Differential regulation of neurogenesis by the two Xenopus GATA-1 genes.

Previously, we have shown that the ventralizing factor bone morphogenetic protein 4 (BMP-4) can inhibit Xenopus neurogenesis. The erythroid transcription factor GATA-1 functions downstream of the BMP-4 signaling pathway and mediates BMP-4-induced erythropoiesis. We have found that similar to BMP-4, GATA-1b inhibits neuralization of Xenopus animal cap (AC) cells. The neural inhibition is not seen with GATA-1a, although both GATA-1a and GATA-1b RNAs are translated at the same efficiency and induce globin expression equally in AC cells. GATA-1b RNA injection into AC cells neither induces expression of Xbra (a general mesoderm marker) nor affects expression of XK81 (epidermal keratin) or BMP-4 and Xvent-1 (two ventral markers). These data suggest that GATA-1b retains the epidermal fate of the AC. Intact GATA-1b protein is required for both inhibition of neurogenesis and induction of globin expression. Our findings indicate that GATA-1b can function in ectoderm to specifically regulate neural inducing mechanisms, apparently related to the expression of chordin, a neuralizing gene. Furthermore, tadpole stage embryos injected with GATA-1b are devoid of all dorsoanterior structures including neural tissue. This report provides evidence that the two transcription factors, derived from a recent genome duplication, share a common biological activity (stimulation of erythropoiesis) while also exhibiting a distinct function (inhibition of neurogenesis).

Characterization of 50wt%SiO2-20wt%P2O5-30wt%CaO glass-ceramic powders prepared by the sol-gel process.

Glass-ceramic powders with the oxide batch formulation 50wt%SiO2, 20wt%P2O5 and 30wt%CaO were prepared by the sol-gel process using tetraethoxysilane, calcium nitrate and phosphoric acid as raw materials. The obtained gels and powders were characterized by TG-DTA thermal analysis, FTIR analyses and XRD measurements. The results indicated that (1) heating to more than 600 degrees C brought about elimination of -OC2H5 side chains from gels, and formation of a SiO2-based glass network, (2) crystallization such as Ca3(PO4)2 notably occurred in the SiO2-based glass matrix when heated to more than 900 degrees C, (3) -OH groups existed on SiO2-based glass-ceramic surfaces in the temperature region less than 900 degrees C, and (4) the melting point of this composition was more than 1400 degrees C. This study suggests that sol-gel-derived SiO2-based glass-ceramic powder containing P and Ca might be useful as bioactive implant materials.

Heterogeneous fluoridated hydroxyapatites partially substituted with fluoride ions.

The partially substituted fluoridated apatites H-FHAp0.5 and FH-HAp0.5 were synthesized heterngeneously by supplying fluoride at a concentration equivalent to half of the maximum value of fluorapatite during the initial or final half of the experimental period. Although X-ray diffraction patterns and SEM photos of H-FHAp0.5 and FH-HAp0.5 apatites were not significantly different, high-resolution electron microscopy showed quite different features; H-FHAp0.5 type apatite was an elongated hexagon with electron beam damage in the core, and FH-HAp0.5 type apatite was a rather wide hexagon analogous to fluorapatite. Energy dispersive spectroscopy attached to the SEM showed H-FHAp0.5 apatite to have a higher fluoride peak intensity than FH-HAp0.5. These results supported the previous speculations on the two different types of heterogeneous fluoridated hydroxyapatites: hydroxyapatite covered with fluorapatite and fluorapatite coverted with hydroxyapatite.

[A case of chronic necrotizing pulmonary aspergillosis successfully treated with combination therapy of antifungal drugs and ulinastatin].

A 64-year-old woman with a history of old tuberculosis, had a fungus ball shadow with meniscus sign in the upper right lung field on a chest X-ray film in 1991. Based on the chest X-ray findings, pulmonary aspergilloma was suspected. Because the size of the intracavitary fungus ball increased, the patient was treated with itraconazole over one year in 1995, but there was no improvement. One month later, she was admitted because of fever, hemoptysis and productive cough, and chest X-ray showed an enlargement of intracavitary mass and infiltrative shadow in the right lung. Chronic necrotizing aspergillosis was diagnosed on the basis of her clinical and radiographic features, and positive serological test. Although itraconazol and amphotericin B were given, cavity and intracavitary fungus ball shadow kept growing. Combination therapy of antifungal drugs and ulinastatin markedly improved symptoms and resulted in complete disappearance of the fungus ball on chest CT scan.

Determination of hepatitis delta virus (HDV)-RNA in asymptomatic cases of HDV infection.

OBJECTIVES: To assess the frequency of hepatitis delta virus (HDV) viremia in asymptomatic cases of HDV infection and the clinical significance of the HDV viremia, we conducted a cross-sectional, community-based study. METHODS: Of 2207 examinees, 210 (9.5%) were found to be positive for hepatitis B surface antigen (HBsAg). Antibody to HDV was detected in 47 (22.4%) of the 210 examinees, and 43 of the 47 were further evaluated for serum HDV-RNA by polymerase chain reaction. RESULTS: Twenty-one (48.8%) of the 43 had detectable levels of HDV-RNA in serum, and 22 (51.2%) were negative for serum HDV-RNA. The majority (61.9%) of the HDV-RNA-positive HBsAg carriers had high levels of serum ALT. In contrast, the frequency of an abnormally high level of serum ALT was only 9.1% in the HBsAg carriers positive for HDV antibody but negative for HDV-RNA, and the frequency did not differ from that seen in the HBsAg-negative individuals. The semiquantified HDV-RNA levels did not correlate with the serum ALT levels. CONCLUSION: Seropositivity of HDV-RNA was strongly associated with liver cell damage, even in asymptomatic cases. The absence of a detectable level of serum HDV-RNA might be related to previous HDV infection.

Interactions of the LIM-domain-binding factor Ldb1 with LIM homeodomain proteins.

The LIM homeodomain (LIM-HD) proteins, which contain two tandem LIM domains followed by a homeodomain, are critical transcriptional regulators of embryonic development. The LIM domain is a conserved cysteine-rich zinc-binding motif found in LIM-HD and LMO (rhombotin or Ttg) proteins, cytoskeletal components, LIM kinases and other proteins. LIM domains are protein-protein interaction motifs, can inhibit binding of LIM-HD proteins to DNA and can negatively regulate LIM-HD protein function. How LIM domains exert these regulatory effects is not known. We have now isolated a new LIM-domain-binding factor, Ldb1, on the basis of its ability to interact with the LIM-HD protein Lhx1 (Lim1). High-affinity binding by Ldb1 requires paired LIM domains and is restricted to the related subgroup of LIM domains found in LIM-HD and LMO proteins. The highly conserved Xenopus Ldb protein XLdb1, interacts with Xlim-1, the Xenopus orthologue of Lhx1. When injected into Xenopus embryos, XLdb1 (or Ldb1) can synergize with Xlim-1 in the formation of partial secondary axes and in activation of the genes encoding goosecoid (gsc), chordin, NCAM and XCG7, demonstrating a functional as well as a physical interaction between the two proteins.

Selectivity of the parietal visual neurones in 3D orientation of surface of stereoscopic stimuli.

We identified a group of neurones in the posterior parietal cortex of the monkey that responded preferentially to a flat stimulus in a particular orientation on a 3D computer graphics display. We designated these neurones surface-orientation selective (SOS) neurones since they showed tuning to the 3D orientation of the surface. Almost all of the SOS neurones were sensitive to binocular disparity and their responses increased with the width or length of the stimulus, but the responses of only some of them decreased with increasing thickness of the stimulus. These results suggest that SOS neurones extract surface orientation signals from the binocular disparity signals and play an important role in the perception of 3D shape and the visual guidance of hand movement.

Characterization of the cDNA and gene encoding human PDE3B, the cGIP1 isoform of the human cyclic GMP-inhibited cyclic nucleotide phosphodiesterase family.

Two distinct PDE3 [cyclic GMP-inhibited cyclic nucleotide phosphodiesterase (cGI PDE)] isoforms, cGIP1 and cGIP2, have been identified. Here we report cloning of the cDNA and gene encoding human (H)cGIP1 (classified as PDE3B). The cDNA encodes a protein of 1112 amino acids (approximately 123 kDa). Northern blots indicate that its mRNA is expressed in several adipose tissue depots. The human PDE3B gene is composed of 16 exons spanning more than 114 kb and was localized to chromosome 11p15 by in situ hybridization. Exon/intron boundaries were determined, and genetic polymorphism, confirmed by single-strand conformational polymorphism of DNA from 25 healthy subjects, was demonstrated in exon 4 at nucleotide 1389 (A/G). Two polymorphic dinucleotide repeat sequences were identified in introns 5 and 12.

Molecular cloning, structure, and chromosomal localization of the mouse LIM/homeobox gene Lhx5.

Lhx5, the mouse ortholog of the Xenopus Xlim-5, is a LIM/homeobox gene expressed in the central nervous system during both embryonic development and adulthood. During development its domain of expression is mainly localized at the most anterior portion of the neural tube, and it precedes the morphological differentiation of the forebrain; for this reason we believe that Lhx5 could play an important role in forebrain patterning. Here we present the structural organization and the chromosomal localization of the Lhx5 gene. The gene is composed of five exons spanning more than 10 kb of genomic sequence. The first and second LIM domains are encoded by the first and second exon, while the codons of the homeobox are split between the third and the fourth exons. The structure of Lhx5 is similar to that of other LIM/homeodomain proteins, Lhx1/lim1 and Lhx3/lim3, but differs from that of other LIM genes, such as mec3 and LMO1/Rbtn1, in which the codons for the LIM domains are interrupted by introns. We have mapped Lhx5 to the central region of mouse chromosome 5.

Characterization of two recombinant PDE3 (cGMP-inhibited cyclic nucleotide phosphodiesterase) isoforms, RcGIP1 and HcGIP2, expressed in NIH 3006 murine fibroblasts and Sf9 insect cells.

cDNAs encoding PDE3 [cGMP-inhibited cyclic nucleotide phosphodiesterase (cGI PDE)] isoforms, cGIP1 and cGIP2, have been cloned from rat (R) and human (H) cDNA libraries. The deduced amino acid sequences of RcGIP1 and HcGIP2 are very similar in their conserved catalytic domains but differ in their N-terminal regulatory domains [Meacci, E., et al. (1992) Proc. Natl. Acad. Sci. U.S.A. 89, 3721-3725; Taira, M., et al. (1993) J. Biol. Chem. 268, 18573-18579]. cDNAs encoding both rat adipocyte RcGIP1 and human myocardial HcGIP2 (full-length forms and truncated forms lacking much of the putative N-terminal domain) were expressed in NIH 3006 fibroblasts and in Sf9 insect cells. The recombinant proteins exhibited the expected subunit molecular mass, immunologic reactivities, and characteristics of native membrane-associated forms of the enzymes, e.g., high affinity for cAMP (Km), sensitivity to the selective cGI PDE inhibitors OPC 3689 and OPC 3911 and to cGMP. The full-length recombinants were predominantly particulate, whereas the truncated HcGIP2 forms were cytosolic suggesting that N-terminal domains contain structural determinants important for membrane association. Both fibroblast RcGIP1 and authentic adipocyte cGI PDE were phosphorylated in vitro by cAMP-dependent protein kinase; tryptic [32P]peptides released from rat adipocyte 32P-cGI PDE and 32P-RcGIP1 exhibited identical electrophoretic profiles suggesting that the same peptides are phosphorylated in both.

Differential expression of cGMP-inhibited cyclic nucleotide phosphodiesterases in human hepatoma cell lines.

PDE3 or cGMP-inhibited cyclic nucleotide phosphodiesterase (cGI PDE) activity was detected in homogenates of HepG2, Hep3B and HuH7, but not SK-Hep-1, human hepatoma cells. In HepG2 and Hep3B cells PDE3 activity was found predominantly in particulate fractions; in HuH7, in both particulate and supernatant fractions. cDNAs encoding two human PDE3s (an 'adipocyte' type, HcGIP1, and a 'cardiovascular' type, HcGIP2) have been cloned. HcGIP1 cDNA hybridized strongly with poly(A)+ RNA species from HepG2 and Hep3B. Both HcGIP1 and HcGIP2 mRNAs were expressed in Hep3B and HuH7 cells. The nucleotide sequence of an approximately 300-bp cDNA fragment, isolated after RT-PCR cloning from HepG2 RNA, was identical to a sequence within the conserved domain of HcGIP1 cDNA, consistent with the presence of HcGIP1 mRNA in HepG2 cells.

On the relations between single cell activity in the motor cortex and the direction and magnitude of three-dimensional static isometric force.

We examined the relations between the steady-state frequency of discharge of cells in the arm area of the motor cortex of the monkey and the direction and magnitude of the three-dimensional static force exerted by the arm on an isometric manipulandum. Data were analyzed from two monkeys (n = 188 cells) using stepwise multiple linear regression. In 154 of 188 (81.9%) cells the regression model was statistically significant (P < 0.05). In 121 of 154 (78.6%) cells the direction but not the magnitude of force had a statistically significant effect on cell activity; in 11 of 154 (7.1%) cells only the magnitude effect was significant; and in 22 of 154 (14.3%) cells both the direction and magnitude effects were significant. The same analysis was used to assess the effect of the direction and magnitude of force on the electromyographic activity of 9 muscles of the arm and shoulder girdle. The regression model was statistically significant. For all the muscles studied in 4 of 9 (44.4%) muscles only the direction effect was significant whereas in the remaining 5 of 9 (55.6%) muscles both the direction and the magnitude were significant. No muscle studied showed a significant effect of force magnitude alone. These differences in the frequency of occurrence of directional and magnitude effects between cells and muscles were statistically significant (P < 0.005, chi 2 test). These findings underscore the fundamental importance of the direction of force in space for both motor cortical cells and proximal muscles and underline the differential relations of the cells and muscles to the direction and magnitude of force. These results indicate that the specification of the magnitude of three-dimensional force is embedded within the directional signal; this combined direction+magnitude effect was 3.9 times more prevalent in the muscles than in the cells studied. In contrast, the pure directional effect was 1.8 times more prevalent in the cells than in the muscles studied. This suggests that the direction of force can be controlled independently of its magnitude and that this direction signal is especially prominent in the motor cortex.

Detection of an amino acid polymorphism in hormone-sensitive lipase in Japanese subjects.

Hormone-sensitive lipase (HSL) plays an important role in energy metabolism by controlling the hydrolysis of triglycerides stored in adipose tissue. To investigate whether mutations in the HSL gene are associated with non-insulin-dependent diabetes mellitus (NIDDM), we screened for mutations of this gene using single-stranded conformation polymorphism (SSCP) in 35 Japanese subjects with NIDDM. SSCP analysis identified a variant pattern in axon 4, and the sequence showed that this variant pattern resulted from amino acid polymorphism (Arg309Cys). Subsequent study showed that this polymorphism was found in 18 of 151 NIDDM patients and 10 of 97 nondiabetic subjects, but allele frequency was not significantly different between the two groups (P = .7). Body mass index, serum triglyceride, and high-density lipoprotein (HDL) cholesterol were not different in subjects with and without the polymorphism. But serum total cholesterol was higher in subjects with the polymorphism than in subjects without it (P = .0005). These data indicate that this HSL polymorphism is not associated with NIDDM, obesity, and serum triglyceride level. However, an effect of the polymorphism to elevate serum total cholesterol has not been excluded, although further study is necessary to resolve its association with cholesterol metabolism.

Cutting effectiveness of diamond points on commercial core composite resins and cements.

In dental clinics, composite resin and cement cores are routinely cut and polished during abutment tooth preparation. To identify their characteristics during cutting, weight-load cutting tests were performed on eight commercial core composites and two cements, using diamond points driven by an air-turbine handpiece. It became evident that the cutting effectiveness of the diamond points on nine workpieces exceeded that on natural tooth dentine, while that on one composite containing Si3,N4 filler was analogous to that on dentine. With continued use, the cutting effectiveness of the diamond point on all workpieces gradually declined. SEM observations revealed that diamond particles of the diamond point wore out with repeated use. It is clinically advised to select the core material with material characteristics during cutting and mechanical strength similar to those of dentin.

The LIM homeodomain protein Lim-1 is widely expressed in neural, neural crest and mesoderm derivatives in vertebrate development.

Polyclonal antibodies to Xlim-1 homeodomain protein of Xenopus laevis were used to study the developmental expression pattern of this protein in Xenopus, rat and mouse. Western blotting of embryo extracts injected with different Xlim-1 constructs confirmed the specificity of the antibody. Beginning at the gastrula stage, Xlim-1 protein was detected in three cell lineages: (i) notochord, (ii) pronephros and (iii) certain regions of the central nervous system, in agreement with earlier studies of the expression of Xlim-1 RNA (Taira et al., Development 120: 1525-1536, 1994a). In addition, several new locations of Xlim-1 expression were found, including the olfactory organ, retina, otic vesicle, dorsal root ganglia and adrenal gland. Similar expression patterns were seen for the Lim-1 protein in frog and rodent tissues. These observations implicate the Xlim-1 gene in the specification of multiple cell lineages, particularly within the nervous system, and emphasize the conserved nature of the role of this gene in different vertebrate animals.

[Pneumonia caused by Saiboku-to].

A 72-year-old man was being treated with Saiboku-to for throat discomfort. Forty-two days after the start of Saiboku-to therapy, dyspnea and fever developed and he was admitted to the hospital. Arterial blood gas analysis showed arterial hypoxemia, and a chest X-ray film and chest CT scan showed diffuse linear-nodular shadows in both lung fields. The results of a lymphocyte-stimulation test were positive for Saiboku-to and Saiboko-to-induced pneumonia was diagnosed. Saiboku-to was stopped and prednisolone was given. The symptoms, respiratory status and chest X-ray findings markedly improved. To our knowledge, this is the fifth case of Saiboku-to-induced pneumonia reported in Japan.

[Six patients with pneumonitis related to blended Chinese traditional medicines].

We encountered six patients with pneumonitis related to blended chinese traditional medicine (Kampo). The duration of treatment with kampo ranged from 14 to 110 days (mean: 38 days). The most common complaints were dyspnea, fever, and dry coughing. Fine crackles were heard at the bases of both lungs. Abnormal laboratory findings included high values of C-reactive protein and glutamic-oxaloacetic transaminase in all patients, lactate dehydrogenase in 5 patients, and eosinophil count in 1 patient. Chest X-ray films and CT films revealed diffuse reticulo-nodular interstitial shadows with consolidation in both lung fields in 3 patients and pleural effusion in 1 patient. Bronchoalveolar lavage was done in 4 patients; examination of the lavage fluid showed lymphocyte alveolitis, either pure or associated with neutrophilia and eosinophilia in 3 patients. Inverted CD4/CD8 lymphocyte ratios were found in 3 patients. Transbronchial lung biopsy was done in 4 patients and specimens from 3 of those 4 showed organizing pneumonitis with thickening of alveolar septa. Lymphocyte stimulation tests were positive in 4 patients. Discontinuation of the drug (2 patients) or administration of corticosteroids (4 patients) was followed by rapid improvement. Patients being treated with kampo preparations should be observed for signs and symptoms of drug-induced pneumonitis.
