Cold-adaptation of human rotavirus.

A human rotavirus strain was cold-adapted for possible future use as a live vaccine. The original strain was isolated in 1980 in primary cynomolgus monkey kidney cells and has a serotype I and subgroup II antigenicity. The virus was serially passaged in African green monkey kidney cells; it was cultivated at 37 degrees C at the first stage of passages, and the cultivation temperature was then shifted down stepwise by 3 degrees C per each 10 passages. Finally the virus was passaged 10 times at 25 degrees C (total passage number of 55). The virus formed small-size plaques with irregular shaped borders at 31 degrees C. Growth at 25 degrees C of the cold-adapted virus was higher than that of the original virus. There was no difference between the migration patterns of 11 dsRNA segments in polyacrylamide gel electrophoresis of the original and the cold-adapted viruses.

The role of radiotherapy in the management of adenoid cystic carcinoma of the head and neck.

Sixty-eight patients with adenoid cystic carcinoma (ACC) of the head and neck treated by radiation with or without surgery were reviewed. The tumors were located in the major salivary glands in 15 patients and in the minor glands of various sites, mostly in the palate and/or upper gum, in 53. The local control rate was naturally better in the patients with elective postoperative radiation and those with recurrent disease who underwent radiation after repeated surgery for the purpose of tumor reduction than in patients with residual and/or inoperable tumors. For localized tumors in the oral cavity or maxillary sinus, local control was excellent using brachytherapy or "trimodal" combined therapy (radiation, drugs, and minor surgery). The effectiveness of radiation seemed to be more significantly related to the size of the tumor than the dose of radiation. The prognosis of the patients with a predominantly "solid" histologic pattern was apparently worse and presented fulminating progression of the disease. Among patients with a histologically typical cribriform pattern and/or undetermined pattern, although distant metastasis developed frequently, the majority survived a relatively long period without functional or cosmetic disturbance to their normal activities associated with slow progression of the disease. In minor gland tumors, as long as the lesion was localized and a solid histologic pattern was unlikely, radical radiation could be considered an effective choice with the advantage of enhancing the quality of life for the patients.

Nucleotide sequencing and characterization of the genes encoding benzene oxidation enzymes of Pseudomonas putida.

The nucleotide sequence of the genes from Pseudomonas putida encoding oxidation of benzene to catechol was determined. Five open reading frames were found in the sequence. Four corresponding protein molecules were detected by a DNA-directed in vitro translation system. Escherichia coli cells containing the fragment with the four open reading frames transformed benzene to cis-benzene glycol, which is an intermediate of the oxidation of benzene to catechol. The relation between the product of each cistron and the components of the benzene oxidation enzyme system is discussed.

Antiarrhythmic effects of a novel diamine derivative, AN-132, on several animal models of cardiac arrhythmias.

The antiarrhythmic activities of AN-132, 3-(diisopropylaminoethylamino)-2',6'-dimethylpropionanilide X 2H3PO4, compared with three reference drugs, were examined in several arrhythmic models of rats, guinea-pigs and dogs. When compared on a weight basis, AN-132 given in oral or i.v. route proved to be more potent and longer-acting than disopyramide, propaphenone and mexiletine against supraventricular and ventricular arrhythmias induced by chloroform, aconitine, ouabain and halothane-adrenaline.

The effect of glycemic control on plasma soluble fibrin monomer complexes and fibronectin in diabetic patients.

We have shown previously that increased concentrations of plasma soluble fibrin monomer complexes (SFMC) and elevated fibronectin (Fn) levels are closely related to the development of diabetic microangiopathy. The purpose of the present study was to explore whether or not changes in plasma glucose levels could have an effect on these protein constituents. Plasma glucose levels of 25 uncontrolled diabetic patients were brought under control with insulin and serial measurements of SFMC and Fn were made over a period of 4 weeks. Glucose values fell from an average of 312 mg/100 ml to 160 mg/100 ml. Ten patients with macroproteinuria (i.e. greater than or equal to 0.5 g/24 hr) showed initially elevated plasma SFMC and Fn concentrations. These levels fell significantly over the 4 week observation period: from 13.6 mg/100 ml to 9.4 mg/100 ml for SFMC and from 38.4 mg/100 ml to 34.5 mg/100 ml for Fn. The remaining 15 patients had nearly normal levels of both SFMC (7.9 mg/100 ml) and Fn (31.1 mg/100 ml) and glycemic control brought no further reduction. The data indicated that a) elevated SFMC and Fn levels are indeed associated with diabetic microangiopathy, especially in the presence of macroproteinuria; and b) adequate glycemic control is capable of normalizing the plasma concentration of these constituents.

Studies in the development of Japanese encephalitis vaccine: expression of virus envelope glycoprotein V3 (E) gene in yeast.

A safe, effective and economical vaccine is required for the prevention of Japanese encephalitis (JE), a disease with high mortality and grave sequelae, which is prevalent in Japan and other countries in east, south-east and southern Asia. As the initial step to produce a second-generation vaccine, recombinant DNA technology was utilized to express the JE virus envelope glycoprotein V3 (E) gene in yeast cells. This report describes the construction of a yeast expression vector in which a cDNA clone covering the V3 gene was connected to the acid-phosphatase promoter of a yeast vector plasmid. Successful expression of the V3 gene was detected by ELISA and Western blotting using monoclonal antibodies against JE V3. Immunization of mice with the V3 antigen expressed in yeast produced limited but definite levels of anti-JE antibodies which could neutralize JE virus. The results are an encouraging step in the development of a practical second-generation JE vaccine.

[The pharmacokinetics of cisplatin and its influence on renal functions based on different infusion methods].

The pharmacokinetics of cisplatin, together with its renal toxicity in relation to different infusion methods were examined. The subjects were 21 in-patients suffering from pulmonary carcinoma treated by anti-neoplastic agents including cisplatin. The patients included 12 given a 5-division dosage, 5 cases given 24-hour continuous infusion, 6 given 12-hour continuous infusion, and 7 given 6-hour continuous infusion. Cisplatin was determined as the concentrations of Pt in plasma and non-protein-bound Pt. Also, as indices of renal function, NAG in urine together with beta 2-microglobulin were examined. No substantial difference could be found in Pt concentration in plasma upon completion of the respective infusion methods. A dual-phase attenuation curve was obtained after the completion of infusion and the half-life period of the beta-phase was around 200 hours. With respect to non-protein-bound Pt concentrations, the 5-division dosage showed the highest value, which was considered to be a temporary phenomenon, while with continuous infusion, the shorter the infusion period, the higher the value became. With respect to the amount of NAG excreted in urine, 12-and 6-hour continuous infusion showed the higher value in comparison with the others in terms of the maximum value and the total amount. With regard to beta 2-microglobulin, 6-hour continuous infusion showed the highest value. The above results therefore suggest that in continuous infusion, the shorter the infusion period the greater the degree of renal tubule injury, and that particularly, 6-hour continuous infusion will possibly cause dysfunction of the renal tubules.

[Laboratory and clinical studies of flomoxef in the pediatric field].

We have carried out laboratory and clinical studies on flomoxef (FMOX, 6315-S). The results were summarized as follows. FMOX was given by 5-minute intravenous administration to 3 children at a single dose of 20 mg/kg. After the intravenous administration, mean serum levels of FMOX were 110.1 +/- 30.95 micrograms/ml at the end of injection, 44.4 +/- 10.55 micrograms/ml at 15 minutes, 11.0 +/- 1.72 micrograms/ml at 1 hour and 0.42 +/- 0.17 microgram/ml at 6 hours. The mean half-life was 1.14 +/- 0.30 hours. The mean urinary excretion rate was 68.8 +/- 17.4% up to 6 hours after the intravenous administration. FMOX was given by 30-minute drip infusion to 2 children at a single dose of 20 mg/kg and to 3 children at a single dose of 40 mg/kg. After the 30-minute drip infusion, mean peak serum levels of FMOX obtained for the 2 dose levels were 45.5 +/- 0.45 micrograms/ml and 87.4 +/- 18.35 micrograms/ml at the end of injection, respectively, and mean half-lives were 0.63 +/- 0.23 hours and 0.70 +/- 0.27 hours, respectively. The mean urinary excretion rate was 53.4 +/- 6.1% up to 6 hours after the 30-minute drip infusion of 40 mg/kg FMOX. Treatment with FMOX was made in 24 cases of pediatric bacterial infections; 5 cases of purulent tonsillitis, 2 cases of bronchopneumonia, 12 cases of pneumonia, and 1 case each of lymphadenitis, pyothorax, purulent meningitis, cellulitis, and abscess. Results obtained were excellent in 15 cases and good in 9 cases. No significant side effect due to the drug was observed in any cases.

[Laboratory and clinical studies of cefuzonam in pediatric field].

We have carried out laboratory and clinical studies of cefuzonam. The results were summarized as follows: The effectiveness of cefuzonam was estimated by a plate dilution method on 26 strains each of S. aureus, E. coli, K. pneumoniae, Salmonella spp. and P. aeruginosa and 27 strains of S. marcescens isolated from patients. The distribution of MIC's of cefuzonam against S. aureus was 0.39 approximately 1.56 micrograms/ml and the peak of the distribution was 0.39 microgram/ml. Strains of 96.2% of E. coli and Salmonella spp. were inhibited at cefuzonam concentrations less than 0.39 microgram/ml. Strains of 92.3% of K. pneumoniae were inhibited at drug concentrations less than 0.20 microgram/ml. The distribution of MIC's of cefuzonam against S. marcescens was less than or equal to 0.025 approximately 12.5 micrograms/ml and the peak of the distribution was 0.2 microgram/ml and 1.56 microgram/ml. MIC's against P. aeruginosa was 12.5 approximately greater than 100 micrograms/ml. Cefuzonam was given by 5-minute intravenous administration to 4 children and 1-hour drip infusion to 1 child at a single dose of 20 mg/kg. After the intravenous administration, mean serum levels of cefuzonam were 30.8 +/- 4.55 microgram/ml at 30 minutes, 13.8 +/- 1.83 micrograms/ml at 1 hour, 0.4 +/- 0.159 microgram/ml at 6 hours. The half-life was 1.12 +/- 0.198 hours. After the drip infusion, the serum levels of the drug were 38.7 micrograms/ml at 1 hour, 5.25 micrograms/ml at 2 hours and 0.087 microgram/ml at 7 hours. The half-life was 0.93 hour. The mean urinary excretion rate was 58.1% and 33.4% up to 6 hours after the intravenous administration and the drip infusion, respectively. Cefuzoname was effective in 4 out of 5 cases with bacterial infections. No side effect due to the drug was observed in any case.

Effect of atrial natriuretic factor on the plasma aldosterone response to potassium infusion in rats--in vivo study.

Previous in vitro studies have shown that atrial natriuretic factor inhibits the secretion of aldosterone stimulated by AII, ACTH, and potassium in adrenal cell suspensions. The present study investigated the effects of atriopeptin II on the plasma aldosterone response to a potassium infusion in conscious unrestrained rats in vivo. The infusion of potassium chloride solution increased plasma aldosterone level from 20.4 +/- 3.7 to 168.4 +/- 27.3 ng/dl. The simultaneous administration of atriopeptin II reduced the increase in plasma aldosterone level (16.0 +/- 2.1 to 63.3 +/- 10.4 ng/dl). There was no significant difference in the plasma renin activity, corticosterone, or serum potassium levels between the two groups. These results suggest that atriopeptin II may be important in the regulation of aldosterone secretion.

A new restriction endonuclease from Spirulina platensis.

Three restriction endonucleases, Sp1I, Sp1II and Sp1III have been purified partially from Spirulina platensis subspecies siamese and named. Sp1I cleaves bacteriophage lambda DNA at one site, phi X 174 RF DNA at two sites, but does not cleave pBR322 DNA. This enzyme recognizes the sequence 5'CGTACG3' 3'GCATCG5' and cuts the site indicated by the arrows. Sp1II is an isoschizomer of Tth111I and Sp1III is an isoschizomer of HaeIII.

Clinicopathologic study of the postoperative maxillary cyst.

Sixty cases of the postoperative maxillary cyst were studied clinicopathologically. The cysts accounted for 19.5% of all oral cystic lesions. Most of the patients were in their 20s and 30s at the time of diagnosis, and the postoperative duration was between 10 and 49 years. Radiographically, most cases revealed a unilocular cystic lesion. In two thirds of the cases, the lesion occupied more than half the volume of the maxillary sinus while the remaining one third of the cases revealed more limited lesions of the sinus. Histopathologically, cuboidal, squamous, and mixed epithelial cyst linings were observed, although the basic epithelial lining was the ciliated columnar type. Epithelial dysplasia was found in two cases. The cysts near the nasal cavity may originate from regenerated nasal mucosa, and the cysts that are located at a distance from the nasal cavity may be lined with epithelium derived from residual mucosa of the sinus entrapped during surgery to relieve sinusitis.
