[Cross protection between an encapsulated strain of Staphylococcus hyicus and an encapsulated strain of Staphylococcus epidermidis].

Strain ST67P of Staphylococcus hyicus was capsular type I (++)/II(+) of S. epidermidis as determined by the method of Ichiman. To mice immunized with heat-killed vaccine of strain ST67P, homologous strain and strain ATCC 31432 (capsular type I), SE-360 (capsular type II) and SE-10 (capsular type III) of S. epidermidis were injected intraperitoneally into mice, then, the viable cell number of the organisms in the peritoneal cavity were enumerated of 30 minutes and 20 hours after the injection. Results showed that the viable cell number of the homologous strain and strain ATCC 31432 was remarkably decreased at 20 hours after the injection, however, the cells were increased with strain SE-360 and SE-10. Passive protective activity of rabbit anti-ATCC 31432 serum was absorbed either with homologous strain or strain ST67P in the mouse, however, protective activity of anti-SE-360 strain serum and anti-SE-10 strain serum was not absorbed with these organisms although the activity was absorbed with homologous organisms. With an ultrathin section preparation of strain ST67P conjugated with ferritin-labelled rabbit anti-homologous strain serum, numerous ferritin granules surrounding the outermost layer of large capsule were electronmicro-scopically demonstrated. In the same organisms treated with ferritin-labelled anti-ATCC31432 strain serum, the all walls were surrounded by a relatively thinner capsule and a number of ferritin granules were located in the outermost layer of the capsule. However, with the organisms treated with ferritin-labelled anti-SE-360 strain serum only a number of ferritin granules were shown on the surface of the cell walls, and neither capsule nor ferritin granules were exhibited in the organisms treated with ferritin-labelled anti-SE-10 strain serum.

Binding of staphylococcal cell surface polysaccharide to human fibrinogen.

The interaction between the binding site of a polysaccharide (called compact colony forming active substance (CCFAS)), obtained from the cell surface of a strain of Staphylococcus, and human fibrinogen (HF) was investigated. The CCFAS was found to bind specifically to both the B beta and gamma chains of HF at pH 7.0 and 8.0, and the A alpha chain at pH 5.0. The binding of CCFAS with fibrinogen fragments obtained by digestion with plasmin were also investigated. Fragments with Mr of 55,000, 24,000, and 19,000 were the major bands precipitated by CCFAS at pH 7.0 and 8.0. Fragments with Mr of 85,000 and 75,000 bound to CCFAS at pH 5.0. Binding of CCFAS (7 micrograms) with fibrinogen could be inhibited by 1.2 micrograms of B beta chain and 1.5 micrograms gamma chain at alkaline pH or 6.2 micrograms of the A alpha chain at pH 5.0. CCFAS was, therefore, assumed to be specifically bonded with HF molecules, in the alkaline range at least, resulting in compact colony forming activity in serum soft agar and paracoagulation.

Specific binding of okadaic acid, a new tumor promoter in mouse skin.

The tumor promoter okadaic acid binds specifically to a particulate as well as a cytosolic fraction of various mouse tissues, e.g., skin, brain, lung and colon. The KD value was 21.7 nM for receptors in the particulate fraction and 1.0 nM for those in the cytosolic fraction of mouse skin. The specific binding of [3H]okadaic acid to the particulate fraction of mouse skin was inhibited dose-dependently by okadaic acid, but not okadaic acid tetramethyl ether, an inactive compound, or by other tumor promoters, such as 12-O-tetradecanoylphorbol-13-acetate and teleocidin. The results suggest a new pathway of tumor promotion mediated through the okadaic acid receptor(s).

Apparent "activation" of protein kinases by okadaic acid class tumor promoters.

A cytosolic fraction of mouse brain gave two peaks of protein kinase activity on DEAE-cellulose column chromatography. The first peak of protein kinase corresponded to protein kinase C. The second peak contained protein kinases that were "activated" dose-dependently by the okadaic acid class tumor promoters, okadaic acid and dinophysistoxin-1. This "activation" was not achieved by other tumor promoters, such as 12-0-tetradecanoyl-phorbol-13-acetate, teleocidin, aplysiatoxin, or palytoxin. In addition, the second peak contained phosphatases. The phosphate liberation from phosphorylated histone type III-S by incubation with the second peak was inhibited by okadaic acid or dinophysistoxin-1, dose-dependently. The resulting apparent "activation" of protein kinases by okadaic acid is indicated and would imply a new pathway of tumor promotion on mouse skin.

Codon 61 mutations in the c-Harvey-ras gene in mouse skin tumors induced by 7,12-dimethylbenz[a]anthracene plus okadaic acid class tumor promoters.

Three okadaic acid class tumor promoters, okadaic acid, dinophysistoxin-1, and calyculin A, have potent tumor-promoting activity in two-stage carcinogenesis experiments on mouse skin. DNA isolated from tumors induced by 7,12-dimethylbenz[a]anthracene (DMBA) and each of these tumor promoters revealed the same mutation at the second nucleotide of codon 61 (CAA----CTA) in the c-Ha-ras gene, determined by the polymerase chain reaction procedure and DNA sequencing. Three potent 12-O-tetradecanoylphorbol-13-acetate (TPA)-type tumor promoters, TPA, teleocidin, and aplysiatoxin, showed the same effects. These results provide strong evidence that this mutation in the c-Ha-ras gene is due to a direct effect of DMBA rather than a selective effect of specific tumor promoters.

Sarcophytols A and B inhibit tumor promotion by teleocidin in two-stage carcinogenesis in mouse skin.

Sarcophytols A and B, isolated from a soft coral, Sarcophyton glaucum, are cembrane-type diterpenes with different numbers of hydroxyl groups. Sarcophytols A and B inhibited tumor promotion by teleocidin in two-stage carcinogenesis experiments on mouse skin. The inhibitory effect of sarcophytol A was demonstrated with two different initiating doses of 7,12-dimethylbenz[a]anthracene (DMBA): 50 micrograms (experiment 1) and 100 micrograms (experiment 2). In experiment 1, three groups of mice were treated with DMBA, and then twice a week with doses (1.6 micrograms, 16 micrograms, and 82 micrograms) of sarcophytol A followed by 2.5 micrograms teleocidin. In week 25, the incidences of tumors in these groups were only 7.1%, 20.0%, and 13.3%, respectively, whereas that in the control group treated with DMBA plus teleocidin was 53.3%. Moreover, at this time, the average numbers of tumors per mouse in these groups were 0.1, 0.3, and 0.3, respectively, while that in the control group was 2.1. In experiment 2 an amount of sarcophytol A (1.6 micrograms) or B (1.7 micrograms) equimolar to 2.5 micrograms teleocidin was applied twice a week, as in experiment 1, and results showed that sarcophytol B also inhibited tumor promotion by teleocidin. Both sarcophytols A and B caused delay in onset of tumor formation, and reduced the percentage of tumor-bearing mice and the average number of tumors per mouse. The effective concentrations of sarcophytols A and B were in the microgram range with an equimolar amount of teleocidin.

[Trypan blue staining capacities of the culture cells (2)--The mechanism of staining].

The cultured cells dispersed by rubber policeman were stained with trypan blue, but these cells were viable at the meaning of keeping viral infective center forming capacity and at the meaning that trypan blue stained cells were transferred to unstained ones by trypsin treatment. This phenomenon may be important not to miscount numbers of the viable cells in cultured cells. Phagocytosis (endocytosis) is well known as a reaction inhibited by NaF or monoiodoacetate, but these metabolic inhibitors did not inhibit trypan blue staining of the cells dispersed both by rubber policeman and 0.05% trypsin treatment. The cells suspended in higher osmotic pressure media were more stainable with trypan blue than lower ones. Addition of K+ to the reaction mixture was not effective on trypan blue staining. These results suggest that "alive" cells dispersed by rubber policeman are stained with trypan blue not to utilize phagocytosis (endocytosis) or Na+-K+ pump, whose reactions couple with energy production. Trypan blue staining capacity of the cultured cells may be depended on pore sizes of protein channels (for example, fixed or gated transport protein channel) in lipid-bilayers of cell membranes. It is concluded that the cells dispersed by trypsin treatment (spherical shape in microscopical observation) and spherical cells in rubber policeman dispersing ones are unstained with trypan blue to contain smaller pore size channels in the cell membranes, but the rest, amoeba-like form cells dispersed by rubber policeman and dead cells are stained to contain larger ones.

Similar, potent tumor-promoting activity of all isomers of teleocidins A and B in a two-stage carcinogenesis experiment on the skin of CD-1 mice.

Teleocidin, isolated from mycelia of Streptomyces mediocidicus is a mixture of two teleocidin A isomers with molecular weights of 437 (A-1 and A-2) and four teleocidin B isomers with molecular weights of 451 (B-1, B-2, B-3, and B-4). Previously we found that each purified isomer of teleocidins A and B had approximately the same activity as teleocidin in an irritant test on mouse ear, in inductions of ornithine decarboxylase in mouse skin and adhesion of human promyelocytic leukemia (HL-60) cells, and in inhibition of the specific binding of [3H]-12-O-tetradecanoylphorbol-13-acetate to a mouse skin particulate fraction. This paper reports the strong activation of protein kinase C in vitro by each isomer of teleocidins A and B at a concentration of 1 microgram/ml. Detailed studies on the potent tumor promoting activities of the two teleocidin A isomers and four teleocidin B isomers in a two-stage carcinogenesis experiment on mouse skin are also reported, including histological findings on the tumors. Treatment of mice with 100 micrograms of 7,12-dimethylbenz(a)anthracene and then 2.5 micrograms of any one of the six isomers of teleocidins A and B twice a week induced tumors in 80.0 to 91.7% of the mice with 2.8 to 5.2 tumors/mouse in week 30. Scarcely any tumors developed in groups treated with 7,12-dimethylbenz(a)anthracene or any one of the isomers of teleocidins A or B alone. The percentages of incidences of mice bearing papillomas and carcinomas in the six groups treated with 7,12-dimethylbenz(a)anthracene plus one isomer of teleocidins A or B were 90.9 to 98.3% and 1.7 to 9.1%, respectively. These results indicate that all of the isomers of teleocidins A and B have potent tumor promoting activity on mouse skin, irrespective of the structural differences between teleocidins A-1 and A-2, and among the four isomers of teleocidin B. The structure-activity relationship of teleocidins A and B is discussed on the basis of our recent results. Based on the structures of related compounds, we propose a revised numbering system for compounds of the teleocidin class.

Specific binding of [3H]retinoids to cellular retinoid-binding proteins.

The specific binding of [3H]retinoids to cellular retinoid-binding proteins was measured directly by the cold acetone filtration method. After incubation of purified cellular retinoid-binding proteins with [3H]retinoids with or without competitors for 2-4 h, bound ligands were separated from free by filtration using cold acetone. Nonspecific binding of the ligands was reduced sufficiently to allow measurement of specific binding of [3H]retinoids to cellular retinoid-binding proteins. This method has the advantages of being rapid and practical and giving reproducible results.

Inhibition of ornithine decarboxylase induction by retinobenzoic acids in relation to their binding affinities to cellular retinoid-binding proteins.

Retinobenzoic acids induce differentiation of human promyelocytic leukemia cells (HL-60). Like retinoic acid, 14 retinobenzoic acids inhibited the induction of ornithine decarboxylase (ODC) by teleocidin in mouse skin. The mechanism(s) of inhibition of ODC induction by 7 retinobenzoic acids, Am 80, Am 81, Am 580, Am 590, Am 68, Sa 80, and Ch 55 was compared with those by all-trans-retinoic acid and the arotinoid compound 19. Application of 114 nmol of Am 80, Am 81, Am 580, Am 590, Am 68, Sa 80, or Ch 55, 10 min before 11.4 nmol of teleocidin, resulted in 76.7%, 82.0%, 76.2%, 28.3%, 48.4%, 58.6%, and 85.1% inhibition of ODC induction, respectively. Since all-trans-retinoic acid and compound 19 were also inhibitory, we determined whether retinobenzoic acids bind to cellular retinoic acid-binding protein (CRABP) isolated from bovine adrenal glands. Am 80 and Am 580 inhibited the specific binding of 3H-retinoic acid to CRABP, but also showed less affinity than authentic unlabeled retinoic acid and compound 19. Am 81, Am 590, Am 68, Sa 80, and Ch 55 at up to 10 microM were not effective competitors of the binding of either 3H-retinoic acid or 3H-retinol. These results suggest that the inhibition of ODC induction can be mediated by pathways that do not involve CRABP or the cellular retinol-binding protein.

Okadaic acid: an additional non-phorbol-12-tetradecanoate-13-acetate-type tumor promoter.

Okadaic acid is a polyether compound of a C38 fatty acid, isolated from a black sponge, Halichondria okadai. Previous studies showed that okadaic acid is a skin irritant and induces ornithine decarboxylase (OrnDCase; 3-hydroxyl-L-glutamate 1-carboxy-lyase, EC 4.1.1.17) in mouse skin 4 hr after its application to the skin. This induction was strongly inhibited by pretreatment of the skin with 13-cis-retinoic acid. A two-stage carcinogenesis experiment in mouse skin initiated by a single application of 100 micrograms of 7,12-dimethylbenz[a]anthracene (DMBA) and followed by application of 10 micrograms of okadaic acid twice a week revealed that okadaic acid is a potent additional tumor promoter: tumors developed in 93% of the mice treated with DMBA and okadaic acid by week 16. In contrast, tumors were found in only one mouse each in the groups treated with DMBA alone or okadaic acid alone. An average of 2.6 tumors per mouse was found in week 30 in the group treated with DMBA and okadaic acid. Unlike phorbol 12-tetradecanoate 13-acetate (TPA), teleocidin, and aplysiatoxin, okadaic acid did not inhibit the specific binding of [3H]TPA to a mouse skin particulate fraction when added up to 100 microM or activate calcium-activated, phospholipid-dependent protein kinase (protein kinase C) in vitro when added up to 1.2 microM. Therefore, the actions of okadaic acid and phorbol ester may be mediated in different ways. These results show that okadaic acid is a non-TPA-type tumor promoter in mouse skin carcinogenesis.

Diarrhetic shellfish toxin, dinophysistoxin-1, is a potent tumor promoter on mouse skin.

Dinophysistoxin-1, 35-methylokadaic acid, is a causative agent of diarrhetic shellfish poisoning. The biological activities and tumor-promoting activity of dinophysistoxin-1 were studied together with those of okadaic acid and 7-O-palmitoyl okadaic acid. Dinophysistoxin-1 is a skin irritant and induces ornithine decarboxylase in mouse skin with the same potency as okadaic acid. 7-O-Palmitoyl okadaic acid induced a lower activity than the other compounds. Dinophysistoxin-1 inhibited the specific [3H]okadaic acid binding to a particulate fraction of mouse epidermis. The binding affinities of dinophysistoxin-1 and okadaic acid to a particulate fraction were almost the same. Dinophysistoxin-1 showed a tumor-promoting activity as strong as that of okadaic acid in a two-stage carcinogenesis experiment on mouse skin. The percentages of tumor-bearing mice in the groups treated with 100 micrograms of 7,12-dimethylbenz[a]anthracene (DMBA) followed by 5 micrograms of dinophysistoxin-1, twice a week, and with DMBA followed by 5 micrograms of okadaic acid twice a week were 86.7% and 80.0% in week 30, respectively. The average number of tumors per mouse was 4.6 in the former group and 3.9 in the latter. Dinophysistoxin-1 and okadaic acid act on cells through different pathways from the 12-O-tetradecanoylphorbol-13-acetate-type tumor promoters.

Tumor promoting activity of teleocidin in skin and forestomach of mice initiated transplacentally with 7,12-dimethylbenz(a)anthracene.

Experiments on the effect of transplacental initiation with 7,12-dimethylbenz(a)anthracene (DMBA) and postnatal promotion with teleocidin were carried out in mice. The percentage of tumor-bearing mice among females treated with DMBA transplacentally on day 17 of gestation and postnatally by topical application of teleocidin to the skin of the back was 73.3% in week 30, whereas that among females treated with DMBA on day 10 of gestation and postnatally by topical application of teleocidin was 20.0%. This indicates that teleocidin shows potent tumor promoting activity on mouse skin in a transplacental initiation and postnatal promotion protocol. Furthermore, in the males treated with DMBA transplacentally on day 17 of gestation and given diet containing 0.01% teleocidin postnatally five tumors of the forestomach were found in 5 of 19 effective mice (26.3%) in week 52. One of these five tumors was a squamous cell carcinoma, and the others were papillomas. This indicates that teleocidin also has tumor promoting activity in the forestomach of mice.

Resistance of human bronchial epithelial cells to teleocidin in a two-stage carcinogenesis experiment.

The tumor-promoting activity of teleocidin in two-stage carcinogenesis was tested using human bronchi grafted into athymic nude mice. On treatment with 7,12-dimethylbenz(a) anthracene (DMBA) plus teleocidin, no bronchial carcinomas were produced in 44 grafted human bronchi, but 8 spindle cell sarcomas developed in mouse tissues. Application of DMBA to grafted human bronchi produced only 3 spindle cell sarcomas in the tissues of 30 mice. Application of teleocidin did not produce any tumors in 24 grafts of bronchi obtained from heavy smokers. Thus, teleocidin had an effect on mouse tissues, but seemed to have no effect on human bronchial cells, although it is uncertain whether the grafted human bronchi were initiated by DMBA.
