Establishment and characterization of HUOT, a human ovarian malignant teratoma cell line producing alpha-fetoprotein.

A cell line designated HUOT was established from a recurrent tumor of human ovarian malignant teratoma. The cell line grew slowly and stably and serial passages were performed 50 times within 35 months. The cells, polygonal or spindle, with neoplastic and pleomorphic features grew in multiple layers and without contact inhibition. Population doubling time was 98 hours and the plating efficiency was less than 6%. The chromosome number varied from 43 to over 256, and the modal number was stable at the hyperdiploid range (52-56). The cultured cells produced anaplastic carcinomas by heterotransplantation into the subcutis of nude mice and were characterized as producing large amounts of alpha-fetoprotein, in vitro, at the stationary growth phase and as forming cystlike structures. Dibutyl cAMP suppressed the cellular proliferation and increased the production of alpha-fetoprotein. Therefore, this HUOT line is expected to have a wide application for various laboratory studies.

Differential effects of in vitro ethanol on prostaglandin E1-sensitive adenylate cyclase from smooth muscle cells and platelets.

In membranes of smooth muscle cells cultured from rabbit mesenteric artery, ethanol dose-dependently activates adenylate cyclase, both basal and PGE1- or GPP(NH)P-sensitive. The alcohol increases the maximal stimulation induced by PGE1 and GPP(NH)P, without greatly affecting their potency. The arrhenius plot displays a discontinuity point, which is only slightly lowered by ethanol. On the contrary, in membranes from human platelets ethanol inhibits basal, GPP(NH)P- and PGE1-sensitive adenylate cyclase, without modification of the prostaglandin or guanine nucleotide potency. The break point present in the Arrhenius plots is definitely lowered by the alcohol. In addition, ethanol decreases the thermostability of the enzyme. Neither in myocytes nor in platelets does ethanol affect the activation energy of the reaction. The data suggest that ethanol probably interacts directly with the membrane proteins, and that its effect is not mediated only through a perturbation of membrane lipid fluidity.

Establishment of human endometrial adenocarcinoma cell line containing estradiol-17 beta and progesterone receptors.

The cell line designated HHUA was established from a well-differentiated endometrial adenocarcinoma (grade 1) of a 54-year-old female on December 10, 1980. The cell line grew well without interruption for 22 months and was subcultivated more than 56 times. The cells were spindle or polygonal in shape, formed multilayers, and did not show contact inhibition. The chromosome number varied widely and showed aneuploidy. The modal number was 46 and no marker chromosome was identified. The cells were transplanted into the subcutis of BALB/C nude mice and produced tumors which were interpreted as adenocarcinoma. The cells contained estradiol-17 beta and progesterone receptors. Administration of estradiol-17 beta successfully depleted cytoplasmic estrogen receptors and brought about increased progesterone receptor levels.

Establishment and characterization of an estrogen-producing human ovarian granulosa tumor cell line.

Cells designated HTOG and HTOT were established by long-term culture from a human ovarian granulosa cell and a theca cell tumor, respectively. The HTOG line grew well forming colonies and multilayered rapidly without contact inhibition; serial passages of HTOG were performed over 100 times successively within 25 months. HTOG were spindle cells, polygonal or spherical in shape, revealed neoplastic and pleomorphic features, and produced estrone (E1) and 17 beta-estradiol (E2). The chromosome number varied considerably and showed hyperploidy; the modal chromosome number was in the hypertriploid-tetraploid range. When HTOG cells were heterotransplanted into the subcutis of BALB/c nude mice, they produced a sarcomatous diffuse type of granulosa cell tumors. In contrast, HTOT cells grew slowly while forming monolayers and underwent five successive passages in about 100 days, but a theca cell tumor line could not be established. HTOT cells were fibroblastic in shape and also produced E1 and E2. The majority of the cells showed diploidy and karyologic normality.

Establishment and characterization of two human mixed mesodermal tumor cell lines from the same patient.

A cell line designated HIRS -BM was established from fluid aspirated from the sternal bone marrow of a 16-year-old female. Another cell line ( HIRS -PB) was derived from the peripheral blood of the same patient. Both lines grew well, multilayering rapidly without contact inhibition, and 62 serial passages were successively done within 28 months. Both cultures contained spindle- or fibrous-shaped cells that revealed neoplastic and pleomorphic features, and these cells were characterized as possessing cross-striations in the cytoplasm. The cross-striations were detected by phosphotungstic acid hematoxylin stain. Some elongated cells were stained positively with anti-myoglobin by use of periodic acid-Schiff methods. The primary tumor in the uterus was diagnosed as a mixed mesodermal tumor composed of adenocarcinoma and rhabdomyosarcoma cells. The karyotype exhibited hyperploidy and large submetacentric marker chromosomes, and the modal chromosome number was 84. No difference was found between the 2 cell lines except for growth behavior and heterotransplantability . HIRS -BM cells grew more rapidly and were highly transplantable. The HIRS -BM cells were transplanted into the subcutis of BALB/c nude mice and produced mixed mesodermal tumors resembling the uterine tumor, while the HIRS -PB cells could not be transplanted. Due to the histogenesis of the mixed mesodermal tumor being's obscure with histologic observations only, this study was performed to obtain data by tissue culture of the tumor and resulted in support of the combination theory reported in the literature in regard to tumor.

On the process of cellular division in Escherichia coli: nucleotide sequence of the gene for penicillin-binding protein 3.

We determined the nucleotide sequence of a DNA fragment containing the ftsI gene coding for the penicillin-binding protein 3 (PBP-3), an indispensable enzyme for cell division of Escherichia coli. The entire ftsI gene was within the 2.8 kilobase PvuII fragment derived from the chromosomal segment on pLC26-6 (Nishimura et al. 1977). The coding region for PBP-3 was identified by comparison with the N-terminal amino acid sequence of in vitro synthesized PBP-3. The structural gene for ftsI consisted of 1,764 base-pairs coding for a 588 amino acid residue-polypeptide with a molecular weight of 63,850. PBP-3 synthesized in vitro showed a lower mobility in SDS-gel electrophoresis than that of the authentic PBP-3, suggesting that the primary translation product of the ftsI gene may be processed to yield mature PBP-3.

Chemical synthesis of a human fibroblast interferon gene and its expression in Escherichia coli.

Using the solid phase phosphotriester method, a gene coding for human fibroblast interferon consisting of 166 amino acid residues was chemically synthesized. The gene obtained by ligation of 61 synthesized oligodeoxyribonucleotide fragments was inserted into the downstream of tryptophan promoter, and expressed in E. coli. Lysate of this E. coli showed the antiviral activity which was specifically neutralized by anti-fibroblast interferon antibody. No particular advantage was observed in the expression efficiency by the synthetic gene over that by the native gene.

Nucleolar organizer regions and constitutive heterochromatin in polyploid species of the genus Odontophrynus (Amphibia, Anura).

Chromosome banding patterns obtained by ammoniacal silver staining (Ag-AS) and alkaline Giemsa (CBG) have been analysed in several amphibian species of the genus Odontophrynus from South America. Ag-AS bands were found at secondary constrictions, mainly of chromosomes 4 and 11. The CBG technique revealed centromeric and telomeric constitutive heterochromatin on almost all chromosomes of these species. Moreover, intercalary bands were found at particular sites of several chromosomes. Some inter- and intra-population polymorphisms were found for the Ag-AS and C-banding patterns. The species variability in the number and position of the Ag-AS bands, known to be regions of active ribosomal cistrons, as well as the specific sites of intercalary heterochromatin, are used to discuss the possible evolutionary relationships among these species.

A synthetic translation-terminator gene. A tool for dissecting the translation direction of a gene.

A 41-nucleotide-long duplex DNA, which contains the translation termination codon TAA in six reading frames and lactose operator sequence of Escherichia coli, has been synthesized. This fragment may be useful not only for producing a truncated protein encoded in a plasmid, but also for the identification of the precise coding region and translation direction of a bacterial gene in the cloned chromosomal segment. The synthetic fragment was inserted into beta-lactamase structural gene in pBR322 in order to test the in vivo activity. The plasmid produced mutant beta-lactamase reduced in size, as expected from the insertion site, and rendered the host bacterium constitutive for beta-galactosidase. Thus, termination codons and lactose operator in synthetic nucleotide appear to be functional in vivo.

A C2077T polymorphism of the type B human natriuretic peptide receptor gene is not associated with myocardial infarction.

INTRODUCTION: We examined previously the genomic structure of the human natriuretic peptide receptor type B (hNPRB) gene and reported a C2077T polymorphism located in exon 11 of the gene. We now have studied the C2077T polymorphism in myocardial infarction [MI] patients and non-MI [control] subjects to evaluate the value of this polymorphism as a marker for MI. MATERIAL AND METHODS: 302 subjects (163 MI patients and 139 non-MI age-matched control subjects) were studied. PCR-restriction fragment length polymorphism analysis (PCR-RFLP) was developed to detect the C2077T transition. RESULTS: The distribution of C2077T polymorphism genotypes in the MI patients (CC:CT:TT, 47.2%:41.1%:11.7%) was not significantly different from that in the control patients (CC:CT:TT, 53.2%:40.3%:6.5%) (chi 2 = 2.73, p = NS). Allele frequencies of the C2077T polymorphism were f(C/T) 68.2%/31.8% in the MI group and 73.4%/26.6% in the control group. However, no association was found between this polymorphism and clinical diagnosis of MI. CONCLUSION: Our data indicate that the C2077T polymorphism is not a useful marker of the relation between the hNPRB gene and MI in the Japanese and variations of the hNPRB gene that may be in linkage disequilibrium with this polymorphism do not play a causative role in MI.
