Temperature dependence of synaptic modulation by a FMRFamide-related neuropeptide in crayfish.

The present study examined the temperature dependence of synaptic transmission and peptidergic modulation of chemical synapses on the phasic abdominal extensor muscles of crayfish. Decreasing the temperature from 25 degrees C to 5 degrees C in saline, decreased the EPSP amplitude by 88% and increased the EPSP half-decay time four-fold. The putative neurohormone DRNFLRFamide (DF2) increased EPSP amplitudes, but was more effective at 7-9 degrees C than at 15-17 degrees C. DF2 might play a hormonal role in counteracting low transmitter release at low temperature.

Large dense-core vesicles in rat adrenal after reserpine: levels of mRNAs of soluble and membrane-bound constituents in chromaffin and ganglion cells indicate a biosynthesis of vesicles with higher secretory quanta.

Rats were injected with a large dose of reserpine known to stimulate the adrenal medulla. Various times after drug treatment the mRNA levels of several constituents of large dense-core vesicles were determined by northern blot analysis and in situ hybridization. The latter method allowed detection of changes in mRNA levels not only in chromaffin cells, but also in the ganglion cells found in adrenal medulla. Levels of the mRNAs of secretory components of large dense-core vesicles (chromogranins A and B, secretogranin II, VGF, and neuropeptide Y) increased in chromaffin cells by 215-857% after 1-3 days of drug treatment. For partly membrane-bound components (dopamine beta-hydroxylase, prohormone convertase 2, carboxypeptidase H, and peptidylglycine alpha-amidating monooxygenase) the changes ranged from 182 to 315%, whereas for glycoprotein III and for intrinsic membrane proteins (cytochrome b561 and vesicle monoamine transporter 2) no change occurred. In ganglion cells the mRNAs that could be detected for VGF, neuropeptide Y, secretogranin II, carboxypeptidase H, and vesicle monoamine transporter 1 showed an analogous pattern of change, with significant increases for the secretory proteins and no change for the membrane components. From these and previous results we suggest the following concept: Long-lasting stimulation of chromaffin cells or neurons does not induce the biosynthesis of a larger number of vesicles but rather leads to the formation of vesicles containing higher secretory quanta of chromogranins and neuropeptides.

Estimation of parity progression ratios from the distribution of births by order and age of the mother.

"This paper presents a methodology for estimating parity progression ratios (the probability that a woman after delivering...any particular birth will ever proceed to the next birth), utilizing data on births tabulated by birth order and age of the mother in a particular year. This approach...does not need estimates of [gross reproductive rate] as well as the infant mortality rate but it requires knowledge of the growth rate of the population." The technique is illustrated with data from the 1984-1987 Pakistan Fertility Survey.

Isoenzyme pattern of clones of Entamoeba histolytica.

The isoenzyme pattern of clones of E. histolytica isolated from symptomatic and asymptomatic patients of amoebiasis were determined by starch gel electrophoresis. Ten parent cultures (uncloned), 5 from symptomatic and 5 from asymptomatic patients were isolated and established in Robinsons medium as xenic cultures. A total of 100 clones were isolated from parent cultures but only 65 could be established. Starch gel electrophoresis was carried out on the uncloned and cloned cultures. The analysis showed that the 5 uncloned culture from symptomatic patients had 4 isolates with pathogenic zymodeme and 1 isolate with non-pathogenic zymodeme pattern. The presence of beta band and absence of alpha band for phosphoglucomutase (PGM) and a fast moving Hexokinase (HK) band were taken as markers of pathogenicity. The 5 cloned cultures from asymptomatic patients had three isolates with pathogenic and 2 with non-pathogenic pattern. The cloned isolates from symptomatic patients (35) had 20 pathogenic zymodemes and asymptomatic patients (30) had 15 pathogenic zymodemes. Non-pathogenic zymodeme obtained in the present study were I, IV, V, X and XIII and the pathogenic zymodemes obtained were II and XIV. The data of the study indicated that (i) asymptomatic patients also harbour pathogenic zymodemes; and (ii) zymodeme II also coexsist with zymodeme XIV in the Indian population.

Differentiation of acute promyelocytic leukemia cell induced by 1,25-dihydroxyvitamin D3 and pentoxifylline (Trental).

Human promyelocytic leukemia cell line HL-60 in tissue culture was treated with various concentrations of 1,25-dihydroxyvitamin D3, (D3) pentoxifylline (Px) or in combinations. Differentiation to normal granulocytes or monocytes was evaluated by nitroblue tetrazolium (NBT) reducing capacity and morphologic criteria. (D3) produced monocytic differentiation, but sub-differentiation inducing doses of (Px) had no effect. In previous studies we found higher doses of (Px) to produce granulocytic differentiation in vitro and in vivo.

Principles and practice of medical audit (medical care evaluation).

1. Medical audit is a philosophy in the field of medical science which has reached to an advanced stage of practice in Western World, but yet to reach and percolate into Indian medical community. 2. Of late, community is getting increasingly aware of its health rights, gradually community participation in health matters including quantum and quality of case, has started increasing. Thus community leaders have started demanding for quality of medical care and accountability of those, responsible for delivery of medical care at various levels. 3. Medical audit or Evaluation of medical care is an answer to ensure the quality of care. But there are misgivings and distrust about medical audit due to its terminology. 4. There is need to education the medical, nursing and paramedical staff regarding medical audit and its sole purpose of self education and improvement of patient care activity. The present paper spells out fundamentals of medical audit, its scope and limitations.

Gonado-modulatory effects of high humidity is pineal dependent in Indian palm squirrel Funambulus pennanti (Wroughton).

F. pennanti presented a clear biphasic pineal mediated seasonal sexual cycle. This sexual cycle was essentially characterised by a very short period of sexual quiescence with an arrest of spermatogenesis during October-November. A small but clear decrease in sexual activity was also observed during March-April. This decrease in sexual activity, however, had no quantifiable effect on spermatogenesis. Sexual recrudescence was observed from December-January. The testes remained sexually active from January till September. Almost an inverse relationship was observed between pineal and testicular weight. Pinealectomy, however, prevented naturally induced gonadal regression during both the periods, i.e. September-November and February-April. Exposure of animals to high RH (80 +/- 4%) during sexually active phase induced a steep regression in testicular weight of sham-operated animals even in the presence of gonad stimulatory long photoperiod (16L:8D) and high temperature (40 degrees +/- 5 degrees C) while exposure of animals to moderate RH (65 +/- 5%) during sexual regression phase partially prevented testicular regression even in presence of inhibitory short photoperiod (11L:13D) and normal environmental temperature (30 degrees +/- 5 degrees C). Pinealectomized animals, neither exhibited testicular regression in February-March nor had involuted testes in September-October, thus, suggesting that the effect of humidity is mediated via the pineal gland.

Alternative splicing of human synexin mRNA in brain, cardiac, and skeletal muscle alters the unique N-terminal domain.

Several synexin (annexin VII) mRNAs have been identified by screening a human fibroblast cDNA library. One type of message contained an alternatively spliced cassette exon, predicting two isoforms of synexin differing in the N-terminal domain. Polymerase chain reaction analysis of synexin mRNA from various fetal and adult tissues, from human and monkey, revealed that the alternative splicing event is tissue-regulated; synexin mRNA containing the cassette exon is prevalent in brain, heart, and skeletal muscle. This is supported by Western blot analysis showing that muscle synexin (annexin VIIb) is larger than synexin from lung (annexin VIIa). The muscle and lung isoforms have the same molecular mass as the recombinant synexins expressed in Escherichia coli using cDNAs containing or lacking the cassette exon, respectively. The difference in size is consistent with the molecular masses predicted from the proteins encoded by the alternatively spliced synexin mRNAs. Another type of synexin mRNA contained a longer 3'-noncoding region generated by the selection of an alternate poly(A) signal. Northern analysis of human fibroblast RNA showed the presence of two bands (2.0- and 2.4-kilobase) when hybridized to a cDNA fragment of the coding region of synexin, but only the 2.4-kilobase band hybridized to a probe made from the longer 3' end.

Transplacental distribution of weak acids in mice: accumulation in compartments of high pH.

The transplacental distribution of three weak acids was measured in mice on gestational day 11. The ratio of drug concentration in maternal, embryonic, and extraembryonic compartments compared to maternal plasma followed a consistent hierarchy such that amniotic fluid greater than or equal to exocoelomic fluid greater than embryo greater than embryo plasma greater than maternal skeletal muscle. This distribution pattern correlates well with pH in these compartments and suggests that pH gradients between compartments are an influential factor in determining weak acid drug disposition during pregnancy.

Genomic organization and chromosomal localization of the human nucleolin gene.

Nucleolin, a eukaryotic nucleolar phosphoprotein, is involved in the synthesis and maturation of ribosomes. To characterize the genomic organization and regulatory sequences of this gene, two overlapping lambda clones containing the human nucleolin gene plus flanking regions were isolated from a genomic library using human nucleolin cDNA. Southern blots of genomic DNA from human, several mammals, chicken, and yeast revealed that the nucleolin gene is well conserved across these species. The gene consists of 14 exons with 13 intervening sequences and spans approximately 11 kilobases of DNA. Analysis of the splice junctions indicated that the amino-terminal domain and the four RNA binding domains plus the nuclear localization signal are split into adjacent exons. Sequences from the 5'-flanking and the first intron contain a high content of GC residues which is consistent with nucleolin being a "housekeeping" gene. Promoter elements include an atypical TATA box (GTTA), one CCAAT box much further from the initiation site, three reverse compliments of CCAAT (ATTGG), and two pyrimidine-rich nucleotide stretches. In addition, this region and the first intron contain numerous potential Sp1, GCF, CRE-fos, GCN, AP-1, AP-2, UCE, and sequences similar to the glucocorticoid receptor binding site. The transcription start site was determined by primer extension and S1 nuclease mapping of RNA from human liver. One Kpn and three Alu repeats were found within two of the middle introns. The 3'-untranslated portion of the gene contains five homology blocks in a 100-base pair region that are highly conserved among human, mouse, and hamster genomes. Finally, we have determined that the human nucleolin gene is located on chromosome 2q12-qter and is present at one copy per haploid genome. A restriction fragment length polymorphism with EcoRI has been detected in the gene.

Circadian periodicity in microfilarial counts.

We propose a second-order harmonic model to describe circadian periodicity in the 24-h cycle of microfilarial counts. Rhesus monkeys (Macaca mulatta) were infected with Brugia malayi. Definite evidence of circadian periodicity was found in the microfilarial counts. The estimates of the parameters, periodicity indices and peak hours were determined.
