Behavioral and adrenocortical responses to stress in neonates and the stabilizing effects of maternal heartbeat on them.

Adrenocortical response to stress in neonates was assessed by estimating salivary concentrations of cortisol, dehydroepiandrosterone (DHEA) and dehydroepiandrosterone-sulfate (DHEA-S). Behavioral responses to stress were also scored clinically, according to the method of Lewis and Thomas [15]. The modulation of these measures by hearing maternal heartbeat was also evaluated. Levels of cortisol and DHEA in saliva in neonates correlated well with those of serum showing significant variation in painful stress. Hearing the maternal heartbeat significantly reduced the grade of variation, but the recorded sound of the Japanese drum with the same rhythm showed no significant effect. Scores of the behavioral response to stress also decreased significantly in neonates who were presented with the maternal heartbeat in comparison with those hearing no sound or the beat of a Japanese drum.

A YAC contig of the human CC chemokine genes clustered on chromosome 17q11.2.

CC chemokines are cytokines that attract and activate leukocytes. The human genes for the CC chemokines are clustered on chromosome 17. To elucidate the genomic organization of the CC chemokine genes, we constructed a YAC contig comprising 34 clones. The contig was shown to contain all 10 CC chemokine genes reported so far, except for one gene whose nucleotide sequence is not available. The contig also contains 4 CC chemokine-like genes, which were deposited in GenBank as ESTs and are here referred to as NCC-1, NCC-2, NCC-3, and NCC-4. Within the contig, the CC chemokine genes were localized in two regions. In addition, the CC chemokine genes were more precisely mapped on chromosome 17q11.2 using a somatic cell hybrid cell DNA panel containing various portions of human chromosome 17. Interestingly, a reciprocal translocation t(Y;17) breakpoint, contained in the hybrid cell line Y1741, lay between the two chromosome 17 chemokine gene regions covered by our YAC contig. From these results, the order and the orientation of CC chemokine genes on chromosome 17 were determined as follows: centromere-neurofibromatosis 1-(MCP-3, MCP-1, NCC-1, I-309)-Y1741 breakpoint-RANTES-(LD78gamma, AT744.2, LD78beta)-(NCC-3, NCC-2, AT744.1, LD78alpha)-NCC-4-retinoic acid receptor alpha- telomere.

Three different mutations of presenilin 1 gene in early-onset Alzheimer's disease families.

Presenilin-1 (PS-1) gene of three Japanese pedigrees with early-onset familial Alzheimer's disease (FAD) disclosed two novel missense mutations resulting in Val96Phe and Ile213Thr, and one mutation resulting in His163Arg. The mean age at onset in a family with His163Arg mutation was similar to those reported in other families with His163Arg. Our results suggested the existence of a variety of PS-1 mutations, and that early-onset FAD with PS-1 mutations is highly penetrant and is only rarely subject to modulation by genetic or environmental modifying factors.

The terminal protein of the linear DNA plasmid pGKL2 shares an N-terminal domain of the plasmid-encoded DNA polymerase.

The 36K protein attached at the 5' end of the linear DNA plasmid pGKL2 from the yeast Kluyveromyces lactis was first purified and characterized. The terminal protein was purified from cells (1 kg wet weight) by ammonium sulphate precipitation and two rounds of centrifugation to equilibrium in CsCl gradients. The pGKL2 was present only in the post-microsomal supernatant. Approximately 10 mg of the purified pGKL2 was recovered and digested with DNase I. The terminal protein (final ca. 0 center dot 8 mg) was homogeneous by electrophoresis and we determined the N-terminal amino acid sequence up to ten residues, showing that it existed in the cryptic N-terminal domain of pGKL2-ORF2 (DNA polymerase) sequence.

T1 and T2 squamous cell carcinomas of the floor of the mouth: results of brachytherapy mainly using 198Au grains.

PURPOSE: To investigate the effectiveness of brachytherapy provided by this institution over a 25-year period, and to specifically verify the efficacy of 198Au grain therapy, this study evaluated the outcomes in patients given brachytherapy for T1 and T2 squamous cell carcinoma (SCCs) of the floor of the mouth. METHODS AND MATERIALS: A retrospective analysis of data from 90 patients with T1 and T2 SCCs of the floor of the mouth who underwent brachytherapy between 1965 and 1989. Therapy mainly consisted of 198Au grain implants with or without external irradiation. As for the brachytherapeutic source, 15 patients were treated with radon seeds, 60 with 198Au grains, 10 with radium needles, 3 with cobalt needles, and 2 with iridium hairpins. Based on the 1987 International Union Against Cancer (UICC) classification, the SCC stagings and number of cases per staging follow: Stage I (T1N0), 21 cases; Stage II (T2N0), 55 cases; and Stage III-IV (T1-2N1-2), 14 cases. The minimum follow-up time was 3 years. RESULTS: The local control rates of these SCCs, based on tumor size, were 89% for T1 lesions, 76% for T2a (< or = 3 cm) lesions, and 56% for T2b (> 3 cm) lesions, and 82% for T1-2 lesions without a gingival involvement, in contrast to 55% for lesions with a gingival involvement (p < 0.05). The 5-year, cause-specific survival rates by staging were 95% for T1N0, 79% for T2N0, and 54% for T1-2N1-2. For patients given 198Au grain brachytherapy, local control was achieved in 93% of the T1 lesions, 79% of the T2a lesions, and in 56% of the T2b lesions. Further, the incidence of severe complications requiring surgery was low (5%). CONCLUSIONS: For T1N0 and T2aN0 SCCs of the floor of the mouth, excluding lesions with a gingival involvement, 198Au grain brachytherapy alone or in combination with external radiotherapy was found to be efficacious.

Dynamics of bovine glial fibrillary acidic protein phosphorylation.

Recently, the dynamic features of the intermediate filaments (IF) have been revealed. The effect of phosphorylation on the dynamics of bovine glial fibrillary acidic protein (GFAP), the astroglial IF, was studied in vitro with fluorescently labeled GFAP. Soluble GFAP in low ionic strength buffer was rapidly and fully phosphorylated to be used as phosphorylated GFAP. Assembly of GFAP was observed to be inhibited in proportion to the extent of phosphorylation by mixing phosphorylated and non-phosphorylated GFAP at various ratios, and phosphorylated GFAP could not be assembled with non-phosphorylated GFAP into filaments at all. Furthermore, the subunit exchange was suppressed in proportion to the extent of phosphorylation. Phosphorylation affects the dynamic equilibrium of GFAP, and contributes to breaking down GFAP frameworks in mitotic glial cells.

Molecular cloning of a 74-kDa regulatory subunit (B" or delta) of human protein phosphatase 2A.

Based on amino acid sequence data of a 74-kDa regulatory subunit (B" or delta) of a human heterotrimeric protein phosphatase 2A, a cDNA encoding the subunit was isolated from a human cerebral cortex library. The cDNA had an open reading frame encoding an M(r) 66,138 protein of 570 amino acids. Bacterial expression of the cDNA yielded a protein immunoreactive with antisera specific to the 74-kDa subunit. The predicted primary structure of the subunit had no similarity to already reported sequences of PP2A regulatory subunits including A, B, and PR72. Potential phosphorylation sites for protein kinases A and C, a bipartite motif of putative nuclear localization signal, and SH3 accessible proline-rich domain, and a unique PQ repeat were found in the sequence. The subunit mRNA of about 2.9 kb was ubiquitously expressed in rat tissues.

Tumor necrosis factor alpha and lymphotoxin stimulate human myeloblastic leukemia cell (ML-1) invasion through a reconstituted basement membrane (Matrigel) with concomitant induction of 92 kDa gelatinase secretion.

We have examined the effects of tumor necrosis factor alpha (TNF) and lymphotoxin (LT) on gelatinase (72 kDa and 92 kDa) and tissue inhibitor of metalloprotease 1 (TIMP1) secretion by human myeloblastic leukemia cells (ML-1) in vitro. TNF (0.1-30 ng/ml) significantly stimulated 92 kDa gelatinase secretion in a dose-dependent manner, but did not significantly stimulate 72 kDa gelatinase secretion. LT also significantly stimulated 92 kDa gelatinase secretion, but the stimulation was less effective compared to TNF. TNF, but not LT, concentrations at 30 ng/ml slightly stimulated TIMP1 secretion. Because 92 kDa gelatinase is thought to play a pivotal role in tumor invasion, we examined the effect of TNF or LT on ML-1 cell invasion through a reconstituted basement membrane (Matrigel). Exposure of ML-1 cells to TNF (3, 10, and 30 ng/ml) or LT (3, 10, and 30 ng/ml) stimulated ML-1 cell invasion through Matrigel in a dose-dependent manner in vitro. The data suggest that TNF- and LT-stimulated 92 kDa gelatinase secretion could play an important role in TNF- or LT-stimulated ML-1 cell invasion.

Telomere shortening in peripheral blood cells was related with aging but not with white blood cell count.

Telomeres in somatic cells are progressively shortened with aging. We investigated the relationship between the telomere length and other factors which may affect the frequency of cell divisions, in peripheral blood cells. Shortening of telomeric repeats was correlated with aging (p < 0.0001), but not with white blood cell count, neutrophil count, and smoking habit. Not only the number of cell divisions, but also some other factors, such as upregulation level of telomerase activity concomitant with the cell division in hematopoietic progenitor cells, might affect the length of telomeric repeats in blood cells.

Apoptosis in mouse taste buds after denervation.

Apoptotic cells in the taste buds of mouse circumvallate papillae after the sectioning of bilateral glossopharyngeal nerves were examined by the method of DNA nick-end labeling (TUNEL), together with standard electron microscopy. The taste buds decreased in number and size 3-11 days after denervation and disappeared at 11 days. The TUNEL method revealed only a few positively stained nuclei in normal taste buds but, in those of mice 1-5 days after denervation, the number of positive nuclei had increased to 3-5 times that of taste buds from normal mice. Electron-microscopic observation after denervation demonstrated taste bud cells containing condensed and fragmentary nuclei in a cytoplasm with increased density. The results show that taste bud cells under normal conditions die by apoptosis at the end of their life span, and that gustatory nerve sectioning causes apoptosis of taste bud cells with taste buds decreasing in number and ultimately disappearing.

Effect of thyroidectomy on photoreceptor cells in adult rat retina.

To evaluate the effect of thyroid hormone on the retina in the adult male rat, morphometric and autoradiographic studies were made. Four days after an intravenous pulse injection of L-[3H]-leucine into adult male rats, the eyeballs were removed and the autoradiogram made. Two weeks after thyroidectomy, the length of photoreceptor cell inner segments was significantly decreased, but that of the outer segments did not change. Daily supplement of T4 (100 microg/kg sc twice for 2 days) before L-[3H]-leucine injection did not restore the decrease in the length. The silver grains of radioactive material distributed in the outer segment were divided into 3 equal portions. Thyroidectomy significantly increased the number of grains in the basal portion near the inner layer indicating a reduction in the renewal rate of the outer segment, but the T4 supplement did not restore the reduction in the renewal rate. These findings suggest that thyroid hormone is required to maintain the rate of renewal of the photoreceptor cell outer segment but the hormone supplement did not rapidly restore the renewal rate.

The requirement for and mobilization of calcium during induction by sodium ascorbate and by hydrogen peroxide of cell death.

The requirement for and mobilization of Ca2+ ions during induction of cell death by sodium ascorbate were compared with those during induction of cell death by hydrogen peroxide. When HL-60 cells were incubated with sodium ascorbate, a rapid increase in the intracellular concentration of Ca2+ ions and subsequent apoptotic cell death, characterized by cell shrinkage, nuclear fragmentation and cleavage of internucleosomal DNA to yield fragments that were multiples of 180-200 base pairs, were induced. However, these effects of sodium ascorbate were significantly reduced in Ca2+-depleted medium. By contrast, hydrogen peroxide induced similar apoptosis associated phenomena in the presence and in the absence of extracellular Ca2+ ions. The intracellular concentration of the reduced form of glutathione was not significantly affected and glutathione disulfide was undetectable during the early stages of apoptosis. These data suggest that sodium ascorbate and hydrogen peroxide initiate cell death by different mechanisms.

Mitral valve replacement after percutaneous transvenous mitral commissurotomy.

Percutaneous transvenous mitral commissurotomy is widely performed as the first choice of the non-pharmacological treatments for mitral stenosis. Five patients have been identified who required mitral valve replacement after percutaneous transvenous mitral commissurotomy. The causes leading to mitral valve replacement were mitral regurgitation in three cases and insufficient commissurotomy in two. Massive mitral regurgitation is one of the most serious complications of percutaneous transvenous mitral commissurotomy. This report aims to elucidate the mechanism of massive mitral regurgitation occurring during percutaneous transvenous mitral commissurotomy. In every such case, there was a large tear in the posterior leaflet without any split in the commissures. The Japanese literature reports that 16 patients have undergone mitral valve replacement for massive regurgitation after percutaneous transvenous mitral commissurotomy: 14 of these cases had a tear in one of the leaflets and no evidence of splitting of the posterior commissure. These facts indicate that relative fragility of the leaflets as compared with rigidity of commissural fusion, especially in the posterior commissure, is an important factor of massive mitral regurgitation during percutaneous transvenous mitral commissurotomy.

Use of temporary atrial pacing in management of patients after cardiac surgery.

The authors' clinical experience with temporary atrial pacing to evaluate its use in the management of patients after cardiac surgery was reviewed. A total of 339 patients undergoing cardiac surgery were studied with regard to postoperative pacing therapy. Postoperative pacing was performed in 186 of 339 patients to treat supraventricular bradycardia or tachyarrhythmias. Rapid atrial pacing was performed to interrupt re-entrant supraventricular tachyarrhythmias. In bradycardic patients, haemodynamics could be improved as the result of significant increase of blood pressure and oxygen saturation in the pulmonary artery (SVO2) caused by atrial pacing. Premature beats could be suppressed in 63% and supraventricular tachyarrhythmias could be interrupted in 66% of the patients only by atrial pacing. Temporary atrial pacing is safe, rapid and effective as the treatment of choice; it is believed that the technique should be applied in preference to pharmacological treatment in the management of patients after cardiac surgery.

Two district alpha(1)-adrenoceptor subtypes in the human prostate: assessment by radioligand binding assay using 3H-prazosin.

1. We showed that there were two distinct alpha(1)-adrenoceptor subtypes (alpha(1H) and alpha(IL)) in the human prostate which show different affinities for 3H-prazosin. 2. WB4101, tamsulosin, 5-methylurapizil, phentolamin, and terazosin, but not nifedipine, had significantly higher pKi values for the alpha(1H)-subtype than for the alpha(IL)-subtypes. 3. There was good correlation (r = 0.92, P < 0.05) between the pKi values obtained for the alpha(1H)-receptors in membrane fractions and the cloned human alpha(1c)-adrenoceptor subtype.

Functional and morphological observations on the microvascular structure of the filiform and fungiform papillae in the cat tongue.

The purpose of this study was to undertake a three-dimensional comparative observation of the microvascular cast specimen (MVCS) of the filiform papillae (FiP) and fungiform papillae (FuP) on the dorsal surface of the cat tongue. By means of the corrosive resin casting technique, the MVCS of the FiP and FuP of the cat tongue were prepared and examined in detail under the scanning electron microscope (SEM). The numerous FiP and FuP distributed sporadically among the different types of the FiP were found on the entire dorsal surface of each cat tongue. A single FiP consisted of a main process (MP) which contained a large spoon-shaped concave network process and the accessory process (AP) contained a bundle of spine-like processes, arranged in a circle at the basal margin of the MP. The FiP can be classified into five types (Type I-V) according to the shape and size of the MP and numbers of the AP. Each FuP consisted of a single MP which contained an ovoid head and many AP surrounding the basal margin of the MP. The FuP can be classified into four types (FuP I-IV) according to the shape and size of the MP and numbers of the AP. The MP of the FuP, a sense organ for taste, of the cat tongue was considered to be a transformed type from the MP of the FiP, which acquires the ability to masticate and swallow functionally.
