposure, and incubated in the equilibrated media at 37 degrees C with 0.1 mM NBT solution for 60 minutes. After incubation, the eyes were embedded in paraffin, sectioned, and mounted without further staining. The same procedure was done to the tocopherol treated retinas. In the light exposed eyes, the NBT staining was markedly observed at the inner retina, and the staining was more remarkable than that of the non-exposed eyes. The tocopherol treated eyes showed weaker NBT staining in the inner retina than the non-treated eyes. The results suggest that free radicals are increased in the inner retina by light exposure and alpha-tocopherol suppresses production of free radicals.
